Dorsal Root Entry Zone

The dorsal root entry zone (DREZ) is the junction where sensory nerve fibers from spinal dorsal roots enter the spinal cord, linking peripheral receptors with central neural circuits. At this interface, incoming afferents pass through the pial boundary, divide into ascending and descending branches, and form synaptic connections within the dorsal horn, where touch, pain, and other sensory signals undergo early processing. In neuroscience and clinical research, the DREZ is important for studying sensory transmission and central sensitization, while targeted procedures such as DREZ lesioning can reduce severe deafferentation pain or spasticity by disrupting pathologically active input pathways.

An in vivo imaging protocol to monitor primary sensory axons following dorsal root crush is described. The procedures utilize wide-field fluorescence microscopy and thy1-YFP transgenic mice, and permit repeated imaging of axon regeneration over 4 cm in the PNS and axon interactions with the interface of the CNS.
