Neurite Volume Measurement

Neurite volume measurement is a quantitative imaging approach used to determine the three-dimensional amount of neuronal processes, including axons and dendrites, and to assess changes in neural structure. The method typically uses microscopy images to distinguish neurites from surrounding background, segment the labeled structures, and calculate their volume from the resulting spatial data. In neuroscience, these measurements help characterize neuronal growth, branching, degeneration, and responses to experimental conditions or treatments. By providing a quantitative structural readout, neurite volume measurement supports studies of development, neurotoxicity, disease mechanisms, and the effects of compounds that alter neuronal morphology.

Immunofluorescence Assay to Evaluate Effect of Target Proteins on Neurite Outgrowth

In this video, we demonstrate an EGFP-based immunofluorescence assay to identify the efficacy of target proteins on the neurite outgrowth of primary cortical neurons. The primary neurons are transfected with a fluorescent protein, EGFP, and a target protein, and then an immunoassay is performed to investigate the effect of the target protein on neurite outgrowth.

Measurement of the Pressure-volume Curve in Mouse Lungs

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung’s structural changes with developing pathology.

Standardization of a Novel Semi-Automatic Software for Neurite Outgrowth Measurement

Neurite outgrowth assays provide a quantitative value about regenerative neuronal processes. The advantage of this semi-automatic software is that it segments cell bodies and neurites separately by creating a mask and measures various parameters such as neurite length, number of branch points, cell-body cluster area, and number of cell clusters.
