Neuronal Cell Assemblies

Neuronal cell assemblies are groups of neurons whose coordinated activity represents information, supports behavior, or contributes to cognitive processes. They form and change through synaptic connectivity and activity-dependent plasticity, allowing neurons to fire together in coordinated patterns that can persist, reappear, or reorganize with experience. In neuroscience, studying these assemblies helps researchers link patterns of neural activity to perception, learning, memory, decision-making, and motor control. Experimental approaches such as electrophysiological recording, calcium imaging, and computational analysis reveal how assembly dynamics encode information and change across brain states, disease conditions, and behavioral tasks.

Compartmentalization of Human Stem Cell-Derived Neurons within Pre-Assembled Plastic Microfluidic Chips

This protocol demonstrates the use of compartmentalized microfluidic chips, injection molded in a cyclic olefin copolymer to cultured neurons differentiated from human stem cells. These chips are preassembled and easier-to-use than traditional compartmentalized poly(dimethylsiloxane) devices. Multiple common experimental paradigms are described here, including viral labeling, fluidic isolation, axotomy, and immunostaining.

Here we describe the use of a self-assembling 3-dimensional scaffold to culture human neural progenitor cells. We present a protocol to release the cells from the scaffolds to be analysed subsequently e.g. by flow cytometry. This protocol might be adapted to other cell types to perform detailed mechanistically studies.
