Organoid Maturation

Organoid maturation is the progressive development of a three-dimensional, stem cell-derived tissue model toward greater cellular organization, physiological function, and resemblance to its in vivo counterpart. In neural organoids, extended culture and directed differentiation promote neuronal and glial maturation, while cell-cell signaling, network activity, and changing tissue conditions shape synapse formation and circuit development. These processes help researchers model human brain development, investigate neurological disease, evaluate therapeutics, and study how genetic or environmental factors alter neural function. Improving maturation remains important for producing organoids that more accurately reproduce adult-like biology and generate reliable, translational findings.

Differentiation and Maturation of Human Embryonic Stem Cells into Neural Organoids

2025

In this video, a stepwise protocol for Human embryonic stem cells into neural organoids is demonstrated. The method involves inducing neuronal rosette formation, followed by controlled maturation and final culturing on a hydrophobic membrane to achieve fully developed neural organoids.

Generating Mature Cerebellar Organoids From Induced Pluripotent Stem Cells Using Single-Use Bioreactors

2025

This video demonstrates a technique for generating mature cerebellar organoids from induced pluripotent stem cells (iPSCs) using a single-use 3D bioreactor. The bioreactor facilitates iPSC aggregate formation, which then undergoes treatment with a series of culture media enriched in growth factors and chemokines that induce the differentiation of the iPSC aggregates into mature cerebellar organoids. Subsequently, the organoids are harvested and cryopreserved for further analysis.

The protocol presents a method to derive human lung organoids from primary lung tissues, expand the lung organoids and induce proximal differentiation to generate 3D and 2D airway organoids that faithfully phenocopy the human airway epithelium.

An In Vitro Method to Generate Human Brain Organoids

2025

The video demonstrates a method for generating human brain organoids from induced pluripotent stem cells. It shows how the neurosphere forms a neuroectodermal layer, which suppresses the formation of other germ layers. The neurospheres are embedded in a 3D gel matrix and form neuroepithelial buds. When incubated with organoid media, these neuroepithelial buds differentiate into specific brain regions, resulting in the formation of mature brain organoids.
