Osmium Tetroxide Staining

Osmium tetroxide staining is a histological method that visualizes lipid-rich structures, making it especially valuable for examining myelin and neural membranes in neuroscience. Osmium tetroxide reacts with unsaturated lipids and is reduced to a dark, electron-dense compound, simultaneously stabilizing membrane architecture and producing strong contrast in tissue sections. Researchers use this approach with light and electron microscopy to assess myelination, axonal organization, membrane ultrastructure, and degenerative changes in nervous tissue. The resulting contrast supports studies of neural development, connectivity, injury, and disease by revealing structural alterations that may be difficult to detect with routine stains.
