Otic Vesicle Injection

Otic vesicle injection is a microsurgical technique that introduces cells, molecules, or genetic materials into the embryonic otic vesicle, the precursor of the inner ear and associated sensory structures. Using a fine glass needle under microscopic guidance, researchers deliver a controlled volume into this fluid-filled compartment, allowing the injected material to contact developing otic epithelium and surrounding tissues. In neuroscience, the method supports studies of inner-ear development, neuronal patterning, hair-cell formation, and auditory or vestibular function. It can also help evaluate developmental signals, gene function, and experimental treatments in models of hearing and balance disorders.

Direct Delivery of MIF Morpholinos Into the Zebrafish Otocyst by Injection and Electroporation Affects Inner Ear Development

A method to deliver morpholinos directly into the zebrafish otocyst at 24hpf has been developed. Using microinjection of morpholinos into the lumen of otic vesicle and electroporation to effect penetration, we were able to bypass the effect of morpholinos on the brain and obtain effects specific to the inner ear.

Differentiating Immortalized Multipotent Otic Progenitors into Spiral Ganglion Neurons and Evaluating the Differentiation

The video demonstrates the differentiation of immortalized multipotent otic progenitors (iMOPs) into spiral ganglion neurons (SGNs) and immunofluorescence-based confirmation of the differentiation. The iMOPs are plated onto culture substrate-coated coverslips in a neuronal differentiation medium for differentiation into SGNs. The differentiated cells are labeled with antibodies specific for neuronal differentiation markers and analyzed under a microscope to confirm differentiation.
