Vesicle Fusion Events

Vesicle fusion events are membrane-merging processes in which intracellular vesicles join a target membrane to release or deliver their contents, making them essential for cellular communication and transport. In neurons, an action potential opens voltage-gated calcium channels at the presynaptic terminal; calcium binds synaptotagmin and activates SNARE proteins, which pull the synaptic vesicle and plasma membranes together to form a fusion pore and release neurotransmitters into the synaptic cleft. Studying these rapid events clarifies how synapses transmit signals and adapt during learning, while also informing research on neurological disorders, neurotoxicity, and therapies that target abnormal neurotransmitter release.

Here, we present a protocol to detect single, SNARE-mediated fusion events between liposomes and supported bilayers in microfluidic channels using polarized TIRFM, with single molecule sensitivity and ~15 msec time resolution. Lipid and soluble cargo release can be detected simultaneously. Liposome size, lipid diffusivity, and fusion pore properties are measured.
