Endosome-lysosome Fusion

Endosome-lysosome fusion is the membrane-merging process that delivers endocytosed cargo to lysosomes for degradation, recycling, or signaling, making it essential for cellular homeostasis and drug response. It occurs as late endosomes mature and acquire fusion competence through Rab7 and HOPS complex activity, followed by SNARE-mediated membrane fusion; lysosomal acidification then supports enzymatic breakdown of cargo. In pharmacology, this pathway influences receptor downregulation, intracellular drug trafficking, antigen processing, and the handling of nanoparticle or biologic therapies. Modulating fusion can therefore alter drug efficacy, toxicity, and diseases associated with defective cargo clearance.
