Crawling Milestones

Crawling milestones are developmental markers that describe how infants progressively use coordinated movement to explore their environment and gain independence. Crawling typically emerges as postural control, muscle strength, balance, and coordination develop, allowing an infant to support the body, shift weight, and move the limbs in a repeated pattern; however, the timing and form of movement can vary widely. In psychology, observing crawling helps researchers and clinicians study motor development, sensorimotor learning, spatial exploration, and interactions between infants and caregivers. Tracking these behaviors can contribute to developmental assessment while supporting research on typical variation and early indicators of motor differences.

We describe a protocol of brightfield intravital microscopy for measuring dynamic neutrophil-endothelial cell interactions during neutrophil recruitment in response to the source of a neutrophil chemoattractant in vivo. Neutrophil intraluminal crawling, transendothelial migration and chemotaxis in mouse cremaster muscle tissue are visualized with time-lapsed video photography and tracked with ImageJ.
