L-glutamate overproduction in Corynebacterium glutamicum, a biotin auxotroph, is induced by biotin limitation or by treatment with certain fatty acid ester surfactants or with penicillin. We have analyzed the relationship between the inductions, 2-oxoglutarate dehydrogenase complex (ODHC) activity, and L-glutamate production. Here we show that a strain deleted for odhA and completely lacking ODHC activity produces L-glutamate as efficiently as the induced wild type (27.8 mmol/g [dry weight] of cells for the ohdA deletion strain compared with only 1.0 mmol/g [dry weight] of cells for the uninduced wild type). This level of production is achieved without any induction or alteration in the fatty acid composition of the cells, showing that L-glutamate overproduction can be caused by the change in metabolic flux alone. Interestingly, the L-glutamate productivity of the odhA-deleted strain is increased about 10% by each of the L-glutamate-producing inductions, showing that the change in metabolic flux resulting from the odhA deletion and the inductions have additive effects on L-glutamate overproduction. Tween 40 was indicated to induce drastic metabolic change leading to L-glutamate overproduction in the odhA-deleted strain. Furthermore, optimizing the metabolic flux from 2-oxoglutarate to L-glutamate by tuning glutamate dehydrogenase activity increased the l-glutamate production of the odhA-deleted strain.

Genetically programmed cell deaths play important roles in unicellular prokaryotes. In postsegregational killing, loss of a gene complex from a cell leads to its descendants' deaths. With type II restriction-modification gene complexes, such death is triggered by restriction endonuclease's attacks on under-methylated chromosomes. Here, we examined how the Escherichia coli transcriptome changes after loss of PaeR7I gene complex. At earlier time points, activation of SOS genes and sigma(E)-regulon was noticeable. With time, more SOS genes, stress-response genes (including sigma(S)-regulon, osmotic-, oxidative- and periplasmic-stress genes), biofilm-related genes, and many hitherto uncharacterized genes were induced, and genes for energy metabolism, motility and outer membrane biogenesis were repressed. As expected from the activation of sigma(E)-regulon, the death was accompanied by cell lysis and release of cellular proteins. Expression of several sigma(E)-regulon genes indeed led to cell lysis. We hypothesize that some signal was transduced, among multiple genes involved, from the damaged genome to the cell surface and led to its disintegration. These results are discussed in comparison with other forms of programmed deaths in bacteria and eukaryotes.

Duchenne muscular dystrophy (DMD) is an X-linked recessive genetic disease caused by mutations in the gene coding for the protein dystrophin. Recent work demonstrates that dystrophin is also found in the vasculature and its absence results in vascular deficiency and abnormal blood flow. This induces a state of ischemia further aggravating the muscular dystrophy pathogenesis. For an effective form of therapy of DMD, both the muscle and the vasculature need to be addressed. To reveal the developmental relationship between muscular dystrophy and vasculature, mdx mice, an animal model for DMD, were crossed with Flt-1 gene knockout mice to create a model with increased vasculature. Flt-1 is a decoy receptor for vascular endothelial growth factor, and therefore both homozygous (Flt-1(-/-)) and heterozygous (Flt-1(+/-)) Flt-1 gene knockout mice display increased endothelial cell proliferation and vascular density during embryogenesis. Here, we show that Flt-1(+/-) and mdx:Flt-1(+/-) adult mice also display a developmentally increased vascular density in skeletal muscle compared with the wild-type and mdx mice, respectively. The mdx:Flt-1(+/-) mice show improved muscle histology compared with the mdx mice with decreased fibrosis, calcification and membrane permeability. Functionally, the mdx:Flt-1(+/-) mice have an increase in muscle blood flow and force production, compared with the mdx mice. Consequently, the mdx:utrophin(-/-):Flt-1(+/-) mice display improved muscle histology and significantly higher survival rates compared with the mdx:utrophin(-/-) mice, which show more severe muscle phenotypes than the mdx mice. These data suggest that increasing the vasculature in DMD may ameliorate the histological and functional phenotypes associated with this disease.

Hier zeigen wir, dass Flt-1(+/-) und mdx:Flt-1(+/-) Erwachsene M&auml;usen auch eine entwicklungspolitisch erh&ouml;hte vaskul&auml;re Dichte in der Skelettmuskulatur, die gegen&uuml;ber der Wildtyp und Mdx-M&auml;use, bzw. anzeigen. Die mdx:Flt-1(+/-)-M&auml;use zeigen verbesserte Muskel Histologie gegen&uuml;ber der Mdx-M&auml;use mit verminderter Fibrose, Verkalkungen und Membran-Durchl&auml;ssigkeit. Funktional, die mdx:Flt-1(+/-) M&auml;use haben eine Steigerung Durchblutung der Muskulatur und Kraft Produktion, verglichen mit der Mdx-M&auml;use. Folglich zeigen die mdx:utrophin(-/-):Flt-1(+/-) M&auml;use verbesserte Muskel Histologie und deutlich h&ouml;heren &Uuml;berlebensraten im Vergleich mit der mdx:utrophin(-/-)-M&auml;use, die schwere Muskel Ph&auml;notypen als die Mdx-M&auml;use zeigen. Diese Daten legen nahe, dass das Gef&auml;&szlig;system in DMD erh&ouml;hen die histologischen und funktionellen Ph&auml;notypen, die diese Krankheit lindern kann.

FLT-1 Haploinsuffizienz aerztlichen Muskeldystrophie Ph&auml;notyp von entwicklungspolitisch erh&ouml;hte Gef&auml;&szlig;system in Mdx-M&auml;use.

Distrofia muscolare di Duchenne (DMD) &egrave; X-linked recessiva genetica malattia causata da mutazioni nel gene che codifica per la proteina distrofina. Lavoro recente dimostra che la distrofina &egrave; presente anche nel sistema vascolare e suoi risultati assenza in deficit vascolare e anormale il flusso di sangue. Distrofia muscular de Duchenne (DMD) es una enfermedad gen&eacute;tica recesiva x-ligada causada por mutaciones en el gen que codifica para la prote&iacute;na distrofina. Trabajo reciente demuestra que la distrofina tambi&eacute;n se encuentra en la vasculatura y flujo de la sangre de sus resultados de ausencia en deficiencia vascular y anormal. Esto induce un estado de isquemia, agravando a&uacute;n m&aacute;s la patogenesia de la distrofia muscular. Para una eficaz forma de terapia de DMD, tanto el m&uacute;sculo y la vasculatura deben abordarse. Para revelar la relaci&oacute;n del desarrollo de distrofia muscular y vasculatura, ratones mdx, un modelo animal para DMD, fueron cruzados con ratones de Flt-1 gene knockout para crear un modelo con mayor vascularizaci&oacute;n. FLT-1 es un receptor de se&ntilde;uelo para el factor de crecimiento endotelial vascular y por lo tanto ambos homocig&oacute;ticos (Flt-1(-/-)) y heterozig&oacute;ticos ratones de knockout de gene (Flt-1 (+/-)) Flt-1 Mostrar la proliferaci&oacute;n creciente de la c&eacute;lula endotelial y la densidad vascular durante la embriog&eacute;nesis. A continuaci&oacute;n, os mostramos que Flt-1(+/-) y mdx:Flt-1(+/-) de ratones adultos tambi&eacute;n presentan una desarrollo creciente densidad vascular en m&uacute;sculo esquel&eacute;tico en comparaci&oacute;n con los ratones de tipo salvaje y mdx, respectivamente. The molecules that regulate the apoptosis cascade are also involved in differentiation and syncytial fusion in skeletal muscle. MyoD is a myogenic transcription factor that plays essential roles in muscle differentiation. We noticed that MyoD(-/-) myoblasts display remarkable resistance to apoptosis by down-regulation of miR-1 (microRNA-1) and miR-206 and by up-regulation of Pax3. This resulted in transcriptional activation of antiapoptotic factors Bcl-2 and Bcl-xL. Forced MyoD expression induces up-regulation of miR-1 and miR-206 and down-regulation of Pax3, Bcl-2, and Bcl-xL along with increased apoptosis in MyoD(-/-) myoblasts. In contrast, MyoD gene knockdown increases cell survival of wild-type myoblasts. The 3' untranslated region of Pax3 mRNA contains two conserved miR-1/miR-206-binding sites, which are required for targeting of these microRNAs (miRNAs). Therefore, these data suggest that MyoD not only regulates terminal differentiation but also apoptosis through miRNA-mediated down-regulation of Pax3. Finally, MyoD, miR-1, and miR-206 are all down-regulated in quiescent satellite cells, which may be required for maintenance of muscle stem cells.

Expression of the four transcription factors, that is, Oct4, Sox2, cMyc, and Klf4 has been shown to generate induced pluripotent stem cells (iPSCs) from many types of specialized differentiated somatic cells. It remains unclear, however, whether fully committed skeletal muscle progenitor cells (myoblasts) have the potency to undergo reprogramming to develop iPSCs in line with previously reported cases. To test this, we have isolated genetically marked myoblasts derived from satellite cell of adult mouse muscles using the Cre-loxP system (Pax7-CreER:R26R and Myf5-Cre:R26R). On infection with retroviral vectors expressing the four factors, these myoblasts gave rise to myogenic lineage tracer lacZ-positive embryonic stem cell (ESC)-like colonies. These cells expressed ESC-specific genes and were competent to differentiate into all three germ layers and germ cells, indicating the successful generation of myoblast-derived iPSCs. Continuous expression of the MyoD gene, a master transcription factor for skeletal muscle specification, inhibited this reprogramming process in myoblasts. In contrast, reprogramming myoblasts isolated from mice lacking the MyoD gene led to an increase in reprogramming efficiency. Our data also indicated that Oct4 acts as a transcriptional suppressor of MyoD gene expression through its interaction with the upstream enhancer region. Taken together, these results indicate that suppression of MyoD gene expression by Oct4 is required for the initial reprogramming step in the development of iPSCs from myoblasts. This data suggests that the skeletal muscle system provides a well-defined differentiation model to further elaborate on the effects of iPSC reprogramming in somatic cells.

Expresi&#xF3;n de los cuatro factores de transcripci&#xF3;n, es decir, Oct4, Sox2, cMYC, y Klf4 se ha demostrado para generar c&#xE9;lulas madre pluripotentes inducidas (iPSCs) de muchos tipos de c&#xE9;lulas som&#xE1;ticas diferenciadas especializados. A&#xFA;n no est&#xE1; claro, sin embargo, si las c&#xE9;lulas esquel&#xE9;ticas totalmente comprometidos progenitoras musculares (mioblastos) tienen la potencia de someterse a una reprogramaci&#xF3;n para desarrollar iPSCs en l&#xED;nea con los casos reportados previamente. La supresi&#xF3;n del gen Oct4 MyoD por el que se requiere para la reprogramaci&#xF3;n de mioblastos para producir c&#xE9;lulas madre pluripotentes inducidas

Modification of complex microbial cellular processes is often necessary to obtain organisms with particularly favorable characteristics, but such experiments can take many generations to achieve. In the present article, we accelerated the experimental evolution of Escherichia coli populations under selection for improved growth using one of the restriction-modification systems, which have shaped bacterial genomes. This resulted in faster evolutionary changes in both the genome and bacterial growth. Transcriptome/genome analysis at various stages enabled prompt identification of sequential genome rearrangements and dynamic gene-expression changes associated with growth improvement. The changes were related to cell-to-cell communication, the cell death program, as well as mass production and energy consumption. These observed changes imply that improvements in microorganism population growth can be achieved by inactivating the cellular mechanisms regulating fraction of active cells in a population. Some of the mutations were shown to have additive effects on growth. These results open the way for the application of evolutionary genome engineering to generate organisms with desirable properties.

Evolutionary genome engineering using a restriction-modification system.

Skeletal myoblast transplantation has therapeutic potential for repairing damaged heart. However, the optimal conditions for this transplantation are still unclear. Recently, we demonstrated that satellite cell-derived myoblasts lacking the MyoD gene (MyoD(-/-)), a master transcription factor for skeletal muscle myogenesis, display increased survival and engraftment compared to wild-type controls following transplantation into murine skeletal muscle. In this study, we compare cell survival between wild-type and MyoD(-/-) myoblasts after transplantation into infarcted heart. We demonstrate that MyoD(-/-) myoblasts display greater resistance to hypoxia, engraft with higher efficacy, and show a larger improvement in ejection fraction than wild-type controls. Following transplantation, the majority of MyoD(-/-) and wild-type myoblasts form skeletal muscle fibers while cardiomyocytes do not. Importantly, the transplantation of MyoD(-/-) myoblasts induces a high degree of angiogenesis in the area of injury. DNA microarray data demonstrate that paracrine angiogenic factors, such as stromal cell-derived factor-1 (SDF-1) and placental growth factor (PlGF), are up-regulated in MyoD(-/-) myoblasts. In addition, over-expression and gene knockdown experiments demonstrate that MyoD negatively regulates gene expression of these angiogenic factors. These results indicate that MyoD(-/-) myoblasts impart beneficial effects after transplantation into an infarcted heart, potentially due to the secretion of paracrine angiogenic factors and enhanced angiogenesis in the area of injury. Therefore, our data provide evidence that a genetically engineered myoblast cell type with suppressed MyoD function is useful for therapeutic stem cell transplantation.
