Deposition of prions into the environment by infected animals may contribute to transmission and spread of chronic wasting disease (CWD) among free-ranging cervids, and identification of such environmental sources may provide an avenue for managing CWD. We evaluated the role that wallow use by elk (Cervus elaphus) may play in CWD transmission by monitoring wallows with animal-activated cameras throughout their period of use. We monitored 39 wallows from 5 August 2005 to 14 October 2005. Elk visited 20 sites; we recorded 22 events when only male elk wallowed and 374 additional events when male and female elk had naso-oral contact with wallow contents. Because wallows are foci of male elk activity, behaviors at wallows could potentially contribute to the maintenance and transmission of CWD. Our findings, however, suggest that because wallows are only used an average of one or two times a season they may not be important in CWD transmission. The data also suggest that mineral licks could be more important in CWD transmission because they were used more frequently and by three species that contract CWD.

Elk use of wallows and potential chronic wasting disease transmission.

Raccoons are the reservoir for the raccoon rabies virus variant in the United States. To combat this threat, oral rabies vaccination (ORV) programs are conducted in many eastern states. To aid in these efforts, the genetic structure of raccoons (Procyon lotor) was assessed in southwestern Pennsylvania to determine if select geographic features (i.e., ridges and valleys) serve as corridors or hindrances to raccoon gene flow (e.g., movement) and, therefore, rabies virus trafficking in this physiographic region. Raccoon DNA samples (n = 185) were collected from one ridge site and two adjacent valleys in southwestern Pennsylvania (Westmoreland, Cambria, Fayette, and Somerset counties). Raccoon genetic structure within and among these study sites was characterized at nine microsatellite loci. Results indicated that there was little population subdivision among any sites sampled. Furthermore, analyses using a model-based clustering approach indicated one essentially panmictic population was present among all the raccoons sampled over a reasonably broad geographic area (e.g., sites up to 36 km apart). However, a signature of isolation by distance was detected, suggesting that widths of ORV zones are critical for success. Combined, these data indicate that geographic features within this landscape influence raccoon gene flow only to a limited extent, suggesting that ridges of this physiographic system will not provide substantial long-term natural barriers to rabies virus trafficking. These results may be of value for future ORV efforts in Pennsylvania and other eastern states with similar landscapes.

Landscape genetics of raccoons (Procyon lotor) associated with ridges and valleys of Pennsylvania: implications for oral rabies vaccination programs.

The raccoon (Procyon lotor) continues to be a prominent terrestrial rabies reservoir in the eastern United States. Describing the dispersal and movements of these animals and determining geographic features that are natural hindrances or corridors to movements could be used to assist oral rabies vaccination efforts. The landscape of the ridge-and-valley system in Pennsylvania exhibits characteristics of both natural potential hindrances and travel corridors to the movements of wildlife. The movements of 49 raccoons were monitored throughout a ridge and two adjacent valley sites to assess their movements related to these landscape features. Results suggest that few raccoons cross the ridge we studied over the short-term and that long-distance movements of these animals are uncommon in this region. Consequently, movement corridors within the ridge were largely confined to spur valleys within the ridge system. These results may be useful in strategic oral rabies vaccination programs in Pennsylvania and other areas where natural hindrances and travel corridors to movement are identified.

Raccoon (Procyon lotor) movements and dispersal associated with ridges and valleys of Pennsylvania: implications for rabies management.

In recent years, elk have begun recolonizing areas east of the Rocky Mountains that are largely agro-forested ecosystems composed of privately owned land where management of elk is an increasing concern due to crop and forage depredation and interspecific disease transmission. We used a Geographic Information System, elk use locations (n = 5013), random locations (n = 25,065), discrete-choice models, and information-theoretic methods to test hypotheses about elk resource selection in an agro-forested landscape located in the Pine Ridge region of northwestern Nebraska, USA. Our objectives were to determine landscape characteristics selected by female elk and identify publicly owned land within the Pine Ridge for potential redistribution of elk. We found distance to edge of cover influenced selection of resources by female elk most and that in areas with light hunting pressure, such as ours, this selection was not driven by an avoidance of roads. Female elk selected resources positioned near ponderosa pine cover types during all seasons, exhibited a slight avoidance of roads during spring and fall, selected areas with increased slope during winter and spring, and selected north- and east-facing aspects over flat areas and areas with south-facing slopes during winter months. We used our models to identified a potential elk redistribution area that had a higher proportion of landcover with characteristics selected by elk in our study area than the current herd areas and more landcover that was publicly owned. With appropriate management plans, we believe elk within the Potential Elk Redistribution Area would predominantly occupy publicly owned land, which would help minimize crop and forage damage on privately owned lands.

Resource selection by elk in an agro-forested landscape of northwestern Nebraska.

Ecological sampling can be labor intensive, and logistically impractical in certain environments. We optimize line intercept sampling and compare estimation methods for assessing feral swine damage within fragile wetland ecosystems in Florida. Sensitive wetland sites, and the swine damage within them, were mapped using GPS technology. Evenly spaced parallel transect lines were simulated across a digital map of each site. The length of each transect and total swine damage under each transect were measured and percent swine damage within each site was estimated by two methods. The total length method (TLM) combined all transects as a single long transect, dividing the sum of all damage lengths across all transects by the combined length of all transect lines. The equal weight method (EWM) calculated the damage proportion for each transect line and averaged these proportions across all transects. Estimation was evaluated using transect spacings of 1, 3, 5, 10, 15, and 20 m. Based on relative root mean squared error and relative bias measures, the TLM produced higher quality estimates than EWM at all transect spacings. Estimation quality decreased as transect spacing increased, especially for TLM. Estimation quality also increased as the true proportion of swine damage increased. Diminishing improvements in estimation quality as transect spacings decreased suggested 5 m as an optimal tradeoff between estimation quality and labor. An inter-transect spacing of 5 m with TLM estimation appeared an optimal starting point when designing a plan for estimating swine damage, with practical, logistical, economic considerations determining final design details.

Optimizing line intercept sampling and estimation for feral swine damage levels in ecologically sensitive wetland plant communities.

Avian scavengers, such as American crows (Corvus brachyrhynchos), have potential to translocate infectious agents (prions) of transmissible spongiform encephalopathy (TSE) diseases including chronic wasting disease, scrapie, and bovine spongiform encephalopathy. We inoculated mice with fecal extracts obtained from 20 American crows that were force-fed material infected with RML-strain scrapie prions. These mice all evinced severe neurological dysfunction 196-231 d postinoculation (x =198; 95% CI: 210-216) and tested positive for prion disease. Our results suggest a large proportion of crows that consume prion-positive tissue are capable of passing infectious prions in their feces (ˆp=1.0; 95% CI: 0.8-1.0). Therefore, this common, migratory North American scavenger could play a role in the geographic spread of TSE diseases.

A striped skunk (Mephitis mephitis) rabies epizootic in northwestern Wyoming was studied from the Index Case in 1988 to the last case in 1993, and possibly is the first rabies epizootic in a previously rabies-free zone monitored from beginning to end. The 843 km(2) study area comprised skunk habitat along 90 km of Shoshone River's floodplain from Bighorn Lake upstream to Cody. Of 1,015 skunks tested, 215 were rabies-positive. Integrating spatial and temporal data from positive cases, we analyzed the epizootic's movements and dynamics at 6-month intervals using multivariate movement maps, a new multivariate descriptive methodology presented here to demonstrate the epizootic's directional flow, while illustrating areas with higher case densities (i.e., wave crests). This approach should help epidemiologists and public health officials to better understand future rabies epizootics.

Graphically Characterizing the Movement of a Rabid Striped Skunk Epizootic Across the Landscape in Northwestern Wyoming.

Mechanisms for the spread of transmissible spongiform encephalopathy diseases, including chronic wasting disease (CWD) in North American cervids, are incompletely understood, but primary routes include horizontal and environmental transmission. Birds have been identified as potential vectors for a number of diseases, where they ingest or are exposed to infected material and later shed the disease agent in new areas after flying substantial distances. We recently identified American crows (Corvus brachyrhynchos) as having the potential to translocate infectious prions in their feces. Our results suggest that this common, migratory North American scavenger is capable of translocating infectious prions to disease-free areas, potentially seeding CWD infection where no other initial source of pathogen establishment is forthcoming. Here we speculate on the role avian scavengers, like American crows, might play in the spatial dissemination of CWD. We also consider the role mammalian scavengers may play in dispersing prions.
