The goal of this work was to shed more light on a preliminary finding about the relationship between the substitution in the thioacyl part of thiobenzanilides and their antituberculous effect. Thus, we prepared a set of 14 derivatives, out of which eight had not yet been reported, and the compounds were evaluated for antimycobacterial activity on a panel of four Mycobacteria species, including Mycobacterium tuberculosis CNCTC My 331/88, Mycobacterium kansasii CNCTC My 235/80, Mycobacterium avium CNCTC My 330/88 and M. kansasii 6509/96. While the contribution of the substituents with differing electronic and lipophilicity characteristics in position 3 to the antituberculous activity was negligible, we found that unsubstituted position 4 in the thioacyl part appears to be a prerequisite for a thiobenzanilide derivative to possess appreciable biological activity.

L&#39;objectif de ce travail &eacute;tait de jeter plus de lumi&egrave;re sur une conclusion pr&eacute;liminaire sur la relation entre la substitution dans le cadre de thioacyles de thiobenzanilides et de leur effet antituberculeux. Ainsi, nous avons pr&eacute;par&eacute; un ensemble de 14 d&eacute;riv&eacute;s, dont huit n&#39;avaient pas encore &eacute;t&eacute; signal&eacute;, et les compos&eacute;s ont &eacute;t&eacute; &eacute;valu&eacute;s pour activit&eacute; antimycobact&eacute;riens sur un panel de quatre esp&egrave;ces de mycobact&eacute;ries, dont Mycobacterium tuberculosis CNCTC My 331/88, Mycobacterium kansasii CNCTC My 235/80, Mycobacterium avium CNCTC My 330/88 et M. kansasii 6509/96. L&#39;obiettivo di questo lavoro &egrave; stato a far luce su un accertamento preliminare circa il rapporto tra la sostituzione nella parte thioacyl del thiobenzanilides e loro effetto antitubercolare. Cos&igrave;, abbiamo preparato una serie di 14 derivati, di cui otto non era ancora stato segnalato, e i composti sono stati valutati per attivit&agrave; antimicobatterica su un pannello di quattro specie di micobatteri, compreso il Mycobacterium tuberculosis CNCTC My 331/88, Mycobacterium kansasii CNCTC My 235/80, Mycobacterium avium CNCTC mio 330/88 e M. kansasii 6509/96. Mentre il contributo dei sostituenti con diverse elettroniche e caratteristiche lipofilia in posizione 3 per l&#39;attivit&agrave; antitubercolare fu trascurabile, abbiamo trovato che posizione 4 nella parte thioacyl sembra essere un prerequisito per un derivato thiobenzanilide di possedere apprezzabili attivit&agrave; biologica.

El objetivo de este trabajo fue a arrojar m&aacute;s luz sobre un hallazgo preliminar sobre la relaci&oacute;n entre la sustituci&oacute;n en la parte de thioacyl de thiobenzanilides y su efecto antituberculosa. As&iacute;, nos prepar&oacute; una serie de 14 derivados, de los cuales ocho a&uacute;n no hab&iacute;an sido denunciada, y se evaluaron los compuestos para actividad antimycobacterial en un panel de cuatro especies de micobacterias, incluyendo Mycobacterium tuberculosis CNCTC My 331/88, Mycobacterium kansasii CNCTC My 235/80, CNCTC mi 330/88 de Mycobacterium avium y M. kansasii 6509/96. Mientras que la contribuci&oacute;n de las caracter&iacute;sticas de la lipofilia en la posici&oacute;n 3 a la actividad antituberculosa y sustitutos con diferentes electr&oacute;nica era insignificante, encontramos que no sustituido posici&oacute;n 4 en la parte de thioacyl parece ser un requisito previo para un derivado thiobenzanilide que poseen actividad biol&oacute;gica apreciable.

Two series of 3-(substituted phenyl)-5-alkyl-2,5-dihydrofuran-2-ones related to a natural product, (-)incrustoporine, were synthesized and their in vitro antifungal activity evaluated. The compounds with halogen substituents on the phenyl ring exhibited selective antifungal activity against the filamentous strains of Absidia corymbifera and Aspergillus fumigatus. On the other hand, the influence of the length of the alkyl chain at C(5) was marginal. The antifungal effect of the most active compound against the above strains was higher than that of ketoconazole, and close to that of amphotericin B. In order to verify the hypothesis about a possible relationship between the Michael-accepting ability of the compounds and their antifungal activity, a series of simple carbanalogues, 2-(substituted phenyl)cyclopent-2-enones, was prepared and subjected to antifungal activity assay as well.

Deux s&eacute;ries de 3-(phenyl)-5-alkyl-2,5-dihydrofuran-2-ones substitu&eacute;s, associ&eacute;s &agrave; un produit naturel, (-) incrustoporine, ont &eacute;t&eacute; synth&eacute;tis&eacute;s et &eacute;valu&eacute; leur activit&eacute; antifongique in vitro. Les compos&eacute;s portant des substituants halog&egrave;nes sur le noyau benz&eacute;nique expos&eacute; s&eacute;lective activit&eacute; antifongique contre les souches filamenteuses de Absidia corymbifera et Aspergillus fumigatus. En revanche, l&#39;influence de la longueur de la cha&icirc;ne alkyle en c (5) a &eacute;t&eacute; marginale. L&#39;effet antifongique du compos&eacute; plus actif contre les souches ci-dessus &eacute;tait sup&eacute;rieur &agrave; celui du k&eacute;toconazole et proche de celle de l&#39;amphot&eacute;ricine b. Due serie di 3-(sostituito con phenyl)-5-alkyl-2,5-dihydrofuran-2-ones relative a un prodotto naturale, incrustoporine (-), sono stati sintetizzati e loro attivit&agrave; antimicotica in vitro valutati. I composti con sostituenti alogeni sull&#39;anello fenil esposto selettiva attivit&agrave; antifungina contro i ceppi filamentosi di Absidia corymbifera e Aspergillus fumigatus. D&#39;altra parte, l&#39;influenza della lunghezza della catena alchilica a C(5) &egrave; stato marginale. L&#39;effetto antimicotico del composto pi&ugrave; attivo contro i ceppi di cui sopra era superiore a quella del ketoconazolo e vicino a quello di amfotericina b. Al fine di verificare l&#39;ipotesi circa una possibile relazione tra la capacit&agrave; di accettare Michael dei composti e la loro attivit&agrave; antimicotica, una serie di semplici carbanalogues,-(substituted phenyl) 2 cyclopent-2-enones, &egrave; stato preparato e sottoposti a dosaggio attivit&agrave; antifungina pure.

Dos series de 3-(phenyl)-5-alkyl-2,5-dihydrofuran-2-ones substituido relacionadas con un producto natural, incrustoporine (-), se sintetizaron y evalu&oacute; su actividad antif&uacute;ngica in vitro. Los compuestos con sustituyentes hal&oacute;geno en el anillo fenilo exhibieron actividad antif&uacute;ngica selectiva contra las cepas filamentosas de Absidia corymbifera y Aspergillus fumigatus. Por otro lado, la influencia de la longitud de la cadena en estudio fue marginal. El efecto antif&uacute;ngico del compuesto m&aacute;s activo frente a las cepas anteriormente fue mayor que la de ketoconazol y cerca de anfotericina B. Con el fin de verificar la hip&oacute;tesis sobre una posible relaci&oacute;n entre la capacidad de aceptar de Michael de los compuestos y su actividad antif&uacute;ngica, una serie de carbanalogues simple,-(substituted phenyl) 2 2-metil-4-oxo-3-(Prop-2-inil)ciclopent-2-en-1-ilo/Praletrin-2-enones, fue preparado y sometido a ensayo de actividad antif&uacute;ngica as&iacute;.

A new fluorogenic substrate was developed for 3alpha-hydroxysteroid dehydrogenases (3alpha-HSD), including the human enzymes implicated in important physiological functions (androgen deactivation, neurosteroid activation). While ketone 5 is nonfluorescent, the corresponding alcohol exhibits high fluorescence with emission maximum at 510 nm, thus constituting a redox optical switch. This study began with a chemical concept of a ketone-alcohol optical switch which guided the synthesis of a focused array of compounds. Subsequently, seven compounds were selected (1-7) on the basis of their optical and chemical (stability) properties and were submitted to a screen against a panel of dehydrogenase enzymes. Probe 5 was found to be highly selective for bacterial, rat, and human 3alpha-HSD enzymes. The kinetic parameters were obtained for human 3alpha-HSD enzyme (type 2 isozyme, AKR 1C3; Km = 2.5 muM, kcat = 8.2 min-1). Remarkably, comparison to 5alpha-dihydrotestosterone (5alpha-DHT, Km = 26 muM, kcat = 0.25 min-1, Figure 4), a likely physiological substrate in prostate, revealed that synthetic probe 5 is in fact a far better substrate for this enzyme. Structure 5 represents an exciting lead for the development of a redox imaging probe.

Un nuovo substrato fluorogenic &egrave; stato sviluppato per 3alfa-idrossisteroide dehydrogenases (3alfa-HSD), compresi gli enzimi umani implicati in importanti funzioni fisiologiche (disattivazione dell&#39;androgeno, neurosteroide attivazione). Mentre chetone 5 &egrave; nonfluorescent, il corrispondente alcol esibisce elevata fluorescenza con emissione massima di 510 nm, costituendo cos&igrave; un interruttore ottico redox. Questo studio ha cominciato con un concetto di un interruttore ottico chetone alcool che guidava la sintesi di una matrice di concentrato di composti chimico. The current arsenal of tools and methods for the continuous monitoring and imaging of redox metabolic pathways in the context of intact cells is limited. Fluorogenic substrates allow for direct measurement of enzyme activity in situ; however, in contrast to proteases and exo-glycosidases, there are no simple guidelines for the design of selective probes for redox metabolic enzymes. Here, we introduce redox probe 1 and demonstrate its high selectivity in living cells for human hydroxysteroid dehydrogenases (HSDs) of the aldo-keto reductase (AKR) superfamily. AKR1C isoforms perform multiple functions among which the metabolism of potent steroid hormones is well documented. Moreover, expression of these enzymes is responsive to cellular stress and pathogenesis, including cancer. Our probe design is based on redox-sensitive optical switches, which couple a ketone-alcohol redox event to a profound change in fluorescence. The high selectivity of phenyl ketone 1 for AKR1C2 over the many endogenous reductases present in mammalian cells was established by a quantitative comparison of the metabolic rates between null control cells (COS-1) and AKR1C2-transfected cells. Phenyl ketone 1 is a cell-permeable fluorogenic probe that permits a direct, real-time, and operationally simple readout of AKR1C2 enzyme activity in intact mammalian cells. Furthermore, it was demonstrated that probe 1 enables the quantitative examination of physiological substrate 5alpha-dihydrotestosterone ("dark substrate") in situ by means of a two-substrate competitive assay. Similarly, inhibitor potency of physiological (ursodeoxycholate) and synthetic inhibitors (flufenamic acid, ibuprofen, and naproxen) was also readily evaluated.

The nervous system transmits signals between neurons via neurotransmitter release during synaptic vesicle fusion. In order to observe neurotransmitter uptake and release from individual presynaptic terminals directly, we designed fluorescent false neurotransmitters as substrates for the synaptic vesicle monoamine transporter. Using these probes to image dopamine release in the striatum, we made several observations pertinent to synaptic plasticity. We found that the fraction of synaptic vesicles releasing neurotransmitter per stimulus was dependent on the stimulus frequency. A kinetically distinct "reserve" synaptic vesicle population was not observed under these experimental conditions. A frequency-dependent heterogeneity of presynaptic terminals was revealed that was dependent in part on D2 dopamine receptors, indicating a mechanism for frequency-dependent coding of presynaptic selection.

Le syst&#xE8;me nerveux transmet des signaux entre les neurones via la lib&#xE9;ration de neurotransmetteurs lors de la fusion des v&#xE9;sicules synaptiques. Afin d&#x26;#39;observer l&#x26;#39;absorption et la lib&#xE9;ration de neurotransmetteurs individuels terminaisons pr&#xE9;synaptiques directement, nous avons con&#xE7;u fluorescentes faux neurotransmetteurs comme substrats pour le transporteur de la v&#xE9;sicule synaptique monoamine. L&#x26;#39;utilisation de ces sondes pour la lib&#xE9;ration de dopamine dans le striatum l&#x26;#39;image, nous avons fait plusieurs observations pertinentes &#xE0; la plasticit&#xE9; synaptique. Nous avons constat&#xE9; que la fraction de v&#xE9;sicules synaptiques lib&#xE9;rant des neurotransmetteurs par stimulus &#xE9;tait d&#xE9;pendante de la fr&#xE9;quence du stimulus. Un cin&#xE9;tiquement distinctes &#xAB;r&#xE9;serve&#xBB; de la population des v&#xE9;sicules synaptiques n&#x26;#39;a pas &#xE9;t&#xE9; observ&#xE9; dans ces conditions exp&#xE9;rimentales. Das Nervensystem &uuml;bertr&auml;gt Signale zwischen Neuronen &uuml;ber Neurotransmitter-Freisetzung w&auml;hrend synaptische Vesikel Verschmelzung. Um Neurotransmitter Aufnahme zu beobachten und Pressemitteilung vom einzelnen pr&auml;synaptischen Klemmen direkt, gestalteten wir fluoreszierende falsche Neurotransmitter als Substrate f&uuml;r die synaptische Vesikel Monoamin-Transporter. Verwenden diese Pr&uuml;fpunkte auf Bild-Dopamin-Release im Striatum, haben wir mehrere Beobachtungen, die synaptische Plastizit&auml;t. Wir fanden, dass der Anteil der synaptische Vesikel, die Freigabe Neurotransmitter pro Impuls die Impuls-Frequenz abh&auml;ngig war. Kinetisch unterschiedliche &quot;Reservieren&quot; synaptische Vesikel Einwohner wurde unter diesen Versuchsbedingungen nicht beobachtet. Il sistema nervoso trasmette segnali tra neuroni tramite rilascio di neurotrasmettitore durante la fusione della vescicola sinaptica. Al fine di osservare assorbimento del neurotrasmettitore e rilasciare direttamente dai singoli terminali presinaptiche, abbiamo progettato fluorescenti falsi neurotrasmettitori come substrati per il trasportatore di monoamine Vescicola sinaptica. Usando queste sonde a immagine rilascio di dopamina nello striato, abbiamo fatto diverse osservazioni pertinenti a sinaptico plasticit&agrave;. Abbiamo trovato che la frazione di vescicole sinaptiche rilasciando il neurotrasmettitore per stimolo era dipende la frequenza di stimolo. Una popolazione di Vescicola sinaptica cineticamente distinti &quot;riserva&quot; non &egrave; stata osservata in queste condizioni sperimentali. Un&#39;eterogeneit&agrave; dipendente dalla frequenza dei terminali presinaptiche &egrave; stata rivelata che era dipendente in parte su ricevitori della dopamina D2, che indica un meccanismo per la codifica di frequenza-dipendente della selezione presinaptica.

El sistema nervioso transmite se&#xF1;ales entre las neuronas a trav&#xE9;s de la liberaci&#xF3;n de neurotransmisores durante la fusi&#xF3;n de ves&#xED;culas sin&#xE1;pticas. Con el fin de observar la captaci&#xF3;n de neurotransmisores y liberaci&#xF3;n de terminales presin&#xE1;pticas individuales directamente, hemos dise&#xF1;ado fluorescentes falsos neurotransmisores como sustratos para el transportador de la ves&#xED;cula sin&#xE1;ptica monoamina. El uso de estas sondas a la liberaci&#xF3;n de la imagen de la dopamina en el estriado, que hizo varias observaciones pertinentes a la plasticidad sin&#xE1;ptica. Hemos encontrado que la fracci&#xF3;n de ves&#xED;culas sin&#xE1;pticas liberaci&#xF3;n de neurotransmisores por est&#xED;mulo era dependiente de la frecuencia del est&#xED;mulo. Una cin&#xE9;tica distinta &#x26;quot;reserva&#x26;quot; de la poblaci&#xF3;n de ves&#xED;culas sin&#xE1;pticas no se observ&#xF3; en estas condiciones experimentales. Una heterogeneidad dependiente de la frecuencia de los terminales presin&#xE1;pticos se revel&#xF3; que era dependiente en parte de los receptores de dopamina D2, lo que indica un mecanismo para dependientes de la frecuencia de codificaci&#xF3;n de selecci&#xF3;n presin&#xE1;ptica.
