Machado-Joseph disease, also known as spinocerebellar ataxia type 3, is the most common of the dominantly inherited ataxias worldwide and is characterized by mutant ataxin-3 misfolding, intracellular accumulation of aggregates and neuronal degeneration. Here we investigated the implication of autophagy, the major pathway for organelle and protein turnover, in the accumulation of mutant ataxin-3 aggregates and neurodegeneration found in Machado-Joseph disease and we assessed whether specific stimulation of this pathway could mitigate the disease. Using tissue from patients with Machado-Joseph disease, transgenic mice and a lentiviral-based rat model, we found an abnormal expression of endogenous autophagic markers, accumulation of autophagosomes and decreased levels of beclin-1, a crucial protein in the early nucleation step of autophagy. Lentiviral vector-mediated overexpression of beclin-1 led to stimulation of autophagic flux, mutant ataxin-3 clearance and overall neuroprotective effects in neuronal cultures and in a lentiviral-based rat model of Machado-Joseph disease. These data demonstrate that autophagy is a key degradation pathway, with beclin-1 playing a significant role in alleviating Machado-Joseph disease pathogenesis.

Transplantation of photoreceptor precursor cells (PPCs) into the retina represents a promising treatment for cell replacement in blinding diseases characterized by photoreceptor loss. In preclinical studies, we and others demonstrated that grafted PPCs integrate into the host outer nuclear layer (ONL) and develop into mature photoreceptors. However, a key feature of light detecting photoreceptors, the outer segment (OS) with natively aligned disc membrane staples, has not been studied in detail following transplantation. Therefore, we used as donor cells PPCs isolated from neonatal double transgenic reporter mice in which OSs are selectively labeled by green fluorescent protein while cell bodies are highlighted by red fluorescent protein. PPCs were enriched using CD73-based magnetic associated cell sorting and subsequently transplanted into either adult wild-type or a model of autosomal-dominant retinal degeneration mice. Three weeks post-transplantation, donor photoreceptors were identified based on fluorescent-reporter expression and OS formation was monitored at light and electron microscopy levels. Donor cells that properly integrated into the host wild-type retina developed OSs with the formation of a connecting cilium and well-aligned disc membrane staples similar to the surrounding native cells of the host. Surprisingly, the majority of not-integrated PPCs that remained in the sub-retinal space also generated native-like OSs in wild-type mice and those affected by retinal degeneration. Moreover, they showed an improved photoreceptor maturation and OS formation by comparison to donor cells located on the vitreous side suggesting that environmental cues influence the PPC differentiation and maturation. We conclude that transplanted PPCs, whether integrated or not into the host ONL, are able to generate the cellular structure for effective light detection, a phenomenon observed in wild-type as well as in degenerated retinas. Given that patients suffering from retinitis pigmentosa lose almost all photoreceptors, our findings are of utmost importance for the development of cell-based therapies.

Outer segment formation of transplanted photoreceptor precursor cells.

La transplantation de cellules photor&eacute;ceptrices de pr&eacute;curseur (SPAC) dans la r&eacute;tine repr&eacute;sente un traitement prometteur pour le remplacement de la cellule en aveuglant les maladies caract&eacute;ris&eacute;es par une perte des photor&eacute;cepteurs. Dans les &eacute;tudes pr&eacute;cliniques, nous et autres ont d&eacute;montr&eacute; que SPAC greff&eacute;es int&eacute;grer la couche nucl&eacute;aire externe h&ocirc;te (ONL) et qu&#39;il devienne matures photor&eacute;cepteurs. Toutefois, un &eacute;l&eacute;ment cl&eacute; de la d&eacute;tection des photor&eacute;cepteurs, le segment externe (OS) avec des agrafes de disque en mode natif align&eacute;s de membrane, la lumi&egrave;re n&#39;a pas &eacute;t&eacute; &eacute;tudi&eacute; en d&eacute;tail apr&egrave;s la transplantation. Par cons&eacute;quent, nous avons utilis&eacute; comme cellules donneuses SPAC isol&eacute;s de souris n&eacute;onatales journaliste transg&eacute;niques double dans laquelle OSs sont s&eacute;lectivement marqu&eacute;s par la prot&eacute;ine fluorescente verte tandis que les corps cellulaires sont mises en &eacute;vidence par une prot&eacute;ine fluorescente rouge. CCP ont &eacute;t&eacute; enrichies &agrave; l&#39;aide de CD73 base magn&eacute;tique cellule associ&eacute;e tri et ensuite transplant&eacute;es sauvage adulte ou un mod&egrave;le de souris de d&eacute;g&eacute;n&eacute;rescence r&eacute;tinienne autosomique dominante. Trois semaines apr&egrave;s, donneur photor&eacute;cepteurs ont &eacute;t&eacute; identifi&eacute;s bas&eacute; sur expression fluorescentes-journaliste et formation de l&#39;OS a &eacute;t&eacute; suivie &agrave; niveaux de microscopie photonique et &eacute;lectronique. Cellules donneuses qui correctement int&eacute;gr&eacute; dans la r&eacute;tine de type sauvage h&ocirc;te d&eacute;velopp&eacute; OSs avec la formation d&#39;un cil de raccordement et bien align&eacute;es disque membrane staples semblables aux cellules environnantes natifs de l&#39;h&ocirc;te. &Eacute;tonnamment, la majorit&eacute; des CCP non int&eacute;gr&eacute; qui est rest&eacute; dans l&#39;espace sous-r&eacute;tinien a &eacute;galement g&eacute;n&eacute;r&eacute; des OSs natif chez les souris de type sauvage et les personnes touch&eacute;es par la d&eacute;g&eacute;n&eacute;rescence r&eacute;tinienne. En outre, ils ont montr&eacute; une maturation photor&eacute;cepteur am&eacute;lior&eacute;e et la formation de l&#39;OS par comparaison aux cellules du donneur situ&eacute;s sur le c&ocirc;t&eacute; vitr&eacute;e, ce qui sugg&egrave;re que les signaux environnementaux influencent la PPC diff&eacute;renciation et la maturation. Transplantation von Sehzellen Vorl&auml;ufer (PPCs) in der Netzhaut stellt eine vielversprechende Behandlung f&uuml;r Zelle Ersatz in blendender Krankheiten mit Photorezeptoren Verlust dar. In pr&auml;klinischen Studien gezeigt wir u.a., dass Aufgepfropfte PPCs die Host nukleare Au&szlig;enschicht (ONL integrieren) und entwickeln sich zu Reifen Photorezeptoren. Ein wesentliches Merkmal des Lichts erkennen von Photorezeptoren, das &auml;u&szlig;ere Segment (OS) mit nativ ausgerichteten CD Membran Staples, ist jedoch nicht im Detail nach Transplantation gepr&uuml;ft worden. Daher haben wir als Spenderzellen PPCs isoliert neonatale Doppel Reporter transgene M&auml;use in die OSs von gr&uuml;n fluoreszierendes Protein selektiv beschriftet werden, w&auml;hrend Zelle K&ouml;rper von rot fluoreszierende Protein hervorgehoben werden verwendet. KKKS wurden angereichert mit CD73-basierte magnetische zugeordnete Zelle Sortieren und anschlie&szlig;end in ein Erwachsener Wildtyp oder ein Modell der autosomal-dominanten Netzhautdegeneration M&auml;use transplantiert. Drei Wochen Posttransplantationsverbesserungen, Spender, die Photorezeptoren identifiziert wurden basierend auf Leuchtstoff-Reporter Ausdruck und OS Bildung wurde auf Licht- und Elektronenmikroskopie Ebenen &uuml;berwacht. Spenderzellen, die ordnungsgem&auml;&szlig; in der Host-Wildtyp-Netzhaut integriert entwickelte OSs mit der Bildung einer verbindenden Zilie und einheitliches CD Membran Staples &auml;hnlich wie die umliegenden nativen Zellen des Wirtes. &Uuml;berraschend, die Mehrheit der nicht-integrierte PPCs, die in den subretinalen Raum blieb auch Native-wie OSs Wildtyp M&auml;usen erzeugt und die von Netzhautdegeneration betroffen. Dar&uuml;ber hinaus zeigten sie eine verbesserte Photorezeptor Reifung und OS-Bildung im Vergleich zu Spenderzellen befindet sich auf der Glask&ouml;rper Seite darauf hindeutet, dass &ouml;kologische Hinweise die PPC Differenzierung und Reifung beeinflussen. Wir schlie&szlig;en, dass PPCs, transplantiert, ob integriert oder nicht in der Host-ONL, k&ouml;nnen die Zellstruktur f&uuml;r effektive leichte Entdeckung, erzeugen ein Ph&auml;nomen im Wildtyp ebenso wie in degenerierten Netzhaut beobachtet. Angesichts der Tatsache, dass Patienten mit Retinitis Pigmentosa fast alle Photorezeptoren verlieren, sind unsere Ergebnisse von gr&ouml;&szlig;ter Bedeutung f&uuml;r die Entwicklung von zellbasierte Therapien.

&Auml;u&szlig;ere Segment Bildung von transplantierten Sehzellen Vorl&auml;ufer.

Il trapianto di cellule precursori di fotorecettori (PPCs) nella retina rappresenta un promettente trattamento per la sostituzione delle cellule nell&#39;accecante malattie caratterizzate da perdita di fotorecettori. In studi preclinici, noi e gli altri hanno dimostrato che innestati PPCs integrare lo strato nucleare esterno (ONL) di host e sviluppare in fotorecettori maturi. Tuttavia, una caratteristica chiave della luce rilevazione fotorecettori, il segmento esterno (OS) con graffette membrana disco allineato in modo nativo, non &egrave; stata studiata in dettaglio dopo trapianto. Trasplante de c&eacute;lulas precursoras de fotorreceptor (PPCs) en la retina representa un tratamiento prometedor para el reemplazo de la c&eacute;lula en cegamiento enfermedades caracterizadas por la p&eacute;rdida de fotorreceptores. En estudios precl&iacute;nicos, nosotros y otros demostraron que PPCs injertados integrar&aacute;n en la capa nuclear externa del anfitri&oacute;n (ONL) y convierten en fotorreceptores maduros. Sin embargo, una caracter&iacute;stica clave de la luz, detectar los fotorreceptores, el segmento externo (OS) con grapas de membrana de disco alineado de forma nativa, no se ha estudiado en detalle despu&eacute;s del trasplante. Por lo tanto, se utiliz&oacute; como c&eacute;lulas de un donante PPCs aislados de ratones transg&eacute;nicos doble neonatal de la reportera en que OSs selectivamente est&aacute;n marcadas por la prote&iacute;na verde fluorescente mientras que los cuerpos celulares se destacan por la prote&iacute;na fluorescente roja. PPCs fueron enriquecidos usando basadas en CD73 de magn&eacute;tico celular asociada clasificaci&oacute;n y posteriormente trasplantado en adultos tipo salvaje o un modelo de ratones de degeneraci&oacute;n retiniana autos&oacute;mica dominante. Poste-trasplante de tres semanas, donante fotorreceptores se identificaron basado en expresi&oacute;n fluorescente-reportero y formaci&oacute;n OS supervisaron en los niveles de luz y microscopia electr&oacute;nica. C&eacute;lulas de un donante que integraron correctamente en la retina de salvaje-tipo anfitri&oacute;n desarrollaron OSs con la formaci&oacute;n de un cilio que conecta y grapas de membrana de disco bien alineada similares a las c&eacute;lulas circundantes nativas del anfitri&oacute;n. Sorprendentemente, la mayor&iacute;a de PPCs no integrado que permaneci&oacute; en el espacio subretinianos tambi&eacute;n genera nativo OSs en ratones de tipo salvaje y los afectados por la degeneraci&oacute;n retiniana. Adem&aacute;s, demostr&oacute; una maduraci&oacute;n fotorreceptor mejorada y formaci&oacute;n OS en comparaci&oacute;n a las c&eacute;lulas del donante situadas en el lado v&iacute;trea sugiriendo que se&ntilde;ales ambientales influyen en la diferenciaci&oacute;n de la PPC y la maduraci&oacute;n. Concluimos que trasplantaron PPCs, ya sea integrado o no en el host ONL, son capaces de generar la estructura celular para la detecci&oacute;n eficaz de luz, un fen&oacute;meno observado en wild-type, as&iacute; como en las retinas degeneradas. Dado que los pacientes que sufren de retinosis pigmentaria pierden casi todos fotorreceptores, nuestros hallazgos son de suma importancia para el desarrollo de terapias basadas en c&eacute;lulas.
