We present the first microfluidic method for extracting proteins from heterogeneous fluids by precipitation. The new method comprises an automated protocol for precipitation of proteins onto surfaces, rinsing the precipitates to remove impurities, and resolubilization in buffer for further analysis. The method is compatible with proteins representing a range of different physicochemical properties, as well as with complex mixtures such as fetal bovine serum and cell lysate. In all cases, the quantitative performance (measured using a fluorescent assay for % recovery) was comparable to that of conventional techniques, which are manual and require more time. Thus, this work represents an important first step in efforts to develop fully automated microfluidic methods for proteomic analyses.

Nous pr&#xE9;sentons la premi&#xE8;re m&#xE9;thode microfluidique pour extraire les prot&#xE9;ines &#xE0; partir de fluides h&#xE9;t&#xE9;rog&#xE8;nes par les pr&#xE9;cipitations. La nouvelle m&#xE9;thode comprend un protocole automatis&#xE9; pour la pr&#xE9;cipitation des prot&#xE9;ines sur des surfaces, le rin&#xE7;age de la pr&#xE9;cipit&#xE9;s pour retirer les impuret&#xE9;s, et resolubilisation dans un tampon pour une analyse ult&#xE9;rieure. Wir pr&auml;sentieren die erste Microfluidic-Methode zur Extraktion von Proteinen aus heterogenen Fl&uuml;ssigkeiten durch Niederschl&auml;ge. Die neue Methode umfasst eine automatisierte Protokoll zur F&auml;llung von Proteinen auf Oberfl&auml;chen, Sp&uuml;lung der Ausscheidungen zum Entfernen von Verunreinigungen und Resolubilization im Puffer zur weiteren Analyse. Die Methode ist kompatibel mit den Proteinen, die eine Palette von unterschiedlichen physikalisch-chemischen Eigenschaften sowie komplexe Mischungen wie Rinderf&ouml;tenserum und Zelle lysate. In allen F&auml;llen war die quantitative Leistung (gemessen mit einem Fluoreszenz-Assay f&uuml;r Wiederaufnahme %) vergleichbar mit denen von herk&ouml;mmlichen Techniken, die manuelle und ben&ouml;tigen mehr Zeit. Daher steht diese Arbeit einen wichtigen ersten Schritt in Bem&uuml;hungen, vollautomatische mikrofluidische Methoden f&uuml;r Proteomic Analysen zu entwickeln.

Vi presentiamo il primo metodo di microfluidica per l&#39;estrazione di proteine da fluidi eterogenei di precipitazione. Il nuovo metodo comprende un protocollo automatizzato per precipitazione delle proteine sulle superfici, risciacquo i precipitati per rimuovere le impurit&agrave; e resolubilization nel buffer per ulteriori analisi. Il metodo &egrave; compatibile con le proteine che rappresentano una vasta gamma di differenti propriet&agrave; fisico-chimiche, cos&igrave; come con miscele complesse come siero bovino fetale e lysate delle cellule. In tutti i casi, le prestazioni quantitative (misurata usando un dosaggio fluorescente per % di recupero) era paragonabile a quella delle tecniche convenzionali, che sono manuali e richiedono pi&ugrave; tempo. Cos&igrave;, questo lavoro rappresenta un primo passo importante negli sforzi per sviluppare metodi di microfluidica completamente automatizzata per le analisi proteomic.

Estrogen is a key hormone in human reproductive physiology, controlling ovulation and secondary sexual characteristics. In addition, it plays an important role in the pathogenesis of breast cancer. Indeed, estrogen receptor antagonists and aromatase inhibitors (which block estrogen biosynthesis) are primary drugs used for treatment and prevention in at-risk populations. Despite its importance, tissue concentrations of estrogen are not routinely measured because conventional techniques require large samples of biopsies for analysis. In response to this need, we have developed a digital microfluidic method and applied it to the extraction and quantification of estrogen in 1-microliter samples of breast tissue homogenate (as would be collected with fine-needle aspiration), as well as in whole blood and serum. This method may be broadly applicable to conditions requiring frequent analysis of hormones in clinical samples (for example, infertility and cancer).

Microchannels can separate analytes faster with higher resolution, higher efficiency and with lower reagent consumption than typical column techniques. Unfortunately, an impediment in the path toward fully integrated microchannel-based laboratories-on-a-chip is the integration of preseparation sample processing. In contrast, the alternative format of digital microfluidics (DMF), in which discrete droplets are manipulated on an array of electrodes, is well-suited for carrying out sequential chemical reactions such as those commonly employed in proteomic sample preparation. We recently reported a new paradigm of "hybrid microfluidics," integrating DMF with microchannels for in-line sample processing and separations. Here, we build on our initial efforts, introducing a second-generation hybrid microfluidic device architecture. In the new multilayer design, droplets are manipulated by DMF in the two-plate format, an improvement that facilitates dispensing samples from reservoirs, as well as droplet splitting and storage for subsequent analysis. To demonstrate the capabilities of the new method, we implemented an on-chip serial dilution experiment, as well as multistep enzymatic digestion. Given the myriad applications requiring preprocessing and chemical separations, the hybrid digital-channel format has the potential to become a powerful new tool for micro total analysis systems.

Ici, nous nous appuyons sur nos efforts initiaux, l&#x26;#39;introduction d&#x26;#39;une deuxi&#xE8;me g&#xE9;n&#xE9;ration d&#x26;#39;architecture hybride dispositif microfluidique. Dans la nouvelle conception multicouche, les gouttelettes sont manipul&#xE9;s par le DMF en forme de deux plaques, une am&#xE9;lioration qui facilite &#xE9;chantillons de distribution &#xE0; partir de r&#xE9;servoirs, ainsi que des gouttelettes de fendage et de stockage pour une analyse ult&#xE9;rieure. Electrospray ionization (ESI) has revolutionized mass spectrometry (MS), providing a facile method for the ionization of macromolecules for analysis by mass. The development of nanoESI-MS has further extended the utility of ESI-MS, permitting the analysis of small-volume samples with enhanced sensitivity over conventional ESI-MS. Traditional nanoESI-MS experiments use pulled-glass capillary emitters, which are expensive to purchase and require specialized instruments and training to fabricate in-house. Furthermore, these emitters suffer from problems including clogging, sample contamination, and irreproducible spray stability. Here, we report a new emitter for nanoESI-MS, made by folding small pieces of polyimide tape. In comparison with conventional pulled-glass capillary emitters, the new emitters are inexpensive and simple to make. Their low cost makes them disposable after a single use, such that sample contamination or clogging is never a problem. Emitter performance has been evaluated for diverse analytes encompassing a large mass range, including small molecules, peptides, proteins, and synthetic polymers. In all cases, the performance is similar to that of pulled-glass capillary emitters, with the advantages of low cost, ease of use, and disposability.

We introduce the marriage of two technologies: digital microfluidics (DMF), a technique in which droplets are manipulated by application of electrostatic forces on an array of electrodes coated by an insulator, and porous polymer monoliths (PPMs), a class of materials that is popular for use for solid-phase extraction and chromatography. In this work, circular PPM discs were formed in situ by dispensing and manipulating droplets of monomer solutions to designated spots on a DMF device followed by UV-initiated polymerization. We used PPM discs formed in this manner to develop a digital microfluidic solid-phase extraction (DMF-SPE) method, in which PPM discs are activated and equilibrated, samples are loaded, PPM discs are washed, and the samples are eluted, all using microliter droplets of samples and reagents. The new method has extraction efficiency (93%) comparable to that of pipet-based ZipTips and is compatible with preparative sample extraction and recovery for on-chip desalting, removal of surfactants, and preconcentration. We anticipate that DMF-SPE may be useful for a wide range of applications requiring preparative sample cleanup and concentration.

A digital microfluidic method for in situ formation of porous polymer monoliths with application to solid-phase extraction.

Nous introduisons le mariage de deux technologies: la microfluidique digitale (DMF), une technique dans laquelle des gouttelettes sont manipul&#xE9;s par l&#x26;#39;application de forces &#xE9;lectrostatiques sur un r&#xE9;seau d&#x26;#39;&#xE9;lectrodes enrob&#xE9;es par un isolant, et les monolithes de polym&#xE8;re poreux (PMP), une classe de mat&#xE9;riaux qui est populaires pour une utilisation pour extraction en phase solide et la chromatographie. Dans ce travail, les disques circulaires PPM ont &#xE9;t&#xE9; form&#xE9; in situ par la distribution et la manipulation des gouttelettes de solutions de monom&#xE8;res &#xE0; sites d&#xE9;sign&#xE9;s sur un dispositif de DMF suivi par polym&#xE9;risation initi&#xE9;e UV. Wir f&uuml;hren die Ehe der beiden Technologien: digitale Biochips (DMF), eine Technik, die in dem Tr&ouml;pfchen werden durch Anwendung der elektrostatische Kr&auml;fte auf ein Array von Elektroden beschichtet durch ein Isolator und por&ouml;sen Polymer Monolithen (PPMs), eine Klasse von Materialien, beliebt f&uuml;r den Einsatz f&uuml;r Festphasen-Extraktion und Chromatographie ist, manipuliert. In dieser Arbeit kreisf&ouml;rmige PPM-Scheiben wurden durch Abgabe in-situ geformt und Bearbeiten von Tr&ouml;pfchen Monomer L&ouml;sungen zu bestimmten Flecken auf einem DMF-Ger&auml;t gefolgt von UV-initiierten Polymerisation. Wir verwendet PPM-Disks, die auf diese Weise gebildet, um eine digitale mikrofluidische Festphasen-Extraktion (DMF-SPE) Methode zu entwickeln, in welche PPM-Disks aktiviert und balanciert werden, Proben werden geladen, PPM-Disks werden gewaschen und Proben sind eluiert, verwenden alle Mikroliter Tr&ouml;pfchen der Proben und Reagenzien. Vi presentiamo il matrimonio di due tecnologie: digital microfluidics (DMF), una tecnica in cui le goccioline sono manipolate da applicazione di forze elettrostatiche su una matrice di elettrodi rivestiti da un isolante e monoliti di polimero poroso (ppm), una classe di materiali che &egrave; popolare per uso per estrazione in fase solida e la cromatografia. In questo lavoro, circolare PPM dischi sono stati formati in situ di erogazione e manipolare le goccioline di soluzioni monomero ai luoghi designati su un dispositivo DMF seguita da polimerizzazione UV-avviato. Abbiamo usato dischi PPM formati in questo modo di sviluppare un metodo di estrazione di fase solida (DMF-SPE) digitale microfluidica, in quali dischi PPM sono attivati ed equilibrati, campioni vengono caricati, PPM dischi vengono lavati e i campioni vengono eluiti, il tutto utilizzando microliter goccioline di campioni e reagenti. Il nuovo metodo ha efficienza di estrazione (93%) paragonabile a quella di dispensare-based ZipTips ed &egrave; compatibile con estrazione campione preparative e recupero per on-chip, dissalazione, rimozione dei tensioattivi e preconcentration. Anticipiamo che DMF-SPE pu&ograve; essere utile per una vasta gamma di applicazioni che richiedono concentrazione e pulitura campione preparative.

Se introduce la uni&#xF3;n de dos tecnolog&#xED;as: digital de microfluidos (DMF), una t&#xE9;cnica en la que las gotas son manipulados por la aplicaci&#xF3;n de las fuerzas electrost&#xE1;ticas de una serie de electrodos recubiertos por un aislante y monolitos de pol&#xED;meros porosos (PMP), una clase de materiales que es popular para el uso de extracci&#xF3;n en fase s&#xF3;lida y cromatograf&#xED;a. En este trabajo, discos circulares PPM se forma in situ por la dispensaci&#xF3;n y la manipulaci&#xF3;n de las gotitas de mon&#xF3;mero soluciones a los puntos designados en un dispositivo de DMF seguido por polimerizaci&#xF3;n de UV-iniciado. Se utiliz&#xF3; discos PPM formados de esta manera para desarrollar un dispositivo digital de microfluidos extracci&#xF3;n en fase s&#xF3;lida (DMF-SPE) m&#xE9;todo, en el que los discos PPM se activan y se equilibra, las muestras se cargan, discos PPM se lavan, y las muestras se eluyeron, todas ellas con gotitas microlitros de las muestras y reactivos. El nuevo m&#xE9;todo tiene una eficiencia de extracci&#xF3;n (93%) comparable a la de base de pipeta-ZipTips y es compatible con la extracci&#xF3;n de la muestra preparativa y la recuperaci&#xF3;n para el chip desalado, la eliminaci&#xF3;n de agentes tensioactivos, y preconcentraci&#xF3;n. Blood samples stored as dried blood spots (DBSs) are emerging as a useful sampling and storage vehicle for a wide range of applications. Unfortunately, the surging popularity of DBS samples has not yet been accompanied by an improvement in automated techniques for extraction and analysis. As a first step towards overcoming this challenge, we have developed a prototype microfluidic system for quantification of amino acids in dried blood spots, in which analytes are extracted, mixed with internal standards, derivatized, and reconstituted for analysis by (off-line and in-line) tandem mass spectrometry. The new method is fast, robust, precise, and most importantly, compatible with automation. We propose that the new method can potentially contribute to a new generation of analytical techniques for quantifying analytes in DBS samples for a wide range of applications.

Dans un premier temps en vue de surmonter ce d&#xE9;fi, nous avons d&#xE9;velopp&#xE9; un prototype de syst&#xE8;me microfluidique pour la quantification des acides amin&#xE9;s dans des taches de sang s&#xE9;ch&#xE9;es, dans lequel les analytes sont extraits, m&#xE9;lang&#xE9;s avec les normes internes, d&#xE9;riv&#xE9;s, et reconstitu&#xE9; pour l&#x26;#39;analyse par (off-line et en en ligne) par spectrom&#xE9;trie de masse en tandem. La nouvelle m&#xE9;thode est rapide, robuste, pr&#xE9;cis, et surtout, compatible avec l&#x26;#39;automatisation. Nous proposons que la nouvelle m&#xE9;thode peut potentiellement contribuer &#xE0; une nouvelle g&#xE9;n&#xE9;ration de techniques d&#x26;#39;analyse permettant de quantifier les analytes dans les &#xE9;chantillons de DBS pour une large gamme d&#x26;#39;applications.

Blutproben als getrocknete Blutpunkte (DBSs) gespeichert entstehen als ein n&uuml;tzliches Instrument f&uuml;r Probenahme und Speicher f&uuml;r eine breite Palette von Anwendungen. Leider wurde die steigende Popularit&auml;t von DBS Proben nicht noch durch eine Verbesserung der automatisierten Techniken zur Extraktion und Analyse begleitet. Als ersten Schritt zur Bew&auml;ltigung dieser Herausforderung entwickelten wir ein Prototyp Microfluidic System zur Quantifizierung der Aminos&auml;uren in getrocknetem Blut Flecken, in denen Analyten werden extrahiert, gemischt mit internen Standards, derivatized und stellte f&uuml;r die Analyse von (offline und in-Line) tandem Massenspektrometrie. Die neue Methode ist schnell, robust, pr&auml;zise, und vor allem mit der Automatisierung kompatibel. Wir schlagen vor, dass die neue Methode m&ouml;glicherweise zu einer neuen Generation von Analyseverfahren zur Quantifizierung von Analyten im DBS-Beispiele f&uuml;r eine Vielzahl von Anwendungen beitragen kann.

Campioni di sangue conservati come macchie di sangue secco (DBSs) stanno emergendo come un utile veicolo di prelievo e stoccaggio per una vasta gamma di applicazioni. Purtroppo, la popolarit&agrave; impetuosa di campioni DBS non ancora stata accompagnata da un miglioramento di tecniche automatizzate per estrazione e analisi. Dried blood spot (DBS) samples on filter paper are surging in popularity as a sampling and storage vehicle for a wide range of clinical and pharmaceutical applications. For example, a DBS sample is collected from every baby born in the province of Ontario, Canada, for quantification of approximately one hundred analytes that are used to screen for 28 conditions, including succinylacetone (SA), a marker for hepatorenal tyrosinemia. Unfortunately, the conventional methods used to evaluate DBS samples for newborn screening and other applications are tedious and slow, with limited options for automated analysis. In response to this challenge, we have developed a method to couple digital microfluidics (DMF) to nanoelectrospray ionization mass spectrometry (nESI-MS) for SA quantification in DBS samples. The new system is formed by sandwiching a pulled glass capillary emitter between the two DMF substrates such that the capillary emitter is immobilized without external seals or gaskets. Moreover, we introduce a new feedback control system that enables high-fidelity droplet manipulation across DBS samples without manual intervention. The system was validated by application to on-chip extraction, derivatization, and analysis of SA and other analytes from DBS samples, with comparable performance to gold-standard methods. We propose that the new methods described here can potentially contribute to a new generation of analytical techniques for quantifying analytes in DBS samples for a wide range of applications.

Dried blood spot analysis by digital microfluidics coupled to nanoelectrospray ionization mass spectrometry.

Digital microfluidics (DMF) has recently emerged as a popular technology for a wide range of applications. In DMF, nanoliter to microliter droplets containing samples and reagents can be manipulated to carry out a range of discrete fluidic operations simply by applying a series of electrical potentials to an array of patterned electrodes coated with a hydrophobic insulator. DMF is distinct from microchannel-based fluidics as it allows for precise control over multiple reagent phases (liquids and solids) in heterogeneous systems with no need for complex networks of connections, microvalves, or pumps. In this review, we discuss the most recent developments in this technology with particular attention to the potential benefits and outstanding challenges for applications in chemistry, biology, and medicine.

Digital microfluidics: a versatile tool for applications in chemistry, biology and medicine.

Next-generation sequencing (NGS) is emerging as a powerful tool for elucidating genetic information for a wide range of applications. Unfortunately, the surging popularity of NGS has not yet been accompanied by an improvement in automated techniques for preparing formatted sequencing libraries. To address this challenge, we have developed a prototype microfluidic system for preparing sequencer-ready DNA libraries for analysis by Illumina sequencing. Our system combines droplet-based digital microfluidic (DMF) sample handling with peripheral modules to create a fully-integrated, sample-in library-out platform. In this report, we use our automated system to prepare NGS libraries from samples of human and bacterial genomic DNA. E. coli libraries prepared on-device from 5 ng of total DNA yielded excellent sequence coverage over the entire bacterial genome, with >99% alignment to the reference genome, even genome coverage, and good quality scores. Furthermore, we produced a de novo assembly on a previously unsequenced multi-drug resistant Klebsiella pneumoniae strain BAA-2146 (KpnNDM). The new method described here is fast, robust, scalable, and automated. Our device for library preparation will assist in the integration of NGS technology into a wide variety of laboratories, including small research laboratories and clinical laboratories.
