&#x4F9D;&#x8D56;&#x4E8E;&#x8D27;&#x7269;&#x7684;&#x5438;&#x6536;&#x548C;&#x751F;&#x7269;&#x5229;&#x7528;&#x5EA6;&#x8FBE;&#x542B;&#x86CB;&#x767D;&#x8D28;&#x548C;&#x80BD;&#x5728;&#x6D3B;&#x7EC6;&#x80DE;&#x4E2D;&#x7684;&#x6A21;&#x5F0F;&#x3002;

Cell-penetrating peptides (CPPs) are capable of introducing a wide range of cargoes into living cells. Descriptions of the internalization process vary from energy-independent cell penetration of membranes to endocytic uptake. To elucidate whether the mechanism of entry of CPP constructs might be influenced by the properties of the cargo, we used time lapse confocal microscopy analysis of living mammalian cells to directly compare the uptake of the well-studied CPP TAT fused to a protein (>50 amino acids) or peptide (<50 amino acids) cargo. We also analyzed various constructs for their subcellular distribution and mobility after the internalization event. TAT fusion proteins were taken up largely into cytoplasmic vesicles whereas peptides fused to TAT entered the cell in a rapid manner that was dependent on membrane potential. Despite their accumulation in the nucleolus, photobleaching of TAT fusion peptides revealed their mobility. The bioavailability of internalized TAT peptides was tested and confirmed by the strong inhibitory effect on cell cycle progression of two TAT fusion peptides derived from the tumor suppressor p21(WAF/Cip) and DNA Ligase I measured in living cells.

Peptides de p&#xE9;n&#xE9;tration cellulaire (CPP) sont capables d&#x26;#39;introduire une vaste gamme de cargaisons dans des cellules vivantes. Les descriptions du processus d&#x26;#39;internalisation varient de p&#xE9;n&#xE9;tration dans la cellule d&#x26;#39;&#xE9;nergie ind&#xE9;pendante des membranes &#xE0; l&#x26;#39;absorption endocytique. Pour &#xE9;lucider si le m&#xE9;canisme d&#x26;#39;entr&#xE9;e des constructions du RPC pourrait &#xEA;tre influenc&#xE9;e par les propri&#xE9;t&#xE9;s de la cargaison, nous avons utilis&#xE9; la microscopie laps de temps d&#x26;#39;analyse confocale de cellules vivantes de mammif&#xE8;res de comparer directement l&#x26;#39;absorption de la TAT bien &#xE9;tudi&#xE9; RPC fusionn&#xE9; &#xE0; une prot&#xE9;ine (&#x26;gt; 50 acides amin&#xE9;s) ou peptide (moins de 50 acides amin&#xE9;s) cargaison. Nous avons &#xE9;galement analys&#xE9; diff&#xE9;rentes constructions pour leur distribution subcellulaire et de la mobilit&#xE9; apr&#xE8;s l&#x26;#39;&#xE9;v&#xE9;nement internalisation. Prot&#xE9;ines de fusion TAT ont &#xE9;t&#xE9; prises en grande partie dans des v&#xE9;sicules cytoplasmiques alors que les peptides fusionn&#xE9;s &#xE0; TAT entr&#xE9; dans la cellule d&#x26;#39;une mani&#xE8;re rapide qui est d&#xE9;pendante du potentiel de membrane. En d&#xE9;pit de leur accumulation dans le nucl&#xE9;ole, le photoblanchiment des peptides de fusion TAT a r&#xE9;v&#xE9;l&#xE9; leur mobilit&#xE9;. La biodisponibilit&#xE9; de la int&#xE9;rioris&#xE9;es peptides TAT &#x200B;&#x200B;a &#xE9;t&#xE9; test&#xE9;e et confirm&#xE9;e par le fort effet inhibiteur sur la progression du cycle cellulaire de deux peptides de fusion d&#xE9;riv&#xE9;s du TAT du suppresseur de tumeur p21 (WAF / CIP) et l&#x26;#39;ADN ligase I mesur&#xE9;e dans les cellules vivantes.

Cargo-d&#xE9;pendante du mode d&#x26;#39;absorption et la biodisponibilit&#xE9; de TAT-contenant des prot&#xE9;ines et peptides dans les cellules vivantes

Zelle durchdringenden Peptide (CPPs) k&ouml;nnen sich darin eine Vielzahl von Ladungen in lebenden Zellen. Beschreibungen des Prozesses Internalisierung variieren von Energie-unabh&auml;ngige Zelle Eindringen von Membranen zu endocytic Aufnahme. Um aufzukl&auml;ren, ob der Mechanismus der Eintrag CPP Konstrukte durch die Eigenschaften der Ladung beeinflusst sein k&ouml;nnten, wir Zeit erl&ouml;schen confocal Mikroskopie Analyse der lebenden S&auml;ugetier-Zellen direkt vergleichen die Aufnahme der gut erforscht CPP TAT zu einem Protein verschmolzen genutzt (&gt; 50 Aminos&auml;uren) oder Peptid (&lt; 50 Aminos&auml;uren) Ladung. Wir analysierten auch verschiedenen Konstrukte f&uuml;r ihre subzellul&auml;re Verteilung und Mobilit&auml;t nach der Internalisierung-Veranstaltung. TAT Schmelzverfahrensproteine wurden weitgehend in zytoplasmatischen Vesikeln &uuml;bernommen, wo Peptide verschmolzen, TAT die Zelle in einer schnellen Weise eingetragen, die Membran abh&auml;ngig war potenzielle. Trotz ihrer Akkumulation in der Nukleolus offenbart Photobleaching TAT Fusion Peptide ihrer Mobilit&auml;t. Die Bioverf&uuml;gbarkeit von verinnerlichten TAT Peptide wurde getestet und best&auml;tigt durch die stark hemmende Wirkung auf die Zellzyklusprogression von zwei TAT Fusion Peptide aus dem Tumor-Suppressor-p21(WAF/Cip) und DNA-Ligase, die ich in lebenden Zellen gemessen.

Abbiamo anche analizzato vari costrutti per la loro distribuzione subcellulare e mobilit&agrave; dopo l&#39;evento di interiorizzazione. Proteine di fusione TAT sono presi in gran parte in vescicole citoplasmatiche che peptidi fuse Tat entrato nella cella in maniera rapida in membrana potenziale. Nonostante la loro accumulazione nel nucleolo, photobleaching di peptidi fusione TAT ha rivelato loro mobilit&agrave;. La biodisponibilit&agrave; di peptidi TAT interiorizzate &egrave; stata testata e confermata dal forte effetto inibitorio sulla progressione del ciclo cellulare dei due peptidi di fusione TAT derivato dal p21(WAF/Cip) soppressore tumorale e DNA ligasi ho misurato in cellule viventi.

Continuous expression of Cre recombinase has the potential to yield toxic side effects in various cell types, thereby limiting applications of the Cre/loxP system for conditional mutagenesis. In this study, we investigate the potential of Cre protein transduction to overcome this limitation. COS-7, CV1-5B, and mouse embryonic stem (ES) cells treated with cell-permeant Cre (HTNCre) maintain a normal growth behavior employing Cre concentrations sufficient to induce recombination in more than 90% of the cells, whereas continuous application of high doses resulted in markedly reduced proliferation. HTNCre-treated ES cells maintain a normal karyotype and are still able to contribute to the germline. Moreover, we present an enhanced HTNCre purification protocol that allows the preparation of a concentrated glycerol stock solution, thereby enabling a considerable simplification of the Cre protein transduction procedure. The protocol described here allows rapid and highly efficient conditional mutagenesis of cultured cells.

Tyrosine site-specific recombinases (SSRs) including Cre and FLP are essential tools for DNA and genome engineering. Cre has long been recognized as the best SSR for genome engineering, particularly in mice. Obtaining another SSR that is as good as Cre will be a valuable addition to the genomic toolbox. To this end, we have developed and validated reagents for the Dre-rox system. These include an Escherichia coli-inducible expression vector based on the temperature-sensitive pSC101 plasmid, a mammalian expression vector based on the CAGGs promoter, a rox-lacZ reporter embryonic stem (ES) cell line based on targeting at the Rosa26 locus, the accompanying Rosa26-rox reporter mouse line, and a CAGGs-Dre deleter mouse line. We also show that a Dre-progesterone receptor shows good ligand-responsive induction properties. Furthermore, we show that there is no crossover recombination between Cre-rox or Dre-loxP. Hence, we add another set of efficient tools to the genomic toolbox, which will enable the development of more sophisticated mouse models for the analysis of gene function and disease.

Tirosina espec&#xED;ficos del sitio recombinaciones (SSR) como Cre y Flp son herramientas esenciales para el ADN y la ingenier&#xED;a del genoma. Cre ha sido reconocida como la mejor SSR para la ingenier&#xED;a del genoma, en particular en los ratones. La obtenci&#xF3;n de otra SSR que es tan bueno como Cre ser&#xE1; una valiosa adici&#xF3;n a la caja de herramientas gen&#xF3;micas. Con este fin, hemos desarrollado y validado los reactivos para el sistema de Dre-rox. Combined application of DNA recombinases Cre and FLP enables tightly controlled independent and/or sequential gene regulations. However, in practice, such dual recombinase strategies are hampered by the comparably low efficiency of the FLP recombinase. Here, we present the engineering of a recombinant cell-permeant FLP protein (TAT-FLP) that induces recombination in >75% of fibroblasts and mouse as well as human embryonic stem (ES) cells. We show that TAT-FLP ideally complements the strength of cell-permeant Cre recombinase for genetic engineering as exemplified by FLP-ON-Cre-OFF, an inducible transgene expression cassette that enables tightly controlled expression in a reversible manner. We exemplify this concept by conditional overexpression of LacZ and the caudal-related homeobox transcription factor CDX2. We expect our FLP transduction system to become widely useful for numerous genetic interventions addressing complex biological questions and the generation of transgene-free therapeutically applicable ES cell-derived cells.

Engineering cell-permeant FLP recombinase for tightly controlled inducible and reversible overexpression in embryonic stem cells.

Hier pr&auml;sentieren wir die Technik eines rekombinanten Zelle-permeant FLP-Proteins (TAT-FLP), die Rekombination in bedingt &gt; 75 % der Fibroblasten sowie Maus und humane embryonale Stammzellen (ES). Protein transduction is based on the ability of certain peptides, designated as cell penetrating peptides (CPPs), to intracellularly deliver cargo molecules, such as peptides and proteins. In combination with site specific recombination, CPP-mediated delivery of recombinases enables a precise and highly efficient control of gene expression in cultured cells and mice. Herein, we provide detailed protocols for engineering and purification of a cell-permeant FLP recombinase protein. Two examples describe the use of cell permeant FLP for excising prespecified fragments from transgenes expressed in fibroblasts and mouse embryonic stem cells. A third example describes the combined use of cell-permeant Cre and FLP recombinases to reversibly induce transgenes in embryonic stem cells. We anticipate that the protocols described herein will be widely used for various genetic interventions addressing complex biological questions.

Dos ejemplos describen el uso de c&#xE9;lulas permeante FLP para extirpar fragmentos preespecificados de transgenes expresados &#x200B;&#x200B;en fibroblastos y c&#xE9;lulas madre embrionarias de rat&#xF3;n. Un tercer ejemplo se describe el uso combinado de la c&#xE9;lula-permeable Cre y recombinaciones de FLP para inducir de forma reversible transgenes en c&#xE9;lulas madre embrionarias. Anticipamos que los protocolos descritos en este documento se utiliza ampliamente para diversas intervenciones gen&#xE9;ticas que abordan cuestiones complejas biol&#xF3;gicas.
