Molecular genetics approaches in zebrafish research are hampered by the lack of a ubiquitous transgene driver element that is active at all developmental stages. Here, we report the isolation and characterization of the zebrafish ubiquitin (ubi) promoter, which drives constitutive transgene expression during all developmental stages and analyzed adult organs. Notably, ubi expresses in all blood cell lineages, and we demonstrate the application of ubi-driven fluorophore transgenics in hematopoietic transplantation experiments to assess true multilineage potential of engrafted cells. We further generated transgenic zebrafish that express ubiquitous 4-hydroxytamoxifen-controlled Cre recombinase activity from a ubi:cre(ERt2) transgene, as well as ubi:loxP-EGFP-loxP-mCherry (ubi:Switch) transgenics and show their use as a constitutive fluorescent lineage tracing reagent. The ubi promoter and the transgenic lines presented here thus provide a broad resource and important advancement for transgenic applications in zebrafish.

Ubiquitous transgene expression and Cre-based recombination driven by the ubiquitin promoter in zebrafish.

Des approches de g&#xE9;n&#xE9;tique mol&#xE9;culaire dans la recherche du poisson z&#xE8;bre sont entrav&#xE9;s par le manque d&#x26;#39;un &#xE9;l&#xE9;ment conducteur transg&#xE8;ne omnipr&#xE9;sente qui est actif &#xE0; tous les stades de d&#xE9;veloppement. Nous rapportons ici l&#x26;#39;isolement et la caract&#xE9;risation du poisson-z&#xE8;bre ubiquitine (ubi) promoteur, qui entra&#xEE;ne l&#x26;#39;expression du transg&#xE8;ne constitutive durant toutes les &#xE9;tapes de d&#xE9;veloppement et analys&#xE9;s organes adultes. Notamment, ubi exprime dans toutes les lign&#xE9;es de cellules sanguines, et nous d&#xE9;montrer l&#x26;#39;application de ubi ax&#xE9;e fluorophore transg&#xE9;niques dans des exp&#xE9;riences de transplantation h&#xE9;matopo&#xEF;&#xE9;tique &#xE0; &#xE9;valuer le potentiel des cellules multilign&#xE9;e vrai greff&#xE9;es. Nous avons en outre g&#xE9;n&#xE9;r&#xE9; le poisson-z&#xE8;bre transg&#xE9;nique qui expriment omnipr&#xE9;sente 4-hydroxytamoxif&#xE8;ne contr&#xF4;l&#xE9;e par l&#x26;#39;activit&#xE9; de la recombinase Cre d&#x26;#39;un ubi: CRE (ERT2) transg&#xE8;ne, ainsi que ubi: loxP-EGFP-loxP-mCherry (ubi: Switch) transg&#xE9;niques et de montrer leur utilisation en tant que constitutive lign&#xE9;e fluorescente tra&#xE7;age r&#xE9;actif. Le promoteur ubi et les lign&#xE9;es transg&#xE9;niques pr&#xE9;sent&#xE9;s ici ainsi fournir une ressource vaste et progr&#xE8;s important pour les applications transg&#xE9;niques chez le poisson z&#xE8;bre.

Molekulargenetik Ans&auml;tze im Zebrafisch-Forschung werden durch das Fehlen eines allgegenw&auml;rtigen Transgene Treiber Elements behindert, die zu allen Entwicklungsstadien aktiv ist. Hier berichten wir die Isolierung und Charakterisierung des Projekttr&auml;gers Zebrafisch Ubiquitin (Ubi), die Laufwerke konstitutive Transgenexpression in allen Entwicklungsstadien und analysiert Erwachsenen Organe. Insbesondere, Ubi dr&uuml;ckt in alle Blutk&ouml;rperchen &Uuml;bertragungslinien und wir demonstrieren die Anwendung der Ubi-gesteuerte Fluorophor Transgenics h&auml;matopoetische Transplantation Experimente auf wahre multiline&auml;rer des engrafted Zellen beurteilen. Wir weiter generiert transgene Zebrafisch, der allgegenw&auml;rtigen 4-Hydroxytamoxifen-Steuerung Cre Recombinase Aktivit&auml;t aus einer ubi:cre(ERt2) Transgene sowie Ubi:loxP auszudr&uuml;cken-EGFP-LoxP-mCherry (Ubi:Switch) Transgenics und ihrer Verwendung als konstitutive fluoreszierende &Uuml;bertragungslinie Ablaufverfolgung Reagenzien zu zeigen. Die Ubi-Projekttr&auml;ger und die transgenen Linien, die hier vorgestellten stellen daher eine breite Ressource und ein wichtiger Fortschritt f&uuml;r transgene Anwendungen im Zebrafisch.

Approcci di genetica molecolare nella ricerca di zebrafish sono ostacolati dalla mancanza di un elemento di driver transgene onnipresente che &egrave; attivo in tutte le fasi dello sviluppo. Riportiamo l&#39;isolamento e la caratterizzazione del promotore zebrafish ubiquitina (ubi), che spinge l&#39;espressione del transgene costitutiva durante tutte le fasi di sviluppo e analizzato organi adulti. In particolare, ubi esprime in tutte le cellule del sangue lignaggi, e dimostreremo l&#39;applicazione del fluoroforo ubi-driven transgenica in esperimenti di trapianto ematopoietico per valutare il vero potenziale multilineare delle cellule engrafted. Abbiamo generato ulteriore zebrafish transgenici che esprimono onnipresente 4-hydroxytamoxifen-controllato Cre ricombinasi attivit&agrave; da un transgene ubi:cre(ERt2), come pure ubi:loxP-EGFP-loxP-transgenetica mCherry (ubi:Switch) e mostrano il loro uso come reagente di tracciatura costitutiva lignaggio fluorescente. Il promotore di ubi e le linee transgeniche presentate qui cos&igrave; forniscono una risorsa ampia e importante avanzamento per applicazioni transgeniche in zebrafish.

Enfoques moleculares en la investigaci&#xF3;n gen&#xE9;tica del pez cebra se ven obstaculizados por la falta de un elemento conductor transg&#xE9;n en todas partes que est&#xE1; activo en todas las etapas de desarrollo. Aqu&#xED; reportamos el aislamiento y caracterizaci&#xF3;n del pez cebra ubiquitina (UBI), el promotor, que lleva a la expresi&#xF3;n del transgen constitutiva durante todas las etapas de desarrollo y se analizaron los &#xF3;rganos adultos. En particular, ubi expresa en todos los linajes de c&#xE9;lulas sangu&#xED;neas, y demostrar la aplicaci&#xF3;n de la ubi-impulsada por fluor&#xF3;foro transg&#xE9;nicos en los experimentos de trasplante hematopoy&#xE9;tico para evaluar el potencial real multilinaje de c&#xE9;lulas injertadas. Hemos generado m&#xE1;s de pez cebra transg&#xE9;nico que expresa en todas partes 4-hydroxytamoxifen controlado por la recombinasa Cre actividad de una ubi: CRE transg&#xE9;n (ERt2), as&#xED; como la ubi: loxP-EGFP-loxP-mCherry (ubi: Switch) transg&#xE9;nicos y demostrar su uso como linaje fluorescente constitutiva rastreo reactivo. El promotor ubi y las l&#xED;neas transg&#xE9;nicas que aqu&#xED; se presentan por lo tanto constituyen un recurso amplio y avance importante para las aplicaciones de transg&#xE9;nicos en el pez cebra.
