Metastasis accounts largely for the high mortality rate of colorectal cancer (CRC) patients. In this study, we performed comparative proteome analysis of primary CRC cell lines HCT-116 and its metastatic derivative E1 using 2-D DIGE. We identified 74 differentially expressed proteins, many of which function in transcription, translation, angiogenesis signal transduction, or cytoskeletal remodeling pathways, which are indispensable cellular processes involved in the metastatic cascade. Among these proteins, stathmin-1 (STMN1) was found to be highly up-regulated in E1 as compared to HCT-116 and was thus selected for further functional studies. Our results showed that perturbations in STMN1 levels resulted in significant changes in cell migration, invasion, adhesion, and colony formation. We further showed that the differential expression of STMN1 correlated with the cells' metastatic potential in other paradigms of CRC models. Using immunohistochemistry, we also showed that STMN1 was highly expressed in colorectal primary tumors and metastatic tissues as compared to the adjacent normal colorectal tissues. Furthermore, we also showed via tissue microarray analyses of 324 CRC tissues and Kaplan-Meier survival plot that CRC patients with higher expression of STMN1 have poorer prognosis.

Proteomic analysis of colorectal cancer metastasis: stathmin-1 revealed as a player in cancer cell migration and prognostic marker.

M&eacute;tastase explique en grande partie le taux &eacute;lev&eacute; de mortalit&eacute; des patients du cancer colorectal (CCR). Dans cette &eacute;tude, nous avons effectu&eacute; l&#39;analyse prot&eacute;omique comparative des lign&eacute;es de cellules primaires CRC HCT-116 et son d&eacute;riv&eacute; m&eacute;tastatique E1 en utilisant le 2D DIGE. Nous avons identifi&eacute; les prot&eacute;ines diff&eacute;rentiellement exprim&eacute;es 74, beaucoup de qui fonctionnent dans la transcription, traduction, angiogen&egrave;se transduction du signal, ou voies de remodelage du cytosquelette, qui sont indispensables processus cellulaires impliqu&eacute;s dans la cascade m&eacute;tastatique. De plus, nous avons montr&eacute; que l&#39;expression diff&eacute;rentielle de STMN1 en corr&eacute;lation avec le potentiel m&eacute;tastatique des cellules dans d&#39;autres paradigmes de mod&egrave;les CRC. Par immunohistochimie, les auteurs montrent &eacute;galement que STMN1 est fortement exprim&eacute;e dans colorectal tumeurs primaires et m&eacute;tastatiques tissus par rapport aux tissus adjacents colorectal normales. En outre, nous avons &eacute;galement montr&eacute; par des analyses de microarray de tissu de 324 tissus CRC et parcelle de survie de Kaplan-Meier que les patients CRC avec une expression plus &eacute;lev&eacute;e de STMN1 aient pronostic moins favorable.

Metastase entfallen weitgehend die hohe Sterblichkeit von Patienten mit Dickdarmkrebs (CRC). In dieser Studie f&uuml;hrten wir vergleichende Proteomanalyse des prim&auml;ren CRC-Zelllinien HCT-116 und ihre metastasierendem Ableitung E1 mit 2-D vorbeikommen. Wir identifizierten 74 differentiell exprimierten Proteinen, viele welche Funktion in Transkription, &Uuml;bersetzung, Angiogenese Signaltransduktion, oder Zytoskelett Umbau Wege, die sind unverzichtbare zellul&auml;re Prozesse der metastasierendem Kaskade beteiligt. Unter dieser Proteine Stathmin-1 (STMN1) wurde festgestellt, dass hoch oben-geregelt in E1 im Vergleich zu HCT-116 und war somit f&uuml;r weitere funktionelle Studien ausgew&auml;hlt. Unsere Ergebnisse zeigten, dass St&ouml;rungen in STMN1 Ebenen wesentliche &Auml;nderungen im Zellbildung Migration, Invasion, Adh&auml;sion und der Kolonie gef&uuml;hrt. Wir zeigten weiter, dass der differentielle Ausdruck des STMN1 mit der Zellen metastasierendem Potenzial in andere Paradigmen der CRC-Modelle korreliert. Verwenden Immunohistochemistry, zeigten wir auch, dass STMN1 kolorektalen Prim&auml;rtumoren und metastasiertem Geweben verglichen mit der angrenzenden normal kolorektalen Gewebe hoch zum Ausdruck gebracht wurde. Dar&uuml;ber hinaus zeigten wir auch &uuml;ber Gewebe Microarray Analysen 324 CRC-Gewebe und Kaplan-Meier-&uuml;berleben-Plot, CRC-Patienten mit h&ouml;heren Ausdruck des STMN1 schlechtere Prognose haben.

Conti di metastasi in gran parte per l&#39;elevato tasso di mortalit&agrave; dei pazienti affetti da cancro del colon-retto (CRC). In questo studio, abbiamo effettuato analisi comparativa del proteoma di linee di cellule primarie CRC HCT-116 e sua derivata metastatico E1 usando DIGE 2-D. Abbiamo individuato le proteine differenzialmente espressione 74, molti dei quali funzione nella trascrizione, traduzione, angiogenesi trasduzione del segnale o del citoscheletro ritoccando i percorsi, che sono indispensabili processi cellulari coinvolti nella cascata metastatica. Tra queste proteine, stathmin-1 (STMN1) &egrave; stato trovato per essere altamente regolamentato in E1 rispetto alla HCT-116 e cos&igrave; &egrave; stato selezionato per studi funzionali. I nostri risultati hanno mostrato che perturbazioni nei livelli di STMN1 ha portato a significativi cambiamenti nella formazione delle cellule di migrazione, invasione, adesione e Colonia. Abbiamo ulteriormente dimostrato che l&#39;espressione differenziale di STMN1 correlata con potenziale metastatico delle cellule in altri paradigmi di modelli CRC. Mediante immunoistochimica, abbiamo anche dimostrato che STMN1 era altamente espresso nei tumori colorettali primari e metastatici tessuti rispetto ai tessuti del colon normali adiacenti. Inoltre, abbiamo anche mostrato tramite analisi microarray tessuto di 324 CRC tessuti e trama di sopravvivenza di Kaplan-Meier che CRC pazienti con pi&ugrave; alta espressione di STMN1 hanno prognosi pi&ugrave; povera.

Met&aacute;stasis en gran parte responsable de la elevada tasa de mortalidad de los pacientes de c&aacute;ncer colorrectal (CCR). En este estudio, realizamos el an&aacute;lisis del proteoma comparativo de l&iacute;neas de c&eacute;lulas primarias de CRC HCT-116 y su derivado metast&aacute;tico E1 utilizando DIGE 2D. Se identificaron 74 prote&iacute;nas expresadas diferencialmente, muchos de los que funcionan en la transcripci&oacute;n, traducci&oacute;n, angiog&eacute;nesis transducci&oacute;n de la se&ntilde;al o citoesqueleto remodelaci&oacute;n de v&iacute;as, que son indispensables procesos celulares implicados en la cascada metast&aacute;sica. Entre estas prote&iacute;nas, estatmina-1 (STMN1) fue encontrado para ser altamente regulados en E1 en comparaci&oacute;n con HCT-116 para arriba y as&iacute; fue seleccionado para futuros estudios funcionales. Nuestros resultados mostraron que las perturbaciones en los niveles de STMN1 dio lugar a cambios significativos en la formaci&oacute;n de Colonia, migraci&oacute;n, invasi&oacute;n y adherencia de la c&eacute;lula. A&uacute;n m&aacute;s, demostramos que la expresi&oacute;n diferencial de STMN1 correlacionados con potencial metast&aacute;sico de las c&eacute;lulas en otros paradigmas de modelos CRC. Usando immunohistochemistry, demostramos tambi&eacute;n que STMN1 fue altamente expresada en colorrectales tumores primarios y metast&aacute;ticos tejidos en comparaci&oacute;n con los tejidos normales adyacentes del colorrectales. Adem&aacute;s, tambi&eacute;n demostramos mediante an&aacute;lisis de microarrays de tejido de trama de supervivencia de Kaplan-Meier y 324 tejidos CRC que pacientes CRC con mayor expresi&oacute;n de STMN1 tienen peor pron&oacute;stico.

Patients with the genetic skin blistering disease recessive dystrophic epidermolysis bullosa (RDEB) develop aggressive cutaneous squamous cell carcinoma (cSCC). Metastasis leading to mortality is greater in RDEB than in other patient groups with cSCC. Here we investigate the dermal component in RDEB using mRNA expression profiling to compare cultured fibroblasts isolated from individuals without cSCC and directly from tumor matrix in RDEB and non-RDEB samples. Although gene expression of RDEB normal skin fibroblasts resembled that of cancer-associated fibroblasts, RDEB cancer-associated fibroblasts exhibited a distinct and divergent gene expression profile, with a large proportion of the differentially expressed genes involved in matrix and cell adhesion. RDEB cancer-associated fibroblasts conferred increased adhesion and invasion to tumor and nontumor keratinocytes. Reduction of COL7A1, the defective gene in RDEB, in normal dermal fibroblasts led to increased type XII collagen, thrombospondin-1, and Wnt-5A, while reexpression of wild type COL7A1 in RDEB fibroblasts decreased type XII collagen, thrombospondin-1, and Wnt-5A expression, reduced tumor cell invasion in organotypic culture, and restricted tumor growth in vivo. Overall, our findings show that matrix composition in patients with RDEB is a permissive environment for tumor development, and type VII collagen directly regulates the composition of matrix proteins secreted by dermal and cancer-associated fibroblasts.
