&#x86CB;&#x767D;&#x916A;&#x6C28;&#x9178;&#x6FC0;&#x9176;&#x6291;&#x5236;&#x5242; SU5614 &#x6291;&#x5236; FLT3 &#x548C;&#x8BF1;&#x5BFC;&#x751F;&#x957F;&#x902E;&#x6355;&#x548C;&#x8868;&#x8FBE;&#x4E00;&#x6FC0;&#x6D3B;&#x7684; FLT3 AML &#x6D3E;&#x751F;&#x7684;&#x7EC6;&#x80DE;&#x51CB;&#x4EA1;&#x3002;

Activating mutations of the protein tyrosine kinase (PTK) FLT3 can be found in approximately 30% of patients with acute myeloid leukemia (AML), thereby representing the most frequent single genetic alteration in AML. These mutations occur in the juxtamembrane (FLT3 length mutations; FLT3-LMs) and the second tyrosine kinase domain of FLT3-TKD and confer interleukin 3 (IL-3)-independent growth to Ba/F3 cells. In the mouse bone marrow transplantation model, FLT3-LMs induce a myeloproliferative syndrome stressing their transforming activity in vivo. In this study, we analyzed the pro-proliferative and antiapoptotic potential of FLT3 in FLT3-LM/TKD-mutation-transformed Ba/F3 cells and AML-derived cell lines. The PTK inhibitor SU5614 has inhibitory activity for FLT3 and selectively induces growth arrest, apoptosis, and cell cycle arrest in Ba/F3 and AML cell lines expressing a constitutively activated FLT3. In addition, the compound reverts the antiapoptotic and pro-proliferative activity of FLT3 ligand (FL) in FL-dependent cells. No cytotoxic activity of SU5614 was found in leukemic cell lines that express a nonactivated FLT3 or no FLT3 protein. At the biochemical level, SU5614 down-regulated the activity of the hyperphosphorylated FLT3 receptor and its downstream targets, signal transducer and activator of (STAT) 3, STAT5, and mitogen-activated protein kinase (MAPK), and the STAT5 target genes BCL-X(L) and p21. Our results show that SU5614 is a PTK inhibitor of FLT3 and has antiproliferative and proapoptotic activity in AML-derived cell lines that endogenously express an activated FLT3 receptor. The selective and potent cytotoxicity of FLT3 PTK inhibitors support a clinical strategy of targeting FLT3 as a new molecular treatment option for patients with FLT3-LM/TKD-mutation(+) AML.

Les mutations de la prot&#xE9;ine tyrosine kinase (PTK) FLT3 peut &#xEA;tre trouv&#xE9;e dans environ 30% des patients atteints de leuc&#xE9;mie my&#xE9;lo&#xEF;de aigu&#xEB; (LMA), repr&#xE9;sentant ainsi la plus fr&#xE9;quente alt&#xE9;ration g&#xE9;n&#xE9;tique unique dans la LMA. Ces mutations se produisent dans la juxtamembrane (FLT3 mutations longueur; FLT3-LMS) et le domaine kinase tyrosine seconde de FLT3-TKD et conf&#xE8;rent interleukine 3 (IL-3) une croissance ind&#xE9;pendante de Ba/F3 cellules. Dans la transplantation de moelle osseuse de souris mod&#xE8;le, FLT3-LM induire un syndrome my&#xE9;loprolif&#xE9;ratif en insistant sur leur activit&#xE9; de transformation in vivo. L&#x26;#39;inhibiteur de PTK SU5614 poss&#xE8;de une activit&#xE9; inhibitrice de FLT3 et induit s&#xE9;lectivement arr&#xEA;t de la croissance, l&#x26;#39;apoptose, et arr&#xEA;t du cycle cellulaire dans les lign&#xE9;es cellulaires Ba/F3 et AML exprimant une FLT3 constitutivement activ&#xE9;e. En outre, le compos&#xE9; revient l&#x26;#39;activit&#xE9; anti-apoptotique et pro-prolif&#xE9;rative de FLT3 ligand (FL) en Floride cellules d&#xE9;pendantes. Au niveau biochimique, SU5614 r&#xE9;gul&#xE9;e &#xE0; la baisse de l&#x26;#39;activit&#xE9; du r&#xE9;cepteur FLT3 hyperphosphoryl&#xE9;e et ses cibles en aval, de transduction de signal et activateur de (STAT) 3, STAT5, et la prot&#xE9;ine kinase activ&#xE9;e par des mitog&#xE8;nes (MAPK), et les STAT5 g&#xE8;nes cibles de BCL- X (L) et p21. Nos r&#xE9;sultats montrent que SU5614 est un inhibiteur de PTK de FLT3 et poss&#xE8;de une activit&#xE9; anti-prolif&#xE9;rative et pro-apoptotique dans des lign&#xE9;es cellulaires d&#xE9;riv&#xE9;es de la LBC qui expriment une mani&#xE8;re endog&#xE8;ne activ&#xE9; FLT3 r&#xE9;cepteur. Mutazioni della chinasi della tirosina della proteina di attivazione FLT3 (PTK) si trovano in circa il 30% dei pazienti con leucemia mieloide acuta (AML), che rappresenta quindi la pi&ugrave; frequente singola alterazione genetica in AML. Queste mutazioni si verificano in juxtamembrane (mutazioni di FLT3 lunghezza; FLT3-LMs) e il secondo dominio tirosin-chinasico di FLT3-TKD e conferire interleuchina 3 (IL-3)-crescita indipendente alle cellule Ba/F3. Nel modello di trapianto del midollo osseo del mouse, FLT3-LMs indurre una sindrome mieloproliferativa sottolineando la loro attivit&agrave; trasformante in vivo. In questo studio, abbiamo analizzato i pro-proliferativa e antiapoptotici potenziale di FLT3 in cellule trasformate FLT3-LM/TKD-mutazione Ba/F3 e linee cellulari derivate da AML. Nessuna attivit&agrave; citotossica di SU5614 &egrave; stato trovato in linee di cellule leucemiche che esprimono un nonactivated FLT3 o senza proteine FLT3. A livello biochimico, SU5614 gi&ugrave;-regolato l&#39;attivit&agrave; del recettore FLT3 iperfosforilata e suoi bersagli a valle, trasduttore di segnale e attivatore di (STAT) 3, STAT5, chinasi di proteina mitogene-attivata (MAPK) e i geni bersaglio STAT5 BCL-X(L) e p21. I nostri risultati mostrano che il SU5614 &egrave; un inibitore PTK di FLT3 e ha attivit&agrave; antiproliferativa e Miselli in linee cellulari derivate da AML che endogena esprimono un recettore FLT3 attivato. La citotossicit&agrave; selettiva e potente di inibitori di FLT3 PTK sostenere una strategia clinica di targeting FLT3 come una nuova opzione di trattamento molecolare per pazienti con leucemia mieloide acuta FLT3-LM/TKD-mutation(+).

Human mutations in or variants of TBX20 are associated with congenital heart disease, cardiomyopathy, and arrhythmias. To investigate whether cardiac disease in patients with these conditions results from an embryonic or ongoing requirement for Tbx20 in myocardium, we ablated Tbx20 specifically in adult cardiomyocytes in mice. This ablation resulted in the onset of severe cardiomyopathy accompanied by arrhythmias, with death ensuing within 1 to 2 weeks of Tbx20 ablation. Accounting for this dramatic phenotype, we identified molecular signatures that posit Tbx20 as a central integrator of a genetic program that maintains cardiomyocyte function in the adult heart. Expression of a number of genes encoding critical transcription factors, ion channels, and cytoskeletal/myofibrillar proteins was downregulated consequent to loss of Tbx20. Genome-wide ChIP analysis of Tbx20-binding regions in the adult heart revealed that many of these genes were direct downstream targets of Tbx20 and uncovered a previously undescribed DNA-binding site for Tbx20. Bioinformatics and in vivo functional analyses revealed a cohort of transcription factors that, working with Tbx20, integrated multiple environmental signals to maintain ion channel gene expression in the adult heart. Our data provide insight into the mechanisms by which mutations in TBX20 cause adult heart disease in humans.

Coordinated release of calcium (Ca(2+) ) from the sarcoplasmic reticulum (SR) through cardiac ryanodine receptor (RYR2) channels is essential for cardiomyocyte function. In catecholaminergic polymorphic ventricular tachycardia (CPVT), an inherited disease characterized by stress-induced ventricular arrhythmias in young patients with structurally normal hearts, autosomal dominant mutations in RYR2 or recessive mutations in calsequestrin lead to aberrant diastolic Ca(2+) release from the SR causing arrhythmogenic delayed after depolarizations (DADs). Here, we report the generation of induced pluripotent stem cells (iPSCs) from a CPVT patient carrying a novel RYR2 S406L mutation. In patient iPSC-derived cardiomyocytes, catecholaminergic stress led to elevated diastolic Ca(2+) concentrations, a reduced SR Ca(2+) content and an increased susceptibility to DADs and arrhythmia as compared to control myocytes. This was due to increased frequency and duration of elementary Ca(2+) release events (Ca(2+) sparks). Dantrolene, a drug effective on malignant hyperthermia, restored normal Ca(2+) spark properties and rescued the arrhythmogenic phenotype. This suggests defective inter-domain interactions within the RYR2 channel as the pathomechanism of the S406L mutation. Our work provides a new in vitro model to study the pathogenesis of human cardiac arrhythmias and develop novel therapies for CPVT.

En la taquicardia ventricular polim&#xF3;rfica catecolamin&#xE9;rgica (TVPC), una enfermedad hereditaria caracterizada por arritmias ventriculares inducidas por el estr&#xE9;s en pacientes j&#xF3;venes con corazones estructuralmente normales, mutaciones autos&#xF3;micas dominantes en RYR2 o mutaciones recesivas en la derivaci&#xF3;n de calsecuestrina aberrante diast&#xF3;lica Ca (2 +) la liberaci&#xF3;n de la SR arritmog&#xE9;nica causando retrasado despu&#xE9;s de despolarizaciones (DADS). Aqu&#xED;, se presenta la generaci&#xF3;n de c&#xE9;lulas madre pluripotentes inducidas (iPSCs) de un paciente que lleva una novela de TVPC RYR2 mutaci&#xF3;n S406L. En los pacientes derivados de IPSC cardiomiocitos, el estr&#xE9;s catecolamin&#xE9;rgica llevado a elevada diast&#xF3;lica Ca (2 +), una reducci&#xF3;n de las concentraciones de Ca 2 + (SR) de contenido y una mayor susceptibilidad a los pap&#xE1;s y las arritmias en comparaci&#xF3;n con el control de los miocitos. Esto se debi&#xF3; al aumento de la frecuencia y duraci&#xF3;n de la primaria Ca (2 +) eventos de liberaci&#xF3;n (Ca (2 +) chispas). El dantroleno, un f&#xE1;rmaco eficaz en la hipertermia maligna, restaurado normales de Ca (2 +) las propiedades de la chispa y rescat&#xF3; el fenotipo arritmog&#xE9;nica. The generation of patient-specific stem cells by reprogramming somatic cells to induced pluripotent stem cells (iPSC) provides the basis for a promising new type of in vitro disease models. Patient-specific iPSC derived from individuals with hereditary disorders can be differentiated into somatic cells in vitro, thus allowing the pathophysiology of the diseases to be studied on a cellular level. Different types of long-QT syndrome have been successfully modeled using this approach, demonstrating that the iPSC-derived patient-specific cardiomyocytes recapitulated key features of the disease in vitro. This approach will likely serve to model other monogenetic or polygenetic cardiovascular disorders in the future. Moreover, test platforms based on patient-specific iPSC could be used to test the potential of drug candidates to induce QT-interval prolongation or other unwanted side effects, screen for novel cardiovascular drugs, or to tailor medical therapy to the specific needs of a single patient.

Recapitulating long-QT syndrome using induced pluripotent stem cell technology.

The discovery that somatic cells can be reprogrammed to induced pluripotent stem cells (iPSC) by overexpression of a combination of transcription factors bears the potential to spawn a wealth of new applications in both preclinical and clinical cardiovascular research. Disease modeling, which is accomplished by deriving iPSC lines from patients affected by heritable diseases and then studying the pathophysiology of the diseases in somatic cells differentiated from these patient-specific iPSC lines, is the so far most advanced of these applications. Long-QT syndrome and catecholaminergic polymorphic ventricular tachycardia are two heart rhythm disorders that have been already successfully modeled by several groups using this approach, which will likely serve to model other mono- or polygenetic cardiovascular disorders in the future. Test systems based on cells derived from iPSC might prove beneficial to screen for novel cardiovascular drugs or unwanted drug side effects and to individualize medical therapy. The application of iPSC for cell therapy of cardiovascular disorders, albeit promising, will only become feasible if the problem of biological safety of these cells will be mastered.

Induced pluripotent stem cells in cardiovascular research.

The generation of induced pluripotent stem cells (iPSC) from human somatic cells bears the possibility to generate patient-specific stem cell lines which can serve as a theoretically unlimited source of somatic cells carrying the genotype of the patients. Different types of the long-QT syndrome have been studied by analyzing the phenotype of cardiomyocytes generated from patient-specific iPSC lines. Major aspects of the pathophysiology of long-QT syndrome, like prolonged action potentials, arrhythmia, and the effects of pro- and antiarrhythmic drugs could be recapitulated in these cells. In the future, patient-specific iPSC-derived cardiomyocytes might be used to screen for new drugs, to avoid unwanted drug side effects, and to deepen our understanding on the pathophysiology of long-QT syndromes.
