It is dogma that action potentials are initiated at the soma/axon hillock of neurons. However, dendrites often exhibit conductances necessary for spike generation and represent functionally independent processing compartments within neurons. GnRH neurons provide an interesting neuronal phenotype with simple, relatively unbranched, unipolar or bipolar dendrites of extensive lengths (>1000 microm) covered in spines. These neurons control fertility and must integrate a variety of internal homeostatic and external environmental cues. We used imaging, electrophysiological, and modeling studies to understand how they integrate and process information along dendrites. Simultaneous recordings from distal dendrites and somata of individual GnRH neurons indicate distal dendrites are the primary site of spike initiation in these cells. Compartmental modeling indicates that sites of spike initiation depend upon location of excitatory input and dendrite geometry. Together, these studies demonstrate a novel pattern of spike generation in mammalian neurons and indicate that afferent inputs within distal dendritic microdomains directly initiate action potentials.

Adult GnRH neurons exhibit a stereotypic morphology with a small soma, single axon, and single dendrite arising from the soma with little branching. The adult morphology of GnRH neurons in mice reflects an anatomical consolidation of dendrites over postnatal development. We examined this issue in rat GnRH neurons with biocytin filling in live hypothalamic slices from infant males, as adult littermates and in gonad-intact males, castrated males, and in males with one of three levels of testosterone (T) treatment. Somatic area and total dendritic length were significantly greater in infant males than in adults. Moreover, total numbers of dendrite branches were greater in infant males as compared with adults. The number of higher order branches and the lengths of higher order branches were also greater in infant males than in adults. Most interestingly, in adults a single dendrite arose from the somata, consistently at 180Â° from the axon. In contrast, prepubertal animals had an average of 2.2 Â± 0.2 primary dendrites arising from somata (range, one to seven primary dendrites). Angles relative to the axon at which dendrites in prepubertal males emanated from GnRH somata were highly variable. Castration at 25 d of age and castration at 25 d of age with one of three levels of T treatment did not influence morphological parameters when GnRH neurons were examined between 40 d and 48 d of age. Thus, a spatially selective remodeling of primary dendrites and consolidation of distal GnRH dendritic arbors occurs during postnatal development and is largely independent of T.

Adultos las neuronas GnRH presentan una morfolog&#xED;a estereotipados con un soma peque&#xF1;o, solo ax&#xF3;n y las dendritas &#xFA;nica derivada de la soma con poca ramificaci&#xF3;n. La morfolog&#xED;a de los adultos de las neuronas GnRH en ratones refleja una consolidaci&#xF3;n anat&#xF3;mica de las dendritas durante el desarrollo postnatal. Hemos examinado este problema en las neuronas de rata con GnRH biocitina rellenar en vivo cortes hipotal&#xE1;micos de los varones infantiles, como sus compa&#xF1;eros de camada de adultos y en las g&#xF3;nadas intactas machos, machos castrados, y en varones con uno de los tres niveles de testosterona (T) de tratamiento. &#xC1;rea som&#xE1;tica y la longitud dendr&#xED;tica total fueron significativamente mayores en varones infantiles que en los adultos. Por otra parte, el n&#xFA;mero total de ramificaciones dendr&#xED;ticas fueron mayores en los varones infantiles, en comparaci&#xF3;n con los adultos. El n&#xFA;mero de sucursales de orden superior y las longitudes de las ramas de orden superior tambi&#xE9;n fueron mayores en los varones infantiles que en los adultos. Lo m&#xE1;s interesante, en los adultos una dendrita &#xFA;nica surgi&#xF3; de la somata, consistente en 180 &#xB0; desde el ax&#xF3;n. En contraste, los animales prep&#xFA;beres ten&#xED;an un promedio de 2,2 &#xB1; 0,2 dendritas primarias derivadas de somata (rango, de uno a siete dendritas primarias). Los &#xE1;ngulos en relaci&#xF3;n con el ax&#xF3;n dendritas a la que en los varones prep&#xFA;beres emanadas de somata GnRH fueron muy variables. La castraci&#xF3;n a 25 d&#xED;as de edad y la castraci&#xF3;n a los 25 d de edad con uno de los tres niveles de tratamiento T no modifican las caracter&#xED;sticas morfol&#xF3;gicas, cuando las neuronas GnRH fueron examinados entre 40 D y D 48 de edad. Por lo tanto, una remodelaci&#xF3;n espacial selectivo de las dendritas primarias y la consolidaci&#xF3;n de los cenadores dendr&#xED;ticas distales de GnRH ocurre durante el desarrollo postnatal y es muy independiente de T.

Gonadotropin-releasing-hormone (GnRH) neurons form part of a central neural oscillator that controls sexual reproduction through intermittent release of the GnRH peptide. Activity of GnRH neurons, and by extension release of GnRH, has been proposed to reflect intrinsic properties and synaptic input of GnRH neurons. To study GnRH neurons, we used traditional electrophysiology and computational methods. These emerging methodologies enhance the elucidation of processing in GnRH neurons. We used dynamic current-clamping to understand how living GnRH somata process input from glutamate and GABA, two key neurotransmitters in the neuroendocrine hypothalamus. In order to study the impact of synaptic integration in dendrites and neuronal morphology, we have developed full-morphology models of GnRH neurons. Using dynamic clamping, we have demonstrated that small-amplitude glutamatergic currents can drive repetitive firing in GnRH neurons. Furthermore, application of simulated GABAergic synapses with a depolarized reversal potential have revealed two functional subpopulations of GnRH neurons: one population in which GABA chronically depolarizes membrane potential (without inducing action potentials) and a second population in which GABAergic excitation results in slow spiking. Finally, when AMPA-type and GABA-type simulated inputs are applied together, action potentials occur when the AMPA-type conductance occurs during the descending phase of GABAergic excitation and at the nadir of GABAergic inhibition. Compartmental computer models have shown that excitatory synapses at >300 microns from somtata are unable to drive spiking with purely passive dendrites. In models with active dendrites, distal synapses are more efficient at driving spiking than somatic inputs. We then used our models to extend the results from dynamic current clamping at GnRH somata to distribute synaptic inputs along the dendrite. We show that propagation delays for dendritic synapses alter synaptic integration in GnRH neurons by widening the temporal window of interaction for the generation of action potentials. Finally, we have shown that changes in dendrite morphology can modulate the output of GnRH neurons by altering the efficacy of action potential generation in response to after-depolarization potentials (ADPs). Taken together, the methodologies of dynamic current clamping and multi-compartmental modeling can make major contributions to the study of synaptic integration and structure-function relationships in hypothalamic GnRH neurons. Use of these methodological approaches will continue to provide keen insights leading to conceptual advances in our understanding of reproductive hormone secretion in normal and pathological physiology and open the door to understanding whether the mechanisms of pulsatile GnRH release are conserved across species.

Gonadotropina hormona liberadora-(GnRH) neuronas forman parte de un oscilador central neural que controla la reproducci&#xF3;n sexual a trav&#xE9;s de la liberaci&#xF3;n intermitente del p&#xE9;ptido GnRH. Actividad de las neuronas GnRH, y por la liberaci&#xF3;n de extensi&#xF3;n de la GnRH, se ha propuesto para reflejar las propiedades intr&#xED;nsecas de entrada y sin&#xE1;ptica de las neuronas GnRH. Para estudiar las neuronas GnRH, se utiliz&#xF3; la electrofisiolog&#xED;a tradicional y los m&#xE9;todos de c&#xE1;lculo. Estas metodolog&#xED;as emergentes aumentar el esclarecimiento de procesamiento en las neuronas GnRH. Utilizamos din&#xE1;mica actual de sujeci&#xF3;n para entender c&#xF3;mo viven somata GnRH entrada del proceso de glutamato y GABA, dos neurotransmisores clave en el hipot&#xE1;lamo neuroendocrino. Con el fin de estudiar el impacto de la integraci&#xF3;n sin&#xE1;ptica en las dendritas y la morfolog&#xED;a neuronal, hemos desarrollado por completo la morfolog&#xED;a de los modelos de las neuronas GnRH. Uso din&#xE1;mico de sujeci&#xF3;n, se ha demostrado que de peque&#xF1;a amplitud corrientes glutamat&#xE9;rgicas puede conducir la acci&#xF3;n repetitiva de las neuronas GnRH. Adem&#xE1;s, la aplicaci&#xF3;n de la simulaci&#xF3;n de las sinapsis GABA&#xE9;rgicas con un potencial de inversi&#xF3;n despolariza han puesto de manifiesto dos subpoblaciones funcionales de las neuronas GnRH: una poblaci&#xF3;n en la que cr&#xF3;nicamente GABA despolariza el potencial de membrana (sin inducir a los potenciales de acci&#xF3;n) y una segunda poblaci&#xF3;n en la que GABA&#xE9;rgicas resultados en un r&#xE1;pido aumento de la excitaci&#xF3;n lenta. Finalmente, cuando el AMPA-tipo y GABA tipo entradas simuladas se aplican juntos, los potenciales de acci&#xF3;n se produce cuando la conductancia AMPA tipo se produce durante la fase descendente de excitaci&#xF3;n GABA&#xE9;rgica y en el nadir de inhibici&#xF3;n GABA&#xE9;rgicas. Modelos compartimentales inform&#xE1;ticos han demostrado que las sinapsis excitadoras a&#x26;gt; 300 micras de somtata no son capaces de conducir con las dendritas de adici&#xF3;n puramente pasivos. En los modelos con las dendritas activos, las sinapsis distales son m&#xE1;s eficientes en la conducci&#xF3;n de un r&#xE1;pido aumento de los insumos som&#xE1;ticos. A continuaci&#xF3;n, utiliza nuestros modelos para ampliar los resultados de la din&#xE1;mica actual de sujeci&#xF3;n en somata GnRH para distribuir entradas sin&#xE1;pticas a lo largo de la dendrita. Se demuestra que los retardos de propagaci&#xF3;n de las sinapsis dendr&#xED;ticas alterar la integraci&#xF3;n sin&#xE1;ptica en las neuronas GnRH mediante la ampliaci&#xF3;n de la ventana temporal de la interacci&#xF3;n para la generaci&#xF3;n de potenciales de acci&#xF3;n. Finalmente, se ha demostrado que los cambios en la morfolog&#xED;a de dendrita puede modular la salida de las neuronas GnRH mediante la alteraci&#xF3;n de la eficacia de la generaci&#xF3;n del potencial de acci&#xF3;n en respuesta a los potenciales despu&#xE9;s de la despolarizaci&#xF3;n (ADP). En conjunto, las metodolog&#xED;as de modelado din&#xE1;mico de corriente de sujeci&#xF3;n y m&#xFA;ltiples compartimentos, puede hacer importantes contribuciones al estudio de la integraci&#xF3;n sin&#xE1;ptica y relaciones estructura-funci&#xF3;n de las neuronas GnRH del hipot&#xE1;lamo.
