The electrophysiological development of hair cells between birth and the eight postnatal day (P8) was studied in the utricular macula of rats gestated in nest boxes mounted upon a centrifuge, subjecting the animals to a gravitational force of 2G. Whole-cell voltage-clamp recordings were made on cells in the acutely isolated epithelium. Cells were accessed through a tear in the epithelium, no enzymatic dissociation procedures were employed. Under artificially enhanced gravity, the whole cell conductance was dramatically altered in the two types of hair cells. Significant increases occurred from P3-4 in the type I cells while in the type II cells, the effect was delayed until P7-8. Fourfold and threefold increases of the mean slope conductance were observed at P7-8 in the type I and type II hair cells, respectively. These results indicate that the electrophysiological properties of a primary transducer such as utricle may be modified by variation of the primary stimulus during development.

Se estudi&oacute; el desarrollo electrofisiol&oacute;gico de las c&eacute;lulas ciliadas entre el nacimiento y los ocho d&iacute;a postnatal (P8) en la m&aacute;cula utricular de ratas gestada en cajas nido descansa sobre una centrifugadora, someter a los animales a una fuerza gravitacional de 2 G. Triple y cu&aacute;druple aumentos de la conductancia de la pendiente media fueron observados en P7-8 en el tipo I y tipo II c&eacute;lulas de pelo, respectivamente. Estos resultados indican que las propiedades electrofisiol&oacute;gicas de un transductor primario como utr&iacute;culo pueden modificarse por la variaci&oacute;n del est&iacute;mulo primario durante el desarrollo.

The mammalian utricular sensory receptors are commonly believed to be non-spiking cells with electrical activity limited to graded membrane potential changes. Here we provide evidence that during the first post-natal week, the sensory hair cells of the rat utricle express a tetrodotoxin (TTX)-sensitive voltage-gated Na+ current that displays most of the biophysical and pharmacological characteristics of neuronal Na+ current. Single-cell RT-PCR reveals that several alpha-subunit isoforms of the Na+ channels are co-expressed within a single hair cell, with a major expression of Nav1.2 and Nav1.6 subunits. In neonatal hair cells, 30 % of the Na+ channels are available for activation at the resting potential. Depolarizing current injections in the range of the transduction currents are able to trigger TTX-sensitive action potentials. We also provide evidence of a TTX-sensitive activity-dependent brain-derived neurotrophic factor (BDNF) release by early post-natal utricle explants. Developmental analysis shows that Na+ currents decrease dramatically from post-natal day 0 (P0) to P8 and become almost undetectable at P21. Concomitantly, depolarizing stimuli fail to induce both action potential and BDNF release at P20. The present findings reveal that vestibular hair cells express neuronal-like TTX-sensitive Na+ channels able to generate Na+-driven action potentials only during the early post-natal period of development. During the same period an activity-dependent BDNF secretion by utricular explants has been demonstrated. This could be an important mechanism involved in vestibular sensory system differentiation and synaptogenesis.

Glutamate is thought to be the main neurotransmitter at the synapse between the type I vestibular hair cell and its cognate calyx afferent. The present study was designed to identify the type of glutamate receptors involved in neurotransmission at this unusual synapse. Immunocytochemistry showed that AMPA GluR2, NMDA NR1 and NR2A/B subunits of the glutamate receptors were confined to the synaptic contact. We then examined the electrical activity at calyx terminals using direct electrophysiological recordings from intact dendritic terminals in explanted turtle posterior crista. We found that sodium-based action potentials support a background discharge that could be modulated by the mechanical stimulation of the hair bundle of the sensory cells. These activities were prevented by blocking both the mechano-electrical transduction channels and L-type voltage-gated Ca(2+) channels involved in synaptic transmission. Although pharmacological analysis revealed that NMDA receptors could operate, our results show that AMPA receptors are mainly involved in synaptic neurotransmission. We conclude that although both AMPA and NMDA glutamate receptor subunits are present at the calyx synapse, only AMPA receptors appear to be involved in the synaptic transmission between the type I vestibular hair cell and the calyx afferent.

Glutamato se cree que es el principal neurotransmisor en la sinapsis entre el tipo me vestibular c&eacute;lula de pelo y su c&aacute;liz cognado aferente. El presente estudio fue dise&ntilde;ado para identificar el tipo de receptores de glutamato implicados en la neurotransmisi&oacute;n en este inusual sinapsis. Immunocytochemistry demostr&oacute; que AMPA GluR2, NMDA NR1 y NR2A/B subunidades de los receptores de glutamato se limitaban al contacto sin&aacute;ptico. Luego examinamos la actividad el&eacute;ctrica en los bornes del c&aacute;liz en directas grabaciones electrofisiol&oacute;gicas de terminales dendr&iacute;ticas intactos de cresta posterior explantado tortuga. We investigated, during the first postnatal week, a voltage-gated sodium current (INa) transiently expressed in neonatal utricular hair cells in rats raised in hypergravity. Its electrophysiological properties did not differ significantly from those recorded from rats raised in normal gravity, but a delay was observed in their developmental expression. In normal gravity conditions, INa expression is maximal at postnatal days 1-2, conferring on the hair cells the ability to fire action potentials, and is down-regulated during the first postnatal week, whereas in hypergravity conditions, the down-regulation is delayed by 4 days. This is the first demonstration showing that development under enhanced gravity affects the transient excitability phase that characterizes neonate utricular hair cells, by delaying a critical period of vestibular development.

Investigamos, durante la primera semana postnatal, voltage-gated sodio actual (INa) transitoriamente expresado en las c&eacute;lulas de pelo utricular neonatales en ratas en hipergravedad. Sus propiedades electrofisiol&oacute;gicas no difiri&oacute; significativamente de los registrados de ratas en gravedad normal, pero se observ&oacute; un retraso en su expresi&oacute;n del desarrollo. En condiciones de gravedad normal, expresi&oacute;n de INa es m&aacute;xima en postnatales d&iacute;as 1-2, confiriendo a las c&eacute;lulas de pelo la capacidad de fuego de potenciales de acci&oacute;n y es regula durante la primera semana postparto, mientras que en condiciones de Hipergravedad, la regulaci&oacute;n se retrasa por 4 d&iacute;as. Se trata de la primera demostraci&oacute;n que muestra que el desarrollo bajo mayor gravedad afecta la fase transitoria de la excitabilidad que caracteriza a las c&eacute;lulas ciliadas utricular reci&eacute;n nacido, retrasando un per&iacute;odo cr&iacute;tico de desarrollo vestibular.

In the rat utricle, synaptic contacts between hair cells and the nerve fibers arising from the vestibular primary neurons form during the first week after birth. During that period, the sodium-based excitability that characterizes neonate utricle sensory cells is switched off. To investigate whether the establishment of synaptic contacts was responsible for the modulation of the hair cell excitability, we used an organotypic culture of rat utricle in which the setting of synapses was prevented. Under this condition, the voltage-gated sodium current and the underlying action potentials persisted in a large proportion of nonafferented hair cells. We then studied whether impairment of nerve terminals in the utricle of adult rats may also affect hair cell excitability. We induced selective and transient damages of afferent terminals using glutamate excitotoxicity in vivo. The efficiency of the excitotoxic injury was attested by selective swellings of the terminals and underlying altered vestibular behavior. Under this condition, the sodium-based excitability transiently recovered in hair cells. These results indicate that the modulation of hair cell excitability depends on the state of the afferent terminals. In adult utricle hair cells, this property may be essential to set the conditions required for restoration of the sensory network after damage. This is achieved via re-expression of a biological process that occurs during synaptogenesis.

Control of hair cell excitability by vestibular primary sensory neurons.

In der Ratte Schlauch, synaptischen Kontakte zwischen den Haarzellen und der Nervenfasern, die infolge der vestibul&auml;ren prim&auml;re Neuronen-Form in der ersten Woche nach der Geburt. W&auml;hrend dieser Zeit ist die Natrium-basierte Reizbarkeit, die Neugeborenen Schlauch Sinneszellen charakterisiert ausgeschaltet. Um zu untersuchen, ob die Herstellung von synaptischen Kontakten f&uuml;r die Modulation der Haarzelle Erregbarkeit verantwortlich war, haben wir eine organotypische Kultur der Ratte-Schlauch, in der die Einstellung der Synapsen verhindert wurde. Unter diesen Bedingungen beibehalten der aktuellen Spannung-Gating Natrium und die zugrunde liegenden Aktionspotentiale in einen Gro&szlig;teil der Nonafferented Haarzellen. Wir untersucht dann, ob die Beeintr&auml;chtigung der Nervenenden in den Schlauch von Erwachsenen Ratten auch Haarzelle Erregbarkeit beeinflussen kann. Wir induzierte selektive und vor&uuml;bergehende Sch&auml;den der afferenten Terminals mit Glutamat Exzitotoxit&auml;t in-vivo. Durante ese per&iacute;odo, la excitabilidad basadas en sodio que caracteriza a las c&eacute;lulas sensoriales de la utr&iacute;culo reci&eacute;n nacido est&eacute; apagado. Para investigar si el establecimiento de contactos sin&aacute;pticos fue responsable de la modulaci&oacute;n de la excitabilidad de la c&eacute;lula de pelo, se utiliz&oacute; una cultura organotypic de utr&iacute;culo de rata en el que el ajuste de sinapsis fue prevenido. Bajo esta condici&oacute;n, el sodio voltaje actual y los potenciales de acci&oacute;n subyacentes persiste en una gran proporci&oacute;n de c&eacute;lulas de pelo nonafferented. Luego estudiamos si deterioro de terminaciones nerviosas en el utr&iacute;culo de ratas adultas tambi&eacute;n puede afectar la excitabilidad de la c&eacute;lula de pelo. Nos induce da&ntilde;os selectivos y transitorias de terminales aferentes con glutamato excitotoxicidad in vivo. La eficacia de la lesi&oacute;n excitatoria fue atestiguada por inflamiento selectivo de los terminales y comportamiento vestibular alterada subyacente. Bajo esta condici&oacute;n, la excitabilidad basadas en sodio transitoriamente recuper&oacute; en c&eacute;lulas de pelo. Estos resultados indican que la modulaci&oacute;n de la excitabilidad de la c&eacute;lula de pelo depende del estado de los terminales aferentes. En c&eacute;lulas de pelo adulto utr&iacute;culo, esta propiedad puede ser esencial para establecer las condiciones necesarias para el restablecimiento de la red sensorial despu&eacute;s del da&ntilde;o. The importance of this functional measure is illustrated by its use in studies of the genetic basis of vestibular function and disease. Head motion is the stimulus for the VsEP. In the bird, it has been established that neurons mediating the linear VsEP respond collectively to the rate of change in linear acceleration during head movement (i.e. jerk) rather than peak acceleration. The kinematic element of motion responsible for triggering mammalian VsEPs has not been characterized in detail. Here we tested the hypothesis that jerk is the kinematic component of head motion responsible for VsEP characteristics. VsEP amplitudes and latencies changed systematically when peak acceleration level was held constant and jerk level was varied from âˆ¼0.9-4.6 g/ms. In contrast, responses remained relatively constant when kinematic jerk was held constant and peak acceleration was varied from âˆ¼0.9 to 5.5 g in mice and âˆ¼0.44 to 2.75 g in rats. Thus the mammalian VsEP depends on jerk levels and not peak acceleration. We conclude that kinematic jerk is the adequate stimulus for the mammalian VsEP. This sheds light on the behavior of neurons generating the response. The results also provide the basis for standardizing the reporting of stimulus levels, which is key to ensuring that response characteristics reported in the literature by many laboratories can be effectively compared and interpreted.

Estado latente corto lineal sensorial potenciales evocados vestibulares (VsEPs) proporcionan un medio para objetivamente y evaluar directamente la funci&oacute;n de receptores de gravedad en mam&iacute;feros y aves. La importancia de esta medida funcional es ilustrada por su uso en estudios de la base gen&eacute;tica de la funci&oacute;n vestibular y la enfermedad. Movimiento de la cabeza es el est&iacute;mulo para la VsEP. En el p&aacute;jaro, se ha establecido que las neuronas que median la VsEP lineal responden colectivamente a la tasa de cambio en aceleraci&oacute;n lineal durante el movimiento de la cabeza (es decir, jerk) en lugar de aceleraci&oacute;n m&aacute;xima. El elemento cinem&aacute;tico del movimiento responsable de desencadenar mam&iacute;feros VsEPs no se ha caracterizado en detalle. Aqu&iacute; probamos la hip&oacute;tesis que tir&oacute;n es el componente cinem&aacute;tico de movimiento principal responsable VsEP caracter&iacute;sticas. VsEP amplitudes y latencias cambiaron sistem&aacute;ticamente cuando nivel de aceleraci&oacute;n pico se llev&oacute; a cabo constante y nivel de tir&oacute;n fue variado de &sim;0.9-4,6 g/Sra. por el contrario, las respuestas permanecieron relativamente constantes cuando tir&oacute;n cinem&aacute;tica se llev&oacute; a cabo constante y aceleraci&oacute;n m&aacute;xima fue oscil&oacute; entre &sim;0.9 y 5.5g en ratones y &sim;0.44 a 2.75 g en ratas. As&iacute; el VsEP mam&iacute;fero depende de niveles del tir&oacute;n y no la aceleraci&oacute;n m&aacute;xima. Concluimos que eso tir&oacute;n cinem&aacute;tica es el est&iacute;mulo adecuado para el VsEP mam&iacute;fero. Esto arroja luz sobre el comportamiento de las neuronas generadoras de la respuesta. Los resultados tambi&eacute;n proporcionan la base para la estandarizaci&oacute;n de la informaci&oacute;n de los niveles de est&iacute;mulo, que es clave para asegurar que las caracter&iacute;sticas de respuesta divulgadas en la literatura por muchos laboratorios efectivamente comparables e interpretadas.
