In yeast and animals, replication of the mitochondrial genome is carried out by the DNA polymerase gamma. In mammals this polymerase is composed of a catalytic and an accessory subunit. Yeast DNA polymerase gamma was purified over 6600-fold from mitochondria. The catalytic polypeptide of this enzyme was identified as a 135-kDa protein by a photochemical crosslinking procedure and its native molecular weight was estimated between 120 and 140 kDa by gel filtration and glycerol gradient sedimentation. These results indicate that yeast DNA polymerase gamma contains only one subunit and thus has a different quaternary structure from its counterpart in animals.

Study of the complexome - all the protein complexes of the cell - is essential for a better understanding and more global vision of cell function. Using two-dimensional blue native/SDS-PAGE (2-D BN/SDS-PAGE) technology, the cytosolic and membrane protein complexes of Escherichia coli were separated. Then, the different partners of each protein complex were identified by LC-MS/MS. In this report, 306 protein complexes were separated and identified. Among these protein complexes, 50 heteromultimeric and 256 homomultimeric protein complexes were found. Among the 50 heteromultimeric protein complexes, 18 previously described protein complexes validate the technology. In this study, 109 new protein complexes were found, providing insight into the function of previously uncharacterized bacterial proteins.

A complexomic study of Escherichia coli using two-dimensional blue native/SDS polyacrylamide gel electrophoresis.

&Eacute;tude de la complexome - tous les complexes prot&eacute;ines de la cellule - est indispensable pour une meilleure compr&eacute;hension et une vision plus globale de la fonction cellulaire. &Agrave; l&#39;aide de deux dimensions bleu natif/SDS-PAGE (2d BN/SDS-PAGE) la technologie, les prot&eacute;ines membranaires et cytosoliques complexes d&#39;Escherichia coli ont &eacute;t&eacute; s&eacute;par&eacute;s. Ensuite, les diff&eacute;rents partenaires de chaque complexe de prot&eacute;ines ont &eacute;t&eacute; identifi&eacute;es par LC-MS/MS. Dans ce rapport, 306 complexes prot&eacute;iques ont &eacute;t&eacute; s&eacute;par&eacute;s et identifi&eacute;s. Parmi ces complexes de prot&eacute;ines, 50 h&eacute;t&eacute;romultim&eacute;rique et 256 homomultimeric complexes de prot&eacute;ines ont &eacute;t&eacute; trouv&eacute;es. Estudio de la complexome - todos los complejos de la prote&iacute;na de la c&eacute;lula - es esencial para una mejor comprensi&oacute;n y una visi&oacute;n m&aacute;s global de la funci&oacute;n celular. Usando dos dimensiones azul nativo/SDS-PAGE (2-D BN/SDS-PAGE) tecnolog&iacute;a, la prote&iacute;na citos&oacute;lica y membrana complejos de Escherichia coli se separaron. Luego, se identificaron los diferentes interlocutores de cada complejo proteico por LC-MS/MS. En este informe, 306 complejos de la prote&iacute;na fueron separados e identificados. Entre estos complejos de la prote&iacute;na, se encontraron 50 heteromultimeric y 256 homomultimeric complejos de la prote&iacute;na. The study of protein interactions constitutes an important domain to understand the physiology and pathogenesis of microorganisms. The two-dimensional blue native/SDS-PAGE was initially reported to analyze membrane protein complexes. In this study, both cytoplasmic and membrane complexes of a bacterium, the strain J99 of the gastric pathogen Helicobacter pylori, were analyzed by this method. It was possible to identify 34 different proteins grouped in 13 multiprotein complexes, 11 from the cytoplasm and two from the membrane, either previously reported partially or totally in the literature. Besides complexes involved in H. pylori physiology, this method allowed the description of interactions involving known pathogenic factors such as (i) urease with the heat shock protein GroEL or with the putative ketol-acid reductoisomerase IlvC and (ii) the cag pathogenicity island CagA protein with the DNA gyrase GyrA as well as insight on the partners of TsaA, a peroxide reductase/stress-dependent molecular chaperone. The two-dimensional blue native/SDS-PAGE combined with mass spectrometry is a potential tool to study the differences in complexes isolated in various situations and also to study the interactions between bacterial and eucaryotic cell proteins.

La deux dimensions bleue natif/SDS-PAGE aurait initialement pour analyser des complexes de prot&eacute;ines membranaires. El azul de dos dimensiones nativo/SDS-PAGE inform&oacute; inicialmente analizar complejos de la prote&iacute;na de membrana. En este estudio, ambos citopl&aacute;smica y complejos de membrana de una bacteria, la cepa J99 de los p&iacute;loros de Helicobacter pat&oacute;geno g&aacute;strico, fueron analizados por este m&eacute;todo. Adem&aacute;s de complejos en la fisiolog&iacute;a de H. pylori, este m&eacute;todo permiti&oacute; la descripci&oacute;n de las interacciones que involucran factores pat&oacute;genos conocidos como (i) ureasa con la prote&iacute;na de choque t&eacute;rmico GroEL o con la supuesta reductoisomerase ketol-&aacute;cido IlvC y (ii) la prote&iacute;na de cag de CagA de Isla de patogenicidad con la ADN girasa GyrA as&iacute; como comprender mejor cu&aacute;les son los socios de TsaA, una chaperona molecular per&oacute;xida de reductasa/estr&eacute;s-dependiente. Patients with cancer produce specific autoantibodies against protein antigens present in limited amount among a large background of immunoglobulins (Igs), nonrelevant as biomarkers, including natural antibodies. Multiple affinity protein profiling (MAPPing) that combines 2-D immunoaffinity chromatography, enzymatic digestion of the isolated proteins, and identification by MS/MS, may facilitate the identification of these so far unknown patient antibodies. The first immunoaffinity chromatography is crucial, as it is used for selectively removing proteins (autoantigens) recognized by natural antibodies. Application of this depletion step to colon cancer cell proteins is specifically described along with the identification of the natural autoantigens, as well as the coupling of this depletion step with the next steps. By enabling to separate antibody-binding proteins recognized by either natural autoantibodies or patient-specific antibodies this approach may contribute significantly towards the definition of autoantibody signatures.

Pacientes con c&aacute;ncer producen autoanticuerpos espec&iacute;ficos contra los ant&iacute;genos de la prote&iacute;na presentes en cantidad limitada entre un gran fondo de inmunoglobulinas (Igs), irrelevantes como biomarcadores, incluyendo anticuerpos naturales. Prote&iacute;na de afinidad m&uacute;ltiples perfiles (MAPPing) combina cromatograf&iacute;a de inmunoafinidad 2-D, digesti&oacute;n enzim&aacute;tica de las prote&iacute;nas aisladas y la identificaci&oacute;n por MS/MS, puede facilitar la identificaci&oacute;n de estos anticuerpos pacientes hasta ahora desconocidos. La cromatograf&iacute;a de inmunoafinidad primera es crucial, ya que es utilizado para eliminar selectivamente las prote&iacute;nas (autoant&iacute;genos) reconocidas por los anticuerpos naturales. Aplicaci&oacute;n de este paso de agotamiento a prote&iacute;nas de c&eacute;lulas de c&aacute;ncer de colon se describe espec&iacute;ficamente junto con la identificaci&oacute;n de los autoant&iacute;genos natural, as&iacute; como el acoplamiento de esta etapa de agotamiento con los siguientes pasos. Habilitando para separar prote&iacute;nas anticuerpo reconocidas por autoanticuerpos naturales o anticuerpos espec&iacute;fico para cada paciente este enfoque puede contribuir significativamente a la definici&oacute;n de firmas autoanticuerpo.

Cancer remains one of the leading causes of death worldwide. Thus, to identify any useful biomarkers is still a need. We performed "cancer immunomics" to identify autoantibody signatures produced in response to the presence of either breast or colorectal cancer. SERological proteome analysis (SERPA) was performed by two-dimensional (2-D) electrophoresis separation, immunoblotting, image analysis, and mass spectrometry. Alternatively, to identify the antigens recognized by the autoantibodies of cancer patients, we developed an approach combining 2-D immunoaffinity chromatography, enzymatic digestion of the isolated antigens, nano flow separation of the resulting peptides, and identification: MAPPing (multiple affinity protein profiling). By these approaches we identified both proteins recognized by autoantibodies independently of a cancer status, and a limited number of proteins reacting preferentially with cancer sera.

Analyse du prot&eacute;ome s&eacute;rologique (SERPA) a &eacute;t&eacute; r&eacute;alis&eacute;e par la s&eacute;paration de l&#39;&eacute;lectrophor&egrave;se bidimensionnelle (2d), immunoblotting, analyse d&#39;image et la spectrom&eacute;trie de masse. Sinon, pour identifier les antig&egrave;nes reconnus par les autoanticorps des patients atteints de cancer, nous avons d&eacute;velopp&eacute; une approche combinant la chromatographie d&#39;immunoaffinit&eacute; 2D, la digestion enzymatique des antig&egrave;nes isol&eacute;s, s&eacute;paration des peptides qui en r&eacute;sulte et identification de l&#39;&eacute;coulement nano : cartographie (profilage plusieurs prot&eacute;ines affinit&eacute;). Par ces approches, nous avons identifi&eacute; deux prot&eacute;ines reconnues par les autoanticorps ind&eacute;pendamment un statut du cancer et un nombre limit&eacute; de prot&eacute;ines r&eacute;agissant pr&eacute;f&eacute;rentiellement avec les s&eacute;rums de cancer.

Alternativ um die Antigene erkannt durch die Autoantik&ouml;rper von Krebspatienten zu identifizieren, entwickelten wir einen Ansatz, bei dem 2-D Immunoaffinity Chromatographie, enzymatische Verdauung der isolierten Antigene, Nano Str&ouml;mung Trennung der resultierende Peptiden und Identifikation: MAPPing (mehrere Affinit&auml;t Protein Profilerstellung). Durch diese Ans&auml;tze haben wir festgestellt, beide Proteine von Autoantik&ouml;rpern unabh&auml;ngig von einer Krebs-Status und eine begrenzte Anzahl von Proteinen reagieren bevorzugt mit Krebs Sera erkannt.

Analisi sierologiche proteome (SERPA) &egrave; stata eseguita da separazione l&#39;elettroforesi bidimensionale (2-D), immunoblotting, analisi di immagine e spettrometria di massa. In alternativa, per identificare gli antigeni riconosciuti da autoanticorpi di pazienti affetti da cancro, abbiamo sviluppato un approccio che unisce cromatografia di immunoaffinity 2-D, digestione enzimatica degli antigeni isolati, separazione di flusso nano dei peptidi risultante e identificazione: MAPPing (pi&ugrave; affinit&agrave; della proteina profiling). Da questi approcci abbiamo identificato due proteine riconosciute da autoanticorpi indipendentemente da uno status di cancro e da un numero limitato di proteine reagiscono preferenzialmente con i sieri di cancro.

C&aacute;ncer sigue siendo una de las principales causas de muerte en todo el mundo. As&iacute; identificar cualquier biomarcadores &uacute;tiles a&uacute;n es necesario. Realizamos &quot;c&aacute;ncer immunomics&quot; para identificar firmas autoanticuerpo producidas en respuesta a la presencia de mama o c&aacute;ncer colorrectal. An&aacute;lisis del proteoma serol&oacute;gica (SERPA) fue realizada por la separaci&oacute;n de la electroforesis de dos dimensiones (2D), immunoblotting, an&aacute;lisis de imagen y espectrometr&iacute;a de masas. Por otra parte, para identificar los ant&iacute;genos reconocidos por los autoanticuerpos de pacientes con c&aacute;ncer, hemos desarrollado un enfoque que combina cromatograf&iacute;a de inmunoafinidad 2-D, digesti&oacute;n enzim&aacute;tica de los ant&iacute;genos aislados, nano separaci&oacute;n del flujo de los p&eacute;ptidos resultantes y la identificaci&oacute;n: MAPPing (m&uacute;ltiples afinidad prote&iacute;na profiling). Por estos enfoques se identificaron ambas prote&iacute;nas reconocidas por autoanticuerpos independientemente de un estado de c&aacute;ncer y un n&uacute;mero limitado de prote&iacute;nas reaccionan preferentemente con este c&aacute;ncer.

Polychlorinated biphenyls (PCBs) and a number of pesticides can act as endocrine disrupting compounds (EDCs). These molecules exhibit hormonal activity in vivo, and can therefore interact and perturb normal physiological functions. Many of these compounds are persistent in the environment, and their bioaccumulation may constitute a significant threat for human health. Physiological abnormalities following exposure to these xenobiotic compounds go along with alterations at the protein level of individual cells. In this study, MCF-7 cells were exposed to environmentally relevant concentrations of atrazine, PCB153 (100 ppb, respectively), 17-beta estradiol (positive control, 10 nM) and a negative control (solvent) for t = 24 h (n = 3 replicates/exposure group). After trizol extraction and protein solubilization, protein expression levels were studied by 2D-DIGE. Proteins differentially expressed were excised, trypsin-digested, and identified by MALDI-ToF-ToF, followed by NCBInr database search. 2D-DIGE experiments demonstrated that 49 spots corresponding to 29 proteins were significantly differentially expressed in MCF-7 cells (>1.5-fold, P < 0.05, Student's paired t test). These proteins belonged to various cellular compartments (nucleus, cytosol, membrane), and varied in function; 88% of proteins were down-regulated during atrazine exposure, whereas 75% of proteins were up-regulated by PCB153. Affected proteins included those regulating oxidative stress such as superoxide dismutase and structural proteins such as actin or tropomyosin, which may explain morphological changes of cells already observed under the microscope. This study highlights the susceptibility of human cells to compounds with endocrine disrupting properties.

Ces mol&#xE9;cules pr&#xE9;sentent une activit&#xE9; hormonale in vivo, et peut donc interagir et de perturber les fonctions physiologiques normales. Beaucoup de ces compos&#xE9;s sont persistants dans l&#x26;#39;environnement, et de leur bioaccumulation peut constituer une menace importante pour la sant&#xE9; humaine. Polychlorierte Biphenyle (PCB) und eine Reihe von Pestiziden k&ouml;nnen als endokrine endokrinsch&auml;dlichen Substanzen (EDCs) fungieren. Diese Molek&uuml;le k&ouml;nnen in-vivo, hormonelle Aktivit&auml;t aufweisen und daher interagieren und normale physiologische Funktionen verl&auml;ngern. Viele dieser Verbindungen sind dauerhaft in der Umwelt und ihrer Bioakkumulation kann eine erhebliche Gefahr f&uuml;r die menschliche Gesundheit darstellen. Physiologische Anomalien nach Exposition gegen&uuml;ber diesen xenobiotic Verbindungen gehen mit Ver&auml;nderungen auf der Protein-Ebene einzelner Zellen. In dieser Studie waren MCF-7 Zellen &ouml;kologisch relevante Konzentrationen von Atrazin, PCB153 ausgesetzt (100 ppb, beziehungsweise), 17-Beta-&Ouml;stradiol (Positivkontrolle, 10 nM) und eine negative Kontrolle (L&ouml;sungsmittel) f&uuml;r t = 24 h (n = 3 Wiederholungen/Expositions-Gruppe). Protein-Ausdruck-Ebene wurden nach Verwendung Trizol Extraktion und Protein Solubilization 2D-DIGE untersucht. Proteine differentiell waren excised Trypsin verdaut und von MALDI-ToF-ToF, gefolgt von der NCBInr-Datenbank-Suche. 2D-DIGE Experimenten nachgewiesen, dass 49 Punkte entspricht 29 Proteine differentiell deutlich in MCF-7 Zellen ausgedr&uuml;ckt wurden (&gt; 1.5-fold, P &lt; 0,05, studentischen gepaarten t-Test). Diese Proteine geh&ouml;rten zu verschiedenen zellularen F&auml;chern (Zytosol, Zellkern, Membran) und variiert in Funktion; 88 % der Proteine wurden unten-geregelt w&auml;hrend Atrazin Belichtung, w&auml;hrend 75 % der Proteine durch PCB153 oben-geregelt waren. I policlorobifenili (PCB) e un numero di pesticidi pu&ograve; agire come composti alleattivit&agrave; endocrini (CDE). Queste molecole presentano attivit&agrave; ormonale in vivo e quindi possono interagire e turbare le normali funzioni fisiologiche. Molti di questi composti sono persistenti nell&#39;ambiente e loro bioaccumulo pu&ograve; costituire una grave minaccia per la salute umana. Anomalie fisiologiche dopo esposizione a questi composti xenobiotici andare insieme con alterazioni a livello della proteina di singole celle. In questo studio, le cellule MCF-7 sono stati esposti a ecologicamente rilevanti concentrazioni di atrazina, PCB153 (100 ppb, rispettivamente), 17-beta estradiolo (controllo positivo, 10 nM) e un controllo negativo (solvente) per t = 24 h (n = 3 gruppo di replica/esposizione). Dopo estrazione trizol e solubilizzazione della proteina, i livelli di espressione della proteina sono stati studiati da 2D-DIGE. Proteine differenzialmente espresse sono stati escisse, digerite tripsina e identificati da MALDI-ToF-ToF, seguita dalla ricerca del database NCBInr. Esperimenti 2D-DIGE ha dimostrato che 49 punti corrispondenti alle 29 proteine significativamente erano differenzialmente espressi in cellule MCF-7 (&amp;gt; 1.5-fold, P &lt; 0.05, test paired t di Student). Despu&#xE9;s de la extracci&#xF3;n y solubilizaci&#xF3;n de las prote&#xED;nas trizol, los niveles de expresi&#xF3;n de prote&#xED;nas se estudiaron por 2D-DIGE. The yeast Yarrowia lipolytica uses hydrophobic substrates, such as alkanes, fatty acids and oils, for its growth. It has developed a strategy for the use of such substrates, involving the production of hydrophobic binding structures called protrusions on the cell surface. These protrusions are resemble channels connecting the cell wall to the inside of the cell, and are probably involved in transport mechanisms that we do not yet fully understand. The complete genome of the haploid Y. lipolytica strain E150 (CLIB99) was sequenced in 2004 by the GÃ©nolevures Consortium. The availability of a complete genome sequence for this species has made it possible to carry out proteomic and other investigations, leading to the characterization of lipid bodies (LB) in terms of (i) their lipid composition, (ii) the major LB proteins, as identified by mass spectrometry, and (iii) differences in protein or lipid composition as a function of the carbon source used. Functional analyses would provide insight into the biological processes associated with these bodies and 2D BN/SDS-PAGE is a highly suitable method for the analysis of protein complexes. This report provides a first description of the analysis and identification of hydrophobic binding protein complexes in Y. lipolytica. For this purpose, we used 2D BN/SDS-PAGE for the separation of protein complexes and HPLC-chip-MS for protein identification. We separated and identified 40 protein complexes (11 heteromultimeric and 29 homomultimeric), providing insight into their function. This study represents a major step forward, as most previous studies identified proteins either on the basis of sequence similarity to proteins from other organisms (44% of the proteins identified in this study) or by prediction (50% of proteins identified in this study) alone.

La levadura lipolytica de Yarrowia utiliza sustratos hidrof&oacute;bicos, tales como alcanos, &aacute;cidos grasos y aceites, para su crecimiento. Se ha desarrollado una estrategia para la utilizaci&oacute;n de estos sustratos, que implica la producci&oacute;n de estructuras de enlace hidrof&oacute;bico llamado protuberancias en la superficie celular. Estas protuberancias son se asemejan a los canales de conexi&oacute;n de la pared celular en el interior de la c&eacute;lula y probablemente participan en los mecanismos de transporte que todav&iacute;a no entendemos completamente. Fue secuenciado el genoma completo de la cepa de lipolytica Y. haploide E150 (CLIB99) en 2004 por el consorcio de G&eacute;nolevures. La disponibilidad de una secuencia completa del genoma de esta especie ha hecho posible llevar a cabo la prote&oacute;mica y otras investigaciones, llevando a la caracterizaci&oacute;n de los cuerpos de l&iacute;pidos (LB) en (i) su composici&oacute;n lip&iacute;dica, (ii) los principal LB prote&iacute;nas, identificado por espectrometr&iacute;a de masas y (iii) las diferencias en la composici&oacute;n de la prote&iacute;na o l&iacute;pido en funci&oacute;n de la fuente de carbono. An&aacute;lisis funcionales proporcionar la penetraci&oacute;n en los procesos biol&oacute;gicos asociados a estos cuerpos y 2D BN/SDS-PAGE es un m&eacute;todo muy adecuado para el an&aacute;lisis de los complejos de la prote&iacute;na. Este informe proporciona una primera descripci&oacute;n del an&aacute;lisis e identificaci&oacute;n de complejos de la prote&iacute;na de enlace hidrof&oacute;bico en Y. lipolytica. Para ello, utilizamos 2D BN/SDS-PAGE para la separaci&oacute;n de los complejos de la prote&iacute;na y chip-HPLC-MS para identificaci&oacute;n de prote&iacute;nas. Se separ&oacute; y se identificaron 40 complejos de la prote&iacute;na (heteromultimeric 11 y 29 homomultimeric), proporcionando informaci&oacute;n sobre su funci&oacute;n. Although many virulence factors have been associated with DU, many questions remain unanswered regarding the evolution of the infection toward this exceptional event, LG-MALT. The present study describes and compares the complexome of two H. pylori strains, strain J99 associated with DU and strain B38 associated with LG-MALT, using the two-dimensional blue native/SDS-PAGE method. It was possible to identify 90 different complexes (49 and 41 in the B38 and J99 strains, respectively); 12 of these complexes were common to both strains (seven and five in the membrane and cytoplasm, respectively), reflecting the variability of H. pylori strains. The 44 membrane complexes included numerous outer membrane proteins, such as the major adhesins BabA and SabA retrieved from a complex in the B38 strain, and also proteins from the hor family rarely studied. BabA and BabB adhesins were found to interact independently with HopM/N in the B38 and J99 strains, respectively. The 46 cytosolic complexes essentially comprised proteins involved in H. pylori physiology. Some orphan proteins were retrieved from heterooligomeric complexes, and a function could be proposed for a number of them via the identification of their partners, such as JHP0119, which may be involved in the flagellar function. Overall, this study gave new insights into the membrane and cytoplasm structure, and those which could help in the design of molecules for vaccine and/or antimicrobial agent development are highlighted.

Complexomics study of two Helicobacter pylori strains of two pathological origins: potential targets for vaccine development and new insight in bacteria metabolism.

Die vorliegende Studie beschreibt und vergleicht die Complexome von zwei H. Infecci&oacute;n por Helicobacter pylori desempe&ntilde;a un papel causal en el desarrollo de linfoma g&aacute;strico tejido linfoide asociado a mucosa (MALT) (LG-Malta) y &uacute;lcera duodenal (DU). Aunque muchos factores de virulencia han sido asociados con DU, muchas preguntas siguen sin respuesta con respecto a la evoluci&oacute;n de la infecci&oacute;n hacia este evento excepcional, LG-Malta. El presente estudio describe y compara la complexome de cepas de dos H. pylori, cepa J99 asociados con DU y cepa B38 asociado con LG-Malta, utilizando el m&eacute;todo azul bidimensional del nativo/SDS-PAGE. Fue posible identificar 90 diversos complejos (49 y 41 en la B38 y J99 cepas, respectivamente); 12 de estos complejos son comunes a ambas cepas (siete y cinco en la membrana y citoplasma, respectivamente), lo que refleja la variabilidad de cepas de H. pylori. Los complejos de 44 membrana incluyen numerosas prote&iacute;nas de membrana externa, como los principales adhesinas BabA y SabA Obtenido de un complejo en la cepa B38 y tambi&eacute;n las prote&iacute;nas de la familia de hor raramente estudiado. BabA y BabB adhesinas encontradas relacionarse independientemente con HopM/N en las cepas B38 y J99, respectivamente. Los 46 complejos citos&oacute;licas esencialmente compuesto por prote&iacute;nas implicadas en la fisiolog&iacute;a de H. pylori. Algunas prote&iacute;nas hu&eacute;rfanos fueron recuperados de heterooligomeric complejos, y una funci&oacute;n podr&iacute;a proponerse para un n&uacute;mero de ellos mediante la identificaci&oacute;n de sus socios, como JHP0119, que pueden estar involucrados en la funci&oacute;n flagelar. The complexome is essential for a better understanding of protein functions. In order to study protein complexes, an approach allowing the extraction and the analysis in native conditions is needed. Two-dimensional blue native/SDS-PAGE (2D BN/SDS-PAGE) technology is thus an interesting and powerful approach for this purpose. This report deals with the analysis and the identification of the organellar protein complexes of Caco-2 human cells using 2D BN/SDS-PAGE and HPLC-chip-MS. We identified 58 protein complexes (26 heteromultimeric and 32 homomultimeric complexes) and 4 monomeric proteins. Among them, 32 protein complexes were pointed out, providing insights into the function of previously uncharacterized human proteins.

Technologie bleu natif/SDS-PAGE &agrave; deux dimensions (2D BN/SDS-PAGE) est donc une approche int&eacute;ressante et puissante &agrave; cet effet. Ce rapport traite de l&#39;analyse et l&#39;identification des complexes prot&eacute;ine organites des cellules humaines Caco-2, en utilisant le 2D BN/SDS-PAGE et puce-HPLC-MS. La complexome es esencial para una mejor comprensi&oacute;n de las funciones de la prote&iacute;na. Con el fin de estudiar los complejos proteicos, es necesario un enfoque que permite la extracci&oacute;n y el an&aacute;lisis en condiciones nativas. Tecnolog&iacute;a bidimensional azul nativo/SDS-PAGE (2D BN/SDS-PAGE) es as&iacute; un enfoque interesante y de gran alcance para ello. Este informe aborda el an&aacute;lisis y la identificaci&oacute;n de los complejos de la prote&iacute;na organellar de Caco-2 c&eacute;lulas humanas usando 2D BN/SDS-PAGE y chip-HPLC-MS. Se identificaron 58 complejos de la prote&iacute;na (heteromultimeric 26 y 32 homomultimeric complejos) y 4 prote&iacute;nas monom&eacute;ricas. Entre ellos, 32 complejos de la prote&iacute;na fueron precisadas, proporcionar penetraciones en la funci&oacute;n de prote&iacute;nas humanas previamente desacostumbradas.
