Un tiers de la population mondiale est infect&#xE9;e par Mycobacterium tuberculosis et multi-souches r&#xE9;sistantes aux m&#xE9;dicaments sont en rapide &#xE9;volution. L&#x26;#39;absence notable d&#x26;#39;un syst&#xE8;me de d&#xE9;pistage organisme entier &#xE0; haut d&#xE9;bit pour l&#x26;#39;&#xE9;tude de la progression de la tuberculose est en passe de devenir le goulot d&#x26;#39;&#xE9;tranglement dans la recherche contre la tuberculose. Nous avons r&#xE9;ussi &#xE0; d&#xE9;velopper un tel syst&#xE8;me en utilisant le mod&#xE8;le de poisson z&#xE8;bre infection &#xE0; Mycobacterium marinum, qui est un mod&#xE8;le bien caract&#xE9;ris&#xE9; pour la progression la tuberculose avec une signification biom&#xE9;dicale, imitant caract&#xE9;ristiques de la tuberculose humaine pathologie. Surtout, nous d&#xE9;montrer la pertinence de notre syst&#xE8;me d&#x26;#39;&#xE9;tudier directement de M. tuberculosis, montrant pour la premi&#xE8;re fois que l&#x26;#39;agent pathog&#xE8;ne humain peut se propager dans ce mod&#xE8;le vert&#xE9;br&#xE9;, ce qui entra&#xEE;ne des sympt&#xF4;mes similaires pr&#xE9;coces de la maladie &#xE0; ceux observ&#xE9;s sur M. marinum infection. Notre syst&#xE8;me est capable de d&#xE9;pistage de la progression de la maladie par injection jaune robotique d&#x26;#39;embryons pr&#xE9;coces et le d&#xE9;pistage flux visuel de la fin du stade larvaire. Nous montrons aussi que ce syst&#xE8;me de mani&#xE8;re fiable r&#xE9;capituler la m&#xE9;thode standard injection dans la veine caudale avec un niveau de d&#xE9;bit de 2.000 embryons par heure. Nous d&#xE9;montrer, en outre la possibilit&#xE9; d&#x26;#39;&#xE9;tudier la transduction du signal conduisant &#xE0; progression de la maladie en utilisant la g&#xE9;n&#xE9;tique inverse &#xE0; haut d&#xE9;bit niveaux. Il est important, nous utilisons des compos&#xE9;s de r&#xE9;f&#xE9;rence pour valider notre syst&#xE8;me &#xE0; l&#x26;#39;essai de mol&#xE9;cules qui emp&#xEA;chent la progression de la tuberculose, qui le rend tr&#xE8;s adapt&#xE9; pour enqu&#xEA;ter sur de nouveaux compos&#xE9;s anti-tuberculeux in vivo.

One-third of the world population is infected with Mycobacterium tuberculosis and multi-drug resistant strains are rapidly evolving. The noticeable absence of a whole organism high-throughput screening system for studying the progression of tuberculosis is fast becoming the bottleneck in tuberculosis research. We successfully developed such a system using the zebrafish Mycobacterium marinum infection model, which is a well-characterized model for tuberculosis progression with biomedical significance, mimicking hallmarks of human tuberculosis pathology. Importantly, we demonstrate the suitability of our system to directly study M. tuberculosis, showing for the first time that the human pathogen can propagate in this vertebrate model, resulting in similar early disease symptoms to those observed upon M. marinum infection. Our system is capable of screening for disease progression via robotic yolk injection of early embryos and visual flow screening of late-stage larvae. We also show that this system can reliably recapitulate the standard caudal vein injection method with a throughput level of 2,000 embryos per hour. We additionally demonstrate the possibility of studying signal transduction leading to disease progression using reverse genetics at high-throughput levels. Importantly, we use reference compounds to validate our system in the testing of molecules that prevent tuberculosis progression, making it highly suited for investigating novel anti-tuberculosis compounds in vivo.

Ein Drittel der Weltbev&ouml;lkerung mit Mycobacterium Tuberculosis infiziert und multiresistente St&auml;mme sind rasant entwickelt. Das sp&uuml;rbare Fehlen eines Gesamtorganismus High-Throughput-Screening-Systems f&uuml;r das Fortschreiten der Tuberkulose zu studieren ist schnell zu den Engpass in der Tuberkulose-Forschung. Wir entwickelten erfolgreich mit dem Zebrafisch Mycobacterium Marinum Infektion Modell, das gut charakterisierten Modell f&uuml;r Tuberkulose Progression mit biomedizinischen Bedeutung ist, ein solches System imitiert Kennzeichen der menschlichen Tuberkulose Pathologie. Wichtig ist, zeigen wir die Eignung unseres Systems direkt studieren M. Tuberkulose, zeigt zum ersten Mal, das die menschliche Krankheitserreger in diesem vertebrate Modell weitergeben kann was &auml;hnlich fr&uuml;hen Symptome bei M. Marinum Infektion beobachtet. Unser System kann Screening f&uuml;r das Fortschreiten der Krankheit mittels Roboter Eigelb Injektion von fr&uuml;hen Embryonen und visuellen Fluss Screening des Sp&auml;tstadiums Larven. Wir zeigen auch, dass dieses System zuverl&auml;ssig die Norm kaudalen Vene-Injektion-Methode mit einem Durchsatz von 2.000 Embryonen pro Stunde rekapitulieren kann. Wir zeigen au&szlig;erdem die M&ouml;glichkeit des Studiums Signaltransduktion zum Fortschreiten der Erkrankung mit Reverser Hochdurchsatz-Ebenen. Wichtig ist, verwenden wir Referenzverbindungen &uuml;berpr&uuml;ft unser System in das Testen von Molek&uuml;len, die Tuberkulose Fortschreiten, verhindern so dass es sehr gut geeignet f&uuml;r die Untersuchung neuartiger Tuberkulosebehandlung Verbindungen in-vivo.

Un terzo della popolazione mondiale &egrave; stato infettato da Mycobacterium tuberculosis e ceppi multifarmacoresistenti stanno rapidamente evolvendo. L&#39;evidente assenza di un sistema di high-throughput screening di tutto l&#39;organismo per studiare la progressione della tubercolosi sta rapidamente diventando il collo di bottiglia nella ricerca di tubercolosi. Abbiamo sviluppato con successo tale sistema utilizzando zebrafish Mycobacterium marinum infezione modello, che &egrave; un modello ben caratterizzato per la progressione di tubercolosi con rilevanza biomedica, mimando i tratti distintivi della patologia tubercolosi umana. Soprattutto, ci dimostrano l&#39;adeguatezza del nostro sistema direttamente lo studio di M. tuberculosis, mostrando per la prima volta che l&#39;agente patogeno umano pu&ograve; propagare in questo modello di vertebrati, con conseguente sintomi di malattia precoce simili a quelle osservate dopo infezione da M. marinum. Il nostro sistema &egrave; in grado di screening per la progressione della malattia mediante iniezione di tuorlo robotici degli embrioni di e lo screening di flusso visivo di larve di stadio avanzato. Mostriamo anche che questo sistema affidabile pu&ograve; riassumere il metodo di iniezione vena caudale standard con un livello di throughput di 2.000 embrioni all&#39;ora. Abbiamo inoltre dimostrare la possibilit&agrave; di studiare la trasduzione del segnale che porta alla progressione della malattia utilizzando genetica inversa a livelli di alto-rendimento. Soprattutto, usiamo composti di riferimento per convalidare il nostro sistema in sperimentazione delle molecole che impediscono la progressione di tubercolosi, che lo rende estremamente adatto per indagare nuovi composti anti-tubercolosi in vivo.

Un tercio de la poblaci&#xF3;n mundial est&#xE1; infectada con Mycobacterium tuberculosis y m&#xFA;ltiples drogas cepas resistentes est&#xE1;n evolucionando r&#xE1;pidamente. La notable ausencia de un sistema de detecci&#xF3;n organismo completo de alto rendimiento para el estudio de la progresi&#xF3;n de la tuberculosis se est&#xE1; convirtiendo en el cuello de botella en la investigaci&#xF3;n de la tuberculosis. Hemos desarrollado con &#xE9;xito un sistema que utiliza el pez cebra Mycobacterium marinum modelo de infecci&#xF3;n, que es un modelo bien caracterizado para la progresi&#xF3;n de la tuberculosis con la importancia biom&#xE9;dica, imitando caracter&#xED;sticas de la patolog&#xED;a humana la tuberculosis. Es importante destacar que demostrar la idoneidad de nuestro sistema para estudiar directamente de M. tuberculosis, mostrando por primera vez que el pat&#xF3;geno humano puede propagar en este modelo de vertebrados, lo que resulta en s&#xED;ntomas iniciales de la enfermedad similares a los observados en la infecci&#xF3;n por M. marinum. Nuestro sistema es capaz de la detecci&#xF3;n de progresi&#xF3;n de la enfermedad a trav&#xE9;s de inyecci&#xF3;n de yema de rob&#xF3;tica de embriones y la detecci&#xF3;n de flujo visual de la &#xFA;ltima etapa de las larvas. Tambi&#xE9;n se muestra que este sistema de forma fiable puede recapitular el m&#xE9;todo est&#xE1;ndar de inyecci&#xF3;n en la vena caudal, con un nivel de rendimiento de 2.000 embriones por hora. Estamos, adem&#xE1;s, demostrar la posibilidad de estudiar la transducci&#xF3;n de se&#xF1;ales que conduce a la progresi&#xF3;n de la enfermedad mediante gen&#xE9;tica inversa en los niveles de alto rendimiento. Es importante destacar que utilizar compuestos de referencia para validar nuestro sistema en el ensayo de mol&#xE9;culas que impiden la progresi&#xF3;n de la tuberculosis, por lo que es muy adecuado para la investigaci&#xF3;n de nuevos compuestos antituberculosos in vivo.

Una pantalla de alto rendimiento para la progresi&#xF3;n de la tuberculosis

Staphylococcus epidermidis bacteria are a major cause of biomaterial-associated infections in modern medicine. Yet there is little known about the host responses against this normally innocent bacterium in the context of infection of biomaterials. In order to better understand the factors involved in this process, a whole animal model with high throughput screening possibilities and markers for studying the host response to S. epidermidis infection are required.

A zebrafish high throughput screening system used for Staphylococcus epidermidis infection marker discovery.

The increasing use of zebrafish larvae for biomedical research applications is resulting in versatile models for a variety of human diseases. These models exploit the optical transparency of zebrafish larvae and the availability of a large genetic tool box. Here we present detailed protocols for the robotic injection of zebrafish embryos at very high accuracy with a speed of up to 2000 embryos per hour. These protocols are benchmarked for several applications: (1) the injection of DNA for obtaining transgenic animals, (2) the injection of antisense morpholinos that can be used for gene knock-down, (3) the injection of microbes for studying infectious disease, and (4) the injection of human cancer cells as a model for tumor progression. We show examples of how the injected embryos can be screened at high-throughput level using fluorescence analysis. Our methods open up new avenues for the use of zebrafish larvae for large compound screens in the search for new medicines.

Robotic injection of zebrafish embryos for high-throughput screening in disease models.

Zebrafish model systems for infectious disease are increasingly used for the functional analysis of molecular pattern recognition processes. These studies benefit from the high conservation level of all innate immune factors in vertebrates. Zebrafish studies are strategically well positioned for this because of the ease of comparisons with studies in other fish species of which the immune system also has been intensively studied, but that are currently still less amendable to detailed genetic or microscopic studies. In this paper we focus on Toll-like receptor (TLR) signalling factors, which currently are the best characterized in mammalian systems. We review the knowledge on TLR signalling in the context of recent advances in zebrafish studies and discuss possibilities for future approaches that can complement studies in cell cultures and rodent models. A focus in these comparisons is the role of negative control mechanisms in immune responses that appear very important in a whole organism to keep adverse systemic responses in check. We also pay much attention to comparisons with studies in common carp that is highly related to zebrafish and that because of its large body mass can complement immune studies in zebrafish.
