The success of plant genetic transformation relies greatly on the strength and specificity of the promoters used to drive genes of interest. In this study, we analyzed gfp gene expression mediated by a polyubiquitin promoter (Gmubi) from soybean (Glycine max) in stably transformed soybean tissues. Strong GFP expression was observed in stably transformed proliferative embryogenic tissues. In whole transgenic plants, GFP expression was observed in root tips, main and lateral roots, cotyledons and plumules in young plants as well as in leaf veins, petioles, flower petals, pollen, pods and developing seeds in mature plants. GFP expression was localized mainly in epidermal cells, leaf mesophyll, procambium and vascular tissues. Introduction of an intron-less version of the Gmubi promoter (Gmupri) displayed almost the same GFP expression pattern albeit at lower intensities. The Gmubi promoter showed high levels of constitutive expression and represents an alternative to viral promoters for driving gene expression in soybean.

Le succ&#xE8;s de la transformation g&#xE9;n&#xE9;tique des plantes repose grandement sur la force et la sp&#xE9;cificit&#xE9; des promoteurs utilis&#xE9;s pour conduire des g&#xE8;nes d&#x26;#39;int&#xE9;r&#xEA;t. Dans cette &#xE9;tude, nous avons analys&#xE9; l&#x26;#39;expression du g&#xE8;ne GFP m&#xE9;di&#xE9;e par un promoteur polyubiquitine (Gmubi) &#xE0; partir de soja (Glycine max) dans stablement tissus de soja transform&#xE9;es. Forte expression de la GFP a &#xE9;t&#xE9; observ&#xE9;e dans transform&#xE9;es de mani&#xE8;re stable prolif&#xE9;ratives tissus embryog&#xE8;nes. Dans l&#x26;#39;ensemble des plantes transg&#xE9;niques, expression de la GFP a &#xE9;t&#xE9; observ&#xE9;e dans les pointes, les racines principales et lat&#xE9;rales, les cotyl&#xE9;dons et plumules des jeunes plantes ainsi que dans les nervures des feuilles, p&#xE9;tioles, p&#xE9;tales de fleurs, le pollen, les gousses et graines dans d&#xE9;veloppement des plantes matures. Expression de la GFP a &#xE9;t&#xE9; localis&#xE9; principalement dans les cellules &#xE9;pidermiques, m&#xE9;sophylle foliaire, procambium et des tissus vasculaires. Pr&#xE9;sentation d&#x26;#39;une version intron-moins du promoteur Gmubi (Gmupri) affich&#xE9; presque le m&#xEA;me sch&#xE9;ma d&#x26;#39;expression de GFP mais &#xE0; des intensit&#xE9;s plus faibles. Le promoteur Gmubi montr&#xE9; des niveaux &#xE9;lev&#xE9;s d&#x26;#39;expression constitutive et repr&#xE9;sente une alternative &#xE0; des promoteurs viraux pour une expression de g&#xE8;ne dans le soja.

Der Erfolg der Pflanze genetische Transformation st&uuml;tzt sich stark auf die St&auml;rke und Besonderheit der Promotoren verwendet, um mit dem Gene. In dieser Studie analysierten wir Gfp-gen-Expression von Polyubiquitin F&ouml;rderer (Gmubi) vermittelt aus Sojabohnen (Glycine max) im Gewebe stabil transformierten Sojabohnen. Strong GFP-Expression wurde in stabil transformierten proliferative embryogenic Gewebe beobachtet. In ganzen transgener Pflanzen wurde GFP-Expression in Wurzel-Tipps, Haupt- und seitlichen Wurzeln Keimbl&auml;tter und Plumules in jungen Pflanzen ebenso wie in Blatt-Adern, Blattstielen, Bl&uuml;tenbl&auml;tter, Pollen, H&uuml;lsen und entwickelnden Samen in den Reifen Pflanzen beobachtet. GFP-Expression wurde haupts&auml;chlich in Epidermiszellen, Blatt Mesophyll, Procambium und vaskul&auml;re Geweben lokalisiert. Einf&uuml;hrung einer Intron-lose Version des Projekttr&auml;gers Gmubi (Gmupri) angezeigten fast das gleiche GFP Ausdrucksmuster allerdings zu niedrigeren Intensit&auml;ten. Der Projekttr&auml;ger Gmubi zeigte hohe konstitutive Ausdruck und stellt eine Alternative virale Promotoren f&uuml;r den Antrieb von Genexpression in Soja.

Il successo della trasformazione genetica vegetale si basa fortemente sulla forza e la specificit&agrave; dei promotori utilizzato per geni di auto di interesse. In questo studio, abbiamo analizzato l&#39;espressione di gene gfp mediata da un promotore di polyubiquitin (Gmubi) da soia (Glycine max) nei tessuti di soia stabilmente trasformato. Espressione forte GFP &egrave; stato osservato nei tessuti embryogenic proliferativi stabilmente trasformati. In piante transgeniche intero, espressione di GFP &egrave; stato osservato nei consigli di radice, radice principale e laterale, cotiledoni e plumules nelle piante giovani, cos&igrave; come nelle vene della foglia, piccioli, petali di fiori, polline, baccelli e semi in via di sviluppo in piante mature. Espressione di GFP &egrave; stato localizzato principalmente nelle cellule epidermiche, mesofillo della foglia, procambium e tessuti vascolari. Introduzione di una versione meno introne del promotore del Gmubi (Gmupri) visualizzato quasi lo stesso schema di espressione GFP seppur a bassa intensit&agrave;. Il promotore di Gmubi ha mostrato elevati livelli di espressione costitutiva e rappresenta un&#39;alternativa ai promotori virali per la guida di espressione genica in soia.

El &#xE9;xito de la transformaci&#xF3;n gen&#xE9;tica de las plantas depende en gran medida de la fuerza y &#x200B;&#x200B;la especificidad de los promotores utilizados para conducir los genes de inter&#xE9;s. En este estudio, se analizaron las buenas pr&#xE1;cticas agrarias expresi&#xF3;n g&#xE9;nica mediada por un promotor poliubiquitina (Gmubi) de soja (Glycine max) en los tejidos de soja transformadas establemente. Fuerte expresi&#xF3;n de GFP se observ&#xF3; en transformadas de forma estable los tejidos embriog&#xE9;nicos proliferantes. En conjunto las plantas transg&#xE9;nicas, expresi&#xF3;n de la GFP se observ&#xF3; en puntas de las ra&#xED;ces, las ra&#xED;ces principales y laterales, los cotiledones y pl&#xFA;mulas en las plantas j&#xF3;venes, as&#xED; como en las venas de las hojas, pec&#xED;olos, p&#xE9;talos de flores, el polen, las vainas y el desarrollo de semillas en las plantas maduras. La expresi&#xF3;n de GFP fue localizada principalmente en las c&#xE9;lulas de la epidermis, mes&#xF3;filo de la hoja, procambium y tejidos vasculares. La introducci&#xF3;n de una versi&#xF3;n menos intr&#xF3;n del promotor Gmubi (Gmupri) que se muestran casi el mismo patr&#xF3;n de expresi&#xF3;n de GFP, aunque a menor intensidad. Although numerous factors can influence gene expression, promoters are perhaps the most important component of the regulatory control process. Promoter regions are often defined as a region upstream of the transcriptional start. They contain regulatory elements that interact with regulatory proteins to modulate gene expression. Most genes possess their own unique promoter and large numbers of promoters are therefore available for study. Unfortunately, relatively few promoters have been isolated and characterized; particularly from soybean (Glycine max).
