&#x6838;&#x5B54;&#x590D;&#x5408;&#x76F8;&#x5173;&#x86CB;&#x767D;&#xFF0C;Mlp2p&#xFF0C;&#x5C06;&#x7ED1;&#x5B9A;&#x5230;&#x9175;&#x6BCD;&#x4E3B;&#x8F74;&#x6746;&#x673A;&#x6784;&#xFF0C;&#x5E76;&#x4FC3;&#x8FDB;&#x5176;&#x6709;&#x6548;&#x7684;&#x7A0B;&#x5E8F;&#x96C6;&#x3002;

The two yeast proteins Mlp1p and Mlp2p (homologues of the vertebrate protein Tpr) are filamentous proteins attached to the nuclear face of nuclear pore complexes. Here we perform a proteomic analysis, which reveals that the two Mlps have strikingly different interacting partners, testifying to their different roles within the cell. We find that Mlp2p binds directly to Spc110p, Spc42p, and Spc29p, which are three core components of the spindle pole body (SPB), the nuclear envelope-associated yeast spindle organizer. We further show that SPB function is compromised in mlp2 mutants. Cells lacking Mlp2p form significantly smaller SPBs, accumulate aberrant SPB component-containing structures inside the nucleus, and have stochastic failures of cell division. In addition, depletion of Mlp2p is synthetically lethal with mutants impaired in SPB assembly. Based on these data, we propose that Mlp2p links the SPB to the peripheral Mlp assembly, and that this linkage is required for efficient incorporation of components into the SPB.

Les deux prot&#xE9;ines de levure et Mlp1p Mlp2p (homologues de l&#x26;#39;Cvr prot&#xE9;ine vert&#xE9;br&#xE9;) sont des prot&#xE9;ines filamenteux attach&#xE9;s &#xE0; la face nucl&#xE9;aire de complexes de pores nucl&#xE9;aires. Ici nous effectuons une analyse prot&#xE9;omique, qui r&#xE9;v&#xE8;le que les deux Mlps ont radicalement diff&#xE9;rentes partenaires d&#x26;#39;interaction, t&#xE9;moignant de leurs diff&#xE9;rents r&#xF4;les au sein de la cellule. Qui effettuiamo un&#39;analisi proteomic, che rivela che i due Mlps hanno sorprendentemente diversi partners interagenti, che testimoniano la loro diversi ruoli all&#39;interno della cellula. La levadura dos prote&#xED;nas Mlp1p y Mlp2p (hom&#xF3;logos de la prote&#xED;na Tpr vertebrado) son prote&#xED;nas filamentosas unidos a la cara nuclear de los complejos de poros nucleares. Aqu&#xED; se realiza un an&#xE1;lisis prote&#xF3;mico, que revela que los dos tienen Mlps socios que interact&#xFA;an sorprendentemente diferentes, lo que demuestra sus diferentes roles dentro de la c&#xE9;lula. Nos parece que se une directamente a Mlp2p Spc110p, Spc42p y Spc29p, que son tres componentes b&#xE1;sicos del cuerpo polo del huso (SPB), la envoltura nuclear asociada a la organizadora del husillo de levadura. Adem&#xE1;s, muestran que la funci&#xF3;n de SPB se ve comprometida en mlp2 mutantes. Las c&#xE9;lulas que carecen de forma significativamente m&#xE1;s peque&#xF1;os Mlp2p SPB, se acumulan aberrantes SPB contienen componentes de estructuras en el interior del n&#xFA;cleo, y tienen fallas estoc&#xE1;sticas de la divisi&#xF3;n celular. Adem&#xE1;s, el agotamiento de Mlp2p es sint&#xE9;tica letal con mutantes con deficiencias en el montaje SPB. Bas&#xE1;ndose en estos datos, se propone que los v&#xED;nculos Mlp2p el SPB a la asamblea Mlp perif&#xE9;rica, y que esta vinculaci&#xF3;n se requiere para la incorporaci&#xF3;n eficiente de los componentes en el SPB.

Protein phosphorylation is estimated to affect 30% of the proteome and is a major regulatory mechanism that controls many basic cellular processes. Until recently, our biochemical understanding of protein phosphorylation on a global scale has been extremely limited; only one half of the yeast kinases have known in vivo substrates and the phosphorylating kinase is known for less than 160 phosphoproteins. Here we describe, with the use of proteome chip technology, the in vitro substrates recognized by most yeast protein kinases: we identified over 4,000 phosphorylation events involving 1,325 different proteins. These substrates represent a broad spectrum of different biochemical functions and cellular roles. Distinct sets of substrates were recognized by each protein kinase, including closely related kinases of the protein kinase A family and four cyclin-dependent kinases that vary only in their cyclin subunits. Although many substrates reside in the same cellular compartment or belong to the same functional category as their phosphorylating kinase, many others do not, indicating possible new roles for several kinases. Furthermore, integration of the phosphorylation results with protein-protein interaction and transcription factor binding data revealed novel regulatory modules. Our phosphorylation results have been assembled into a first-generation phosphorylation map for yeast. Because many yeast proteins and pathways are conserved, these results will provide insights into the mechanisms and roles of protein phosphorylation in many eukaryotes.

La fosforilaci&#xF3;n de prote&#xED;nas se calcula que afecta a un 30% del proteoma y es un importante mecanismo de regulaci&#xF3;n que controla muchos procesos celulares b&#xE1;sicos. Hasta hace poco tiempo, nuestra comprensi&#xF3;n bioqu&#xED;mica de la fosforilaci&#xF3;n de prote&#xED;nas en una escala global ha sido muy limitado: s&#xF3;lo la mitad de las quinasas de levadura han conocido en sustratos in vivo y la fosforilaci&#xF3;n de la quinasa es conocida por menos de 160 fosfoprote&#xED;nas. They are typically prepared by overexpression and high-throughput purification and printing onto microscope slides. The arrays can be used to screen nearly the entire proteome in an unbiased fashion and have enormous utility for a variety of applications. These include protein-protein interactions, identification of novel lipid- and nucleic acid-binding proteins, and finding targets of small molecules, protein kinases, and other modification enzymes. Protein microarrays are thus powerful tools for individual studies as well as systematic characterization of proteins and their biochemical activities and regulation.

Protein kinases are key regulators of cellular processes. In spite of considerable effort, a full understanding of the pathways they participate in remains elusive. We globally investigated the proteins that interact with the majority of yeast protein kinases using protein microarrays. Eighty-five kinases were purified and used to probe yeast proteome microarrays. One-thousand-twenty-three interactions were identified, and the vast majority were novel. Coimmunoprecipitation experiments indicate that many of these interactions occurred in vivo. Many novel links of kinases to previously distinct cellular pathways were discovered. For example, the well-studied Kss1 filamentous pathway was found to bind components of diverse cellular pathways, such as those of the stress response pathway and the Ccr4-Not transcriptional/translational regulatory complex; genetic tests revealed that these different components operate in the filamentation pathway in vivo. Overall, our results indicate that kinases operate in a highly interconnected network that coordinates many activities of the proteome. Our results further demonstrate that protein microarrays uncover a diverse set of interactions not observed previously.

Prot&eacute;ines kinases sont des r&eacute;gulateurs cl&eacute;s de processus cellulaires. En d&eacute;pit des efforts consid&eacute;rables, une pleine compr&eacute;hension des voies qu&#39;ils participent demeure insaisissable. Globalement, nous avons &eacute;tudi&eacute; les prot&eacute;ines qui interagissent avec la majorit&eacute; des kinases de prot&eacute;ine de levure &agrave; l&#39;aide de puces &agrave; prot&eacute;ines. Quatre-vingt-cinq kinases ont purifi&eacute; et utilis&eacute; pour sonder la levure proteome microarrays. Les interactions de mille-vingt-trois ont &eacute;t&eacute; identifi&eacute;es, et la grande majorit&eacute; &eacute;taient roman. Co-immunopr&eacute;cipitation exp&eacute;riences indiquent que plusieurs de ces interactions est produite in vivo.
