In order to clarify the biochemical basis to the divergence of sex pheromones in the genus Ostrinia (Lepidoptera: Crambidae), the pheromone biosynthetic pathway in O. zaguliaevi, a close relative of the European corn borer O. nubilalis, was investigated. Deuterium-labeled hexadecanoic or tetradecanoic acids were topically applied to the surface of the pheromone gland, and the incorporation of the label into pheromone components and their putative precursors was determined. It was suggested that the two components shared by O. zaguliaevi and O. nubilalis, (E)-11- and (Z)-11-tetradecenyl acetates, are biosynthesized from hexadecanoic acid through one round of chain shortening, Delta11 desaturation, reduction, and acetylation. An additional component specifically found in O. zaguliaevi, (Z)-9-tetradecenyl acetate, is likely to be produced by delta11 desaturation of hexadecanoic acid, one round of chain shortening, reduction, and acetylation. Non-production of (Z)-9-tetradecenyl acetate in O. nubilalis was suggested to be due to the blockage of chain shortening from (Z)-11-hexadecenoate to (Z)-9-tetradecenoate.

Female sex pheromones are considered to be produced in a "pheromone gland" located in the terminal abdominal segments (8th-10th, TAS) of a moth; however, in many moth species, the cells that produce pheromones have not actually been specified. We investigated cells in the TAS that synthesize pheromones in the adzuki bean borer Ostrinia scapulalis, by locating pheromones and their precursors, and mRNA for Delta11-desaturase, a key enzyme in pheromone biosynthesis. We demonstrated that the pheromone components, (E)-11- and (Z)-11-tetradecenyl acetates, and their fatty acyl precursors were specifically contained in the dorsal part of the TAS. A cDNA (OscaZ/E11) that encodes a Delta11-desaturase was cloned from the TAS. RT-PCR and in situ hybridization unequivocally showed that OscaZ/E11 is specifically expressed in the modified epidermal cells located at the dorsal end of the 8th-9th intersegmental membrane.

In order to prepare pure and well-defined oligosaccharides from agarose in a rapid and simple manner, an enzymatic degradation method was developed, which includes degradation with either recombinant beta-agarase (EC 3.2.1.81) AgaA or AgaB and gel permeation chromatography. Agarose was degraded with AgaA at the optimized conditions, yielding 47% and 45% of neoagarotetraose and neoagarohexaose, respectively. These neoagaro-oligosaccharides were conveniently separated by consecutive column chromatography on Bio-Gel P2 or P6 and were identified by FACE. The structure of these neoagaro-oligosaccharides was confirmed by MALDI-TOF MS and (13)C NMR spectroscopy.

A simple method of preparing diverse neoagaro-oligosaccharides with beta-agarase.

Afin de pr&eacute;parer des oligosaccharides pures et bien d&eacute;finies de l&#39;agarose de mani&egrave;re rapide et simple, une m&eacute;thode de d&eacute;gradation enzymatique a &eacute;t&eacute; d&eacute;velopp&eacute;e, qui comprend la d&eacute;gradation avec soit chromatographie &agrave; perm&eacute;ation AgaA ou AgaB et gel recombinant beta-l&#39;agarase (EC 3.2.1.81). Um rein und klar definierte Oligosaccharide aus Agarose in eine schnelle und einfache Weise vorzubereiten, wurde eine enzymatische Abbau-Methode entwickelt die Verschlechterung mit entweder rekombinantes Beta-Agarase (EG 3.2.1.81)-AgaA oder AgaB und Gel-Permeations-Chromatografie. Among mice, pheromones and other social odor cues convey information about sex, social status, and identity; however, the molecular nature of these cues is essentially unknown. To identify these cues, we screened chromatographic fractions of female mouse urine for their ability to cause reproducible firing rate increases in the pheromone-detecting vomeronasal sensory neurons (VSNs) using multielectrode array (MEA) recording. Active compounds were found to be remarkably homogenous in their basic properties, with most being of low molecular weight, moderate hydrophobicity, low volatility, and possessing a negative electric charge. Purification and structural analysis of active compounds revealed multiple sulfated steroids, of which two were identified as sulfated glucocorticoids, including corticosterone 21-sulfate. Sulfatase-treated urine extracts lost >80% of their activity, indicating that sulfated compounds are the predominant VSN ligands in female mouse urine. As measured by MEA recording, a collection of 31 synthetic sulfated steroids triggered responses 30-fold more frequently than did a similarly sized stimulus set containing the majority of all previously reported VSN ligands. Collectively, VSNs detected all major classes of sulfated steroids, but individual neurons were sensitive to small variations in chemical structure. VSNs from both males and females detected sulfated steroids, but knock-outs for the sensory transduction channel TRPC2 did not detect these compounds. Urine concentrations of the two sulfated glucocorticoids increased many fold in stressed animals, indicating that information about physiological status is encoded by the urine concentration of particular sulfated steroids. To increase the thermostability of beta-agarase AgaB by directed evolution, the mutant gene libraries were generated by error-prone polymerase chain reaction (PCR) and deoxyribonucleic acid (DNA) shuffling. Mutants with high thermostability were screened by a simple method based on agarase-degrading agar to generate a clear zone on the agar plate. A mutant S2 was obtained through two rounds of error-prone PCR and a single round of DNA shuffling and selection. It has higher thermostability and slightly increased catalytic activity than wild-type AgaB. Melting temperature (T(m)) of S2, as determined by circular dichroism, is 4.6 degrees C higher than that of wild-type AgaB, and the half-life of S2 is 350 min at 40 degrees C, which is 18.4-fold longer than that of the wild-type enzyme. Saturation mutagenesis and hydrophobic cluster analysis indicated that hydrophobic interaction might be the key factor that enhances the enzyme stability.

Para aumentar la termoestabilidad de beta-agarase AgaB por evoluci&oacute;n dirigida, las bibliotecas del gen mutante fueron generadas por la reacci&oacute;n en cadena de errores de la polimerasa (PCR) y el &aacute;cido desoxirribonucleico (ADN) arrastrando los pies. In many species, social behavior is organized via chemical signaling. While many of these signals have been identified for insects, the chemical identity of these social cues (often called pheromones) for mammals is largely unknown. We recently isolated these chemical cues that caused firing in the pheromone-sensing neurons of the vomeronasal organ from female mouse urine [Nodari, F., et al. (2008) J. Neurosci. 28, 6407-6418]. Here, we report their structural characterization. Mass spectrometric approaches, including tandem quadrupole, multiple-stage linear ion trap, high-resolution mass spectrometry, and H-D exchange followed by ESI mass spectrometry, along with (1)H and (13)C nuclear magnetic resonance spectroscopy, including two-dimensional correlation spectroscopy, total correlation spectroscopy, heteronuclear multiple-quantum coherence, and NOE, were used to identify two sulfated steroids, 4-pregnene-11beta,20,21-triol-3-one 21-sulfate (I) (the configuration at C20 was not deduced) and 4-pregnene-11beta,21-diol-3,20-dione 21-sulfate (II), whose presence is sex-specific. The identification of this novel class of mammalian social signaling compounds suggests that steroid hormones, upon conjugation, assume a new biological role, conveying information about the organism's identity and physiological state.

To obtain a high level expression of phytase with favorable characteristics, a codon-optimized phytase gene from Citrobacter freundii was synthesized and transferred into Pichia pastoris. Small-scale expression experiments and activity assays were used to screen positive colonies. After purified by NiÂ²+-NTA agarose affinity column, the characterizations of the recombinant phytase were determined. The recombinant phytase (r-phyC) had two distinct pH optima at 2.5 and 4.5 and an optimal temperature at 50 Â°C. It retained more than 80% activity after being incubated under various buffer (pH 1.5-8.0) at 37 Â°C for 1 h. The specific activity, Km, and Vmax values of r-phyC for sodium phytate were 2,072â€‰Â±â€‰18 U mgâ»Â¹, 0.52â€‰Â±â€‰0.04 mM, and 2,380â€‰Â±â€‰84 U mgâ»Â¹ minâ»Â¹, respectively. Time-resolved luminescence detection technique using lanthanide chelates as luminescent probes or sensors is a highly sensitive and widely used tool for the luminescence detections of various biological and bioactive molecules. The essential of this technique is the developments of various functional luminescent probes or sensors that can selectively recognize the biological targets. In this work, a dual-chelating ligand N,N,N(1),N(1)-{2,6-bis(3'-aminomethyl-1'-pyrazolyl)-4-[N,N-bis(2-picolyl)amino-methylenepyridine]} tetrakis(acetic acid) (BBATA) has been designed and synthesized. The luminescence of its Tb(3+) chelate is very weak, but can be selectively and strongly enhanced upon reaction with Zn(2+) ions. Thus a Tb(3+) chelate-based luminescent chemosensor, BBATA-Tb(3+), for highly selective and sensitive time-resolved luminescence detection of Zn(2+) ions was developed. To confirm the utility of new chemosensor for the detection of intracellular Zn(2+) ions, the performance of BBATA-Tb(3+) as a chemosensor for time-resolved luminescent imaging detection of Zn(2+) ions in living cells was investigated. The results demonstrated the efficacy and advantage of the new luminescent chemosensor for time-resolved luminescence detection of intracellular Zn(2+) ions.

In dieser Arbeit, ein Dual-Chelat-Ligand N, N, N (1), N (1)-{2,6-Bis (3&#39;-Aminomethyl-1&#39;-Pyrazolyl)-4-[N, N-bis(2-picolyl) amino-Methylenepyridine]} Tetrakis(acetic acid) (BBATA) entwickelt und synthetisiert wurde. Die Resultate zeigten die Wirksamkeit und Nutzen der neuen leuchtenden Chemorezeptor f&uuml;r Zeit aufgel&ouml;st Lumineszenz Nachweis der intrazellul&auml;ren Zn(2+) Ionen.

Tecnica di rilevazione Time-resolved luminescenza con chelati lantanidi come sonde luminescenti o sensori &egrave; uno strumento altamente sensibile e ampiamente usato per i rilevamenti di luminescenza di varie molecole biologiche e bioattivi. L&#39;essenziale di questa tecnica &egrave; gli sviluppi di varie sonde luminescenti funzionale o sensori in grado di riconoscere selettivamente i bersagli biologici. In questo lavoro, un ligando dual-chelanti N, N, N (1), N (1)-{2,6-bis (3 - amminometil - 1-pyrazolyl) -4-[N, N-bis(2-picolyl) ammino-methylenepyridine]} tetrakis(acetic acid) (BBATA) &egrave; stato progettato e sintetizzato. La luminescenza dei suoi Tb(3+) chelato &egrave; molto debole, ma pu&ograve; essere selettivamente e fortemente migliorata su reazione con gli ioni Zn(2+). Lo esencial de esta t&eacute;cnica es la evoluci&oacute;n de varias sondas luminiscentes funcionales o sensores que pueden reconocer selectivamente los objetivos biol&oacute;gicos. En este trabajo, un ligando quelantes del doble N, N, N (1), N (1)-{2, 6-bis (3&#39;-aminomethyl-1&#39;-compuestos) -4-[N, N-bis(2-picolyl) amino-methylenepyridine]} tetrakis(acetic acid) (BBATA) ha sido dise&ntilde;ado y sintetizado. La luminiscencia de su quelato de Tb(3+) es muy d&eacute;bil, pero puede mejorarse selectivamente y fuertemente a la reacci&oacute;n con los iones del Zn(2+). As&iacute; se desarroll&oacute; una Tb(3+) base de quelato luminiscente chemosensor, BBATA-Tb(3+), para la detecci&oacute;n de tiempo resuelto altamente selectivo y sensible de la luminiscencia de iones Zn(2+). Para confirmar la utilidad de nuevo chemosensor para la detecci&oacute;n de iones intracelulares de Zn(2+), se investig&oacute; el desempe&ntilde;o de BBATA-Tb(3+) como un chemosensor para la detecci&oacute;n de imagen luminiscente resuelta en el tiempo de Zn(2+) iones en las c&eacute;lulas vivas. The gene encoding a cold-active and xylose-stimulated Î²-glucosidase of Marinomonas MWYL1 was synthesized and expressed in Escherichia coli. The recombinant enzyme (reBglM1) was purified and characterized. The molecular mass of the purified reBglM1 determined by SDS-PAGE agree with the calculated values (50.6Â Da). Optima of temperature and pH for enzyme activity were 40Â°C and 7.0, respectively. The enzyme exhibited about 20% activity at 5Â°C and was stable over the range of pH 5.5-10.0. The presence of xylose significantly enhanced enzyme activity even at higher concentrations up to 600Â mM, with maximal stimulatory effect (about 1.45-fold) around 300Â mM. The enzyme is active with both glucosides and galactosides and showed high catalytic efficiency (k (cat)Â =Â 500.5Â s(-1)) for oNPGlc. These characterizations enable the enzyme to be a promising candidate for industrial applications.

Gen f&uuml;r eine K&auml;lte-aktiven und Xylose-stimuliert &beta;-Glucosidase von Marinomonas MWYL1 wurde synthetisiert und ausgedr&uuml;ckt in Escherichia coli. Das rekombinante Enzym (reBglM1) wurde gereinigt und charakterisiert. Die molare Masse von den gereinigten reBglM1 bestimmt durch SDS-PAGE Stimmen die berechneten Werte (50,6 Da). Optima von Temperatur und pH-Wert f&uuml;r Enzymaktivit&auml;t betrugen 40 &deg; C und 7.0. Das Enzym stellte etwa 20 % Aktivit&auml;t bei 5 &deg; C und &uuml;ber den Bereich von pH-Wert 5,5-10,0 stabil war. Das Vorhandensein von Xylose erheblich verbessert worden Enzym-Aktivit&auml;t auch bei h&ouml;heren Konzentrationen bis zu 600 mM, mit Maximal stimulierende Wirkung (etwa 1.45-fold) etwa 300 mM. Das Enzym ist mit Glukoside und Galactosides aktiv und zeigte hohen katalytischen Effizienz (k (Kat) = 500.5 s(-1)) f&uuml;r oNPGlc. Il gene che codifica per un freddo-attivo e stimolato xilosio &beta;-glucosidasi di Marinomonas MWYL1 &egrave; stato sintetizzato ed espresso in Escherichia coli. L&#39;enzima ricombinante (reBglM1) &egrave; stata purificata e caratterizzata. La massa molecolare del reBglM1 purificato determinato da SDS-PAGE sono d&#39;accordo con i valori calcolati (Da 50,6). Optimum di temperatura e pH per l&#39;attivit&agrave; dell&#39;enzima erano 40 &deg; C e 7.0, rispettivamente. L&#39;enzima esposto circa il 20% attivit&agrave; a 5 &deg; C ed &egrave; stabile nell&#39;intervallo di pH 5.5-10.0.
