Little is known about the formation of niches, local micro-environments required for stem-cell maintenance. Here we develop an in vivo assay for adult haematopoietic stem-cell (HSC) niche formation. With this assay, we identified a population of progenitor cells with surface markers CD45(-)Tie2(-)alpha(V)(+)CD105(+)Thy1.1(-) (CD105(+)Thy1(-)) that, when sorted from 15.5 days post-coitum fetal bones and transplanted under the adult mouse kidney capsule, could recruit host-derived blood vessels, produce donor-derived ectopic bones through a cartilage intermediate and generate a marrow cavity populated by host-derived long-term reconstituting HSC (LT-HSC). In contrast, CD45(-)Tie2(-)alpha(V)(+)CD105(+)Thy1(+) (CD105(+)Thy1(+)) fetal bone progenitors form bone that does not contain a marrow cavity. Suppressing expression of factors involved in endochondral ossification, such as osterix and vascular endothelial growth factor (VEGF), inhibited niche generation. CD105(+)Thy1(-) progenitor populations derived from regions of the fetal mandible or calvaria that do not undergo endochondral ossification formed only bone without marrow in our assay. Collectively, our data implicate endochondral ossification, bone formation that proceeds through a cartilage intermediate, as a requirement for adult HSC niche formation.

En revanche, CD45(-)Tie2(-)alpha(V)(+)CD105(+)Thy1(+) (CD105(+)Thy1(+)) OS foetal prog&eacute;niteurs forme osseuse qui ne contient-elle pas une cavit&eacute; de la moelle osseuse. Suppression de l&#39;expression des facteurs impliqu&eacute;s dans l&#39;ossification endochondrale, tels qu&#39;osterix et facteur de croissance endoth&eacute;lial vasculaire (VEGF), inhibe la g&eacute;n&eacute;ration de niche. CD105(+)Thy1(-) les populations de souches provenant de r&eacute;gions de la mandibule foetale ou la calotte cranienne qui ne subit pas l&#39;ossification endochondrale forment uniquement d&#39;OS sans moelle dans notre test. Collectivement, nos donn&eacute;es indiquent que l&#39;ossification endochondrale, la formation osseuse qui proc&egrave;de par un interm&eacute;diaire de cartilage, comme une exigence pour adulte formation de niche HSC.

&Uuml;ber die Bildung von Nischen, lokalen Mikro-Umgebungen f&uuml;r Stammzell Wartung erforderlich ist wenig bekannt. Im Gegensatz dazu CD45(-)Tie2(-)alpha(V)(+)CD105(+)Thy1(+) (CD105(+)Thy1(+)) fetalen Knochen Vorl&auml;uferzellen Form Knochen, die keinen Knochenmark-Hohlraum enth&auml;lt. Unterdr&uuml;cken von Ausdruck der Faktoren, die in Endochondral Verkn&ouml;cherung, wie Osterix und vaskul&auml;ren endothelialen Wachstumsfaktor (VEGF), gehemmt Nischen Erzeugung. CD105(+)Thy1(-) Stammvater Populationen abgeleitet aus den Regionen des fetalen Unterkiefer oder der Sch&auml;delknochen, die nicht Endochondral Verkn&ouml;cherung erfolgt nur Knochen ohne Knochenmark in unserem Assay gebildet. Kollektiv, implizieren unsere Daten Endochondral Verkn&ouml;cherung, Knochenneubildung, das durch eine Knorpel-Mittelstufe, als Voraussetzung f&uuml;r Erwachsene HSC Nische Bildung geht.

Poco si sa circa la formazione di nicchie, micro-ambienti locali necessari per la manutenzione delle cellule staminali. Qui possiamo sviluppare un test in vivo per la formazione di nicchia (HSC) di cellule staminali emopoietiche adulte. Sopprimere l&#39;espressione di fattori coinvolti nella ossificazione endocondrale, quali osterix e fattore di crescita endoteliale vascolare (VEGF), ha inibito la generazione di nicchia. Popolazioni progenitore CD105(+)Thy1(-) derivate dalle regioni della mandibola fetale o calvaria che non subiscono ossificazione endocondrale formano solo osso senza midollo nel nostro saggio. Collettivamente, i nostri dati coinvolgono ossificazione endocondrale, formazione ossea che procede attraverso un intermedio di cartilagine, come requisito per la formazione di nicchia adulti HSC.

Poco se sabe sobre la formaci&oacute;n de nichos, microambientes locales necesarios para el mantenimiento de c&eacute;lulas madre. Aqu&iacute; desarrollamos un ensayo in vivo para formaci&oacute;n de nicho (HSC) c&eacute;lulas madre adultas hematopoy&eacute;ticas. Con este ensayo, hemos identificado una poblaci&oacute;n de c&eacute;lulas progenitoras con marcadores de superficie CD45(-)Tie2(-)alpha(V)(+)CD105(+)Thy1.1(-) (CD105(+)Thy1(-)) que, cuando trasplantado en la c&aacute;psula del ri&ntilde;&oacute;n de rat&oacute;n adulto y ordenada de los huesos fetales de la post-coitum 15.5 d&iacute;as podr&iacute;a reclutar derivado del anfitri&oacute;n de vasos sangu&iacute;neos, producen donante-derivadas ect&oacute;picos huesos por un cart&iacute;lago intermedio y generar una cavidad de la m&eacute;dula poblada por HSC reconstituyente a largo plazo derivados del anfitri&oacute;n (LT-HSC). En contraste, CD45(-)Tie2(-)alpha(V)(+)CD105(+)Thy1(+) (CD105(+)Thy1(+)) fetal progenitores forma hueso que no contiene una cavidad de la m&eacute;dula. Supresi&oacute;n de la expresi&oacute;n de factores implicados en la osificaci&oacute;n endocondral, osterix como factor de crecimiento endotelial vascular (VEGF), inhibe la generaci&oacute;n de nicho. CD105(+)Thy1(-) poblaciones de progenitoras derivadas de las regiones de la mand&iacute;bula fetal o calvaria no sometidos a la osificaci&oacute;n endocondral forman solo hueso sin m&eacute;dula en nuestro an&aacute;lisis. Colectivamente, nuestros datos implican la osificaci&oacute;n endocondral, formaci&oacute;n de hueso que procede a trav&eacute;s de un intermediario del cart&iacute;lago, como requisito para la formaci&oacute;n de nicho adultos de HSC.

A common genetic mutation found in clear cell renal cell carcinoma (CC-RCC) is the loss of the von Hippel-Lindau (VHL) gene, which results in stabilization of hypoxia-inducible factors (HIFs), and contributes to cancer progression and metastasis. CUB-domain-containing protein 1 (CDCP1) was shown to promote metastasis in scirrhous and lung adenocarcinomas as well as in prostate cancer. In this study, we established a molecular mechanism linking VHL loss to induction of the CDCP1 gene through the HIF-1/2 pathway in renal cancer. Also, we report that Fyn, which forms a complex with CDCP1 and mediates its signaling to PKCÎ´, is a HIF-1 target gene. Mechanistically, we found that CDCP1 specifically regulates phosphorylation of PKCÎ´, but not of focal adhesion kinase or Crk-associated substrate. Signal transduction from CDCP1 to PKCÎ´ leads to its activation, increasing migration of CC-RCC. Furthermore, patient survival can be stratified by CDCP1 expression at the cell surface of the tumor. Taken together, our data indicates that CDCP1 protein might serve as a therapeutic target for CC-RCC.

VHL loss in renal cell carcinoma leads to up-regulation of CUB domain-containing protein 1 to stimulate PKC{delta}-driven migration.

Une mutation g&eacute;n&eacute;tique commune trouv&eacute; &agrave; cellules claires renal cell carcinoma (CC-RCC) est la perte du g&egrave;ne von Hippel-Lindau (VHL), qui aboutit &agrave; la stabilisation des facteurs inductibles par l&#39;hypoxie (HIFs) et contribue &agrave; la progression du cancer et des m&eacute;tastases. CUB-domaine contenant la prot&eacute;ine 1 (CDCP1) s&#39;est av&eacute;r&eacute;e favoriser des m&eacute;tastases dans squirrheux et ad&eacute;nocarcinomes pulmonaires ainsi que dans la prostate. Dans cette &eacute;tude, nous avons &eacute;tabli un m&eacute;canisme mol&eacute;culaire qui relie la perte VHL &agrave; l&#39;induction du g&egrave;ne CDCP1 par la voie de l&#39;HIF-1/2 dans le cancer du rein. En outre, nous rapportons que Fyn, qui forme un complexe avec CDCP1 et m&eacute;diatise sa signalisation &agrave; PKC&delta;, est un g&egrave;ne cible de HIF-1. Pluripotent cells represent a powerful tool for tissue regeneration, but their clinical utility is limited by their propensity to form teratomas. Little is known about their interaction with the surrounding niche following implantation and how this may be applied to promote survival and functional engraftment. In this study, we evaluated the ability of an osteogenic microniche consisting of a hydroxyapatite-coated, bone morphogenetic protein-2-releasing poly-l-lactic acid scaffold placed within the context of a macroenvironmental skeletal defect to guide in vivo differentiation of both embryonic and induced pluripotent stem cells. In this setting, we found de novo bone formation and participation by implanted cells in skeletal regeneration without the formation of a teratoma. This finding suggests that local cues from both the implanted scaffold/cell micro- and surrounding macroniche may act in concert to promote cellular survival and the in vivo acquisition of a terminal cell fate, thereby allowing for functional engraftment of pluripotent cells into regenerating tissue.

In vivo directed differentiation of pluripotent stem cells for skeletal regeneration.

Les cellules pluripotentes repr&eacute;sentent un outil puissant pour la r&eacute;g&eacute;n&eacute;ration des tissus, mais leur utilit&eacute; clinique est limit&eacute;e par leur propension &agrave; t&eacute;ratomes de forme. On conna&icirc;t leur interaction avec la niche environnante apr&egrave;s implantation et comment cela peut s&#39;appliquer pour promouvoir la survie et la prise de greffe fonctionnelle. Dans cette &eacute;tude, nous avons &eacute;valu&eacute; la possibilit&eacute; d&#39;une parcelle ost&eacute;og&eacute;nique consistant en un enduit hydroxyapatite, osseuse morphog&eacute;n&eacute;tique lib&eacute;rant prot&eacute;ine-2 l-polylactique acide &eacute;chafaudage plac&eacute; dans le contexte d&#39;une anomalie squelettique macro-environnementaux pour orienter la diff&eacute;renciation in vivo de deux des cellules souches pluripotentes embryonnaires et induits. Dans ce cadre, nous avons constat&eacute; la formation osseuse de novo et la participation de cellules implant&eacute;es dans la r&eacute;g&eacute;n&eacute;ration squelettique sans la formation d&#39;un t&eacute;ratome. Pluripotenten Zellen stellen ein m&auml;chtiges Werkzeug zur Geweberegeneration, sondern ihre klinischen Nutzen ist durch ihre Neigung, Form Teratome begrenzt. Wenig ist bekannt &uuml;ber ihre Interaktion mit der umgebenden Nische nach Einpflanzung und wie diese angewendet werden kann, &Uuml;berleben und funktionale Verteilung zu f&ouml;rdern. In dieser Studie ausgewertet wir die F&auml;higkeit einer osteogenen Microniche, bestehend aus einer Hydroxylapatit beschichtete, Knochen morphogenetische Protein-2-Freigabe Poly-l-Milchs&auml;ure saure Ger&uuml;st platziert im Rahmen eines Makroumwelt Skelett Mangels, in-vivo Differenzierung der beiden embryonalen und induzierten pluripotenten Stammzellen zu f&uuml;hren. In dieser Einstellung fanden wir de Novo Knochenbildung und die Teilnahme von implantierten Zellen Skelett Regeneration ohne die Bildung von ein Teratom. Dieser Befund legt nahe, dass lokale Hinweise aus dem implantierten Ger&uuml;st/Zelle Mikro- und umliegenden Macroniche zur F&ouml;rderung der zellulare &uuml;berleben und der in-vivo Erwerb von ein terminal Zelle Schicksal, wodurch f&uuml;r funktionale Verteilung von pluripotenten Zellen in Gewebe regenerieren zusammenwirken k&ouml;nnen.

Cellule pluripotenti rappresentano un potente strumento per la rigenerazione dei tessuti, ma la loro utilit&agrave; clinica &egrave; limitata dalla loro propensione a teratomi forma. Poco si sa circa la loro interazione con la nicchia circostante seguendo l&#39;impianto e come questo pu&ograve; essere applicato per promuovere la sopravvivenza e l&#39;attecchimento funzionale. In questo studio abbiamo valutato la possibilit&agrave; di un microniche osteogeniche costituito da un rivestimento di idrossiapatite, osso morphogenetic protein-2-rilasciando poly-l-lattico acido impalcatura collocato all&#39;interno del contesto di un difetto scheletrico senza per guidare la differenziazione in vivo di entrambe le cellule staminali pluripotenti indotte ed embrionale. In questa impostazione, abbiamo trovato la formazione ossea de novo e partecipazione da parte delle cellule impiantate nella rigenerazione scheletrica senza la formazione di un teratoma. Questa scoperta suggerisce che spunti locali sia dal patibolo impiantato per cella micro - e macroniche circostante possono agire di concerto per promuovere la sopravvivenza cellulare e l&#39;acquisizione di un destino di cellula terminale, consentendo in tal modo per attecchimento funzionale delle cellule pluripotenti nella rigenerazione dei tessuti in vivo.

C&eacute;lulas pluripotentes representan una herramienta poderosa para la regeneraci&oacute;n de los tejidos, pero su utilidad cl&iacute;nica es limitada por su propensi&oacute;n a los teratomas de forma. Poco se sabe sobre su interacci&oacute;n con el nicho circundante despu&eacute;s de la implantaci&oacute;n y c&oacute;mo esto puede aplicarse para promover la supervivencia y el injerto funcional. Organs are composites of tissue types with diverse developmental origins, and they rely on distinct stem and progenitor cells to meet physiological demands for cellular production and homeostasis. How diverse stem cell activity is coordinated within organs is not well understood. Here we describe a lineage-restricted, self-renewing common skeletal progenitor (bone, cartilage, stromal progenitor; BCSP) isolated from limb bones and bone marrow tissue of fetal, neonatal, and adult mice. The BCSP clonally produces chondrocytes (cartilage-forming) and osteogenic (bone-forming) cells and at least three subsets of stromal cells that exhibit differential expression of cell surface markers, including CD105 (or endoglin), Thy1 [or CD90 (cluster of differentiation 90)], and 6C3 [ENPEP glutamyl aminopeptidase (aminopeptidase A)]. These three stromal subsets exhibit differential capacities to support hematopoietic (blood-forming) stem and progenitor cells. Although the 6C3-expressing subset demonstrates functional stem cell niche activity by maintaining primitive hematopoietic stem cell (HSC) renewal in vitro, the other stromal populations promote HSC differentiation to more committed lines of hematopoiesis, such as the B-cell lineage. Gene expression analysis and microscopic studies further reveal a microenvironment in which CD105-, Thy1-, and 6C3-expressing marrow stroma collaborate to provide cytokine signaling to HSCs and more committed hematopoietic progenitors. As a result, within the context of bone as a blood-forming organ, the BCSP plays a critical role in supporting hematopoiesis through its generation of diverse osteogenic and hematopoietic-promoting stroma, including HSC supportive 6C3(+) niche cells.

Clonal precursor of bone, cartilage, and hematopoietic niche stromal cells.

Very small embryonic-like stem cells (VSELs) isolated from bone marrow (BM) have been reported to be pluripotent. Given their nonembryonic source, they could replace blastocyst-derived embryonic stem cells in research and medicine. However, their multiple-germ-layer potential has been incompletely studied. Here, we show that we cannot find VSELs in mouse BM with any of the reported stem cell potentials, specifically for hematopoiesis. We found that: (1) most events within the "VSEL" flow-cytometry gate had little DNA and the cells corresponding to these events (2) could not form spheres, (3) did not express Oct4, and (4) could not differentiate into blood cells. These results provide a failure to confirm the existence of pluripotent VSELs.
