Atherosclerosis is closely associated with disturbed flow characterized by low and oscillatory shear stress, but studies directly linking disturbed flow to atherogenesis is lacking. The major reason for this has been a lack of an animal model in which disturbed flow can be acutely induced and cause atherosclerosis. Here, we characterize partial carotid ligation as a model of disturbed flow with characteristics of low and oscillatory wall shear stress. We also describe a method of isolating intimal RNA in sufficient quantity from mouse carotid arteries. Using this model and method, we found that partial ligation causes upregulation of proatherogenic genes, downregulation of antiatherogenic genes, endothelial dysfunction, and rapid atherosclerosis in 2 wk in a p47(phox)-dependent manner and advanced lesions by 4 wk. We found that partial ligation results in endothelial dysfunction, rapid atherosclerosis, and advanced lesion development in a physiologically relevant model of disturbed flow. It also allows for easy and rapid intimal RNA isolation. This novel model and method could be used for genome-wide studies to determine molecular mechanisms underlying flow-dependent regulation of vascular biology and diseases.

Ici, nous caract&#xE9;risons partielle ligature carotidienne comme un mod&#xE8;le de flux perturb&#xE9; avec des caract&#xE9;ristiques de stress de la paroi &#xE0; faible cisaillement et oscillatoire. Nous d&#xE9;crivons &#xE9;galement une m&#xE9;thode d&#x26;#39;isolement d&#x26;#39;ARN intima en quantit&#xE9; suffisante &#xE0; partir des art&#xE8;res carotides de souris. L&#x26;#39;utilisation de ce mod&#xE8;le et la m&#xE9;thode, nous avons constat&#xE9; que la ligature partielle r&#xE9;gulation &#xE0; la hausse les causes de g&#xE8;nes proath&#xE9;rog&#xE8;nes, la r&#xE9;gulation n&#xE9;gative des g&#xE8;nes antiath&#xE9;rog&#xE8;nes, la dysfonction endoth&#xE9;liale et l&#x26;#39;ath&#xE9;roscl&#xE9;rose rapide en 2 semaines dans un p47 (phox) de mani&#xE8;re d&#xE9;pendante et des l&#xE9;sions avanc&#xE9;es par 4 sem. Nous avons trouv&#xE9; que les r&#xE9;sultats partiels dans la ligature la dysfonction endoth&#xE9;liale, l&#x26;#39;ath&#xE9;roscl&#xE9;rose rapide, et le d&#xE9;veloppement des l&#xE9;sions avanc&#xE9;es dans un mod&#xE8;le physiologiquement pertinent de flux perturb&#xE9;. Il permet &#xE9;galement de facile et rapide d&#x26;#39;isolement d&#x26;#39;ARN intimale. Ce nouveau mod&#xE8;le et la m&#xE9;thode pourrait &#xEA;tre utilis&#xE9;e pour l&#x26;#39;ensemble du g&#xE9;nome des &#xE9;tudes pour d&#xE9;terminer les m&#xE9;canismes mol&#xE9;culaires qui sous-tendent la r&#xE9;gulation d&#xE9;pendante de flux de la biologie vasculaire et maladies.

Atherosklerose ist eng verbunden mit gest&ouml;rtem Flow zeichnet sich durch niedrige und oszillierende Schubspannung, aber Studien direkt verbindet gest&ouml;rten Str&ouml;mung zu Atherogenesis fehlt. Der wesentliche Grund hierf&uuml;r wurde ein Mangel an einem Tiermodell gest&ouml;rt hereinstr&ouml;men k&ouml;nnen akut induzierte werden und verursachen Arteriosklerose. Hier pr&auml;gen wir teilweise Carotis Ligaturen als Modell einer gest&ouml;rten Str&ouml;mung mit Merkmalen der niedrigen und oszillierende Wand Schubspannung. Wir beschreiben auch eine Methode zur Isolierung von Hypertrophien RNS in ausreichender Menge von Maus Halsarterien. Mit diesem Modell und Methode, wir fanden, dass partielle Verbindung verursacht Hochregulation von Genen, Proatherogenic, Medikament antiatherogenic Gene, endothelialer Dysfunktion und schnelle Atherosklerose in 2 wk in eine p47 (Phox)-abh&auml;ngigen Weise und erweiterte L&auml;sionen von 4 wk. Wir fanden, dass die partielle Ligaturen endothelialen Dysfunktion, schnelle Atherosklerose und erweiterte L&auml;sion Entwicklung in einem physiologisch relevanten Modell des gest&ouml;rten Fluss f&uuml;hrt. Dar&uuml;ber hinaus k&ouml;nnen f&uuml;r die einfache und schnelle Hypertrophien RNA-Isolierung. Dieses neue Modell und Methode k&ouml;nnte f&uuml;r genomweiten Studien verwendet werden, molekulare Mechanismen flie&szlig;en-abh&auml;ngige Verordnung f&uuml;r vaskul&auml;re Biologie und Krankheiten zu bestimmen.

Il motivo principale per questo &egrave; stata la mancanza di un modello animale in cui flusso disturbato pu&ograve; essere indotta acutamente e causare l&#39;aterosclerosi. Qui, ci caratterizzano legatura carotidea parziale come un modello di flusso disturbato con le caratteristiche di sollecitazione di taglio muro basso e oscillatorio. Abbiamo anche descrivere un metodo di isolamento di RNA intimale in quantit&agrave; sufficiente da arterie carotidee del mouse. Utilizzando questo modello e metodo, abbiamo trovato quella legatura parziale provoca upregulation dei geni proatherogenic, downregulation dei geni antiatherogenic, disfunzione endoteliale e aterosclerosi rapida in 2 sett in un p47 (phox)-modo dipendente e lesioni avanzate da 4 sett. Abbiamo trovato che legatura parziale provoca la disfunzione endoteliale, aterosclerosi rapida e sviluppo di lesione avanzata in un modello di flusso disturbato fisiologicamente rilevante. Permette inoltre di isolamento del RNA intimale facile e rapido. Questo modello di romanzo e il metodo potrebbe essere utilizzati per studi genome-wide per determinare i meccanismi molecolari sottostanti di regolazione di flusso-dipendente della biologia vascolare e malattie.

La aterosclerosis se asocia estrechamente con la alteraci&#xF3;n del flujo caracterizado por la tensi&#xF3;n de corte baja y oscilante, pero los estudios que vinculen directamente alteraci&#xF3;n del flujo a la aterog&#xE9;nesis es insuficiente. La raz&#xF3;n principal de ello ha sido la falta de un modelo animal en el que el flujo puede ser perturbado gravemente inducida y la aterosclerosis causa. En este sentido, caracterizan la ligadura de la car&#xF3;tida parcial como un modelo de alteraci&#xF3;n del flujo con las caracter&#xED;sticas de la tensi&#xF3;n de cizallamiento bajo y oscilante. Tambi&#xE9;n describe un m&#xE9;todo para aislar ARN intimales en cantidad suficiente de rat&#xF3;n arterias car&#xF3;tidas. El uso de este modelo y m&#xE9;todo, se encontr&#xF3; que la ligadura parcial de la regulaci&#xF3;n positiva de genes causas proaterog&#xE9;nicos, regulaci&#xF3;n a la baja de los genes antiaterog&#xE9;nicas, la disfunci&#xF3;n endotelial y la aterosclerosis r&#xE1;pido en 2 semanas en un p47 (phox) dependiente de la forma y las lesiones avanzadas a las 4 semanas. Hemos encontrado que los resultados parciales en la ligadura de la disfunci&#xF3;n endotelial, la aterosclerosis acelerada y el desarrollo de lesiones avanzadas en un modelo fisiol&#xF3;gicamente relevantes de la alteraci&#xF3;n del flujo. Tambi&#xE9;n permite el aislamiento &#xED;ntima f&#xE1;cil y r&#xE1;pida de ARN. Este nuevo modelo y el m&#xE9;todo podr&#xED;a ser utilizado para todo el genoma estudios para determinar el flujo de los mecanismos moleculares que subyacen a la regulaci&#xF3;n dependiente de la biolog&#xED;a vascular y las enfermedades.
