Amyotrophe Lateralsklerose (ALS) ist verbunden mit einer erh&ouml;hten Anzahl von Mikroglia innerhalb des Zentralnervensystems (ZNS). Allerdings ist es unbekannt, ob die Microgliosis aus CNS resident Mikroglia Verbreitung oder Einstellung von Knochenmark (BM) Ergebnisse-Microglial Vorstufen abgeleitet. Hier bewerten wir die Verteilung und die Anzahl der BM-abgeleitete Zellen im R&uuml;ckenmark mit Transplantation von gr&uuml;n fluoreszierendes Protein (GFP)-BM Zellen in Myelo-ablated M&auml;use menschlichen mutant Superoxiddismutase 1 (mSOD), einem murinen Modell ALS over-expressing gekennzeichnet. Transplantation von GFP + BM keinen Einfluss auf die Rate der Fortschreiten der Krankheit bei mSOD M&auml;usen. Meine Zahlen von Mikroglia und GFP + Zellen im R&uuml;ckenmark Schn&uuml;re des Steuerelements M&auml;use waren nicht wesentlich von denen bei asymptomatischen mSOD M&auml;usen und zeigte keine &Auml;nderung mit Tier Alter. Die Anzahl der GFP + Zellen und Mikroglia (F4 / 80 + und CD11b +-Zellen) innerhalb des R&uuml;ckenmarks von mSOD M&auml;use erh&ouml;ht im Vergleich zu den altersbezogenen Steuerelemente zu einem Zeitpunkt, wann mSOD M&auml;use Krankheitssymptome, bis Krankheit Endstadium ausgestellt. Obwohl wir eine Zunahme der Anzahl der GFP + Zellen im R&uuml;ckenmark Schn&uuml;re mSOD M&auml;use mit Krankheitssymptomen, mittlere Anzahl von GFP + F4 beobachteten / 80 + Zellen weniger als 20 % von allen F4 bestehen / 80 + Zellen und nicht mit Fortschreiten der Krankheit zu erh&ouml;hen. Toutefois, on ignore si les r&#xE9;sultats microgliose de la prolif&#xE9;ration de SNC r&#xE9;sidant microglie, ou le recrutement de la moelle osseuse (BM) d&#xE9;riv&#xE9;s de pr&#xE9;curseurs microgliales. Amyotrophic lateral sclerosis (ALS) is associated with increased numbers of microglia within the central nervous system (CNS). However, it is unknown whether the microgliosis results from proliferation of CNS resident microglia, or recruitment of bone marrow (BM)-derived microglial precursors. Here we assess the distribution and number of BM-derived cells in spinal cord using transplantation of green fluorescent protein (GFP)-labeled BM cells into myelo-ablated mice over-expressing human mutant superoxide dismutase 1 (mSOD), a murine model of ALS. Transplantation of GFP+ BM did not affect the rate of disease progression in mSOD mice. Mean numbers of microglia and GFP+ cells in spinal cords of control mice were not significantly different from those in asymptomatic mSOD mice and showed no change with animal age. The number of GFP+ cells and microglia (F4/80+ and CD11b+ cells) within the spinal cord of mSOD mice increased compared to age-matched controls at a time when mSOD mice exhibited disease symptoms, continuing up to disease end-stage. Although we observed an increase in the number of GFP+ cells in spinal cords of mSOD mice with disease symptoms, mean numbers of GFP+ F4/80+ cells comprised less than 20% of all F4/80+ cells and did not increase with disease progression. Furthermore, the relative rates of proliferation in CD45+GFP- and CD45+GFP+ cells were comparable. Thus, we demonstrate that the microgliosis present in spinal cord tissue of mSOD mice is primarily due to an expansion of resident microglia and not to the recruitment of microglial precursors from the circulation.

Sclerosi laterale amiotrofica (SLA) &egrave; associata all&#39;aumento del numero delle microglia all&#39;interno del sistema nervoso centrale (SNC). Tuttavia, non &egrave; noto se la microgliosis deriva dalla proliferazione di microglia residente CNS, o assunzione di midollo osseo (BM)-derivato microgliali precursori. La esclerosis lateral amiotr&#xF3;fica (ALS) est&#xE1; asociada con un mayor n&#xFA;mero de microgl&#xED;a en el sistema nervioso central (SNC). Sin embargo, se desconoce si los resultados microgliosis de proliferaci&#xF3;n de la microgl&#xED;a residentes del SNC, o la contrataci&#xF3;n de la m&#xE9;dula &#xF3;sea (MO) derivados de los precursores microgliales. Aqu&#xED; se eval&#xFA;a la distribuci&#xF3;n y el n&#xFA;mero de evacuaciones intestinales derivados de las c&#xE9;lulas en la m&#xE9;dula espinal mediante el trasplante de la prote&#xED;na verde fluorescente (GFP)-BM en c&#xE9;lulas marcadas con mielo-ablacionadas los ratones que sobreexpresan el super&#xF3;xido dismutasa humana mutante 1 (mSOD), un modelo murino de esclerosis lateral amiotr&#xF3;fica . El trasplante de las buenas pr&#xE1;cticas agrarias + BM no afect&#xF3; la tasa de progresi&#xF3;n de la enfermedad en ratones mSOD. La media de n&#xFA;mero de microglia y las buenas pr&#xE1;cticas agrarias + c&#xE9;lulas en la m&#xE9;dula espinal de los ratones de control no fueron significativamente diferentes de los de los ratones mSOD asintom&#xE1;ticos y no mostr&#xF3; ning&#xFA;n cambio con la edad del animal. El n&#xFA;mero de buenas pr&#xE1;cticas agrarias + c&#xE9;lulas y microglia (F4/80 + y CD11b + en las c&#xE9;lulas) en la m&#xE9;dula espinal de los ratones mSOD mayor en comparaci&#xF3;n con controles pareados por edad en el momento en los ratones mostraron s&#xED;ntomas de la enfermedad mSOD, continuando hasta el final de la enfermedad. Amyotrophic lateral sclerosis (ALS) is associated with increased numbers of microglia within the CNS. However, it is unclear to what extent bone marrow (BM)-derived cells contribute to this microgliosis. We have studied the adoptive transfer of green fluorescent protein (GFP)-labeled whole BM cells and BM from mice that express GFP only in CX(3)CR1+ cells (CX(3)CR1(+/GFP)) into the CNS of a murine model of ALS having over-expression of mutant superoxide dismutase (mSOD), and wt littermates. We find that most GFP+ and CX(3)CR1(+/GFP) cells are found adjacent to the microvasculature within the CNS, both in mSOD and wt mice. GFP+ and CX(3)CR1(+/GFP) cells within the CNS have a variety of morphologies, including cells with an elongated appearance, weak Iba-1 immunoreactivity, and often mannose receptor immunoreactivity, indicating that these cells are perivascular microglia. Typically, less than 10% of BM-derived cells had a stellate-shape and expressed strong Iba-1 immunoreactivity, as expected for parenchymal microglia, indicating that BM-derived cells uncommonly generate parenchymal microglia. Adoptive transfer of BM-derived cells from CX(3)CR1(+/GFP) mice revealed that many elongated cells are GFP+, demonstrating that some perivascular cells are derived from BM cells of the CX(3)CR1+ lineage. The significantly greater numbers of BM cells in mSOD than in control mice indicate that the presence of these BM cells in the spinal cord is regulated by conditioning stimuli that may include irradiation and inflammatory factors within the CNS.

Bone marrow-derived cells in the central nervous system of a mouse model of amyotrophic lateral sclerosis are associated with blood vessels and express CX(3)CR1.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by upper and lower motoneuron death. Mutations in the gene for superoxide dismutase 1 (SOD1) cause a familial form of ALS and have been used to develop transgenic mice which overexpress human mutant SOD1 (mSOD) and these mice exhibit a motoneuron disease which is pathologically and phenotypically similar to ALS. Neuroinflammation is a pathological hallmark of many neurodegenerative diseases including ALS and is typified by the activation and proliferation of microglia and the infiltration of T cells into the brain and spinal cord. Although the neuroinflammatory response has been considered a consequence of neuronal dysfunction and death, evidence indicates that manipulation of this response can alter disease progression. Previously viewed as deleterious to neuronal survival, recent reports suggest a trophic role for activated microglia in the mSOD mouse during the early stages of disease that is dependent on instructive signals from infiltrating T cells. However, at advanced stages of disease, activated microglia acquire increased neurotoxic potential, warranting further investigation into factors capable of skewing microglial activation towards a neurotrophic phenotype as a means of therapeutic intervention in ALS.

The Neuroinflammatory Response in ALS: The Roles of Microglia and T Cells.

Myeloablative preconditioning using irradiation is the most commonly used technique to generate rodents having chimeric bone marrow, employed for the study of bone marrow-derived cell accumulation in the healthy and diseased central nervous system. However, irradiation has been shown to alter the blood-brain barrier, potentially creating confounding artefacts. To better study the potential of bone marrow-derived cells to function as treatment vehicles for neurodegenerative diseases alternative preconditioning regimens must be developed. We treated transgenic mice that over-express human mutant superoxide dismutase 1, a model of amyotrophic lateral sclerosis, with busulfan to determine whether this commonly used chemotherapeutic leads to stable chimerism and promotes the entry of bone marrow-derived cells into spinal cord. Intraperitoneal treatment with busulfan at 60 mg/kg or 80 mg/kg followed by intravenous injection of green fluorescent protein-expressing bone marrow resulted in sustained levels of chimerism (~80%). Bone marrow-derived cells accumulated in the lumbar spinal cord of diseased mice at advanced stages of pathology at both doses, with limited numbers of bone marrow derived cells observed in the spinal cords of similarly treated, age-matched controls; the majority of bone marrow-derived cells in spinal cord immunolabelled for macrophage antigens. Comparatively, significantly greater numbers of bone marrow-derived cells were observed in lumbar spinal cord following irradiative myeloablation. These results demonstrate bone marrow-derived cell accumulation in diseased spinal cord is possible without irradiative preconditioning.

Microglia have long been the focus of much attention due to their strong proliferative response (microgliosis) to essentially any kind of damage to the CNS. More recently, we reached the realization that these cells play specific roles in determining progression and outcomes of essentially all CNS disease. Thus, microglia has ceased to be viewed as an accessory to underlying pathologies and has now taken center stage as a therapeutic target. Here, we review how our understanding of microglia's involvement in promoting or limiting the pathogenesis of diseases such as amyotrophic lateral sclerosis, Alzheimer's disease, Huntington's disease, multiple sclerosis, X-linked adrenoleukodystrophy (X-ALD) and lysosomal storage diseases (LSD) has changed over time. While strategies to suppress the deleterious and promote the virtuous functions of microglia will undoubtedly be forthcoming, replacement of these cells has already proven its usefulness in a clinical setting. Over the past few years, we have reached the realization that microglia have a developmental origin that is distinct from that of bone marrow-derived myelomonocytic cells. Nevertheless, microglia can be replaced, in specific situations, by the progeny of hematopoietic stem cells (HSCs), pointing to a strategy to engineer the CNS environment through the transplantation of modified HSCs. Thus, microglia replacement has been successfully exploited to deliver therapeutics to the CNS in human diseases such as X-ALD and LSD. With this outlook in mind, we will discuss the evidence existing so far for microglial involvement in the pathogenesis and the therapy of specific CNS disease.

The role of microglia in human disease: therapeutic tool or target?

Previous work has suggested that bone marrow (BM)-derived cells (BMDCs) accumulate within the CNS and could potentially associate with β-amyloid plaques in Alzheimer's disease (AD). To explore the accumulation of BMDCs in murine AD, we transplanted green fluorescent protein (GFP)-labeled BM cells into triple transgenic (3×Tg) and wild-type (wt) mice using non-irradiative myelosuppresive conditioning with busulfan (BU). We find that BU (80mg/kg) is sufficient to obtain adequate chimerism (>85%) in wt mice. In order to obtain appreciable non-irradiative chimerism in the 3×Tg mice (>80%), anti-asialo ganglio-N-tetraosylceramide (α-ASGM-1) antibody was also used to reduce natural killer cell function and thereby abrogate the hybrid resistance of the 3×Tg mouse strain. Using BU conditioning and α-ASGM-1 together, we observed sustained BM chimerism and BMDC accumulation within the CNS of the 3×Tg and wt mice. In cortex and hippocampus, BMDC accumulation was perivascular in distribution and similar between 3×Tg and wt mice, with no clear association between BMDCs and AD plaques. We conclude that non-irradiative BM chimerism can be achieved with BU in 3×Tg mice, but requires α-ASGM-1 (or similar appropriate NK-cell depletion). Use of this chimerism protocol permits BMDCs accumulation in the CNS of mixed strain recipient mice although BMDCs appear to be largely perivascular within cortex and hippocampus.
