Chemoreceptors are key components of the high-performance signal transduction system that controls bacterial chemotaxis. Chemoreceptors are typically localized in a cluster at the cell pole, where interactions among the receptors in the cluster are thought to contribute to the high sensitivity, wide dynamic range, and precise adaptation of the signaling system. Previous structural and genomic studies have produced conflicting models, however, for the arrangement of the chemoreceptors in the clusters. Using whole-cell electron cryo-tomography, here we show that chemoreceptors of different classes and in many different species representing several major bacterial phyla are all arranged into a highly conserved, 12-nm hexagonal array consistent with the proposed "trimer of dimers" organization. The various observed lengths of the receptors confirm current models for the methylation, flexible bundle, signaling, and linker sub-domains in vivo. Our results suggest that the basic mechanism and function of receptor clustering is universal among bacterial species and was thus conserved during evolution.

Ch&#xE9;mor&#xE9;cepteurs sont g&#xE9;n&#xE9;ralement localis&#xE9;s dans un cluster au p&#xF4;le cellulaire, o&#xF9; les interactions entre les r&#xE9;cepteurs de la grappe sont consid&#xE9;r&#xE9;s comme contribuant &#xE0; la haute sensibilit&#xE9;, une large gamme dynamique, et l&#x26;#39;adaptation pr&#xE9;cise du syst&#xE8;me de signalisation. Chemoreceptors sind Schl&uuml;sselkomponenten des Hochleistungs-Signaltransduktion Systems, die bakterielle Chemotaxis steuert. Chemoreceptors sind in der Regel in einem Cluster am Pol Zelle, lokalisiert, wo Interaktionen zwischen den Rezeptoren im Cluster gedacht werden, um die hohe Empfindlichkeit und Dynamikbereich pr&auml;zise Anpassung des Signalling Systems beitragen. Bisherige strukturelle und genomische Untersuchungen haben Modelle, jedoch f&uuml;r die Anordnung der Chemoreceptors in den Clustern gebracht. Mit ganzen-Zelle Elektronen Cryo-Tomographie, zeigen hier wir, dass alle Chemoreceptors verschiedener Klassen und in vielen verschiedenen Arten, die mehrere bedeutende bakterielle St&auml;mme in einem stark konservierten, 12-nm sechseckigen Array mit der vorgeschlagenen &quot;Trimer des Dimer&quot; Organisation angeordnet sind. Die verschiedenen beobachteten L&auml;ngen der Rezeptoren best&auml;tigen aktuelle Modelle f&uuml;r die Methylierung, flexible B&uuml;ndel, Signalisierung und Linker Subdomains in-vivo. Chemocettori sono componenti chiave del sistema di trasduzione di segnale ad alte prestazioni che controlla la chemiotassi batterica. Chemocettori sono tipicamente localizzate in un cluster presso il polo della cellula, dove le interazioni tra i recettori del cluster sono pensate per contribuire all&#39;alta sensibilit&agrave;, ampia gamma dinamica e preciso adattamento del sistema di segnalazione. Precedenti studi di genomici e strutturali hanno prodotto modelli in conflitto, tuttavia, per la disposizione dei chemocettori in grappoli. The bacterial flagellum is one of nature's most amazing and well-studied nanomachines. Its cell-wall-anchored motor uses chemical energy to rotate a microns-long filament and propel the bacterium towards nutrients and away from toxins. While much is known about flagellar motors from certain model organisms, their diversity across the bacterial kingdom is less well characterized, allowing the occasional misrepresentation of the motor as an invariant, ideal machine. Here, we present an electron cryotomographical survey of flagellar motor architectures throughout the Bacteria. While a conserved structural core was observed in all 11 bacteria imaged, surprisingly novel and divergent structures as well as different symmetries were observed surrounding the core. Correlating the motor structures with the presence and absence of particular motor genes in each organism suggested the locations of five proteins involved in the export apparatus including FliI, whose position below the C-ring was confirmed by imaging a deletion strain. The combination of conserved and specially-adapted structures seen here sheds light on how this complex protein nanomachine has evolved to meet the needs of different species.

Korrelation der motorischen Strukturen mit der Anwesenheit und Abwesenheit von bestimmten motor Gene in jedem Organismus vorgeschlagen, die Positionen der f&uuml;nf-Proteine in der Export-Apparat, einschlie&szlig;lich FliI, deren Position unterhalb der C-Ring Imaging eine L&ouml;schung Belastung best&auml;tigt wurde. Die Kombination von konservierten und speziell angepassten Strukturen gesehen hier beleuchtet wie dieses komplexe Protein Nanomaschine entwickelt hat, um die Bed&uuml;rfnisse der verschiedenen Arten.

El flagelo bacteriano es una de las nanom&#xE1;quinas m&#xE1;s sorprendente y bien estudiado de la naturaleza. Su pared celular anclado el motor utiliza la energ&#xED;a qu&#xED;mica para hacer girar un filamento de micras de largo y propulsar a la bacteria a los alimentos y lejos de las toxinas. Aunque se sabe mucho sobre los motores flagelares de ciertos organismos modelo, su diversidad en todo el reino bacteriano es muy bien caracterizada, que permite la tergiversaci&#xF3;n ocasional del motor como una m&#xE1;quina de todos los idiomas, ideal. A continuaci&#xF3;n, presentamos una encuesta electr&#xF3;nica cryotomographical de las arquitecturas de motor flagelar a trav&#xE9;s de las bacterias. Mientras que un n&#xFA;cleo estructural conservada se observ&#xF3; en todas las 11 bacterias con imagen, las estructuras sorprendentemente nuevos y divergentes, as&#xED; como diferentes simetr&#xED;as se observ&#xF3; que rodea al n&#xFA;cleo. Correlacionar las estructuras de motor con la presencia y ausencia de genes de motor particulares de cada organismo sugiri&#xF3; la ubicaci&#xF3;n de cinco prote&#xED;nas implicadas en el aparato de exportaci&#xF3;n incluyendo flii, cuya posici&#xF3;n por debajo del anillo C fue confirmada por im&#xE1;genes una supresi&#xF3;n cepa. La combinaci&#xF3;n de estructuras conservadas y adaptados especialmente ven en la foto arroja luz sobre c&#xF3;mo este complejo de prote&#xED;nas nanom&#xE1;quina ha evolucionado para satisfacer las necesidades de las diferentes especies.
