Loss of synaptic activity or innervation induces sprouting of intact motor nerve terminals that adds or restores nerve-muscle connectivity. Ciliary neurotrophic factor (CNTF) and terminal Schwann cells (tSCs) have been implicated as molecular and cellular mediators of the compensatory process. We wondered if the previously reported lack of terminal sprouting in CNTF null mice was due to abnormal reactivity of tSCs. To this end, we examined nerve terminal and tSC responses in CNTF null mice using experimental systems that elicited extensive sprouting in wildtype mice. Contrary to the previous report, we found that motor nerve terminals in the null mice sprout extensively in response to major sprouting-stimuli such as exogenously applied CNTF per se, botulinum toxin-elicited paralysis, and partial denervation by L4 spinal root transection. In addition, the number, length and growth patterns of terminal sprouts, and the extent of reinnervation by terminal or nodal sprouts, were similar in wildtype and null mice. tSCs in the null mice were also reactive to the sprouting-stimuli, elaborating cellular processes that accompanied terminal sprouts or guided reinnervation of denervated muscle fibers. Lastly, CNTF was absent in quiescent tSCs in intact, wildtype muscles and little if any was detected in reactive tSCs in denervated muscles. Thus, CNTF is not required for induction of nerve terminal sprouting, for reactivation of tSCs, and for compensatory reinnervation after nerve injury. We interpret these results to support the notion that compensatory sprouting in adult muscles is induced primarily by contact-mediated mechanisms, rather than by diffusible factors.

Facteur neurotrophique ciliaire (CNTF) et de terminaux des cellules de Schwann (TSCS) ont &#xE9;t&#xE9; impliqu&#xE9;s en tant que m&#xE9;diateurs mol&#xE9;culaires et cellulaires du processus compensatoire. Nous nous sommes demand&#xE9; si le manque signal&#xE9; auparavant du terminal de germination chez les souris nulles CNTF &#xE9;tait due &#xE0; une r&#xE9;action anormale du TSC. &#xC0; cette fin, nous avons examin&#xE9; terminaison nerveuse et les r&#xE9;ponses TSC dans CNTF souris nulles en utilisant des syst&#xE8;mes exp&#xE9;rimentaux qui ont suscit&#xE9; une vaste germination chez les souris de type sauvage. Nous interpr&#xE9;tons ces r&#xE9;sultats pour soutenir l&#x26;#39;id&#xE9;e que la germination compensatoire dans les muscles adultes est induite principalement par contact des m&#xE9;canismes faisant intervenir, plut&#xF4;t que par des facteurs diffusibles.

Dar&uuml;ber hinaus waren die Anzahl, L&auml;nge und Wachstumsmuster der terminal Sprossen und das Ausma&szlig; der Reinnervation von terminal oder nodal Sprossen in Wildtyp und Null-M&auml;use. tSCs in die null M&auml;use waren auch reaktiv auf die sprie&szlig;ende-Reize, Ausarbeitung von zellul&auml;re Prozessen, die begleitet terminal Sprossen oder gef&uuml;hrte Reinnervation denervated Muskelfasern. Schlie&szlig;lich CNTF fehlte im Ruhezustand tSCs in intakt, Wildtyp Muskeln und wenig wenn jeder in reaktiven tSCs in denervated Muskeln erkannt wurde. CNTF ist daher nicht erforderlich, f&uuml;r die Induktion von Nerve terminal sprie&szlig;en, Reaktivierung des tSCs und f&uuml;r Ausgleichsma&szlig;nahmen Reinnervation nach Nervenverletzung. Wir interpretieren diese Ergebnisse um die Vorstellung zu unterst&uuml;tzen, dass Ausgleichsma&szlig;nahmen sprie&szlig;en in Erwachsenen Muskeln haupts&auml;chlich durch Kontakt-vermittelten Mechanismen statt durch diffusible Faktoren hervorgerufen wird.

La p&#xE9;rdida de la actividad sin&#xE1;ptica o inervaci&#xF3;n induce la brotaci&#xF3;n de las terminales nerviosas motoras intactas que a&#xF1;ade o restaura la conectividad de los m&#xFA;sculos y nervios. Factor neurotr&#xF3;fico ciliar (CNTF) y la terminal de las c&#xE9;lulas de Schwann (TSC) han sido implicados como mediadores moleculares y celulares del proceso de compensaci&#xF3;n. Nos preguntamos si la falta se ha informado de brotes terminales en ratones nulos CNTF se debi&#xF3; a una reacci&#xF3;n anormal del TSC. Con este fin, hemos examinado las respuestas del nervio terminal y el CET en CNTF ratones nulos que utilizan sistemas experimentales que suscit&#xF3; amplia que brota en los ratones de tipo salvaje. Ambas mol&#xE9;culas difusibles y la superficie envolvente-se cree que inducen la brotaci&#xF3;n de las terminaciones nerviosas motoras en respuesta a la par&#xE1;lisis. Both diffusible and surface-bound molecules are thought to induce sprouting of motor nerve terminals in response to paralysis. Here we report that the sprouting induced by ciliary neurotrophic factor (CNTF) is qualitatively different from the sprouting induced by botulinum toxin (BoTX). We show first that subcutaneous application of CNTF to levator auris longus muscles of adult mice evokes sprouting from nearly all nerve terminals. Surprisingly, however, most terminal sprouts remain within the boundaries of the endplate region and rarely grow extrasynaptically even if CNTF is administered chronically. In contrast, terminal sprouts induced by BoTX extend vigorously along the extrasynaptic muscle surface. The different patterns of sprout elongation are attributable in part to different patterns of initiation: whereas CNTF-induced sprouts emerge randomly from the surface of terminal branches, BoTX-induced sprouts emerge exclusively along the perimeter of terminal branches in direct apposition to muscle fiber membranes. Combined treatment with CNTF and BoTX produces exceptionally robust extraterminal sprouting with little if any intrasynaptic growth of terminal sprouts. We interpret these results as showing that paralysis induces sprouting primarily by muscle-associated, surface-bound molecules rather than by diffusible factors. Our findings may be useful in defining the physiological role of the numerous candidate sprouting-inducers and in promoting compensatory sprouting after nerve injury for therapeutic benefit.

Les deux mol&#xE9;cules diffusibles et li&#xE9;e &#xE0; la surface sont consid&#xE9;r&#xE9;s pour induire la germination des terminaisons nerveuses en r&#xE9;ponse &#xE0; la paralysie. Nous rapportons ici que la germination induite par le facteur neurotrophique ciliaire (CNTF) est qualitativement diff&#xE9;rente de la germination induite par la toxine botulique (BoTX). Nous montrons d&#x26;#39;abord que l&#x26;#39;application sous-cutan&#xE9;e de CNTF &#xE0; muscles releveurs longus auris de souris adultes &#xE9;voque la germination de presque tous les terminaisons nerveuses. &#xC9;tonnamment, cependant, la plupart des germes de terminaux rester dans les limites de la r&#xE9;gion flasque et cultivent rarement extrasynaptically m&#xEA;me si le CNTF est administr&#xE9; de fa&#xE7;on chronique. Nous interpr&#xE9;tons ces r&#xE9;sultats comme indiquant que la paralysie induit la germination principalement par le muscle associ&#xE9;e, li&#xE9;e &#xE0; la surface des mol&#xE9;cules plut&#xF4;t que par des facteurs diffusibles. Hier berichten wir, dass das sprie&szlig;en von Zilien Neurotrophic Faktor (CNTF) induziert qualitativ anders als das sprie&szlig;en von Botulinum-Toxin (BoTX) induziert. Wir zeigen zun&auml;chst, dass subkutane Anwendung CNTF auf Musculus Levator Auris Longus Muskeln des Erwachsenen M&auml;usen hervorruft sprie&szlig;en aus fast allen Nervenenden. &Uuml;berraschend, aber die meisten terminal Sprossen bleiben innerhalb der Grenzen des Gebietes Endplatte und selten extrasynaptically zu wachsen, auch wenn CNTF chronisch verwaltet wird. Im Gegensatz dazu erweitern terminal Sprossen induziert durch BoTX kr&auml;ftig entlang der extrasynaptic Muskel-Fl&auml;che. Die unterschiedlichen Muster der spriessen Dehnung entfallen teilweise auf unterschiedliche Muster der Initiation: w&auml;hrend CNTF induziert Sprossen zuf&auml;llig von der Oberfl&auml;che der terminal Zweige entstehen, BoTX induziert Sprossen entstehen ausschlie&szlig;lich entlang der Umfang der terminal Niederlassungen in direkte Apposition zu Muskelfaser-Membranen. Kombinierte Behandlung mit CNTF und BoTX produziert besonders robuste extraterminal sprie&szlig;en mit wenig bis gar keinen intrasynaptic Wachstum von terminal Sprossen. Wir interpretieren diese Ergebnisse als Ergebnis, da&szlig; L&auml;hmung hervorruft, sprie&szlig;en in erster Linie von Muskel-assoziierte, Oberfl&auml;che gebundene Molek&uuml;le nicht von diffusible Faktoren. Unsere Erkenntnisse n&uuml;tzlich in die physiologische Rolle von den zahlreichen Kandidaten-sprie&szlig;en-Induktoren definieren und bei der F&ouml;rderung der kompensatorischen sprie&szlig;en nach Nervenverletzung f&uuml;r therapeutischen Nutzen.

I diversi modelli di allungamento del germoglio sono attribuibili in parte a diversi modelli di iniziazione: considerando che germogli CNTF-indotta casualmente emergono dalla superficie del terminale rami, germogli di BoTX-indotta emergono esclusivamente lungo il perimetro dei rami terminali in diretta apposizione delle membrane della fibra muscolare. Trattamento combinato con CNTF e BoTX produce extraterminal eccezionalmente robusto che spuntano con poco se qualsiasi crescita agisce di germogli terminali. Dobbiamo interpretare questi risultati come mostrando che induce la paralisi spuntano principalmente dal muscolo-associate, associato a superficie molecole piuttosto che da fattori diffusibile. I nostri risultati possono essere utili nel definire il ruolo fisiologico dei numerosi candidati spuntano-induttori e nella promozione di germogliatura compensativa dopo lesione del nervo per beneficio terapeutico.

Muscarinic acetylcholine receptors (mAChRs) modulate synaptic function, but whether they influence synaptic structure remains unknown. At neuromuscular junctions (NMJs), mAChRs have been implicated in compensatory sprouting of axon terminals in paralyzed or denervated muscles. Here we used pharmacological and genetic inhibition and localization studies of mAChR subtypes at mouse NMJs to demonstrate their roles in synaptic stability and growth but not in compensatory sprouting. M(2) mAChRs were present solely in motor neurons, whereas M(1), M(3), and M(5) mAChRs were associated with Schwann cells and/or muscle fibers. Blockade of all five mAChR subtypes with atropine evoked pronounced effects, including terminal sprouting, terminal withdrawal, and muscle fiber atrophy. In contrast, methoctramine, an M(2/4)-preferring antagonist, induced terminal sprouting and terminal withdrawal, but no muscle fiber atrophy. Consistent with this observation, M(2)(-/-) but no other mAChR mutant mice exhibited spontaneous sprouting accompanied by extensive loss of parental terminal arbors. Terminal sprouting, however, seemed not to be the causative defect because partial loss of terminal branches was common even in the M(2)(-/-) NMJs without sprouting. Moreover, compensatory sprouting after paralysis or partial denervation was normal in mice deficient in M(2) or other mAChR subtypes. We also found that many NMJs of M(5)(-/-) mice were exceptionally small and reduced in proportion to the size of parental muscle fibers. These findings show that axon terminals are unstable without M(2) and that muscle fiber growth is defective without M(5). Subtype-specific muscarinic signaling provides a novel means for coordinating activity-dependent development and maintenance of the tripartite synapse.

Blocco di tutti i cinque sottotipi di mAChR con atropina evocato pronunciati effetti, tra cui germinazione terminale terminale ritiro e atrofia della fibra muscolare. Al contrario, methoctramine, M(2/4)-preferendo antagonista, indotta germogliatura terminale e terminale ritiro, ma nessuna atrofia della fibra muscolare. Coerente con questa osservazione, M(2)(-/-), ma non altri topi mutanti mAChR esposte spontanea germinazione accompagnata da perdita di pergole terminale parentale. Germogliatura terminale, tuttavia, non sembrava essere il difetto causa, perch&eacute; parziale perdita dei rami terminali era comune anche in NMJs il M(2)(-/-) senza che spuntano. Inoltre, era normale nei topi carenti di M(2) o altri sottotipi mAChR compensativi che spuntano dopo paralisi o denervazione parziale. Abbiamo anche trovato che molti NMJs di M(5)(-/-) topi erano eccezionalmente piccolo e ridotto in proporzione alle dimensioni delle fibre muscolari dei genitori. Questi risultati mostrano che assoni terminali sono instabili senza M(2) e che la crescita della fibra muscolare &egrave; difettoso senza M(5). Specifico del sottotipo muscarinico segnalazione fornisce che un romanzo significa per coordinare lo sviluppo di attivit&agrave;-dipendente e manutenzione della sinapsi tripartita.

Los receptores muscar&#xED;nicos de acetilcolina (mAChRs) modulan la funci&#xF3;n sin&#xE1;ptica, pero si ellos influyen en la estructura sin&#xE1;ptica sigue siendo desconocido. En las uniones neuromusculares (NMJs), mAChRs han sido implicados en brotes de compensaci&#xF3;n de los terminales de los axones en los m&#xFA;sculos paralizados o denervado. Aqu&#xED; hemos utilizado los estudios de inhibici&#xF3;n y la localizaci&#xF3;n farmacol&#xF3;gicas y gen&#xE9;ticas de los subtipos mAchR en NMJs rat&#xF3;n para demostrar su papel en la estabilidad y el crecimiento sin&#xE1;ptico, pero no en compensaci&#xF3;n de germinaci&#xF3;n. M (2) mAChRs estaban presentes &#xFA;nicamente en las neuronas motoras, mientras que M (1), M (3), y mAChRs M (5) se asociaron con las c&#xE9;lulas de Schwann y / o fibras musculares. El bloqueo de los cinco subtipos de mAchR con atropina evocado efectos pronunciados, incluyendo terminal de germinaci&#xF3;n, la retirada del terminal y atrofia de las fibras musculares. Por el contrario, methoctramina, una M (4.2) que prefieren el antagonista, el terminal induce la germinaci&#xF3;n y la retirada del terminal, pero no la atrofia de las fibras musculares. De acuerdo con esta observaci&#xF3;n, M (2) (- / -), pero no los ratones mutantes mAchR otros exhiben espont&#xE1;nea brotaci&#xF3;n acompa&#xF1;ado por la p&#xE9;rdida de muchas glorietas terminales padres. Terminal de brotes, sin embargo, no parece ser el defecto causal porque la p&#xE9;rdida parcial de las ramas terminales es com&#xFA;n incluso en las M (2) (- / -) NMJs sin brotar. In amyotrophic lateral sclerosis (ALS), the exogenous temporal triggers that result in initial motor neuron death are not understood. Overactivation and consequent accelerated loss of vulnerable motor neurons is one theory of disease initiation. The vulnerability of motor neurons in response to chronic peripheral nerve hyperstimulation was tested in the SOD1(G93A) rat model of ALS. A novel in vivo technique for peripheral phrenic nerve stimulation was developed via intra-diaphragm muscle electrode implantation at the phrenic motor endpoint. Chronic bilateral phrenic nerve hyperstimulation in SOD1(G93A) rats accelerated disease progression, including shortened lifespan, hastened motor neuron loss and increased denervation at diaphragm neuromuscular junctions. Hyperstimulation also resulted in focal decline in adjacent forelimb function. These results show that peripheral phrenic nerve hyperstimulation accelerates cell death of vulnerable spinal motor neurons, modifies both temporal and anatomical onset of disease, and leads to involvement of disease in adjacent anatomical regions in this ALS model.

Respiratory dysfunction is the leading cause of morbidity and mortality following traumatic spinal cord injury (SCI). Injuries targeting mid-cervical spinal cord regions affect the phrenic motor neuron pool that innervates the diaphragm, the primary respiratory muscle of inspiration. Contusion-type injury in the cervical spinal cord is one of the most common forms of human SCI; however, few studies have evaluated mid-cervical contusion in animal models or characterized consequent histopathological and functional effects of degeneration of phrenic motor neuron-diaphragm circuitry. In an attempt to target the phrenic motor neuron pool, two unilateral contusion injury paradigms were tested, a single injury at level C4 and a double injury both at levels C3 and C4, and animals were followed for up to 6 weeks post-injury. Both unilateral cervical injury paradigms are reproducible with no mortality or need for breathing assistance, and are accompanied by phrenic motor neuron loss, phrenic nerve axon degeneration, diaphragm atrophy, denervation and subsequent partial reinnervation at the diaphragm neuromuscular junction, changes in spontaneous diaphragm EMG recordings, and reduction in phrenic nerve compound muscle action potential amplitude. These findings demonstrate significant and chronically persistent respiratory compromise following mid-cervical SCI due to phrenic motor neuron degeneration. These injury paradigms and accompanying analyses provide important tools both for understanding mechanisms of phrenic motor neuron and diaphragm pathology following SCI and for evaluating therapeutic strategies in clinically relevant cervical SCI models.

Phrenic motor neuron degeneration compromises phrenic axonal circuitry and diaphragm activity in a unilateral cervical contusion model of spinal cord injury.

A primary cause of morbidity and mortality following cervical spinal cord injury (SCI) is respiratory compromise, regardless of the level of trauma. In particular, SCI at mid-cervical regions targets degeneration of both descending bulbospinal respiratory axons and cell bodies of phrenic motor neurons, resulting in deficits in the function of the diaphragm, the primary muscle of inspiration. Contusion-type trauma to the cervical spinal cord is one of the most common forms of human SCI; however, few studies have evaluated mid-cervical contusion in animal models or characterized consequent histopathological and functional effects of degeneration of phrenic motor neuron-diaphragm circuitry. We have generated a mouse model of cervical contusion SCI that unilaterally targets both C4 and C5 levels, the location of the phrenic motor neuron pool, and have examined histological and functional outcomes for up to 6 weeks post-injury. We report that phrenic motor neuron loss in cervical spinal cord, phrenic nerve axonal degeneration, and denervation at diaphragm neuromuscular junctions (NMJ) resulted in compromised ipsilateral diaphragm function, as demonstrated by persistent reduction in diaphragm compound muscle action potential amplitudes following phrenic nerve stimulation and abnormalities in spontaneous diaphragm electromyography (EMG) recordings. This injury paradigm is reproducible, does not require ventilatory assistance, and provides proof-of-principle that generation of unilateral cervical contusion is a feasible strategy for modeling diaphragmatic/respiratory deficits in mice. This study and its accompanying analyses pave the way for using transgenic mouse technology to explore the function of specific genes in the pathophysiology of phrenic motor neuron degeneration and respiratory dysfunction following cervical SCI.

Degeneration of phrenic motor neurons induces long-term diaphragm deficits following mid-cervical spinal contusion in mice.

Contusion-type cervical spinal cord injury (SCI) is one of the most common forms of SCI observed in patients. In particular, injuries targeting the C3-C5 region affect the pool of phrenic motor neurons (PhMNs) that innervates the diaphragm, resulting in significant and often chronic respiratory dysfunction. Using a previously described rat model of unilateral midcervical C4 contusion with the Infinite Horizon Impactor, we have characterized the early time course of PhMN degeneration and consequent respiratory deficits following injury, as this knowledge is important for designing relevant treatment strategies targeting protection and plasticity of PhMN circuitry. PhMN loss (48% of the ipsilateral pool) occurred almost entirely during the first 24 h post-injury, resulting in persistent phrenic nerve axonal degeneration and denervation at the diaphragm neuromuscular junction (NMJ). Reduced diaphragm compound muscle action potential amplitudes following phrenic nerve stimulation were observed as early as the first day post-injury (30% of pre-injury maximum amplitude), with slow functional improvement over time that was associated with partial reinnervation at the diaphragm NMJ. Consistent with ipsilateral diaphragmatic compromise, the injury resulted in rapid, yet only transient, changes in overall ventilatory parameters measured via whole-body plethysmography, including increased respiratory rate, decreased tidal volume, and decreased peak inspiratory flow. Despite significant ipsilateral PhMN loss, the respiratory system has the capacity to quickly compensate for partially impaired hemidiaphragm function, suggesting that C4 hemicontusion in rats is a model of SCI that manifests subacute respiratory abnormalities. Collectively, these findings demonstrate significant and persistent diaphragm compromise in a clinically relevant model of midcervical contusion SCI; however, the therapeutic window for PhMN protection is restricted to early time points post-injury. On the contrary, preventing loss of innervation by PhMNs and/or inducing plasticity in spared PhMN axons at the diaphragm NMJ are relevant long-term targets.

Early phrenic motor neuron loss and transient respiratory abnormalities after unilateral cervical spinal cord contusion.

Previous studies demonstrated that Schwann cells (SCs) express distinct motor and sensory phenotypes, which impact the ability of these pathways to selectively support regenerating neurons. In the present study, unbiased microarray analysis was used to examine differential gene expression in denervated motor and sensory pathways in rats. Several genes that were significantly upregulated in either denervated sensory or motor pathways were identified and two secreted factors were selected for further analysis: osteopontin (OPN) and clusterin (CLU) which were upregulated in denervated motor and sensory pathways, respectively. Sciatic nerve transection induced upregulation of OPN and CLU and expression of both returned to baseline levels with ensuing regeneration. In vitro analysis using exogenously applied OPN induced outgrowth of motor but not sensory neurons. CLU, however, induced outgrowth of sensory neurons, but not motor neurons. To assess the functional importance of OPN and CLU, peripheral nerve regeneration was examined in OPN and CLU(-/-) mice. When compared with OPN(+/+) mice, motor neuron regeneration was reduced in OPN(-/-) mice. Impaired regeneration through OPN(-/-) peripheral nerves grafted into OPN(+/+) mice indicated that loss of OPN in SCs was responsible for reduced motor regeneration. Sensory neuron regeneration was impaired in CLU(-/-) mice following sciatic nerve crush and impaired regeneration nerve fibers through CLU(-/-) nerve grafts transplanted into CLU(+/+) mice indicated that reduced sensory regeneration is likely due to SC-derived CLU. Together, these studies suggest unique roles for SC-derived OPN and CLU in regeneration of peripheral motor and sensory axons.

Novel roles for osteopontin and clusterin in peripheral motor and sensory axon regeneration.

A major portion of spinal cord injury (SCI) cases affect midcervical levels, the location of the phrenic motor neuron (PhMN) pool that innervates the diaphragm. While initial trauma is uncontrollable, a valuable opportunity exists in the hours to days following SCI for preventing PhMN loss and consequent respiratory dysfunction that occurs during secondary degeneration. One of the primary causes of secondary injury is excitotoxic cell death due to dysregulation of extracellular glutamate homeostasis. GLT1, mainly expressed by astrocytes, is responsible for the vast majority of functional uptake of extracellular glutamate in the CNS, particularly in spinal cord. We found that, in bacterial artificial chromosome-GLT1-enhanced green fluorescent protein reporter mice following unilateral midcervical (C4) contusion SCI, numbers of GLT1-expressing astrocytes in ventral horn and total intraspinal GLT1 protein expression were reduced soon after injury and the decrease persisted for ≥6 weeks. We used intraspinal delivery of adeno-associated virus type 8 (AAV8)-Gfa2 vector to rat cervical spinal cord ventral horn for targeting focal astrocyte GLT1 overexpression in areas of PhMN loss. Intraspinal delivery of AAV8-Gfa2-GLT1 resulted in transduction primarily of GFAP(+) astrocytes that persisted for ≥6 weeks postinjury, as well as increased intraspinal GLT1 protein expression. Surprisingly, we found that astrocyte-targeted GLT1 overexpression increased lesion size, PhMN loss, phrenic nerve axonal degeneration, and diaphragm neuromuscular junction denervation, and resulted in reduced functional diaphragm innervation as assessed by phrenic nerve-diaphragm compound muscle action potential recordings. These results demonstrate that GLT1 overexpression via intraspinal AAV-Gfa2-GLT1 delivery exacerbates neuronal damage and increases respiratory impairment following cervical SCI.

Overexpression of the astrocyte glutamate transporter GLT1 exacerbates phrenic motor neuron degeneration, diaphragm compromise, and forelimb motor dysfunction following cervical contusion spinal cord injury.

Approximately half of traumatic spinal cord injury (SCI) cases affect cervical regions, resulting in chronic respiratory compromise. The majority of these injuries affect midcervical levels, the location of phrenic motor neurons (PMNs) that innervate the diaphragm. A valuable opportunity exists following SCI for preventing PMN loss that occurs during secondary degeneration. One of the primary causes of secondary injury is excitotoxicity due to dysregulation of extracellular glutamate homeostasis. Astrocytes express glutamate transporter 1 (GLT1), which is responsible for the majority of CNS glutamate clearance. Given our observations of GLT1 dysfunction post-SCI, we evaluated intraspinal transplantation of Glial-Restricted Precursors (GRPs)--a class of lineage-restricted astrocyte progenitors--into ventral horn following cervical hemicontusion as a novel strategy for reconstituting GLT1 function, preventing excitotoxicity and protecting PMNs in the acutely injured spinal cord. We find that unmodified transplants express low levels of GLT1 in the injured spinal cord. To enhance their therapeutic properties, we engineered GRPs with AAV8 to overexpress GLT1 only in astrocytes using the GFA2 promoter, resulting in significantly increased GLT1 protein expression and functional glutamate uptake following astrocyte differentiation in vitro and after transplantation into C4 hemicontusion. Compared to medium-only control and unmodified GRPs, GLT1-overexpressing transplants reduced lesion size, diaphragm denervation and diaphragm dysfunction. Our findings demonstrate transplantation-based replacement of astrocyte GLT1 is a promising approach for SCI.

Transplantation of glial progenitors that overexpress glutamate transporter GLT1 preserves diaphragm function following cervical SCI.

Amyotrophic lateral sclerosis (ALS) is characterized by relatively rapid degeneration of both upper and lower motor neurons, with death normally occurring 2-5years following diagnosis primarily due to respiratory paralysis resulting from phrenic motor neuron (PhMN) loss and consequent diaphragm denervation. In ALS, cellular abnormalities are not limited to MNs. For example, decreased levels and aberrant functioning of the major central nervous system (CNS) glutamate transporter, GLT1, occur in spinal cord and motor cortex astrocytes of both humans with ALS and in SOD1(G93A) rodents, a widely studied ALS animal model. This results in dysregulation of extracellular glutamate homeostasis and consequent glutamate excitotoxicity, a primary mechanism responsible for MN loss in ALS animal models and in the human disease. Given these observations of GLT1 dysfunction in areas of MN loss, as well as the importance of testing therapeutic strategies for preserving PhMNs in ALS, we evaluated intraspinal delivery of an adeno-associated virus type 8 (AAV8)-Gfa2 vector to the cervical spinal cord ventral horn of SOD1(G93A) ALS mice for focally restoring intraspinal GLT1 expression. AAV8 was specifically injected into the ventral horn bilaterally throughout the cervical enlargement at 110days of age, a clinically-relevant time point coinciding with phenotypic/symptomatic disease onset. Intraspinal delivery of AAV8-Gfa2-GLT1 resulted in robust transduction primarily of GFAP(+) astrocytes that persisted until disease endstage, as well as a 2-3-fold increase in total intraspinal GLT1 protein expression in the ventral horn. Despite this robust level of astrocyte transduction and GLT1 elevation, GLT1 overexpression did not protect PhMNs, preserve histological PhMN innervation of the diaphragm NMJ, or prevent decline in diaphragmatic respiratory function as assessed by phrenic nerve-diaphragm compound muscle action potential (CMAP) recordings compared to control AAV8-Gfa2-eGFP injected mice. In addition, AAV-Gfa2-GLT1 did not delay forelimb disease onset, extend disease duration (i.e. time from either forelimb or hindlimb disease onsets to endstage) or prolong overall animal survival. These findings suggest that focal restoration of GLT1 expression in astrocytes of the cervical spinal cord using AAV delivery is not an effective therapy for ALS.
