The major structural elements of retroviruses are contained in a single polyprotein, Gag, which in human immunodeficiency virus type 1 (HIV-1) comprises the MA, CA, spacer peptide 1 (SP1), NC, SP2, and p6 polypeptides. In the immature HIV-1 virion, the domains of Gag are arranged radially with the N-terminal MA domain at the membrane and C-terminal NC-SP2-p6 region nearest to the center. Here, we report the three-dimensional structures of individual immature HIV-1 virions, as obtained by electron cryotomography. The concentric shells of the Gag polyprotein are clearly visible, and radial projections of the different Gag layers reveal patches of hexagonal order within the CA and SP1 shells. Averaging well-ordered unit cells leads to a model in which each CA hexamer is stabilized by a bundle of six SP1 helices. This model suggests why the SP1 spacer is essential for assembly of the Gag lattice and how cleavage between SP1 and CA acts as a structural switch controlling maturation.

Carboxysomes are organelle-like polyhedral bodies found in cyanobacteria and many chemoautotrophic bacteria that are thought to facilitate carbon fixation. Carboxysomes are bounded by a proteinaceous outer shell and filled with ribulose 1,5-bisphosphate carboxylase/oxygenase (RuBisCO), the first enzyme in the CO(2) fixation pathway, but exactly how they enhance carbon fixation is unclear. Here we report the three-dimensional structure of purified carboxysomes from Synechococcus species strain WH8102 as revealed by electron cryotomography. We found that while the sizes of individual carboxysomes in this organism varied from 114 nm to 137 nm, surprisingly, all were approximately icosahedral. There were on average approximately 250 RuBisCOs per carboxysome, organized into three to four concentric layers. Some models of carboxysome function depend on specific contacts between individual RuBisCOs and the shell, but no evidence of such contacts was found: no systematic patterns of connecting densities or RuBisCO positions against the shell's presumed hexagonal lattice could be discerned, and simulations showed that packing forces alone could account for the layered organization of RuBisCOs.

Carboxysomes sind begrenzt durch eine proteinogene Au&szlig;enh&uuml;lle und gef&uuml;llt mit Ribulose 1, 5-bisphosphat-Carboxylase/Oxygenase (RuBisCO), das erste Enzym der CO(2) Fixierung, aber genau, wie sie CO2-Fixierung erh&ouml;hen, ist unklar. Hier berichten wir die dreidimensionale Struktur des gereinigten Carboxysomes von Synechococcus Arten Stamm WH8102, wie durch Elektronen Cryotomography aufgedeckt. Wir fanden, dass w&auml;hrend die Gr&ouml;&szlig;en der einzelnen Carboxysomes in diesem Organismus schwankte zwischen 114 nm bis 137 nm, &uuml;berrascht waren alle ungef&auml;hr ikosaedrischen. Im Durchschnitt gab es etwa 250 RuBisCOs pro Carboxysome, in drei bis vier konzentrischen Schichten organisiert. Carboxysomes est&#xE1;n delimitadas por una carcasa exterior prote&#xED;nico y lleno de ribulosa 1,5-bisfosfato carboxilasa / oxigenasa (Rubisco), la primera enzima en el CO (2) v&#xED;a la fijaci&#xF3;n, pero exactamente c&#xF3;mo mejorar la fijaci&#xF3;n de carbono no est&#xE1; claro. A new method for recording both fluorescence and cryo-EM images of small bacterial cells was developed and used to identify chemoreceptor arrays in cryotomograms of intact Caulobacter crescentus cells. We show that in wild-type cells preserved in a near-native state, the chemoreceptors are hexagonally packed with a lattice spacing of 12 nm, just a few tens of nanometers away from the flagellar motor that they control. The arrays were always found on the convex side of the cell, further demonstrating that Caulobacter cells maintain dorsal/ventral as well as anterior/posterior asymmetry. Placing the known crystal structure of a trimer of receptor dimers at each vertex of the lattice accounts well for the density and agrees with other constraints. Based on this model for the arrangement of receptors, there are between one and two thousand receptors per array.

Eine neue Methode zur Erfassung von Fluoreszenz und Cryo-EM-Bilder der kleinen bakteriellen Zellen entwickelt und Chemorezeptor-Arrays in Cryotomograms intakt Caulobacter Crescentus Zellen identifiziert. Wir zeigen, dass in Wildtyp Zellen in einem fast Native Zustand beibehalten, die Chemoreceptors hexagonal mit Gitter Abstand von 12 verpackt sind nm, nur ein paar Dutzend Nanometer abseits der Flimmerband Motor, die sie kontrollieren. Die Arrays wurden immer gefunden, auf der konvexen Seite der Zelle, zeigt, dass Caulobacter Zellen dorsal/Ventral sowie anterior/Posterior Asymmetrie aufrechterhalten. Platzieren die bekannten Kristallstruktur von einem Trimer des Rezeptor-Dimer an jeden Eckpunkt des Gitters entfallen auch die Dichte und stimmt mit anderen Einschr&auml;nkungen. Auf der Grundlage dieses Modells f&uuml;r die Anordnung der Rezeptoren, gibt es zwischen einem und zwei tausend Rezeptoren pro Array.

Un nuovo metodo per registrazione immagini cryo-EM di piccole cellule batteriche e di fluorescenza &egrave; stato sviluppato e utilizzato per identificare matrici chemoreceptor in cryotomograms di Caulobacter ha celle intatte. Mostriamo che in cellule wild-type conservate in uno stato quasi nativa, i chemocettori esagonale sono confezionati con una spaziatura di reticolo di 12 nm, solo poche decine di nanometri dal motore flagellari che essi controllano. Le matrici sono state trovate sempre sul lato convesso della cella, ulteriore dimostrazione che le cellule Caulobacter mantengano dorsale/ventrale cos&igrave; come asimmetria anteriore/posteriore. Posizionando la struttura di cristallo conosciuto di un trimero di dimeri di recettore per ogni vertice del reticolo rappresenta bene la densit&agrave; e concorda con altri vincoli. Basato su questo modello per la disposizione dei recettori, ci sono tra uno e due mila recettori per array.

Nos muestran que en c&#xE9;lulas de tipo salvaje se conservan en un estado casi nativo, los quimiorreceptores se hexagonal lleno de un espaciado reticular de 12 nm, a s&#xF3;lo unas pocas decenas de nan&#xF3;metros de distancia del motor flagelar que ellos controlan. Las matrices se encuentran siempre en el lado convexo de la c&#xE9;lula, lo que demuestra que las c&#xE9;lulas Caulobacter mantener dorsal / ventral as&#xED; como anterior / posterior asimetr&#xED;a.
