We have recently devised a method to quantify interactions between a membrane protein ("bait") and a fluorophore-labeled protein ("prey") directly in the live-cell plasma membrane (Schwarzenbacher et al. Nature Methods 5:1053-1060 2008). The idea is to seed cells on surfaces containing micro-patterned antibodies against the exoplasmic domain of the bait, and monitor the co-patterning of the fluorescent prey via fluorescence microscopy. Here, we characterized the time course of bait and prey micropattern formation upon seeding the cells onto the micro-biochip. Patterns were formed immediately after contact of the cells with the surface. Cells were able to migrate over the chip surface without affecting the micropattern contrast, which remained constant over hours. On single cells, bait contrast may be subject to fluctuations, indicating that the bait can be released from and recaptured on the micropatterns. We conclude that interaction studies can be performed at any time-point ranging from 5 min to several hours post seeding. Monitoring interactions with time opens up the possibility for new assays, which are briefly sketched in the discussion section.

Recientemente, hemos ideado un m&#xE9;todo para cuantificar las interacciones entre una prote&#xED;na de membrana (&#x26;quot;carnada&#x26;quot;) y una prote&#xED;na marcada con fluor&#xF3;foro (&#x26;quot;presa&#x26;quot;) directamente en la membrana plasm&#xE1;tica de c&#xE9;lulas vivas (Schwarzenbacher et al. Nature Methods 5:1053-1060 2008 ). La idea es que las c&#xE9;lulas de semilla en las superficies que contienen micro-estampados anticuerpos contra el dominio exoplasmic del cebo, y monitorear la co-patr&#xF3;n de la presa fluorescente a trav&#xE9;s de microscop&#xED;a de fluorescencia. En este caso, se caracteriz&#xF3; la evoluci&#xF3;n en el tiempo de cebo y la formaci&#xF3;n de presas micropattern a la siembra de las c&#xE9;lulas en el micro-biochip. Los patrones se formaron inmediatamente despu&#xE9;s del contacto de las c&#xE9;lulas con la superficie. Las c&#xE9;lulas fueron capaces de migrar a la superficie del chip sin afectar el contraste micropattern, que se mantuvo constante durante horas. En las c&#xE9;lulas individuales, contraste cebo puede estar sujeto a fluctuaciones, lo que indica que el cebo puede ser liberado desde y recapturados en las micropatrones. Se concluye que los estudios de interacci&#xF3;n se puede realizar en cualquier punto de tiempo que oscila desde 5 minutos a varias horas despu&#xE9;s de la siembra. We investigated the mobility and phase-partitioning of the fluorescent oxidized phospholipid analogue 1-palmitoyl-2-glutaroyl-sn-glycero-3-phospho-N-Alexa647-ethanolamine (PGPE-Alexa647) in supported lipid bilayers. Compared to the conventional phospholipid dihexadecanoylphosphoethanolamine (DHPE)-Bodipy we found consistently higher diffusion constants. The effect became dramatic when immobile obstacles were inserted into the bilayer, which essentially blocked the diffusion of DHPE-Bodipy but hardly influenced the movements of PGPE-Alexa647. In a supported lipid bilayer made of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), the differences in probe mobility leveled off with increasing cholesterol content. Using coarse-grained molecular dynamics simulations, we could ascribe this effect to increased interactions between the oxidized phospholipid and the membrane matrix, concomitant with a translation in the headgroup position of the oxidized phospholipid: at zero cholesterol content, its headgroup is shifted to the outside of the DOPC headgroup region, whereas increasing cholesterol concentrations pulls the headgroup into the bilayer plane.

Utilisation grossiers simulations de dynamique mol&#xE9;culaire, nous avons pu attribuer cet effet &#xE0; des interactions accrues entre le phospholipide oxyd&#xE9; et la matrice de la membrane, concomitante avec une traduction dans la position groupe de t&#xEA;te du phospholipide oxyd&#xE9;: &#xE0; z&#xE9;ro le taux de cholest&#xE9;rol, de son groupe de t&#xEA;te se d&#xE9;place vers l&#x26;#39;ext&#xE9;rieur de la r&#xE9;gion DOPC groupe de t&#xEA;te, alors que l&#x26;#39;augmentation des concentrations de cholest&#xE9;rol tire le groupe de t&#xEA;te dans le plan bicouche.

Wir untersuchten die Mobilit&auml;t und die Phase-Partitionierung der fluoreszierenden oxidierte Phospholipide analogen 1-palmitoyl-2-glutaroyl-sn-glycero-3-phospho-N-Alexa647-ethanolamine (PGPE-Alexa647) in unterst&uuml;tzten Lipid-Doppelschichten. Im Vergleich zu den konventionellen Phospholipid-Dihexadecanoylphosphoethanolamine (DHPE)-Bodipy fanden wir durchweg h&ouml;here Verbreitung-Konstanten. Der Effekt wurde dramatisch, als unbewegliche Hemmnisse in der Bilayer eingef&uuml;gt wurden, die im Wesentlichen die Verbreitung der DHPE-Bodipy blockiert, aber kaum beeinflusst die Bewegungen der PGPE-Alexa647. In einem unterst&uuml;tzten Lipid-Bilayer hergestellt aus 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) die Unterschiede in der Sonde Mobilit&auml;t Sonnenhelligkeit mit zunehmenden Cholesteringehalt. Mit grobk&ouml;rnigen Molekulardynamik-Simulationen, konnten wir diesen Effekt f&uuml;hren, auf die erh&ouml;hte Interaktionen zwischen die oxidierte Phospholipide und die Membran-Matrix, damit einhergehende mit einer &Uuml;bersetzung in die Headgroup Position oxidierte Phospholipide: bei Null Cholesteringehalt, seine Headgroup an der Au&szlig;enseite der DOPC Headgroup Region, verschoben ist, w&auml;hrend erh&ouml;hen Cholesterin-Konzentrationen der Headgroup in die Bilayer-Ebene zieht.

Cholesterin verlangsamt die seitlichen Mobilit&auml;t eine oxidierte Phospholipide in einer unterst&uuml;tzten Lipid Bilayer.

Abbiamo studiato la mobilit&agrave; e la fase di partizionamento del fluorescente fosfolipidi ossidati analogo 1-palmitoyl-2-glutaroyl-sn-glycero-3-phospho-N-Alexa647-ethanolamine (PGPE-Alexa647) in lipidici supportati. Rispetto alla dihexadecanoylphosphoethanolamine fosfolipide convenzionale (DHPE)-Bodipy abbiamo trovato costanti di diffusione sempre maggiore. L&#39;effetto &egrave; diventato drammatico quando ostacoli immobili sono stati inseriti nel doppio strato, che sostanzialmente bloccato la diffusione del DHPE-Bodipy ma difficilmente influenzato i movimenti di PGPE-Alexa647. In un doppio strato lipidico supportati di 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), le differenze di mobilit&agrave; sonda livellata con maggiore contenuto di colesterolo. Utilizzando simulazioni dinamiche molecolari grossolana, potremmo attribuire questo effetto aumentato interazioni tra i fosfolipidi ossidati e la matrice della membrana, concomitante con una traduzione nella posizione dei fosfolipidi ossidati capigruppo: a zero contenuto di colesterolo, suoi capigruppo &egrave; spostato all&#39;esterno della regione DOPC capigruppo, mentre aumentando le concentrazioni di colesterolo tira i capigruppo nel piano doppio strato.

Hemos investigado la movilidad y la fase-particionamiento del an&#xE1;logo de fosfol&#xED;pido fluorescente oxidado 1-palmitoil-2-glutaroyl-sn-glicero-3-fosfato-N-Alexa647-etanolamina (PGPE-Alexa647) en bicapas lip&#xED;dicas soportadas. En comparaci&#xF3;n con el fosfol&#xED;pido dihexadecanoylphosphoethanolamine convencional (DHPE)-Bodipy hemos encontrado consistentemente m&#xE1;s altos constantes de difusi&#xF3;n. El efecto fue dram&#xE1;tico cuando los obst&#xE1;culos inm&#xF3;viles fueron insertadas en la bicapa, que esencialmente bloqueado la difusi&#xF3;n de DHPE-Bodipy pero apenas influy&#xF3; en los movimientos de PGPE-Alexa647. En una bicapa lip&#xED;dica apoyado hecha de 1,2-dioleoil-sn-glicero-3-fosfocolina (DOPC), las diferencias en la movilidad de la sonda se nivel&#xF3; con contenido de colesterol en aumento. Uso de grano grueso simulaciones de din&#xE1;mica molecular, se podr&#xED;a atribuir este efecto a las interacciones entre el aumento de la fosfol&#xED;pido oxidado y la matriz de la membrana, concomitante con una traducci&#xF3;n en la posici&#xF3;n de grupo de cabeza del fosfol&#xED;pido oxidado: contenido de colesterol en cero, su grupo de cabeza se desplaza hacia el exterior de la regi&#xF3;n DOPC grupo de cabeza, mientras que aumentando las concentraciones de colesterol tira del grupo de cabeza en el plano bicapa.

Cationic antimicrobial peptides (CAMPs) selectively target bacterial membranes by electrostatic interactions with negatively charged lipids. It turned out that for inhibition of microbial growth a high CAMP membrane concentration is required, which can be realized by the incorporation of hydrophobic groups within the peptide. Increasing hydrophobicity, however, reduces the CAMP selectivity for bacterial over eukaryotic host membranes, thereby causing the risk of detrimental side-effects. In this study we addressed how cationic amphipathic peptides-in particular a CAMP with Lysine-Leucine-Lysine repeats (termed KLK)-affect the localization and dynamics of molecules in eukaryotic membranes. We found KLK to selectively inhibit the endocytosis of a subgroup of membrane proteins and lipids by electrostatically interacting with negatively charged sialic acid moieties. Ultrastructural characterization revealed the formation of membrane invaginations representing fission or fusion intermediates, in which the sialylated proteins and lipids were immobilized. Experiments on structurally different cationic amphipathic peptides (KLK, 6-MO-LF11-322 and NK14-2) indicated a cooperation of electrostatic and hydrophobic forces that selectively arrest sialylated membrane constituents.

Kationische antimikrobielle Peptide (CAMPs) Ziel selektiv bakterielle Membranen durch elektrostatische Wechselwirkungen mit negativ geladenen Lipiden. Es stellte sich heraus, die f&uuml;r die Hemmung der mikrobiellen Wachstum eine hohe Lager-Membran-Konzentration erforderlich ist, kann durch eine hydrophobe Gruppen innerhalb der Peptid realisiert werden. Erh&ouml;hung der Hydrophobie, reduziert jedoch die CAMP Selektivit&auml;t f&uuml;r bakteriell &uuml;ber eukaryotischen Host Membranen, wodurch das Risiko von sch&auml;dlichen Nebenwirkungen. In dieser Studie, die wir angesprochen wie kationische AMPHIPHILE Peptide-in bestimmten CAMP mit Lysin-Leucin-Lysin wird wiederholt (genannt KLK)-beeinflussen die Lokalisierung und die Dynamik von Molek&uuml;len in eukaryotischen Membranen. Wir fanden die KLK selektiv hemmen der Endozytose eine Untergruppe der Membranproteine und Lipide durch die elektrostatisch Interaktion mit negativ geladenen Sialins&auml;ure Polymerharze. Ultrastrukturelle Charakterisierung ergab die Bildung von Membran-einst&uuml;lpungen, die Kernspaltung oder Fusion Zwischenprodukte, in denen die Sialylated Proteinen und Lipiden immobilisiert wurden darstellt. Experimente auf strukturell unterschiedliche kationische AMPHIPHILE Peptide (KLK, 6-MO-LF11-322 und NK14-2) angegeben, eine Kooperation von elektrostatischen und hydrophobe Kr&auml;fte, die Sialylated Membrane Bestandteile selektiv zu verhaften.

Kationische AMPHIPHILE Peptide ansammeln Sialylated Proteinen und Lipiden in der Plasmamembran der eukaryotischen Wirtszellen.

Peptidi antimicrobici cationici (accampamenti) bersaglio in modo selettivo membrane batteriche da interazioni elettrostatiche con i lipidi caricati negativamente. Si &egrave; scoperto che per l&#39;inibizione della crescita microbica una concentrazione elevata membrana CAMP &egrave; richiesto, che pu&ograve; essere realizzato per l&#39;incorporazione di gruppi idrofobi all&#39;interno del peptide. Aumentando idrofobicit&agrave;, tuttavia, si riduce la selettivit&agrave; CAMP per batterico sopra membrane eucariote ospite, causando cos&igrave; il rischio di effetti collaterali dannosi. In questo studio abbiamo affrontato come cationico amphipathic peptidi-in particolare un CAMP con lisina-leucina-lisina ripete (chiamato KLK)-possono influenzare la localizzazione e la dinamica delle molecole nelle membrane eucariote. Abbiamo trovato KLK di inibire selettivamente l&#39;endocitosi di un sottogruppo di proteine di membrana e lipidi interagendo elettrostaticamente con acido sialico caricato negativamente moiety. Caratterizzazione ultrastrutturale ha rivelato la formazione dei invaginations membrana che rappresentano intermedi di fissione o fusione, in cui il sialilato proteine e lipidi sono stati immobilizzati. Esperimenti su amphipathic cationico strutturalmente diversi peptidi (KLK, 6-MO-LF11-322 e NK14-2) ha indicato una cooperazione delle forze elettrostatiche e idrofobiche che arrestare selettivamente costituenti di membrana sialilato.

P&#xE9;ptidos cati&#xF3;nicos antimicrobianos (campamentos) atacar selectivamente las membranas bacterianas por interacciones electrost&#xE1;ticas con los l&#xED;pidos cargados negativamente. Result&#xF3; que para la inhibici&#xF3;n del crecimiento microbiano una concentraci&#xF3;n CAMP membrana se requiere una alta, que puede ser realizado por la incorporaci&#xF3;n de grupos hidr&#xF3;fobos en el p&#xE9;ptido. El aumento de la hidrofobicidad, sin embargo, reduce la selectividad CAMP para bacteriana sobre membranas hu&#xE9;sped eucari&#xF3;ticas, causando con ello el riesgo de efectos secundarios perjudiciales. En este estudio se abord&#xF3; la forma de p&#xE9;ptidos cati&#xF3;nicos anfip&#xE1;ticos en particular, un campamento con lisina-leucina-lisina se repite (denominado KLK)-afecta a la localizaci&#xF3;n y la din&#xE1;mica de las mol&#xE9;culas en las membranas eucariotas. Hemos encontrado KLK para inhibir selectivamente la endocitosis de un subgrupo de prote&#xED;nas y l&#xED;pidos de membrana por electrost&#xE1;ticamente interactuar con carga negativa restos de &#xE1;cido si&#xE1;lico. Caracterizaci&#xF3;n ultraestructural revel&#xF3; la formaci&#xF3;n de invaginaciones de membrana que representan compuestos intermedios de fisi&#xF3;n o fusi&#xF3;n, en los que las prote&#xED;nas y los l&#xED;pidos sialiladas fueron inmovilizados. Here, we measure the actin cytoskeleton arrangement of different morphological states of human platelets using a new protocol for photo-switching of rhodamine class fluorophores. A new medium composition was established for imaging the cytoskeleton using Alexa Fluor 488 conjugated to phalloidin. Morphological states of platelets bound to a glass substrate are visualized and quantified by two-dimensional localization microscopy at nanoscopic resolution. Marker-less drift correction yields localization of individual Alexa 488 conjugated to phalloidin with a positional accuracy of 12 nm.

Localization Microscopy of Actin Cytoskeleton in Human Platelets.

Mobility of proteins and lipids plays a major role in physiological processes. Platforms which were developed to study protein interaction between immobilized and mobile proteins suffer from shortcomings such as fluorescence quenching or complicated fabrication methods. Here we report a versatile platform comprising immobilized histidine-tagged proteins and biotinylated proteins in a mobile phase. Importantly, multiphoton photolithography was used for easy and fast fabrication of the platform and allows, in principle, extension of its application to three dimensions. The platform, which is made up of functionalized polymer structures embedded in a mobile lipid bilayer, shows low background fluorescence and allows for mobility of arbitrary proteins.

Proteins on Supported Lipid Bilayers Diffusing around Proteins Fixed on Acrylate Anchors.

microRNAs (miRNAs) are post-transcriptional regulators of messenger RNA (mRNA), and transported through the whole organism by-but not limited to-lipoprotein particles. Here, we address the miRNA profile in serum and lipoprotein particles of healthy individuals in comparison with patients with uremia. Moreover, we quantitatively determined the cellular lipoprotein-particle-uptake dependence on the density of lipoprotein particle receptors and present a method for enhancement of the transfer efficiency. We observed a significant increase of the cellular miRNA level using reconstituted high-density lipoprotein (HDL) particles artificially loaded with miRNA, whereas incubation with native HDL particles yielded no measurable effect. Thus, we conclude that no relevant effect of lipoprotein-particle-mediated miRNA-transfer exists under in vivo conditions though the miRNA profile of lipoprotein particles can be used as a diagnostic marker.
