We demonstrate tunable, fluidic, two-dimensional diffraction gratings based on a microfluidic platform comprising a flow-focusing bubble generator and flowing, regular lattices of bubbles formed by dynamic self-assembly. The structure of these lattices can be tuned with switching times of less than ten seconds by changing the pressures and rates of flow applied to the device. These diffraction gratings exhibit high stability (over hours of operation if properly designed and operated). For our devices, we achieved tunable ranges in pitch from 12 to 51 microm, corresponding to first-order diffraction angles from 3.2 degrees to 0.7 degrees for light with a wavelength of 632 nm.

A method is described for the formation of stable, composite aqueous emulsions of 1) combinations of distinct families of bubbles of nitrogen, 2) combinations of distinct families of droplets of an organic fluid (either perfluoro(methyl)decalin or hexadecane), and 3) combinations of bubbles and droplets. A system of two or three microfluidic flow-focusing units is coupled to a single outlet channel. The composite emulsions can be precisely tuned, both in their composition and in the number fraction of components--either bubbles or droplets--of different types. The use of microfluidic technology, with closely coupled flow-focusing units, guarantees that the emulsions are mixed locally at a controlled local stoichiometry. The emulsions self-assemble in a nonequilibrium process to form a wide variety of highly organized periodic lattices.

Synthesis of composite emulsions and complex foams with the use of microfluidic flow-focusing devices.

This paper describes the mechanism of formation of bubbles of nitrogen in water containing Tween 20 as a surfactant, and of droplets of water in hexadecane containing Span 80 as a surfactant. The study of these microfluidic systems compares two or four flow-focusing generators coupled through shared inlets, supplying the continuous phase, and through a common outlet channel. The processes that form bubbles in neighboring generators interact for a wide range of flow parameters; the formation of bubbles alternates in time and space, and the bubbles assemble into complex patterns in the outlet channel. The dynamics of formation of bubbles in these systems are stable for long time (at least 10 min). For a certain range of flow parameters, the coupled flow-focusing generators exhibit two stable modes of operation for a single set of flow parameters. The dynamics of formation of droplets of water in hexadecane by the coupled flow-focusing generators are simpler--the adjacent generators produce only monodisperse droplets over the entire range of flow parameters that are explored. These observations suggest that the mechanism of interaction between coupled flow-focusing generators relies on the compressibility of the dispersed phase (e.g., the gas or liquid), and on variations in pressure at the flow-focusing orifices induced by the breakup of bubbles or droplets.

Degradable microparticles have broad utility as vehicles for drug delivery and form the basis of several therapies approved by the US Food and Drug Administration. Conventional emulsion-based methods of manufacturing produce particles with a wide range of diameters (and thus kinetics of release) in each batch. This paper describes the fabrication of monodisperse, drug-loaded microparticles from biodegradable polymers using the microfluidic flow-focusing (FF) devices and the drug-delivery properties of those particles. Particles are engineered with defined sizes, ranging from 10 microm to 50 microm. These particles are nearly monodisperse (polydispersity index = 3.9%). A model amphiphilic drug (bupivacaine) is incorporated within the biodegradable matrix of the particles. Kinetic analysis shows that the release of the drug from these monodisperse particles is slower than that from conventional methods of the same average size but a broader distribution of sizes and, most importantly, exhibit a significantly lower initial burst than that observed with conventional particles. The difference in the initial kinetics of drug release is attributed to the uniform distribution of the drug inside the particles generated using the microfluidic methods. These results demonstrate the utility of microfluidic FF for the generation of homogenous systems of particles for the delivery of drugs.

Preparation of monodisperse biodegradable polymer microparticles using a microfluidic flow-focusing device for controlled drug delivery.

Les m&eacute;thodes conventionnelles &agrave; base d&#39;&eacute;mulsion de fabrication produisent des particules avec une large gamme de diam&egrave;tres (et donc la cin&eacute;tique de lib&eacute;ration) dans chaque lot. Cet article d&eacute;crit la fabrication de monodispers&eacute;, drogue-charg&eacute; des microparticules de polym&egrave;res biod&eacute;gradables en utilisant les dispositifs microfluidiques flux de focalisation (FF) et les propri&eacute;t&eacute;s de l&#39;administration de m&eacute;dicaments de ces particules. This paper describes a microfluidic instrument that produces drops of supercooled water suspended in a moving stream of liquid fluorocarbon, and measures the temperatures at which ice nucleates in the drops. A microfluidic chip containing a monodisperse drop generator and a straight channel with 38 embedded resistance thermometers was placed in contact with a seven-zone temperature-control plate and imaged under a microscope with a high-speed camera. This instrument can record the freezing temperatures of tens of thousands of drops within minutes, with an accuracy of 0.4 degrees C. The ice-nucleation temperatures in approximately 80-microm drops were reported for the freezing of 37 061 drops of pure water, and of 8898 drops of water seeded with silver iodide. Nucleation of ice in pure water was homogenous and occurred at temperatures between -36 and -37.8 degrees C, while water containing silver iodide froze between -10 and -19 degrees C. The instrument recorded the largest sets of individual freezing temperatures (37 061), had the fastest data acquisition rate (75 measurements/s), and the best optical (3 microm) and temporal (70 micros) resolutions among instruments designed to study nucleation of ice. The dendritic growth of ice in 150-microm drops of supercooled water at -35 degrees C was observed and imaged at a rate of 16 000 frames/s.

This article describes a new procedure for generating and transmitting a message--a sequence of optical pulses--by aligning a mask (an opaque sheet containing transparent "windows") below a microfluidic channel in which flows an opaque continuous fluid containing transparent droplets. The optical mask encodes the message as a unique sequence of windows that can transmit or block light; the flow of transparent droplets in the channel converts this message into a sequence of optical pulses. The properties of the windows on the mask (e.g., their size, wavelength of transmittance, orientation of polarization) determine the information carried in these optical pulses (e.g., intensity, color, polarization). The structure of a transmitted signal depends on the number and spacing of droplets in the channel. Fourier transformation can deconvolve superimposed signals created by the flow of multiple droplets into the message that a single droplet would transmit. The research described in this contribution explores a new field at the intersection of chemistry, materials science, and information technology: infochemistry.

Infochemistry: encoding information as optical pulses using droplets in a microfluidic device.

This Account describes a strategy for fabricating multicomponent microsystems in which the structures of essentially all of the components are formed in a single step of micromolding. This strategy, which we call "cofabrication", is an alternative to multilayer microfabrication, in which multiple layers of components are sequentially aligned ("registered") and deposited on a substrate by photolithography. Cofabrication has several characteristics that make it an especially useful approach for building multicomponent microsystems. It rapidly and inexpensively generates correctly aligned components (for example, wires, heaters, magnetic field generators, optical waveguides, and microfluidic channels) over very large surface areas. By avoiding registration, the technique does not impose on substrates the size limitations of common registrations tools, such as steppers and contact aligners. We have demonstrated multicomponent microsystems with surface areas exceeding 100 cm(2), but in principle, device size is only limited by the requirements of generating the original master. In addition, cofabrication can serve as a low-cost strategy for building microsystems. The technique is amenable to a variety of laboratory settings and uses fabrication tools that are less expensive than those used for multistep microfabrication. Moreover, the process requires only small amounts of solvent and photoresist, a costly chemical required for photolithography; in cofabrication, photoresist is applied and developed only once to produce a master, which is then used to produce multiple copies of molds containing the microfluidic channels. From a broad perspective, cofabrication represents a new processing paradigm in which the exterior (or shell) of the desired structures are produced before the interior (or core). This approach, generating the insulation or packaging structure first and injecting materials that provide function in channels in liquid phase, makes it possible to design and build microsystems with component materials that cannot be easily manipulated conventionally (such as solid materials with low melting points, liquid metals, liquid crystals, fused salts, foams, emulsions, gases, polymers, biomaterials, and fragile organics). Moreover, materials can be altered, removed, or replaced after the manufacturing stage. For example, cofabrication allows one to build devices in which a liquid flows through the device during use, or is replaced after use. Metal wires can be melted and reset by heating (in principle, repairing a break). This method leads to certain kinds of structures, such as integrated metallic wires with large cross-sectional areas or optical waveguides aligned in the same plane as microfluidic channels, that would be difficult or impossible to make with techniques such as sputter deposition or evaporation. This Account outlines the strategy of cofabrication and describes several applications. Specifically, we highlight cofabricated systems that combine microfluidics with (i) electrical wires for microheaters, electromagnets, and organic electrodes, (ii) fluidic optical components, such as optical waveguides, lenses, and light sources, (iii) gels for biological cell cultures, and (iv) droplets for compartmentalized chemical reactions, such as protein crystallization.

Cofabrication: a strategy for building multicomponent microsystems.

The formation of bubbles in a flow-focusing (FF) junction comprising multiple rectangular sections is described. The simplest junctions comprise two sections (throat and orifice). Systematic investigation of the influence on the formation of bubbles of the flow of liquid and the geometry of the junction identifies regimes that generate monodisperse, bidisperse, and tridisperse trains of bubbles. The mechanisms by which these junctions form monodisperse and bidisperse bubbles are inferred from the shapes of the gas thread during breakup: these mechanisms differ primarily by the process in which the gas thread collapses in the throat and/or orifice. The dynamic self-assembly of bidisperse bubbles leads to unexpected groupings of bubbles during their flow along the outlet channel.

This paper describes the behavior of bubbles suspended in a carrier liquid and moving within microfluidic networks of different connectivities. A single-phase continuum fluid, when flowing in a network of channels, partitions itself among all possible paths connecting the inlet and outlet. The flow rates along different paths are determined by the interaction between the fluid and the global structure of the network. That is, the distribution of flows depends on the fluidic resistances of all channels of the network. The movement of bubbles of gas, or droplets of liquid, suspended in a liquid can be quite different from the movement of a single-phase liquid, especially when they have sizes slightly larger than the channels, so that the bubbles (or droplets) contribute to the fluidic resistance of a channel when they are transiting it. This paper examines bubbles in this size range; in the size range examined, the bubbles are discrete and do not divide at junctions. As a consequence, a single bubble traverses only one of the possible paths through the network, and makes a sequence of binary choices ("left" or "right") at each branching intersection it encounters. We designed networks so that, at each junction, a bubble enters the channel into which the volumetric flow rate of the carrier liquid is highest. When there is only a single bubble inside a network at a time, the path taken by the bubble is, counter-intuitively, not necessarily the shortest or the fastest connecting the inlet and outlet. When a small number of bubbles move simultaneously through a network, they interact with one another by modifying fluidic resistances and flows in a time dependent manner; such groups of bubbles show very complex behaviors. When a large number of bubbles (sufficiently large that the volume of the bubbles occupies a significant fraction of the volume of the network) flow simultaneously through a network, however, the collective behavior of bubbles-the fluxes of bubbles through different paths of the network-can resemble the distribution of flows of a single-phase fluid.

En este trabajo se describe el comportamiento de las burbujas suspendidas en un l&#xED;quido portador y se desplazan dentro de las redes de microfluidos de conectividades diferentes. Una sola fase fluido continuo, cuando fluye en una red de canales, las particiones en s&#xED; entre todos los posibles caminos que conectan la entrada y la salida. Las tasas de flujo a lo largo de caminos diferentes est&#xE1;n determinadas por la interacci&#xF3;n entre el fluido y la estructura global de la red. Esto es, la distribuci&#xF3;n de los flujos depende de las resistencias flu&#xED;dicas de todos los canales de la red. This paper describes a general scheme to fabricate microchannels from curable polymers on a laboratory benchtop. Using the scheme described here, benchtop fabrication of SU-8 microfluidic systems was demonstrated for the first time, and their compatibility with organic solvents was demonstrated. The fabrication process has three major stages: 1) transferring patterns of microchannels to polymer films by molding, 2) releasing the patterned film and creating inlets and outlets for fluids, and 3) sealing two films together to create a closed channel system. Addition of a PDMS slab supporting the polymer film provided structural integrity during and after fabrication, allowing manipulation of the polymer films without fracturing or deformation. SU-8 channels fabricated according to this scheme exhibited solvent compatibility against continuous exposure to acetone and ethylacetate, which are incompatible with native PDMS. Using the SU-8 channels, continuous generation of droplets of ethylacetate, and templated synthesis of poly (lactic-co-glycolic acid) (PLGA) microparticles, both with stable size, were demonstrated continuously over 24 h, and at intervals over 75 days.

Benchtop fabrication of microfluidic systems based on curable polymers with improved solvent compatibility.

Bacterial sepsis is a serious clinical condition that can lead to multiple organ dysfunction and death despite timely treatment with antibiotics and fluid resuscitation. We have developed an approach to clearing bacteria and endotoxin from the bloodstream, using magnetic nanoparticles (MNPs) modified with bis-Zn-DPA, a synthetic ligand that binds to both Gram-positive and Gram-negative bacteria. Magnetic microfluidic devices were used to remove MNPs bound to Escherichia coli , a Gram-negative bacterium commonly implicated in bacterial sepsis, from bovine whole blood at flows as high as 60 mL/h, resulting in almost 100% clearance. Such devices could be adapted to clear bacteria from septicemic patients.

Synthetic ligand-coated magnetic nanoparticles for microfluidic bacterial separation from blood.

This review surveys selected methods of manufacture and applications of microdevices-miniaturized functional devices capable of handling cell and tissue cultures or producing particles-and discusses their potential relevance to nanomedicine. Many characteristics of microdevices such as miniaturization, increased throughput, and the ability to mimic organ-specific microenvironments are promising for the rapid, low-cost evaluation of the efficacy and toxicity of nanomaterials. Their potential to accurately reproduce the physiological environments that occur in vivo could reduce dependence on animal models in pharmacological testing. Technologies in microfabrications and microfluidics are widely applicable for nanomaterial synthesis and for the development of diagnostic devices. Although the use of microdevices in nanomedicine is still in its infancy, these technologies show promise for enhancing fundamental and applied research in nanomedicine.
