The complexity of micropatterned cell constructs has been limited by difficulties in patterning more than two surface components on a culture substrate. Photolithography using multiple aligned masks is well established for generalized multicomponent patterning, but is often too harsh for biomolecules. We report a two-mask photolithographic process that is tuned to preserve bioactivity in patterns composed of covalently coupled poly(ethylene glycol) (PEG), adsorbed extracellular matrix protein (e.g., collagen I), and adsorbed serum proteins (e.g., vitronectin). Thereby, we pattern two cell types-primary hepatocytes and 3T3 fibroblasts-demonstrating control over contact and spacing (20-200 microm) between the two cell types for over one week. This method is applicable to the study of intercellular communication in cell biology and tissue engineering.

The development and function of living tissues depends largely on interactions between cells that can vary in both time and space; however, temporal control of cell-cell interaction is experimentally challenging. By using a micromachined silicon substrate with moving parts, we demonstrate the dynamic regulation of cell-cell interactions via direct manipulation of adherent cells with micrometer-scale precision. We thereby achieve mechanical control of both tissue composition and spatial organization. As a case study, we demonstrate the utility of this tool in deconstructing the dynamics of intercellular communication between hepatocytes and supportive stromal cells in coculture. Our findings indicate that the maintenance of the hepatocellular phenotype by stroma requires direct contact for a limited time ( approximately hours) followed by a sustained soluble signal that has an effective range of <400 microm. This platform enables investigation of dynamic cell-cell interaction in a multitude of applications, spanning embryogenesis, homeostasis, and pathogenic processes.

Mit einem Mikromaschinen-Silizium-Substrat mit beweglichen Teilen, zeigen wir die dynamische Regelung der Zell-Zell-Interaktionen &uuml;ber direkte Manipulation adh&auml;renter Zellen mit Mikrometer-Ma&szlig;stab Pr&auml;zision. Wir erreichen damit mechanische Kontrolle der Gewebe Zusammensetzung und der r&auml;umlichen Organisation. Als Fallstudie zeigen wir die N&uuml;tzlichkeit dieses Tool in der Dekonstruktion der Dynamik der interzellul&auml;ren Kommunikation zwischen Hepatozyten und unterst&uuml;tzenden Stromazellen Zellen in Coculture. Unsere Ergebnisse zeigen, dass die Wartung des hepatozellul&auml;ren Ph&auml;notyps von Stroma direkten Kontakt f&uuml;r eine begrenzte Zeit erfordert (ca. Stunden) gefolgt von ein nachhaltiges l&ouml;sliche Signal, das eine effektive Reichweite von hat &lt; 400 Microm. Extracellular matrix (ECM) molecules have been shown to function as cues for neurite guidance in various populations of neurons. Here we show that laminin (LN) and fibronectin (FN) presented in stripe micro-patterns can provide guidance cues to neonatal (P5) inner ear spiral ganglion (SG) neurites. The response to both ECM molecules was dose-dependent. In a LN versus poly-L-lysine (PLL) assay, neurites were more often observed on PLL at low coating concentrations (5 and 10 microg/mL), while they were more often on LN at a high concentration (80 microg/mL). In a FN versus PLL assay, neurites were more often on PLL than on FN stripes at high coating concentrations (40 and 80 microg/mL). In a direct competition between LN and FN, neurites were observed on LN significantly more often than on FN at both 10 and 40 microg/mL. The data suggest a preference by SG neurites for LN at high concentrations, as well as avoidance of both LN at low and FN at high concentrations. The results also support a potential model for neurite guidance in the developing inner ear in vivo. LN, in the SG and osseus spiral lamina may promote SG dendrite growth toward the organ of Corti. Within the organ of Corti, lower concentrations of LN may slow neurite growth, with FN beneath each row of hair cells providing a stop or avoidance signal. This could allow growth cone filopodia increased time to sample their cellular targets, or direct the fibers upward toward the hair cells.

Liver sinusoidal endothelial cells (LSECs) differ, both structurally and functionally, from endothelial cells (ECs) lining blood vessels of other tissues. In probing the role of the underlying extracellular matrix, we identified collagen I and collagen III as well as mixtures of collagen I/collagen IV/fibronectin as having a positive effect on LSEC survival. Furthermore, using a stable hepatocellular model (hepatocyte-fibroblast) we were able to prolong the expression of both SE-1 and phenotypic functions of LSEC such as factor VIII activity and AcLOL uptake in cocultured LSECs through the production of short-range paracrine signals. In the course of these experiments, we identified the antigen recognized by SE-1 as CD32b. H&#xED;gado c&#xE9;lulas endoteliales sinusoidales (LSECs) difieren, tanto estructural como funcionalmente, de las c&#xE9;lulas endoteliales (ECS) los vasos sangu&#xED;neos de otros tejidos de revestimiento. Por ejemplo, en contraste con otros CE, LSECs poseen fenestraciones, tienen bajos niveles detectables de plaquetas adhesi&#xF3;n de c&#xE9;lulas endoteliales mol&#xE9;cula 1 de expresi&#xF3;n, y en el tejido de rata, que caracter&#xED;sticamente expresar un marcador de superficie celular reconocido por el anticuerpo SE-1.
