The dynamic properties of the regulatory network governing the choice between lytic and lysogenic growths of coliphage lambda is studied using a Markov chain stochastic model. Our computer simulation confirms the finding by Li et al. [Proc. Natl. Acad. Sci. USA 101, 4781 (2004)] on the dynamics of budding yeast: that the biological stationary states are global attractors; the biological pathways of lytic and lysogenic growths are attracting trajectories; and the network functions are robustly designed against structural perturbations. In addition, our model shows the stochastic switch from lysogen to lytic growth, which has been observed in experiments. A definition of pseudoenergy is introduced in the network dynamics to reveal a transitionlike behavior in the system.

The main product of the conversion of puerarin by unpermeabilized cells of bacterium Microbacterium oxydans CGMCC 1788 was puerarin-7-O-glucoside (241 +/- 31.9 microM). Permeabilization with 40% ethanol could not increase conversion yield, whereas it resulted in change of main product; a previous trace product became a main product (213 +/- 48.0 microM) which was identified as a novel puerarin-7-O-fructoside by electrospray ionization time-of-flight MS, (13)C NMR, (1)H NMR, and GC-MS analysis of sugar composition, and puerarin-7-O-glucoside became a trace product (14.8 +/- 5.4 microM). However, the extract from cells of M. oxydans CGMCC 1788 permeabilized with ethanol converted puerarin to form 113.9 +/- 27.7 microM puerarin-7-O-glucoside and 187.8 +/- 29.5 microM puerarin-7-O-fructoside under the same conditions. When unpermeabilized intact cells were recovered and used repeatedly for the conversion of puerarin, with increase of reuse times, the yield of puerarin-7-O-glucoside gradually decreased, whereas the yield of puerarin-7-O-fructoside increased gradually in the conversion mixture. The main product of the conversion of puerarin by the tenth recycled unpremerbilized cells was puerarin-7-O-fructoside (288.4 +/- 24.0 microM). Therefore, the change of permeability of cell membrane of bacterium M. oxydans CGMCC 1788 contributed to the change of conversion of the product's composition.

A filamentous cytoskeleton largely governs the physical shape and mechanical properties of eukaryotic cells. In bacteria, proteins homologous to all three classes of eukaryotic cytoskeletal filaments have recently been discovered. These proteins are essential for the maintenance of bacterial cell shape and have been shown to guide the localization of key cell-wall-modifying enzymes. However, whether the bacterial cytoskeleton is stiff enough to affect the overall mechanical rigidity of a cell has not been probed. Here, we used an optical trap to measure the bending rigidity of live Escherichia coli cells. We find that the actin-homolog MreB contributes nearly as much to the stiffness of a cell as the peptidoglycan cell wall. By quantitatively modeling these measurements, our data indicate that the MreB is rigidly linked to the cell wall, increasing the mechanical stiffness of the overall system. These data are the first evidence that the bacterial cytoskeleton contributes to the mechanical integrity of a cell in much the same way as it does in eukaryotes.

In Bakterien sind f&uuml;r alle drei Klassen eukaryotische Zytoskelett Filamente homologe Proteine vor kurzem entdeckt worden. Diese Proteine sind essentiell f&uuml;r die Aufrechterhaltung der Bakterienzelle Form und sind gezeigt worden, um die Lokalisierung der zentralen Zelle-Wand-&auml;ndern Enzyme zu f&uuml;hren. Jedoch hat ob bakterielle Zytoskeletts ist steif genug, um die insgesamt mechanische Steifigkeit einer Zelle beeinflussen nicht untersucht worden. Puerarin-7-O-fructoside was transformed by Trichoderma harzianum CGMCC 1523 into 3'-hydroxypuerarin-7-O-fructoside; this was identified by MS and NMR. However, puerarin-7-O-glucoside was not directly hydroxylated but hydrolyzed back into puerarin, which was transformed into 3'-hydroxypuerarin by the same fungi. Comparative analysis of free radical scavenging activity of DPPH showed that the free radical scavenging activity of puerarin-7-O-glucoside was reduced to approximately 1/2 of that of puerarin, while the free radical scavenging activity of puerarin-7-O-fructoside was increased to approximately 1.5 times of that of puerarin. The free radical scavenging activity of 3'-hydroxypuerarin-7-O-fructoside was further increased by 2.2 times of that of puerarin-7-O-fructoside, which was close to that of 3'-hydroxypuerarin.

The plasma samples were deproteinised with methanol after the addition of an internal standard, hesperidin. HPLC analysis was performed on a Diamonsil C(18) analytical column, using methanol -0.5% aqueous phosphoric acid (45:55, v/v) as the mobile phase with ultraviolet detection at 330â€‰nm. The calibration curve was linear over the range of 5-450â€‰Âµgâ€‰mL(-1) in rat plasma. The average extraction recovery of VOG was 98.74%â€‰Â±â€‰0.44%, and the relative standard deviations of the intra- and inter-day precisions were not greater than 4.1% and 2.0%, respectively. The validated method was successfully applied during a pharmacokinetic study in rats after oral administration of VOG at different doses, and all the results indicated that the pharmacokinetics of VOG in rats obeyed nonlinear processes.

A simple and sensitive HPLC method was developed to simultaneously determine three active compounds, vitexin-4â€³-O-glucoside (VG), vitexin-2â€³-O-rhamnoside (VR) and hyperoside (HP), in rat plasma after administering the hawthorn leaves extract (HLE). An HPLC assay with baicalin as the internal standard was carried out using a Phenomsil C(18) analytical column with UV detection at 332â€‰nm. The mobile phase consisted of methanol-acetonitrile-tetrahydrofuran-1% glacial acetic acid (6â€‰:â€‰1.5â€‰:â€‰18.5â€‰:â€‰74, v/v/v/v). The calibration curves were linear over the range of 2.5-500, 0.2-25 and 0.25-12.5â€‰Âµgâ€‰mL(-1) for VG, VR and HP, respectively. The method was reproducible and reliable, with relative standard deviations of the intra- and inter-day precision between 1.2% and 13.2% for the analysis of the three analytes. The validated HPLC method herein described was successfully applied to the pharmacokinetic study of VG, VR and HP after oral administration of HLE to rats over the dose range of 2.5-10â€‰mLâ€‰kg(-1).

Se desarroll&oacute; un m&eacute;todo HPLC simple y sensible para determinar simult&aacute;neamente tres compuestos activos, vitexina-4&Prime;-O-gluc&oacute;sido (VG), vitexina-2&Prime;-O-rhamn&oacute;sido (VR) y hyperoside (HP), en plasma de rata despu&eacute;s de administrar el espino deja extracto (HLE). Un an&aacute;lisis HPLC con Baicalina como est&aacute;ndar interno se llev&oacute; a cabo utilizando una columna anal&iacute;tica Phenomsil C(18) con detecci&oacute;n UV en 332 nm. La fase m&oacute;vil consisti&oacute; en metanol-acetonitrilo-tetrahidrofurano - 1% de &aacute;cido ac&eacute;tico glacial (6: 1.5: 18,5: 74, v/v/v/v). Las curvas de calibraci&oacute;n fueron lineales en el rango de 2.5-500, 0.2-25 y 12,5-0.25 &micro;g ml de VG, VR y HP, respectivamente. El m&eacute;todo era reproducible y confiable, con desviaciones est&aacute;ndar relativas de la precisi&oacute;n intra - y inter - d&iacute;a entre 1.2% y 13,2% para el an&aacute;lisis de los tres analitos. Bacterial cells possess multiple cytoskeletal proteins involved in a wide range of cellular processes. These cytoskeletal proteins are dynamic, but the driving forces and cellular functions of these dynamics remain poorly understood. Eukaryotic cytoskeletal dynamics are often driven by motor proteins, but in bacteria no motors that drive cytoskeletal motion have been identified to date. Here, we quantitatively study the dynamics of the Escherichia coli actin homolog MreB, which is essential for the maintenance of rod-like cell shape in bacteria. We find that MreB rotates around the long axis of the cell in a persistent manner. Whereas previous studies have suggested that MreB dynamics are driven by its own polymerization, we show that MreB rotation does not depend on its own polymerization but rather requires the assembly of the peptidoglycan cell wall. The cell-wall synthesis machinery thus either constitutes a novel type of extracellular motor that exerts force on cytoplasmic MreB, or is indirectly required for an as-yet-unidentified motor. Biophysical simulations suggest that one function of MreB rotation is to ensure a uniform distribution of new peptidoglycan insertion sites, a necessary condition to maintain rod shape during growth. These findings both broaden the view of cytoskeletal motors and deepen our understanding of the physical basis of bacterial morphogenesis.

Eukaryotische Zytoskelett Dynamik sind oft angetrieben von motor Proteine, aber in Bakterien keine Motoren, die Zytoskelett Bewegung fahren bis heute identifiziert wurden. Hier untersuchen wir quantitativ die Dynamik der Escherichia-coli-Aktin-Homolog MreB, die f&uuml;r die Aufrechterhaltung der Stab-wie Zelle Form in Bakterien erforderlich ist. Wir finden, dass MreB um die L&auml;ngsachse der Zelle dauerhaft gedreht. W&auml;hrend fr&uuml;here Studien suggerieren, dass MreB Dynamik durch eigene Polymerisation bestimmt sind, zeigen wir, dass MreB Drehung nicht eine eigene Polymerisation h&auml;ngt, sondern die Assembly der Zellwand Peptidoglycan verlangt. Die Zellwand Synthese Maschine bildet somit entweder eine neuartige Art von extrazellul&auml;ren Motor, der &uuml;bt Kraft auf zytoplasmatischen MreB oder indirekt erforderlich f&uuml;r eine als-noch-unbekannte Motor. Spironolactone is a medicinally important molecule that is clinically used in the treatment and management of many diseases such as oedema and ascites in cirrhosis of the liver, malignant ascites, nephrotic syndrome, chronic lung disease, resistant hypertension, congestive heart failure, and primary hyperaldosteronism. Microbial transformations of spironolactone by Cunninghamella elegans ATCC 9245 was carried out. Two new hydroxylated derivatives, 12β-hydroxy-spironolactone and 2α-hydroxy-spironolactone, were synthesized. Their structures were characterized on the basis of the spectroscopic data. The substrate can be efficiently converted into the products within 72 h after its addition to the fermentation broth of C. elegans ATCC 9245.

Synthesis of two new hydroxylated derivatives of spironolactone by microbial transformation.

FBXW7 acts as a typical tumor suppressor, with loss-of-function alterations in human cancers, by promoting ubiquitylation and degradation of many oncoproteins. Lysine-specific demethylase 1 (LSD1) is a well-characterized histone demethylase. Whether LSD1 has demethylase-independent activity remains elusive. Here we report that LSD1 directly binds to FBXW7 to destabilize FBXW7 independent of its demethylase activity. Specifically, LSD1 is a pseudosubstrate of FBXW7 and LSD1-FBXW7 binding does not trigger LSD1 ubiquitylation, but instead promotes FBXW7 self-ubiquitylation by preventing FBXW7 dimerization. The self-ubiquitylated FBXW7 is subjected to degradation by proteasome as well as lysosome in a manner dependent on autophagy protein p62/SQSTM1. Biologically, LSD1 destabilizes FBXW7 to abrogate its functions in growth suppression, nonhomologous end-joining repair, and radioprotection. Collectively, our study revealed a previously unknown activity of LSD1, which likely contributes to its oncogenic function. Targeting LSD1 protein, not only its demethylase activity, might be a unique approach for LSD1-based drug discovery for anticancer application.

Immunoassays such as the enzyme-linked immunosorbent assay (ELISA) are utilized extensively for detecting protein biomarkers and small molecules in healthcare, environmental monitoring, and food analysis. Unfortunately, the current strategies for immunoassays often require sophisticated apparatus such as a microplate reader, which might not be available in resource-limited areas. To mitigate this problem, we designed a compact smartphone based-device and a multicolor response immunosensor. First, we designed a compact and cost-effective 3D-printed attachment, where a light-emitting diode was used as a light excitation source and a smartphone captured the fluorescent emission signals. Second, by combining quantum dots hybrid and chemical redox reaction, multiple color responses were displayed in the presence of the analyte at different concentrations. Third, solutions with distinct tonality could be readily distinguished by the naked eye and they were suitable for quantitative analysis using the hue-saturation-lightness color space based on a smartphone application. The versatility of the proposed sensing system was demonstrated by implementing an indirect competitive ELISA for analyzing trace drug residues in foodstuffs. The multicolor response of this sensing strategy allows us to visually quantify drug residues in foodstuffs. Moreover, the smartphone-based immunosensor can assess the exact concentration of the analyte by using a self-designed mobile application. The proposed assay provides a highly sensitive performance that the limit of detection was 0.37 ng/mL by visual detection and 0.057 ng/mL using the compact device. Due to its advantages in terms of portability, straightforward visual detection, high sensitivity, and cost effectiveness, the proposed immunosensor has great potential for applications in areas without access to laboratories or expensive infrastructure.

Head and neck squamous cell carcinoma (HNSCC) is one of the most malignant cancers with poor prognosis worldwide. Emerging evidence indicates that competing endogenous RNAs (ceRNAs) are involved in various diseases, however, the regulatory mechanisms of ceRNAs underlying HNSCC remain unclear. In this study, we retrieved differentially expressed long non-coding RNAs (DElncRNAs), messenger RNAs (DEmRNAs) and microRANs (DEmiRNAs) from The Cancer Genome Atlas database and constructed a ceRNA-based risk model in HNSCC by integrated bioinformatics approaches. Functional enrichment analyses showed that DEmRNAs might be involved in extracellular matrix related biological processes, and protein-protein interaction network further selected out prognostic genes, including MYL1 and ACTN2. Importantly, co-expressed RNAs identified by weighted co-expression gene network analysis constructed the ceRNA networks. Moreover, AC114730.3, AC136375.3, LAT and RYR3 were highly correlated to overall survival of HNSCC by Kaplan-Meier method and univariate Cox regression analysis, which were subsequently implemented multivariate Cox regression analysis to build the risk model. Our study provides a deeper understanding of ceRNAs on the regulatory mechanisms, which will facilitate the expansion of the roles on the ceRNAs in the tumorigenesis, development and treatment of HNSCC.
