The zebrafish mutant violet beauregarde (vbg) can be identified at two days post-fertilization by an abnormal circulation pattern in which most blood cells flow through a limited number of dilated cranial vessels and fail to perfuse the trunk and tail. This phenotype cannot be explained by caudal vessel abnormalities or by a defect in cranial vessel patterning, but instead stems from an increase in endothelial cell number in specific cranial vessels. We show that vbg encodes activin receptor-like kinase 1 (Acvrl1; also known as Alk1), a TGFbeta type I receptor that is expressed predominantly in the endothelium of the vessels that become dilated in vbg mutants. Thus, vbg provides a model for the human autosomal dominant disorder, hereditary hemorrhagic telangiectasia type 2, in which disruption of ACVRL1 causes vessel malformations that may result in hemorrhage or stroke. ALK1 belongs to the type I receptor family for transforming growth factor-beta family ligands. Heterozygous ALK1 mutations cause hereditary hemorrhagic telangiectasia type 2 (HHT2), a multisystemic vascular disorder. Based largely on in vitro studies, TGF-beta1 has been considered as the most likely ALK1 ligand related to HHT, yet the identity of the physiologic ALK1 ligand remains controversial. In cultured endothelial cells, ALK1 and another TGF-beta type I receptor, ALK5, regulate angiogenesis by controlling TGF-beta signal transduction, and ALK5 is required for ALK1 signaling. However, the extent to which such interactions between these 2 receptors play a role in pathogenesis of HHT is unknown. We directly addressed these issues in vivo by comparing the phenotypes of mice in which the Alk1, Alk5, or Tgfbr2 gene was conditionally deleted in restricted vascular endothelia using a novel endothelial Cre transgenic line. Alk1-conditional deletion resulted in severe vascular malformations mimicking all pathologic features of HHT. Yet Alk5- or Tgfbr2-conditional deletion in mice, or Alk5 inhibition in zebrafish, did not affect vessel morphogenesis. These data indicate that neither ALK5 nor TGFBR2 is required for ALK1 signaling pertinent to the pathogenesis of HHT and suggest that HHT might not be a TGF-beta subfamily disease.

ALK5- and TGFBR2-independent role of ALK1 in the pathogenesis of hereditary hemorrhagic telangiectasia type 2.

Suppression conditionnelle Alk1 a entra&icirc;n&eacute; des malformations vasculaires graves imitant toutes les caract&eacute;ristiques pathologiques de VRD. Encore suppression conditionnelle-Alk5 ou Tgfbr2 chez la souris, ou inhibition Alk5 chez le poisson z&egrave;bre, n&#39;a pas affect&eacute; la morphogen&egrave;se de navire. Ces donn&eacute;es indiquent que ni ALK5 ni TGFBR2 est requis pour ALK1 signalisation pertinents &agrave; la pathogen&egrave;se de VRD et sugg&egrave;rent que VRD pourrait ne pas &ecirc;tre une maladie de sous-famille des TGF-b&ecirc;ta.

ALK1 geh&ouml;rt zum Typ I Rezeptor-Familie f&uuml;r die Umwandlung von Wachstumsfaktor-Beta-Familie-Liganden. Heterozygote ALK1-Mutationen verursachen heredit&auml;re h&auml;morrhagische Teleangiektasie Typ 2 (HHT2), eine multisystemische vaskul&auml;re Erkrankung. Basiert weitgehend auf in-vitro-Studien, TGF-beta1 wurde als als das wahrscheinlichste ALK1-Ligand mit Bezug zu HHT, noch die Identit&auml;t des physiologischen Liganden ALK1 bleibt umstritten. In kultivierten Endothelzellen, ALK1 und anderen TGF-Beta Typ I Rezeptor, ALK5, Angiogenese regulieren, indem steuernde TGF-Beta Signaltransduktion und ALK5 f&uuml;r ALK1 Signalisierung erforderlich ist. Das Ausma&szlig;, zu dem diese Wechselwirkungen zwischen diesen 2 Rezeptoren in der Pathogenese der HHT eine Rolle spielen, ist jedoch unbekannt. Wir adressiert direkt diese Probleme, die in-vivo durch den Vergleich der Ph&auml;notypen von M&auml;usen, in denen das Alk1, Alk5 oder Tgfbr2-Gen bedingt in eingeschr&auml;nkten vaskul&auml;re Endothelia mithilfe einer neuartigen endothelialen Cre transgenen Linie gel&ouml;scht wurde. Alk1-bedingte L&ouml;schung f&uuml;hrte zu schweren vaskul&auml;ren Fehlbildungen imitieren alle pathologischen Merkmale der HHT. Alk5 - oder Tgfbr2-bedingte L&ouml;schung bei M&auml;usen oder Alk5-Hemmung in Zebrafisch, hat noch nicht Schiff Morphogenese beeinflusst. Diese Daten zeigen, dass weder ALK5 noch TGFBR2 ist erforderlich f&uuml;r ALK1-Steuerinformationen f&uuml;r die Pathogenese der HHT und Vermutung nahe, dass HHT keine Krankheit des TGF-Beta-Unterfamilie.

Basato in gran parte su studi in vitro, TGF-beta1 &egrave; stato considerato come il pi&ugrave; probabile ligando ALK1 correlato a HHT, eppure l&#39;identit&agrave; del ligando fisiologico ALK1 rimane controversa. Nella colture delle cellule endoteliali ALK1 e TGF-beta un altro tipo I recettori, ALK5, regolano l&#39;angiogenesi di controllo trasduzione del segnale TGF-beta, e ALK5 &egrave; necessaria per ALK1 di segnalazione. Tuttavia, nella misura in cui tali interazioni tra questi 2 recettori giocano un ruolo nella patogenesi di HHT &egrave; sconosciuta. Abbiamo affrontato direttamente questi aspetti in vivo confrontando i fenotipi di topi in cui il gene Alk1, Alk5 o Tgfbr2 &egrave; stato eliminato in modo condizionale nell&#39;endotelio vascolare limitato utilizzando un romanzo endoteliali Cre transgenici. Eliminazione Alk1-condizionale ha provocato gravi malformazioni vascolari che mimano tutte le caratteristiche patologiche di HHT. Ancora Alk5 o Tgfbr2 condizionale eliminazione nei topi o Alk5 inibizione in zebrafish, non ha influito morfogenesi nave. Questi dati indicano che n&eacute; ALK5 n&eacute; TGFBR2 &egrave; richiesto per ALK1 segnalazione pertinente alla patogenesi della HHT e suggeriscono che HHT potrebbe non essere una malattia della sottofamiglia TGF-beta.

ALK1 pertenece al tipo I familia del receptor para la transformaci&oacute;n de ligandos familia del factor de crecimiento-beta. Las mutaciones heterozig&oacute;ticas ALK1 causan telangiectasia hemorr&aacute;gica hereditaria tipo 2 (HHT2), un desorden vascular multisist&eacute;mico. Basado fundamentalmente en estudios in vitro, TGF-beta1 ha sido considerado como el m&aacute;s probable ligand ALK1 relacionadas con HHT, sin embargo, la identidad del ligando fisiol&oacute;gico ALK1 sigue siendo pol&eacute;mica. Aortic arch malformations are common congenital disorders that are frequently of unknown etiology. To gain insight into the factors that guide branchial aortic arch development, we examined the process by which these vessels assemble in wild type zebrafish embryos and in kurzschluss(tr12) (kus(tr12)) mutants. In wild type embryos, each branchial aortic arch first appears as an island of angioblasts in the lateral pharyngeal mesoderm, then elaborates by angiogenesis to connect to the lateral dorsal aorta and ventral aorta. In kus(tr12) mutants, angioblast formation and initial sprouting are normal, but aortic arches 5 and 6 fail to form a lumenized connection to the lateral dorsal aorta. Blood enters these blind-ending vessels from the ventral aorta, distending the arteries and precipitating fusion with an adjacent vein. This arteriovenous malformation (AVM), which shunts nearly all blood directly back to the heart, is not exclusively genetically programmed, as its formation correlates with blood flow and aortic arch enlargement. By positional cloning, we have identified a nonsense mutation in unc45a in kus(tr12) mutants. Our results are the first to ascribe a role for Unc45a, a putative myosin chaperone, in vertebrate development, and identify a novel mechanism by which an AVM can form.

Arco a&oacute;rtico malformaciones son trastornos cong&eacute;nitos comunes que suelen ser de etiolog&iacute;a desconocida. Para profundizar en los factores que gu&iacute;an el desarrollo branquial arco a&oacute;rtico, examinamos el proceso por el cual estos vasos montan en embriones de pez cebra de tipo salvaje y en kurzschluss(tr12) (kus(tr12)) mutantes. En embriones de tipo salvaje, cada arco a&oacute;rtico branquial primero aparece como una isla de angioblastos en el mesodermo far&iacute;ngeo lateral y luego elabora por angiog&eacute;nesis para conectar a la aorta dorsal lateral y la aorta ventral. En mutantes kus(tr12), formaci&oacute;n angioblast y brotaci&oacute;n inicial son normales, pero no forman una conexi&oacute;n lumenized a la aorta dorsal lateral arcos a&oacute;rticos 5 y 6. Sangre entra en estos vasos oculto-conclusi&oacute;n de la aorta ventral, distensi&oacute;n de las arterias y precipitando la fusi&oacute;n con una vena adyacente. Esta malformaci&oacute;n arteriovenosa (MAV), que los shunts casi toda la sangre directamente hacia el coraz&oacute;n, no exclusivamente gen&eacute;ticamente programada, como su formaci&oacute;n se correlaciona con la ampliaci&oacute;n de flujo y arco a&oacute;rtico de sangre. Por clonaje posicional, hemos identificado una mutaci&oacute;n de absurdo en unc45a en kus(tr12) mutantes. Bone morphogenetic proteins (BMPs) are critical players in development and disease, regulating such diverse processes as dorsoventral patterning, palate formation, and ossification. These ligands are classically considered to signal via BMP receptor-specific Smad proteins 1, 5, and 8. To determine the spatiotemporal pattern of Smad1/5/8 activity and thus canonical BMP signaling in the developing zebrafish embryo, we generated a transgenic line expressing EGFP under the control of a BMP-responsive element. EGFP is expressed in many established BMP signaling domains and is responsive to alterations in BMP type I receptor activity and smad1 and smad5 expression. This transgenic Smad1/5/8 reporter line will be useful for determining ligand and receptor requirements for specific domains of BMP activity, as well as for genetic and pharmacological screens aimed at identifying enhancers or suppressors of canonical BMP signaling.

Prote&iacute;nas morfogen&eacute;ticas &oacute;seas (BMPs) son actores cr&iacute;ticos en el desarrollo y la enfermedad, regulan estos procesos diversos como patr&oacute;n dorsoventral, formaci&oacute;n de paladar y osificaci&oacute;n. Estos ligandos se consideran cl&aacute;sicamente se&ntilde;al mediante prote&iacute;nas de Smad BMP del receptor espec&iacute;fico 1, 5 y 8. Para determinar el patr&oacute;n espacio-temporal de la actividad de Smad1/5/8 y as&iacute; BMP se&ntilde;alizaci&oacute;n can&oacute;nica en el embri&oacute;n de pez cebra en desarrollo, hemos generado una l&iacute;nea transg&eacute;nica expresando EGFP bajo el control de un elemento sensible al BMP. EGFP se expresa en BMP establecido muchos dominios de se&ntilde;alizaci&oacute;n y es sensible a alteraciones en BMP tipo I smad1 y smad5 de la expresi&oacute;n y la actividad del receptor. Esta l&iacute;nea transg&eacute;nica de reportero Smad1/5/8 ser&aacute; &uacute;til para determinar los requisitos de ligando y receptor para dominios espec&iacute;ficos de actividad BMP, as&iacute; como para pantallas gen&eacute;ticas y farmacol&oacute;gicas destinadas a determinar potenciadores o supresores de se&ntilde;alizaci&oacute;n de BMP can&oacute;nica.

Arteriovenous malformations (AVMs) are fragile direct connections between arteries and veins that arise during times of active angiogenesis. To understand the etiology of AVMs and the role of blood flow in their development, we analyzed AVM development in zebrafish embryos harboring a mutation in activin receptor-like kinase I (alk1), which encodes a TGFÎ² family type I receptor implicated in the human vascular disorder hereditary hemorrhagic telangiectasia type 2 (HHT2). Our analyses demonstrate that increases in arterial caliber, which stem in part from increased cell number and in part from decreased cell density, precede AVM development, and that AVMs represent enlargement and stabilization of normally transient arteriovenous connections. Whereas initial increases in endothelial cell number are independent of blood flow, later increases, as well as AVMs, are dependent on flow. Furthermore, we demonstrate that alk1 expression requires blood flow, and despite normal levels of shear stress, some flow-responsive genes are dysregulated in alk1 mutant arterial endothelial cells. Taken together, our results suggest that Alk1 plays a role in transducing hemodynamic forces into a biochemical signal required to limit nascent vessel caliber, and support a novel two-step model for HHT-associated AVM development in which pathological arterial enlargement and consequent altered blood flow precipitate a flow-dependent adaptive response involving retention of normally transient arteriovenous connections, thereby generating AVMs.

Interaction between alk1 and blood flow in the development of arteriovenous malformations.

Malformations art&eacute;rioveineuses (MAV) sont fragiles liaisons directes entre les art&egrave;res et les veines qui surviennent pendant les p&eacute;riodes d&#39;active angiogen&egrave;se. Pour comprendre l&#39;&eacute;tiologie de MAV et le r&ocirc;le de la circulation sanguine dans leur d&eacute;veloppement, nous avons analys&eacute; le d&eacute;veloppement AVM dans des embryons de poisson-z&egrave;bre porteur d&#39;une mutation dans l&#39;activine receptor-like kinase (alk1), qui code pour une famille TGF&beta; type I j&#39;ai r&eacute;cepteurs impliqu&eacute;s dans la pathologie vasculaire humain t&eacute;langiectasie h&eacute;morragique h&eacute;r&eacute;ditaire type 2 (HHT2). Nos analyses montrent que l&#39;augmentation de calibre art&eacute;riel, qui proviennent en partie du nombre accru de cellules et, en partie, de la densit&eacute; cellulaire r&eacute;duite, pr&eacute;c&egrave;de d&eacute;veloppement AVM et que MAV repr&eacute;sentent l&#39;&eacute;largissement et la stabilisation des connexions art&eacute;rio-veineuses normalement transitoires. Consid&eacute;rant que des augmentations initiales nombre de cellules endoth&eacute;liales sont ind&eacute;pendantes de la circulation sanguine, plus tard augmente, ainsi que MAV, d&eacute;pendent de flux. En outre, nous d&eacute;montrons qu&#39;alk1 expression requiert la circulation sanguine, malgr&eacute; les niveaux normaux de la contrainte de cisaillement, certains g&egrave;nes favorisant l&#39;&eacute;coulement sont dysr&eacute;gulation dans les cellules endoth&eacute;liales art&eacute;rielles mutant alk1. Arterio-ven&ouml;se Fehlbildungen (AVMD) sind fragile direkte Verbindungen zwischen Arterien und Venen, die in Zeiten der aktiven Angiogenese entstehen. Zum Verst&auml;ndnis der &Auml;tiologie der AVMD und die Rolle des Blutflusses in ihrer Entwicklung analysierten wir AVM-Entwicklung in den Zebrafish Embryos beherbergen eine Mutation in Activin Rezeptor-wie Kinase I (alk1), das eine TGF&beta;-Familie kodiert Typ I Rezeptor verwickelt in den menschlichen Gef&auml;&szlig;st&ouml;rung heredit&auml;re h&auml;morrhagische Teleangiektasie Typ 2 (HHT2). Unsere Analysen zeigen, dass die Erh&ouml;hung der arteriellen Kaliber, die teilweise aus erh&ouml;hte Zellenzahl und teilweise verminderten Zelldichte Stiel, AVM Entwicklung vorausgehen und AVMD Erweiterung und Stabilisierung des normalerweise transiente arterioven&ouml;sen Verbindungen darstellen. W&auml;hrend der anf&auml;nglichen Anstieg der endothelial Zelle Nummer Blutfluss, sp&auml;ter erh&ouml;ht, sowie AVMD unabh&auml;ngig sind, sind Str&ouml;mung abh&auml;ngig. Dar&uuml;ber hinaus zeigen wir, dass alk1 Ausdruck Blutfluss erfordert und trotz eines normalen Niveaus der Scherspannung, einige Fluss-responsive Gene defekten in alk1 mutant arterielle Endothelzellen sind. Genommen ben&ouml;tigt zusammen, unsere Ergebnisse legen nahe, dass Alk1 eine Rolle spielt bei h&auml;modynamischen Kr&auml;fte in eine biochemische Signal transducing, um entstehende Schiff Kaliber zu begrenzen und die Unterst&uuml;tzung einer neuartigen zweistufigen Modell AVM HHT-assoziierte Entwicklung in dem pathologische arterielle Erweiterung und daraus resultierende ver&auml;nderte Durchblutung eine Fluss-abh&auml;ngige adaptive Antwort, die Aufbewahrung der normalerweise transiente arterioven&ouml;sen Verbindungen, damit einen AVMD auszuf&auml;llen.

Malformazioni artero-venose (MAV) sono fragili collegamenti diretti tra le arterie e le vene che si presentano durante i periodi di angiogenesi attivo. Per comprendere l&#39;eziologia della SMA e il ruolo del flusso di sangue nel loro sviluppo, abbiamo analizzato lo sviluppo di AVM in embrioni di zebrafish harboring una mutazione in activin receptor-like chinasi (alk1), che codifica per una famiglia TGF&beta; tipo I I recettori implicati nella patologia vascolare umano teleangiectasia emorragica ereditaria tipo 2 (HHT2). Le nostre analisi dimostrano che aumenta in calibro arterioso, che derivano in parte dal numero aumentato di cellule e in parte dalla densit&agrave; delle cellule &egrave; diminuito, precede lo sviluppo AVM e che SMA rappresenta l&#39;allargamento e la stabilizzazione delle connessioni artero-norma transitorie. Considerando che iniziali aumenti nel numero delle cellule endoteliali sono indipendenti del flusso di sangue, poi aumenta, cos&igrave; come SMA, dipendono dal flusso. Inoltre, dimostriamo che alk1 espressione richiede il flusso di sangue, e nonostante normali livelli di sollecitazione di taglio, alcuni geni flusso-sensibli a reagire sono in alk1 mutante arterioso cellule endoteliali. Presi insieme, che i nostri risultati suggeriscono che Alk1 gioca un ruolo in transducing forze emodinamiche in un segnale biochimico necessario per limitare il calibro del vaso nascente e sostenere un modello di romanzo in due fasi per lo sviluppo di AVM HHT-associati in cui patologico allargamento arteriosa e del flusso sanguigno alterato conseguente precipitare una risposta adattativa di flusso-dipendente che comportano ritenzione di norma transitorie connessioni artero-venose, generando quindi SMA.

Las malformaciones arteriovenosas (MAV) son fr&aacute;giles conexiones directas entre las arterias y venas que se presentan durante la &eacute;poca de la angiog&eacute;nesis activa. Para entender la etiolog&iacute;a de las malformaciones arteriovenosas y el papel del flujo sangu&iacute;neo en su desarrollo, hemos analizado el desarrollo de AVM en embriones de pez cebra que una mutaci&oacute;n en quinasa como receptor de activina I (alk1), que codifica una familia TGF&beta; tipo I receptores implicados en el trastorno vascular humano telangiectasia hemorr&aacute;gica hereditaria tipo 2 (HHT2). Nuestros an&aacute;lisis demuestran que aumenta en calibre arterial, derivadas en parte del n&uacute;mero creciente de la c&eacute;lula y en parte de densidad celular disminuida, precede al desarrollo de AVM, y que las malformaciones arteriovenosas representan ampliaci&oacute;n y estabilizaci&oacute;n de conexiones arteriovenosas normalmente transitorias. Considerando que el aumento inicial en el n&uacute;mero de c&eacute;lulas endoteliales es independiente del flujo sangu&iacute;neo, m&aacute;s adelante aumenta, as&iacute; como malformaciones arteriovenosas, son dependientes en flujo. Adem&aacute;s, demostramos que alk1 expresi&oacute;n requiere flujo sangu&iacute;neo, y a pesar de niveles normales de la tensi&oacute;n de esquileo, algunos genes sensibles al flujo dysregulated en alk1 mutante arterial c&eacute;lulas endoteliales. Tomados juntos, que nuestros resultados sugieren que Alk1 desempe&ntilde;a un papel en la transducci&oacute;n de las fuerzas hemodin&aacute;micas en una se&ntilde;al bioqu&iacute;mica necesaria para limitar el calibre del vaso naciente y apoyar un modelo de dos etapas de la novela para el desarrollo de AVM HHT-asociado en que patol&oacute;gico ampliaci&oacute;n arterial y flujo sangu&iacute;neo alterado consiguiente precipitan una respuesta adaptativa de flujo dependiente con retenci&oacute;n de conexiones arteriovenosas normalmente transitorias, generando malformaciones arteriovenosas.

The cranial vasculature is essential for the survival and development of the central nervous system and is important in stroke and other brain pathologies. Cranial vessels form in a reproducible and evolutionarily conserved manner, but the process by which these vessels assemble and acquire their stereotypic patterning remains unclear. Here, we examine the stepwise assembly and patterning of the vascular network of the zebrafish hindbrain. The major artery supplying the hindbrain, the basilar artery, runs along the ventral keel of the hindbrain in all vertebrates. We show that this artery forms by a novel process of medial sprouting and migration of endothelial cells from a bilateral pair of primitive veins, the primordial hindbrain channels. Subsequently, a second wave of dorsal sprouting from the primordial hindbrain channels gives rise to angiogenic central arteries that penetrate into and innervate the hindbrain. The chemokine receptor cxcr4a is expressed in migrating endothelial cells of the primordial hindbrain channels, whereas its ligand cxcl12b is expressed in the hindbrain neural keel immediately adjacent to the assembling basilar artery. Knockdown of either cxcl12b or cxcr4a results in defects in basilar artery formation, showing that the assembly and patterning of this crucial artery depends on chemokine signaling.
