Members of the green fluorescent protein (GFP) family share sequence similarity and the 11-stranded beta-barrel fold. Fluorescence or bright coloration, observed in many members of this family, is enabled by the intrinsic properties of the polypeptide chain itself, without the requirement for cofactors. Amino acid sequence of fluorescent proteins can be altered by genetic engineering to produce variants with different spectral properties, suitable for direct visualization of molecular and cellular processes. Naturally occurring GFP-like proteins include fluorescent proteins from cnidarians of the Hydrozoa and Anthozoa classes, and from copepods of the Pontellidae family, as well as non-fluorescent proteins from Anthozoa. Recently, an mRNA encoding a fluorescent GFP-like protein AmphiGFP, related to GFP from Pontellidae, has been isolated from the lancelet Branchiostoma floridae, a cephalochordate (Deheyn et al., Biol Bull, 2007 213:95).

Gr&uuml;n fluoreszierendes Protein (GFP) Familienmitglieder Teilen Sequenz &Auml;hnlichkeit und die 11-angeschwemmte Beta-Fass-Klappe. Fluoreszenz oder helle F&auml;rbung, beobachtet in viele Mitglieder dieser Familie, ist von der inh&auml;renten Eigenschaften der Polypeptid-Kette selbst, ohne dass Kofaktoren aktiviert. Aminos&auml;uresequenz fluoreszierende Proteine kann durch Gentechnik Varianten mit unterschiedlichen spektralen Eigenschaften, geeignet f&uuml;r direkte Visualisierung der molekularen und zellul&auml;ren Prozesse produzieren ge&auml;ndert werden. The saltwater rotifer, Brachionus plicatilis, is widely used in the aquaculture industry as a prey item for first-feeding fishes due to its ease of culture, small size, rapid reproductive rate, and amenability to enrichment with nutrients. Despite the distinct advantages of this approach, rotifers have only been sporadically utilized for rearing larval zebrafish, primarily because of the common misconception that maintaining cultures of rotifers is difficult and excessively time-consuming. Here we present simple methods for maintaining continuous cultures of rotifers capable of supporting even the very largest zebrafish aquaculture facility, with minimal investments in materials, time, labor, and space. Examples of the methods' application in one large, existing facility is provided, and troubleshooting of common problems is discussed.
