Growing evidence supports the hypothesis that soluble, diffusible forms of the amyloid Î²-peptide (AÎ²) are pathogenically important in Alzheimer's disease (AD) and thus have both diagnostic and therapeutic salience. To learn more about the dynamics of soluble AÎ² economy in vivo, we used microdialysis to sample the brain interstitial fluid (ISF), which contains the most soluble AÎ² species in brain at steady state, in >40 wake, behaving APP transgenic mice before and during the process of AÎ² plaque formation (age 3-28 months). Diffusible forms of AÎ², especially AÎ²(42), declined significantly in ISF as mice underwent progressive parenchymal deposition of AÎ². Moreover, radiolabeled AÎ² administered at physiological concentrations into ISF revealed a striking difference in the fate of soluble AÎ² in plaque-rich (vs plaque-free) mice: it clears more rapidly from the ISF and becomes more associated with the TBS-extractable pool, suggesting that cerebral amyloid deposits can rapidly sequester soluble AÎ² from the ISF. Likewise, acute Î³-secretase inhibition in plaque-free mice showed a marked decline of AÎ²(38), AÎ²(40), and AÎ²(42), whereas in plaque-rich mice, AÎ²(42) declined significantly less. These results suggest that most of the AÎ²(42) that populates the ISF in plaque-rich mice is derived not from new AÎ² biosynthesis but rather from the large reservoir of less soluble AÎ²(42) in brain parenchyma. Together, these and other findings herein illuminate the in vivo dynamics of soluble AÎ² during the development of AD-type neuropathology and after Î³-secretase inhibition and help explain the apparent paradox that CSF AÎ²(42) levels fall as humans develop AD.

Dynamic analysis of amyloid Î²-protein in behaving mice reveals opposing changes in ISF versus parenchymal AÎ² during age-related plaque formation.

Preuves de plus en plus supporte l&#39;hypoth&egrave;se que des formes solubles, diffusibles de la &beta;-peptide b&ecirc;ta-amylo&iuml;de (A&szlig;) sont r&ocirc;le importants dans la maladie d&#39;Alzheimer (ma) et donc saillance tant diagnostique que th&eacute;rapeutique. Pour en savoir plus sur la dynamique de l&#39;&eacute;conomie de b&ecirc;ta-amylo&iuml;des soluble in vivo, nous avons utilis&eacute; la microdialyse pour go&ucirc;ter le liquide interstitiel de cerveau (ISF), qui renseigne sur les esp&egrave;ces de b&ecirc;ta-amylo&iuml;des plus solubles dans le cerveau &agrave; l&#39;&eacute;tat stationnaire, dans &gt; 40 se r&eacute;veiller, se comportant des souris transg&eacute;niques APP avant et pendant le processus de formation de plaques b&ecirc;ta-amylo&iuml;des (&acirc;ge 3-28 mois). Formes diffusibles de b&ecirc;ta-amylo&iuml;des, surtout A&beta;(42), a baiss&eacute; en ISF comme souris ont subi des d&eacute;p&ocirc;ts parenchymateux progressive d&#39;A&beta;. En outre, A&beta; radioactif administr&eacute;e &agrave; des concentrations physiologiques dans ISF a r&eacute;v&eacute;l&eacute; une diff&eacute;rence frappante dans le destin de l&#39;A&szlig; soluble chez les souris de la plaque-rich (vs exempt de plaque): elle efface plus rapidement de l&#39;ISF et devient plus associ&eacute; &agrave; la piscine du SCT extractible, sugg&eacute;rant que les d&eacute;p&ocirc;ts amylo&iuml;des c&eacute;r&eacute;brales peuvent s&eacute;questrer rapidement soluble A&beta; de l&#39;ISF. De m&ecirc;me, inhibition de &gamma;-s&eacute;cr&eacute;tase aigu&euml; chez les souris exempt de plaque a montr&eacute; une baisse marqu&eacute;e de la A&beta;(38), A&beta;(40) et A&beta;(42), tandis que chez les souris de la plaque-rich, A&beta;(42) baiss&eacute; significativement moins. Ces r&eacute;sultats sugg&egrave;rent que la majeure partie de la A&beta;(42) qui remplit l&#39;ISF plaque riche en souris provient pas de biosynth&egrave;se d&#39;A&beta; nouvelle mais plut&ocirc;t de l&#39;important r&eacute;servoir de A&beta;(42) moins solubles dans le parenchyme du cerveau. Ensemble, ces et autres constatations ci-apr&egrave;s illuminent la dynamique in vivo d&#39;A&szlig; soluble au cours du d&eacute;veloppement de la neuropathologie type d&#39;annonce et apr&egrave;s l&#39;inhibition de la &gamma;-secretase et aident &agrave; expliquer le paradoxe apparent que les niveaux de CSF A&beta;(42) tombent comme les humains d&eacute;veloppent AD.

F&uuml;r weitere Informationen &uuml;ber die Dynamik der l&ouml;slichen A&beta;-Wirtschaft, die in-vivo, verwendeten wir Microdialysis Gehirn interstitielle Fl&uuml;ssigkeit (ISF), zu probieren, die enth&auml;lt die meisten l&ouml;slichen A&beta;-Arten in Brain bei Steady-State, in &gt; 40 wake, verh&auml;lt sich die APP transgene M&auml;usen, vor und w&auml;hrend des Prozesses der A&beta; Plaquebildung (Alter 3-28 Monate). Diffusible Formen der A&beta;, besonders A&beta;(42), sanken deutlich im ISF wie M&auml;use progressive Lungenparenchym Ablagerung von A&beta; unterzog. Dar&uuml;ber hinaus offenbart radioaktive A&beta; verabreicht bei physiologischen Konzentrationen in ISF einen auffallenden Unterschied in das Schicksal der l&ouml;slichen A&beta; Plakette-Reich (gegen Zahnbelag-freie) M&auml;usen: es l&ouml;scht schneller von der ISF und der TBS extrahierbaren-Pool, was darauf hindeutet, dass zerebrale Amyloid-Ablagerungen schnell l&ouml;slichen A&beta; aus dem ISF absondern k&ouml;nnen mehr zugeordnet wird. Ebenso akute &gamma;-Sekretase-Hemmung in Plakette-freie M&auml;use zeigten einen deutlichen R&uuml;ckgang der A&beta;(38), A&beta;(40) und A&beta;(42), w&auml;hrend im Zahnbelag-reiche M&auml;use, A&beta;(42) deutlich zur&uuml;ckgegangen weniger. Diese Ergebnisse legen nahe, dass die A&beta;(42), die das ISF in Plakette-Reich f&uuml;llt die meisten M&auml;use leitet sich nicht von neuen A&beta;-Biosynthese, sondern von den gro&szlig;en Reservoir an weniger l&ouml;slichen A&beta;(42) im Gehirn Parenchym. Zusammen, diese und andere Befunde hierin beleuchten die in-vivo Dynamik der l&ouml;slichen A&beta; w&auml;hrend der Entwicklung von AD-Typ Neuropathologie und nach &gamma;-Sekretase-Hemmung und helfen das scheinbare Paradoxon zu erkl&auml;ren, das CSF A&beta;(42) Ebenen fallen wie Menschen AD entwickeln.

Dynamischer Analyse von Amyloid-&beta;-Protein verhaltensgleiche M&auml;usen zeigt gegnerische &Auml;nderungen im ISF gegen&uuml;ber Lungenparenchym A&beta; w&auml;hrend altersbedingte Plaquebildung.

Per saperne di pi&ugrave; circa le dinamiche dell&#39;economia A&beta; solubile in vivo, abbiamo usato per Microdialisi per assaggiare il liquido interstiziale (ISF), cervello che contiene la specie A&beta; pi&ugrave; solubile nel cervello ad uno stato di costante, in &amp;gt; 40 wake, comportando topi transgenici APP prima e durante il processo di formazione di placche A&beta; (et&agrave; 3-28 mesi). Questi risultati suggeriscono che la maggior parte delle A&beta;(42) che popola l&#39;ISF in placca-ricchi topi &egrave; derivato non da biosintesi A&beta; nuova ma piuttosto da grande serbatoio di A&beta;(42) meno solubili nel parenchima cerebrale. Insieme, queste ed altre scoperte qui illuminano le dinamiche in vivo di A&beta; solubile durante lo sviluppo di Neuropatologia annuncio-tipo e dopo inibizione &gamma;-secretasi e aiutano a spiegare l&#39;apparente paradosso che livelli di CSF A&beta;(42) cadono come gli esseri umani sviluppano annuncio.

Creciente evidencia apoya la hip&oacute;tesis que formas solubles y difusibles del &beta;-p&eacute;ptido amiloide (A&szlig;) son pat&oacute;geno importantes en la enfermedad de Alzheimer (EA) y por lo tanto tienen prominencia tanto diagn&oacute;stica y terap&eacute;utica. Para obtener m&aacute;s informaci&oacute;n sobre la din&aacute;mica de la econom&iacute;a de A&szlig; soluble en vivo, utilizamos microdialysis para probar el l&iacute;quido intersticial del cerebro (ISF), que contiene las especies de A&szlig; m&aacute;s solubles en el cerebro en estado estacionario, en &gt; 40 Estela, comport&aacute;ndose ratones transg&eacute;nicos APP antes y durante el proceso de formaci&oacute;n de la placa A&beta; (edad 3-28 meses). Difusibles formas de A&szlig;, especialmente A&beta;(42), disminuyeron significativamente en ISF como ratones experimentaron la deposici&oacute;n parenquimal progresiva de A&szlig;. Por otra parte, A&szlig; radiactiva administrada en concentraciones fisiol&oacute;gicas en ISF revel&oacute; una notable diferencia en el destino de A&szlig; soluble en ratones ricos en placa (vs placa libre): se elimina m&aacute;s r&aacute;pidamente de la ISF y se convierte m&aacute;s asociado con la piscina TBS extra&iacute;bles, sugiriendo que los dep&oacute;sitos de amiloide cerebrales pueden secuestrar r&aacute;pidamente A&beta; soluble de la ISF. Adem&aacute;s, la inhibici&oacute;n de la &gamma;-secretasa aguda en ratones libres de placa demostr&oacute; una marcada disminuci&oacute;n de A&beta;(38), A&beta;(40) y A&beta;(42), mientras que en ratones ricos en placa, A&beta;(42) disminuido significativamente menos. Estos resultados sugieren que la mayor&iacute;a de los A&beta;(42) que rellena la ISF en placa-ricos ratones se deriva no de bios&iacute;ntesis de A&szlig; nuevo sino de la gran reserva de A&beta;(42) menos solubles en el tejido cerebral.
