&Beta; 2 Syntrophin &#x662F; Cdk5 &#x57FA;&#x4F53;&#x7684;&#x6291;&#x5236;&#x80F0;&#x5C9B;&#x7D20;&#x5206;&#x6CCC;&#x9897;&#x7C92;&#x7684;&#x52A8;&#x529B;&#x3002;

The molecular basis for the interaction of insulin granules with the cortical cytoskeleton of pancreatic Î²-cells remains unknown. We have proposed that binding of the granule protein ICA512 to the PDZ domain of Î²2-syntrophin anchors granules to actin filaments and that the phosphorylation/dephosphorylation of Î²2-syntrophin regulates this association. Here we tested this hypothesis by analyzing INS-1 cells expressing GFP-Î²2-syntrophin through the combined use of biochemical approaches, imaging studies by confocal and total internal reflection fluorescence microscopy as well as electron microscopy. Our results support the notion that Î²2-syntrophin restrains the mobility of cortical granules in insulinoma INS-1 cells, thereby reducing insulin secretion and increasing insulin stores in resting cells, while increasing insulin release upon stimulation. Using mass spectrometry, in vitro phosphorylation assays and Î²2-syntrophin phosphomutants we found that phosphorylation of Î²2-syntrophin on S75 near the PDZ domain decreases its binding to ICA512 and correlates with increased granule motility, while phosphorylation of S90 has opposite effects. We further show that Cdk5, which regulates insulin secretion, phosphorylates S75. These findings provide mechanistic insight into how stimulation displaces insulin granules from cortical actin, thus promoting their motility and exocytosis.

La base mol&eacute;culaire de l&#39;interaction des granules d&#39;insuline avec le cytosquelette cortical des cellules &beta; pancr&eacute;atiques reste inconnue. Nous avons propos&eacute; que la liaison de la prot&eacute;ine de granule ICA512 pour le PDZ domaine de &beta;2-syntrophin ancres granules aux filaments d&#39;actine et que la phosphorylation/d&eacute;phosphorylation de &beta;2-syntrophin r&eacute;glemente cette association. Ici, nous avons test&eacute; cette hypoth&egrave;se en analysant les cellules INS-1 exprimant GFP-&beta;2-syntrophin par l&#39;utilisation combin&eacute;e des approches biochimiques, imagerie par microscopie de fluorescence confocale et total internal reflection ainsi que la microscopie &eacute;lectronique. Nos r&eacute;sultats confirment la notion que &beta;2-syntrophin retient la mobilit&eacute; des granules corticaux insulinome INS-1 cellules, ce qui r&eacute;duira la s&eacute;cr&eacute;tion d&#39;insuline et augmentation des magasins de l&#39;insuline dans les cellules au repos, tout en augmentant la lib&eacute;ration d&#39;insuline lors de la stimulation. &Agrave; l&#39;aide de la spectrom&eacute;trie de masse, &eacute;preuves de phosphorylation in vitro et &beta;2-syntrophin phosphomutants nous avons trouv&eacute; que la phosphorylation de &beta;2-syntrophin sur S75 pr&egrave;s du domaine PDZ diminue sa liaison &agrave; ICA512 et est corr&eacute;l&eacute;e avec la motilit&eacute; accrue de granule, tandis que la phosphorylation de la S90 a effets oppos&eacute;s. De plus, nous montrons que Cdk5, qui r&eacute;gule la s&eacute;cr&eacute;tion d&#39;insuline, phosphoryle S75. Ces r&eacute;sultats donnent un aper&ccedil;u m&eacute;caniste de comment la stimulation d&eacute;place des granules d&#39;insuline d&#39;actine corticale, favorisant ainsi leur motilit&eacute; et l&#39;exocytose.

Die molekulare Grundlage f&uuml;r das Zusammenspiel von Insulin-Granulat mit kortikalen Zytoskeletts pankreatischer &beta;-Zellen ist noch nicht bekannt. Wir haben vorgeschlagen, dass die Bindung des Proteins Untermodul ICA512 an der PDZ Dom&auml;ne des &szlig;2-Syntrophin Granulate nach Aktin-Filamente verankert und die Phosphorylierung/Dephosphorylierung von &szlig;2-Syntrophin dieses Vereins regelt. Hier testeten wir diese Hypothese durch die Analyse von INS-1 Zellen GFP-&szlig;2-Syntrophin durch den kombinierten Einsatz von biochemischen Ans&auml;tze zum Ausdruck zu bringen, imaging-Studien durch konfokale und total internal Reflection-Fluoreszenzmikroskopie sowie Elektronenmikroskopie. Unsere Resultate unterst&uuml;tzen das Konzept, dass die &szlig;2-Syntrophin die Mobilit&auml;t der kortikalen K&ouml;rnchen in Insulinom INS-1 Zellen, dadurch Verringerung der Insulinsekretion und erh&ouml;hen Insulin L&auml;den in Ruhestellung Zellen, w&auml;hrend die Erh&ouml;hung der Insulin-Freisetzung auf Anregung zur&uuml;ckh&auml;lt. Mit Massenspektrometrie, in-vitro-Phosphorylierung-Assays und &szlig;2-Syntrophin-Phosphomutants, die wir fanden, dass Phosphorylierung von &szlig;2-Syntrophin auf S75 nahe der PDZ-Dom&auml;nen verringert seine Bindung an ICA512 und korreliert mit erh&ouml;hter Untermodul Motilit&auml;t, w&auml;hrend Phosphorylierung von S90 gegen&uuml;ber Auswirkungen hat. Wir zeigen weiter, dass Cdk5, die Insulin-Sekretion regelt, S75 phosphorylates. Diese Erkenntnisse einen mechanistischen Einblick in wie Stimulierung Insulin Granulat aus kortikalen Actin, f&ouml;rdern ihre Beweglichkeit und Exozytose verdr&auml;ngt.

Le basi molecolari per l&#39;interazione dei granuli di insulina con il citoscheletro corticale delle &beta;-cellule pancreatiche rimane sconosciuta. Abbiamo proposto che vincolante della proteina granulo ICA512 per il PDZ dominio di &beta;2-syntrophin ancore granuli a filamenti di actina e che la fosforilazione/defosforilazione di &beta;2-syntrophin regola questa associazione. Qui abbiamo testato questa ipotesi analizzando INS-1 cellule che esprimono GFP-&beta;2-syntrophin attraverso l&#39;uso combinato di approcci biochimici, gli studi di microscopia di fluorescenza confocale riflessione interna totale, cos&igrave; come la microscopia elettronica di imaging. I nostri risultati supportano la nozione che &beta;2-syntrophin trattiene la mobilit&agrave; dei granuli corticali nell&#39;insulinoma INS-1 celle, quindi riducendo la secrezione di insulina e aumento negozi di insulina nelle cellule a riposo, mentre aumentando il rilascio di insulina dopo stimolazione. Mediante spettrometria di massa, saggi in vitro fosforilazione e &beta;2-syntrophin phosphomutants abbiamo trovato che la fosforilazione della &beta;2-syntrophin su S75 vicino il dominio PDZ diminuisce la sua associazione a ICA512 e correla con motilit&agrave; granulo aumentata, mentre la fosforilazione della S90 ha opposto gli effetti. Mostriamo ulteriormente che Cdk5, che regola la secrezione di insulina, tirosine S75. Questi risultati forniscono la visione meccanicistica come stimolazione si sposta granelli insulina da actina corticale, promuovendo cos&igrave; la loro motilit&agrave; ed esocitosi.

&Beta;2 Syntrophin &#xC778;&#xC290;&#xB9B0; &#xBD84; &#xC54C;&#xAC31;&#xC774;&#xC758; &#xC6B4;&#xB3D9; &#xC131;&#xC744; &#xC5B5;&#xC81C; Cdk5 &#xAE30;&#xD310; &#xC774;&#xB2E4;.

Las bases moleculares de la interacci&oacute;n de gr&aacute;nulos de insulina con el citoesqueleto cortical de c&eacute;lulas &beta; pancre&aacute;ticas sigue siendo desconocida. Hemos propuesto que Uni&oacute;n de la prote&iacute;na ICA512 del gr&aacute;nulo para el PDZ dominio de &beta;2-syntrophin anclas de gr&aacute;nulos a filamentos de actina y que la fosforilaci&oacute;n/desfosforilaci&oacute;n de &beta;2-syntrophin regula esta asociaci&oacute;n. Aqu&iacute; probamos esta hip&oacute;tesis analizando las c&eacute;lulas de la INS-1 expresando GFP-&beta;2-syntrophin mediante el uso combinado de m&eacute;todos bioqu&iacute;micos, proyecci&oacute;n de imagen estudia por microscop&iacute;a de fluorescencia confocal y total reflexi&oacute;n interna as&iacute; como microscopia electr&oacute;nica. Nuestros resultados apoyan la noci&oacute;n que &beta;2-syntrophin refrena la movilidad de los gr&aacute;nulos corticales en las c&eacute;lulas de la INS-1 de insulinoma, lo que reduce la secreci&oacute;n de insulina y aumento de tiendas de insulina en las c&eacute;lulas de reclinaci&oacute;n, al tiempo que aumenta la secreci&oacute;n de la insulina sobre el est&iacute;mulo. Mediante espectrometr&iacute;a de masas, ensayos in vitro de la fosforilaci&oacute;n y &beta;2-syntrophin phosphomutants encontramos que la fosforilaci&oacute;n de &beta;2-syntrophin en S75 cerca del dominio PDZ disminuye su uni&oacute;n a ICA512 y se correlaciona con la motilidad del gr&aacute;nulo mayor, mientras que la fosforilaci&oacute;n de S90 tiene enfrente de efectos. Adem&aacute;s, mostramos que Cdk5, que regula la secreci&oacute;n de insulina, fosforila S75. Estos resultados proporcionan la visi&oacute;n mecanicista como estimulaci&oacute;n desplaza gr&aacute;nulos de insulina de actina cortical, promoviendo su motilidad y exocitosis.
