&#x4E2D;&#x65AD;&#x7684;&#x8F74;&#x7A81;&#x8FD0;&#x8F93;&#x7531; huntingtin &#x635F;&#x5931;&#x6216;&#x679C;&#x8747;&#x4E2D;&#x7684;&#x81F4;&#x75C5;&#x6027; polyQ &#x86CB;&#x767D;&#x7684;&#x8868;&#x8FBE;&#x3002;

We tested whether proteins implicated in Huntington's and other polyglutamine (polyQ) expansion diseases can cause axonal transport defects. Reduction of Drosophila huntingtin and expression of proteins containing pathogenic polyQ repeats disrupt axonal transport. Pathogenic polyQ proteins accumulate in axonal and nuclear inclusions, titrate soluble motor proteins, and cause neuronal apoptosis and organismal death. Expression of a cytoplasmic polyQ repeat protein causes adult retinal degeneration, axonal blockages in larval neurons, and larval lethality, but not neuronal apoptosis or nuclear inclusions. A nuclear polyQ repeat protein induces neuronal apoptosis and larval lethality but no axonal blockages. We suggest that pathogenic polyQ proteins cause neuronal dysfunction and organismal death by two non-mutually exclusive mechanisms. One mechanism requires nuclear accumulation and induces apoptosis; the other interferes with axonal transport. Thus, disruption of axonal transport by pathogenic polyQ proteins could contribute to early neuropathology in Huntington's and other polyQ expansion diseases.

Abbiamo testato se le proteine implicate nella Corea di Huntington e altre malattie di espansione delle poliglutamine (polyQ) possono causare difetti di trasporto assonale. Riduzione della drosofila huntingtina ed espressione delle proteine contenenti patogeni polyQ ripete interrompere il trasporto assonale. Patogeni polyQ proteine si accumulano in inclusioni assonale e nucleare, titolo proteine solubili motore e causano apoptosi neuronale e morte organismica. Espressione di una proteina di ripetizione polyQ citoplasmatico provoca degenerazione retinica adulta, blocchi assonali in neuroni larvale e mortalit&agrave; larvale, ma apoptosi neuronale non o inclusioni nucleari. Una proteina di ripetizione polyQ nucleare induce apoptosi neuronale e mortalit&agrave; larvale ma senza blocchi assonali. Suggeriamo che le proteine patogene polyQ causano disfunzione neuronale e morte organismica da due meccanismi non escludono. Un meccanismo richiede accumulo nucleare e induce l&#39;apoptosi; l&#39;altra interferisce con il trasporto assonale. Cos&igrave;, interruzione del trasporto assonale da patogeni polyQ proteine poteva contribuire all&#39;inizio neuropatologia in Corea di Huntington e altre malattie di espansione polyQ.

Essential to this system are the molecular motors, kinesin and dynein, which transport a variety of neuronal cargoes. Elucidating the transport pathways, the identity of the cargoes transported, and the regulation of motor-cargo complexes are areas of intense investigation. Evidence suggests that essential components, including signaling proteins, neuroprotective and repair molecules, and vesicular and cytoskeletal components are all transported. In addition newly emerging data indicate that defects in axonal transport pathways may contribute to the initiation or progression of chronic neuronal dysfunction. In this review we concentrate on microtubule-based motor proteins, their linkers, and cargoes and discuss how factors in the axonal transport pathway contribute to disease states. As additional cargo complexes and transport pathways are identified, an understanding of the role these pathways play in the development of human disease will hopefully lead to new diagnostic and treatment strategies.

Overexpression of amyloid precursor protein (APP), as well as mutations in the APP and presenilin genes, causes rare forms of Alzheimer's disease (AD). These genetic changes have been proposed to cause AD by elevating levels of amyloid-beta peptides (Abeta), which are thought to be neurotoxic. Since overexpression of APP also causes defects in axonal transport, we tested whether defects in axonal transport were the result of Abeta poisoning of the axonal transport machinery. Because directly varying APP levels also alters APP domains in addition to Abeta, we perturbed Abeta generation selectively by combining APP transgenes in Drosophila and mice with presenilin-1 (PS1) transgenes harboring mutations that cause familial AD (FAD). We found that combining FAD mutant PS1 with FAD mutant APP increased Abeta42/Abeta40 ratios and enhanced amyloid deposition as previously reported. Surprisingly, however, this combination suppressed rather than increased APP-induced axonal transport defects in both Drosophila and mice. In addition, neuronal apoptosis induced by expression of FAD mutant human APP in Drosophila was suppressed by co-expressing FAD mutant PS1. We also observed that directly elevating Abeta with fusions to the Familial British and Danish Dementia-related BRI protein did not enhance axonal transport phenotypes in APP transgenic mice. Finally, we observed that perturbing Abeta ratios in the mouse by combining FAD mutant PS1 with FAD mutant APP did not enhance APP-induced behavioral defects. A potential mechanism to explain these findings was suggested by direct analysis of axonal transport in the mouse, which revealed that axonal transport or entry of APP into axons is reduced by FAD mutant PS1. Thus, we suggest that APP-induced axonal defects are not caused by Abeta.

Eine &Uuml;berexpression von Amyloid Vorl&auml;uferprotein (APP) sowie Mutationen in den Genen APP und Presenilin f&uuml;hrt dazu, dass die seltene Formen der Alzheimer-Krankheit (AD). Diese genetischen Ver&auml;nderungen sind vorgeschlagen worden, um AD erh&ouml;hen Ebenen des Amyloid-Beta Peptid (Abeta), verursachen, die neurotoxische beimessen. La sobreexpresi&#xF3;n de la prote&#xED;na precursora amiloide (APP), as&#xED; como las mutaciones en los genes APP y la presenilina, hace que las formas raras de la enfermedad de Alzheimer (EA). Estos cambios gen&#xE9;ticos han sido propuestos para causar la DA mediante la elevaci&#xF3;n de los niveles de los p&#xE9;ptidos beta-amiloide (Abeta), que se cree que son neurot&#xF3;xicos. Dado que la sobreexpresi&#xF3;n de APP tambi&#xE9;n causa defectos en el transporte axonal, hemos probado si los defectos en el transporte axonal fueron el resultado de la intoxicaci&#xF3;n por Abeta de la maquinaria de transporte axonal. Debido a que directamente diferentes niveles de APP tambi&#xE9;n altera dominios de aplicaci&#xF3;n, adem&#xE1;s de Abeta, que perturba la generaci&#xF3;n de Abeta selectivamente mediante la combinaci&#xF3;n de transgenes de APP en Drosophila y ratones con la presenilina-1 (PS1) transgenes albergan mutaciones que causan Alzheimer familiar (FAD). Neurodegeneration induced by abnormal hyperphosphorylation and aggregation of the microtubule-associated protein tau defines neurodegenerative tauopathies. Destabilization of microtubules by loss of tau function and filament formation by toxic gain of function are two mechanisms suggested for how abnormal tau triggers neuronal loss. Recent experiments in kinesin-1 deficient mice suggested that axonal transport defects can initiate biochemical changes that induce activation of axonal stress kinase pathways leading to abnormal tau hyperphosphorylation. Here we show using Drosophila and mouse models of tauopathies that reductions in axonal transport can exacerbate human tau protein hyperphosphorylation, formation of insoluble aggregates and tau-dependent neurodegeneration. Together with previous work, our results suggest that non-lethal reductions in axonal transport, and perhaps other types of minor axonal stress, are sufficient to induce and/or accelerate abnormal tau behavior characteristic of Alzheimer's disease and other neurodegenerative tauopathies.

Hier zeigen wir mit Drosophila und Maus Modelle der Tauopathien, dass K&uuml;rzungen im axonale Transport menschlichen Tau Protein Hyperphosphorylation, Bildung von unl&ouml;slichen Aggregaten und Tau-abh&auml;ngige Neurodegeneration versch&auml;rfen k&ouml;nnen. Zusammen mit fr&uuml;heren Arbeiten zufolge unseren Ergebnissen nicht-t&ouml;dlichen Verringerungen der axonale Transport und vielleicht andere kleinere axonale Stress, ausreichend sind zu induzieren und/oder abnorme Tau Verhalten charakteristisch f&uuml;r die Alzheimer-Krankheit und anderen neurodegenerativen Tauopathien zu beschleunigen.

Neurodegenerazione indotta da anormale iperfosforilazione e aggregazione del tau microtubule-associated protein definisce tauopatie neurodegenerative. The application of nanotechnology in biological research is beginning to have a major impact leading to the development of new types of tools for human health. One focus of nanobiotechnology is the development of nanoparticle-based formulations for use in drug or gene delivery systems. However most of the nano probes currently in use have varying levels of toxicity in cells or whole organisms and therefore are not suitable for in vivo application or long-term use. Here we test the potential of a novel silica based nanoparticle (organically modified silica, ORMOSIL) in living neurons within a whole organism. We show that feeding ORMOSIL nanoparticles to Drosophila has no effect on viability. ORMOSIL nanoparticles penetrate into living brains, neuronal cell bodies and axonal projections. In the neuronal cell body, nanoparticles are present in the cytoplasm, but not in the nucleus. Strikingly, incorporation of ORMOSIL nanoparticles into the brain did not induce aberrant neuronal death or interfered with normal neuronal processes. Our results in Drosophila indicate that these novel silica based nanoparticles are biocompatible and not toxic to whole organisms, and has potential for the development of long-term applications.

Die Anwendung der Nanotechnologie in der biologischen Forschung beginnt stark zur Entwicklung neuer Arten von Werkzeugen f&uuml;r die menschliche Gesundheit auswirken. Ein Schwerpunkt der Nanobiotechnologie ist die Entwicklung von Nanopartikel-basierte Formulierungen f&uuml;r den Einsatz in Drogen- oder gen-Delivery-Systeme. Jedoch die meisten die Nano-Sonden zur Zeit im Einsatz haben unterschiedliche Ebenen der Toxizit&auml;t in Zellen oder ganze Organismen und sind daher nicht f&uuml;r in-vivo-Anwendung oder langfristigen Einsatz geeignet. Hier testen wir das Potenzial der eine neuartige Kiesels&auml;ure basierte Nanopartikel (organisch modifizierte Silika, ORMOSIL) in lebenden Neuronen innerhalb eines Organismus. Wir zeigen, dass die Verf&uuml;tterung von ORMOSIL Nanopartikel an Drosophila hat keinen Einfluss auf die Lebensf&auml;higkeit. ORMOSIL Nanopartikel dringen in die lebenden Gehirn, neuronalen Zelle K&ouml;rper und axonale Projektionen. In neuronale Zellk&ouml;rper, Nanopartikel sind vorhanden im Zytoplasma, aber nicht in den Zellkern. La aplicaci&#xF3;n de la nanotecnolog&#xED;a en la investigaci&#xF3;n biol&#xF3;gica est&#xE1; empezando a tener un impacto importante que lleva al desarrollo de nuevos tipos de herramientas para la salud humana. Uno de los objetivos de la nanobiotecnolog&#xED;a es el desarrollo de formulaciones a base de nanopart&#xED;culas para su uso en sistemas de administraci&#xF3;n de f&#xE1;rmacos o genes. Sin embargo la mayor&#xED;a de las sondas nano actualmente en uso tienen diferentes niveles de toxicidad en c&#xE9;lulas u organismos enteros y por lo tanto no son adecuados para su aplicaci&#xF3;n in vivo o el uso a largo plazo. Aqu&#xED; ponemos a prueba el potencial de las nanopart&#xED;culas de s&#xED;lice una novela basada en (s&#xED;lice modificada org&#xE1;nicamente, ormosil) en neuronas vivas dentro de un organismo completo. Se demuestra que la alimentaci&#xF3;n de nanopart&#xED;culas ormosil a Drosophila tiene ning&#xFA;n efecto sobre la viabilidad. Ormosil nanopart&#xED;culas penetran en los cerebros de vida, los &#xF3;rganos de c&#xE9;lulas neuronales y las proyecciones axonales. En el cuerpo de las c&#xE9;lulas neuronales, nanopart&#xED;culas est&#xE1;n presentes en el citoplasma, pero no en el n&#xFA;cleo. Sorprendentemente, la incorporaci&#xF3;n de nanopart&#xED;culas ormosil en el cerebro no induce muerte neuronal aberrante o interferido con los procesos normales de las neuronas. Nuestros resultados en Drosophila indican que estos nuevos basados &#x200B;&#x200B;en nanopart&#xED;culas de s&#xED;lice son biocompatibles y no t&#xF3;xico para los organismos enteros, y tiene un potencial para el desarrollo de aplicaciones a largo plazo.
