Chronic in vivo imaging studies of the brain require a labeling method that is fast, long-lasting, efficient, nontoxic, and cell-type specific. Over the last decade, adeno-associated virus (AAV) has been used to stably express fluorescent proteins in neurons in vivo. However, AAV's main limitation for many studies (such as those of neuronal development) is the necessity of second-strand DNA synthesis, which delays peak transgene expression. The development of double-stranded AAV (dsAAV) vectors has overcome this limitation, allowing rapid transgene expression. Here, we have injected different serotypes (1, 2, 6, 7, 8, and 9) of a dsAAV vector carrying the green fluorescent protein (GFP) gene into the developing and adult mouse visual cortex and characterized its expression. We observed labeling of both neurons and astrocytes with serotype-specific tropism. dsAAV-GFP labeling showed high levels of neuronal GFP expression as early as 2 days postinjection and as long as a month, surpassing conventional AAV's onset of expression and matching its longevity. Neurons labeled with dsAAV-GFP appeared structurally and electrophysiologically identical to nonlabeled neurons, suggesting that dsAAV-GFP is neither cytotoxic nor alters normal neuronal function. We also demonstrated that dsAAV-labeled cells can be imaged with subcellular resolution in vivo over multiple days. We conclude that dsAAV is an excellent vector for rapid labeling and long-term in vivo imaging studies of astrocytes and neurons on the single cell level within the developing and adult visual cortex.

Chronique dans les &#xE9;tudes d&#x26;#39;imagerie in vivo du cerveau exigent une m&#xE9;thode de marquage qui est rapide, durable, efficace, non toxique, et un type de cellule sp&#xE9;cifique. Au cours de la derni&#xE8;re d&#xE9;cennie, virus ad&#xE9;no-associ&#xE9; (AAV) a &#xE9;t&#xE9; utilis&#xE9; pour exprimer de fa&#xE7;on stable des prot&#xE9;ines fluorescentes dans les neurones in vivo. Wir zeigte auch, dass DsAAV-markierten Zellen &uuml;ber mehrere Tage mit in-vivo subzellulare Aufl&ouml;sung abgebildet werden k&ouml;nnen. Cr&#xF3;nica en vivo en los estudios por im&#xE1;genes del cerebro requieren un m&#xE9;todo de etiquetado que es r&#xE1;pido, duradero, eficiente, no t&#xF3;xico, y la espec&#xED;fica de tipo celular. Durante la &#xFA;ltima d&#xE9;cada, virus adeno-asociado (AAV) se ha utilizado para expresar de forma estable las prote&#xED;nas fluorescentes en las neuronas in vivo. Sin embargo, la limitaci&#xF3;n principal de AAV para muchos estudios (tales como los de desarrollo neuronal) es la necesidad de segunda l&#xED;nea de s&#xED;ntesis de ADN, que retrasa la expresi&#xF3;n del transg&#xE9;n pico. El desarrollo de la doble cadena (AAV dsAAV) vectores ha superado esta limitaci&#xF3;n, lo que permite la expresi&#xF3;n del transgen r&#xE1;pida. En este sentido, se han inyectado diferentes serotipos (1, 2, 6, 7, 8 y 9) de un vector que lleva la dsAAV prote&#xED;na verde fluorescente (GFP) de genes en el desarrollo y la corteza visual del rat&#xF3;n adulto y se caracteriza su expresi&#xF3;n. Hemos observado el etiquetado de las neuronas y astrocitos con el serotipo espec&#xED;fico del tropismo. dsAAV-GFP etiquetado mostraron altos niveles de expresi&#xF3;n de GFP neuronal ya en dos d&#xED;as despu&#xE9;s de la inyecci&#xF3;n y el tiempo como un mes, superando el inicio convencional de AAV de expresi&#xF3;n y la congruencia de su longevidad. Las neuronas marcadas con dsAAV-GFP apareci&#xF3; estructural y electrofisiol&#xF3;gica id&#xE9;ntica a las neuronas nonlabeled, lo que sugiere que dsAAV-GFP no es citot&#xF3;xico ni altera la funci&#xF3;n neuronal normal. Tambi&#xE9;n hemos demostrado que las c&#xE9;lulas marcadas con dsAAV se pueden obtener im&#xE1;genes con resoluci&#xF3;n subcelular en vivo durante varios d&#xED;as. Microglia are the immune cells of the brain. In the absence of pathological insult, their highly motile processes continually survey the brain parenchyma and transiently contact synaptic elements. Aside from monitoring, their physiological roles at synapses are not known. To gain insight into possible roles of microglia in the modification of synaptic structures, we used immunocytochemical electron microscopy, serial section electron microscopy with three-dimensional reconstructions, and two-photon in vivo imaging to characterize microglial interactions with synapses during normal and altered sensory experience, in the visual cortex of juvenile mice. During normal visual experience, most microglial processes displayed direct apposition with multiple synapse-associated elements, including synaptic clefts. Microglial processes were also distinctively surrounded by pockets of extracellular space. In terms of dynamics, microglial processes localized to the vicinity of small and transiently growing dendritic spines, which were typically lost over 2 d. When experience was manipulated through light deprivation and reexposure, microglial processes changed their morphology, showed altered distributions of extracellular space, displayed phagocytic structures, apposed synaptic clefts more frequently, and enveloped synapse-associated elements more extensively. While light deprivation induced microglia to become less motile and changed their preference of localization to the vicinity of a subset of larger dendritic spines that persistently shrank, light reexposure reversed these behaviors. Taken together, these findings reveal different modalities of microglial interactions with synapses that are subtly altered by sensory experience. These findings suggest that microglia may actively contribute to the experience-dependent modification or elimination of a specific subset of synapses in the healthy brain.

Microglia son las c&#xE9;lulas inmunitarias del cerebro. En ausencia de insulto patol&#xF3;gico, sus procesos altamente m&#xF3;viles continuamente examinar el par&#xE9;nquima cerebral y transitoriamente en contacto con los elementos sin&#xE1;pticos. Aparte de la supervisi&#xF3;n, sus funciones fisiol&#xF3;gicas en las sinapsis no se conocen. Para conocer las posibles funciones de la microgl&#xED;a en la modificaci&#xF3;n de las estructuras sin&#xE1;pticas, se utiliz&#xF3; la microscop&#xED;a de electrones inmunocitoqu&#xED;mica, microscop&#xED;a electr&#xF3;nica de serie de la secci&#xF3;n con reconstrucciones tridimensionales, y dos fotones de im&#xE1;genes in vivo para caracterizar las interacciones microgliales con las sinapsis durante la experiencia sensorial normal y alterada , en la corteza visual de ratones j&#xF3;venes. Durante la experiencia visual normal, la mayor&#xED;a de los procesos microgliales muestra aposici&#xF3;n directa con m&#xFA;ltiples sinapsis asociadas a los elementos, incluyendo las hendiduras sin&#xE1;pticas. Procesos microgliales tambi&#xE9;n estaban claramente rodeado de bolsas de espacio extracelular. En t&#xE9;rminos de la din&#xE1;mica, procesos microgliales localizada en las cercan&#xED;as de las espinas dendr&#xED;ticas peque&#xF1;as y creciente transitoriamente, que se perdieron t&#xED;picamente durante 2 d.
