Mitochondria are key regulators of cellular energy and are the focus of a large number of studies examining the regulation of mitochondrial dynamics and biogenesis in healthy and diseased conditions. One approach for monitoring mitochondrial biogenesis is to measure the rate of mitochondrial DNA (mtDNA) replication. We developed a sensitive technique to visualize newly synthesized mtDNA in individual cells in order to study mtDNA replication within subcellular compartments of neurons. The technique combines the incorporation of BrdU and/or EdU into mtDNA together with a tyramide signal amplification protocol. Employing this technique, we visualized and measured mtDNA biogenesis in individual cells. The labeling procedure for EdU allows for more comprehensive results by allowing the comparison of its incorporation with other intracellular markers because it does not require harsh acid or enzyme digests necessary to recover the BrdU epitope. In addition, the utilization of both BrdU and EdU permits sequential pulse-chase experiments to follow the intracellular localization of mtDNA replication. The ability to quantify mitochondrial biogenesis provides an essential tool for investigating alterations in mitochondrial dynamics involved in the pathogenesis of multiple cellular disorders including neuropathies and neurodegenerative diseases.

Mitochondrial DNA (mtDNA) Biogenesis: Visualization and Duel Incorporation of BrdU and EdU Into Newly Synthesized mtDNA In Vitro.

Les mitochondries sont des r&#xE9;gulateurs cl&#xE9;s de l&#x26;#39;&#xE9;nergie cellulaire et font l&#x26;#39;objet d&#x26;#39;un grand nombre d&#x26;#39;&#xE9;tudes portant sur la r&#xE9;gulation de la dynamique mitochondriale et la biogen&#xE8;se dans des conditions saines et malades. Une approche pour la surveillance biogen&#xE8;se mitochondriale est de mesurer le taux de l&#x26;#39;ADN mitochondrial (ADNmt) de r&#xE9;plication. Nous avons d&#xE9;velopp&#xE9; une technique sensible pour visualiser l&#x26;#39;ADN mitochondrial nouvellement synth&#xE9;tis&#xE9; dans des cellules individuelles dans le but d&#x26;#39;&#xE9;tudier la r&#xE9;plication au sein de l&#x26;#39;ADNmt compartiments subcellulaires de neurones. La technique combine l&#x26;#39;incorporation de BrdU et / ou EdU en ADNmt avec un protocole d&#x26;#39;amplification de signal tyramide. En employant cette technique, nous avons visualis&#xE9; et mesur&#xE9; la biogen&#xE8;se ADNmt dans des cellules individuelles. La proc&#xE9;dure d&#x26;#39;&#xE9;tiquetage pour les EdU permet des r&#xE9;sultats plus complets, en permettant la comparaison de sa constitution avec d&#x26;#39;autres marqueurs intracellulaires, car il ne n&#xE9;cessite pas d&#x26;#39;acide dur ou enzyme dig&#xE8;re n&#xE9;cessaires pour r&#xE9;cup&#xE9;rer l&#x26;#39;&#xE9;pitope BrdU. En outre, l&#x26;#39;utilisation de deux BrdU et Edu permet s&#xE9;quentielles pulse-chase exp&#xE9;riences de suivre la localisation intracellulaire de la r&#xE9;plication ADNmt. Mitochondrien sind wichtige Regulatoren der zellularen Energie und stehen im Mittelpunkt einer gro&szlig;en Anzahl von Studien, die die Regulierung der mitochondrialen Dynamik und Biogenese in gesunden und Kranken Bedingungen zu untersuchen. Ein Ansatz zur &Uuml;berwachung der mitochondrialen Biogenese ist die Rate der mitochondriale DNA (MtDNA) zu messen. Wir entwickelten eine sensible Technik um eben synthetisierten MtDNA in einzelne Zellen zu visualisieren, um MtDNA-Replikation innerhalb subzellulare Depots von Neuronen zu studieren. Die Technik kombiniert die Einbeziehung von BrdU und/oder EdU in MtDNA gemeinsam mit einem Tyramide Signal-Verst&auml;rkung-Protokoll. Mit dieser Technik, wir visualisiert und MtDNA Biogenese in einzelnen Zellen gemessen. Das Kennzeichenrecht Verfahren f&uuml;r EdU erlaubt umfassendere Ergebnisse dadurch, dass des Vergleichs der Gr&uuml;ndung mit anderen intrazellul&auml;ren Markern weil es keine harten S&auml;ure erfordert oder Enzym verdaut der BrdU-Epitope Wiederherstellung erforderlich. Las mitocondrias son los principales reguladores de la energ&#xED;a celular y son el centro de un gran n&#xFA;mero de estudios que examinan la regulaci&#xF3;n de la biog&#xE9;nesis mitocondrial y la din&#xE1;mica en condiciones sanas y enfermas. Un enfoque para el seguimiento de la biog&#xE9;nesis mitocondrial es medir la velocidad de ADN mitocondrial (ADNmt) de replicaci&#xF3;n. Hemos desarrollado una t&#xE9;cnica sensible para visualizar ADNmt reci&#xE9;n sintetizada en las c&#xE9;lulas individuales con el fin de estudiar la replicaci&#xF3;n del ADN mitocondrial dentro de los compartimentos subcelulares de las neuronas. La t&#xE9;cnica combina la incorporaci&#xF3;n de BrdU y / o EDU en ADNmt junto con un protocolo de amplificaci&#xF3;n de la se&#xF1;al tyramide. El empleo de esta t&#xE9;cnica, visualizar y medir la biog&#xE9;nesis de ADNmt en las c&#xE9;lulas individuales. El procedimiento de etiquetado para EDU permite obtener resultados m&#xE1;s amplios, al permitir la comparaci&#xF3;n de su incorporaci&#xF3;n con otros marcadores intracelulares, ya que no requieren &#xE1;cido &#xE1;spero o enzima digiere necesario recuperar el ep&#xED;topo BrdU.
