Directed cell migration is a physical process that requires dramatic changes in cell shape and adhesion to the extracellular matrix. For efficient movement, these processes must be spatiotemporally coordinated. To a large degree, the morphological changes and physical forces that occur during migration are generated by a dynamic filamentous actin (F-actin) cytoskeleton. Adhesion is regulated by dynamic assemblies of structural and signaling proteins that couple the F-actin cytoskeleton to the extracellular matrix. Here, we review current knowledge of the dynamic organization of the F-actin cytoskeleton in cell migration and the regulation of focal adhesion assembly and disassembly with an emphasis on how mechanical and biochemical signaling between these two systems regulate the coordination of physical processes in cell migration.

Per efficiente movimento, questi processi devono essere spatiotemporally coordinati. In larga misura, i cambiamenti morfologici e le forze fisiche che si verificano durante la migrazione sono generate da un citoscheletro dinamico actina filamentosa (F-actina). Adesione &egrave; regolato da assembly dinamici strutturali e proteine di segnalazione che citoscheletro coppia F-actina alla matrice extracellulare. The regulation of cellular traction forces on the extracellular matrix is critical to cell adhesion, migration, proliferation, and differentiation. Diverse lamellar actin organizations ranging from contractile lamellar networks to stress fibers are observed in adherent cells. Although lamellar organization is thought to reflect the extent of cellular force generation, understanding of the physical behaviors of the lamellar actin cytoskeleton is lacking. To elucidate these properties, we visualized the actomyosin dynamics and organization in U2OS cells over a broad range of forces. At low forces, contractile lamellar networks predominate and force generation is strongly correlated to actomyosin retrograde flow dynamics with nominal change in organization. Lamellar networks build âˆ¼60% of cellular tension over rapid time scales. At high forces, reorganization of the lamellar network into stress fibers results in moderate changes in cellular tension over slower time scales. As stress fibers build and tension increases, myosin band spacing decreases and Î±-actinin bands form. On soft matrices, force generation by lamellar networks is unaffected, whereas tension-dependent stress fiber assembly is abrogated. These data elucidate the dynamic and structural signatures of the actomyosin cytoskeleton at different levels of tension and set a foundation for quantitative models of cell and tissue mechanics.

En las fuerzas bajas, contr&#xE1;ctiles redes laminares predominan y la generaci&#xF3;n de la fuerza est&#xE1; fuertemente correlacionada con la din&#xE1;mica de la actomiosina flujo retr&#xF3;grado con el cambio nominal de la organizaci&#xF3;n. Redes de l&#xE1;minas de construir alrededor del 60% de la tensi&#xF3;n celular en escalas de tiempo r&#xE1;pidos. En grandes fuerzas, la reorganizaci&#xF3;n de la red de laminar en fibras de estr&#xE9;s da lugar a cambios moderados en la tensi&#xF3;n celular en escalas de tiempo m&#xE1;s lento. Como las fibras de estr&#xE9;s construir y la tensi&#xF3;n aumenta, disminuye la miosina de la banda con espacio y &#x3B1;-actinina formar bandas. En las matrices suaves, la generaci&#xF3;n de la fuerza por las redes laminares no se ve afectada, mientras que la tensi&#xF3;n dependiente de la Asamblea de fibras de estr&#xE9;s se suprime. Focal adhesions (FAs) are the predominant mechanism by which cells mechanically couple to and exert traction forces on their extracellular matrix (ECM). It is widely presumed that FA size is modulated by force to mediate changes in adhesion strength at different levels of cellular tension. However, previous studies seeking correlations between force and FA morphology have yielded variable and often conflicting results. Here we show that a strong correlation between adhesion size and traction force exists only during the initial stages of myosin-mediated adhesion maturation and growth. For mature adhesions, no correlation between traction stress and size is observed. Rather, the tension that is sustained at mature adhesions is more strongly influenced by proximity to the cell edge, with peripheral adhesions transmitting higher tension than adhesions near the cell center. Finally, we show that mature adhesions can withstand sixfold increases in tension without changes in size. Thus, although a strong correlation between adhesion size and mechanical tension is observed during the initial stages of myosin-mediated adhesion maturation, no correlation is observed in mature, elongated adhesions. This work places spatiotemporal constraints on the force-dependent growth of adhesions and provides insight into the mechanical regulation of cell-ECM adhesion.

Adh&#xE9;sions focales (AF) sont le m&#xE9;canisme pr&#xE9;dominant par lequel les cellules m&#xE9;caniquement un couple &#xE0; exercer des forces de traction et de leur matrice extracellulaire (MEC). Il est largement suppos&#xE9; que la taille FA est modul&#xE9;e par la force &#xE0; la m&#xE9;diation des changements dans la force d&#x26;#39;adh&#xE9;rence &#xE0; diff&#xE9;rents niveaux de tension cellulaire. Cependant, des &#xE9;tudes ant&#xE9;rieures qui cherchent des corr&#xE9;lations entre la force et la morphologie FA ont donn&#xE9; des r&#xE9;sultats variables et souvent contradictoires. Es wird allgemein vermutet, dass FA-Gr&ouml;&szlig;e durch Kraft Ver&auml;nderungen Adh&auml;sion St&auml;rke auf verschiedenen Ebenen der zellul&auml;ren Spannung vermitteln moduliert wird. Schlie&szlig;lich zeigen wir, dass Reife Adh&auml;sionen Sechsfache Anstieg der Spannung ohne &Auml;nderungen in der Gr&ouml;&szlig;e gewachsen sind. Zwar eine starke Korrelation zwischen Haftung und mechanische Spannungen in der Anfangsphase der Myosin-vermittelte Adh&auml;sion Reifung beobachtet wird, ist somit keine Korrelation in Reife, langgestreckte Adh&auml;sionen beobachtet. Diese Arbeit raumzeitliche Einschr&auml;nkungen auf das Kraft-abh&auml;ngiges Wachstum von Adh&auml;sionen und bietet Einblick in die mechanische Regelung der Zelle-ECM-Adh&auml;sion.

Adhesiones focales (AF) son el principal mecanismo por el cual las c&#xE9;lulas mec&#xE1;nicamente a la par y ejercer fuerzas de tracci&#xF3;n en su matriz extracelular (MEC). Es ampliamente presume que el tama&#xF1;o de FA es modulada por la fuerza para mediar cambios en la fuerza de adhesi&#xF3;n a diferentes niveles de tensi&#xF3;n celular. Sin embargo, los estudios anteriores en busca de correlaciones entre la fuerza y &#x200B;&#x200B;la morfolog&#xED;a de la FA han dado resultados variables y en ocasiones contradictorias. As&#xED;, aunque una fuerte correlaci&#xF3;n entre el tama&#xF1;o de la adhesi&#xF3;n y la tensi&#xF3;n mec&#xE1;nica se observa durante las etapas iniciales de maduraci&#xF3;n adherencia mediada por la miosina, ninguna correlaci&#xF3;n se observa en las adherencias maduros, alargados. Este trabajo pone restricciones espacio-temporales en el crecimiento de la fuerza dependiente de las adherencias y da una idea de la regulaci&#xF3;n mec&#xE1;nica de la adhesi&#xF3;n c&#xE9;lula-MEC.
