TDP-43 is an RNA/DNA-binding protein implicated in transcriptional repression and mRNA processing. Inclusions of TDP-43 are hallmarks of frontotemporal dementia and amyotrophic lateral sclerosis. Besides aggregation of TDP-43, loss of nuclear localization is observed in disease. To identify relevant targets of TDP-43, we performed expression profiling. Thereby, histone deacetylase 6 (HDAC6) downregulation was discovered on TDP-43 silencing and confirmed at the mRNA and protein level in human embryonic kidney HEK293E and neuronal SH-SY5Y cells. This was accompanied by accumulation of the major HDAC6 substrate, acetyl-tubulin. HDAC6 levels were restored by re-expression of TDP-43, dependent on RNA binding and the C-terminal protein interaction domains. Moreover, TDP-43 bound specifically to HDAC6 mRNA arguing for a direct functional interaction. Importantly, in vivo validation in TDP-43 knockout Drosophila melanogaster confirmed the specific downregulation of HDAC6. HDAC6 is necessary for protein aggregate formation and degradation. Indeed, HDAC6-dependent reduction of cellular aggregate formation and increased cytotoxicity of polyQ-expanded ataxin-3 were found in TDP-43 silenced cells. In conclusion, loss of functional TDP-43 causes HDAC6 downregulation and might thereby contribute to pathogenesis.

TDP-43 es una prote&#xED;na de ARN / ADN de uni&#xF3;n implicados en la represi&#xF3;n de la transcripci&#xF3;n y procesamiento del ARNm. Las inclusiones de TDP-43 son caracter&#xED;sticas de la demencia frontotemporal y la esclerosis lateral amiotr&#xF3;fica. Adem&#xE1;s de la agregaci&#xF3;n de TDP-43, la p&#xE9;rdida de localizaci&#xF3;n nuclear se observa en la enfermedad. Para identificar los objetivos pertinentes de la TDP-43, se realiz&#xF3; un perfil de expresi&#xF3;n. De esta manera, regulaci&#xF3;n a la baja histona deacetilasa 6 (HDAC6) fue descubierta en la TDP-43 silenciamiento y confirmada en el nivel de ARNm y prote&#xED;nas en HEK293E de ri&#xF1;&#xF3;n embrionario humano y de las c&#xE9;lulas neuronales SH-SY5Y. Esto estuvo acompa&#xF1;ado por la acumulaci&#xF3;n de los principales HDAC6 sustrato, acetil-tubulina. HDAC6 niveles fueron restaurados por la re-expresi&#xF3;n de la TDP-43, depende de la uni&#xF3;n de la ARN y los dominios de la prote&#xED;na C-terminal de la interacci&#xF3;n. Por otra parte, la TDP-43 vinculado espec&#xED;ficamente a HDAC6 ARNm argumentando a favor de una interacci&#xF3;n funcional directa. Es importante destacar que, en la validaci&#xF3;n in vivo en el TDP-43 nocaut Drosophila melanogaster confirm&#xF3; la regulaci&#xF3;n a la baja espec&#xED;fica de HDAC6. HDAC6 es necesario para la formaci&#xF3;n de prote&#xED;na agregada y la degradaci&#xF3;n. En efecto, dependiente de HDAC6 reducci&#xF3;n de la formaci&#xF3;n y la citotoxicidad celular total mayor de polyQ-expandida ataxina-3 se encuentra en TDP-43 c&#xE9;lulas silenciados. En conclusi&#xF3;n, la p&#xE9;rdida de funcionamiento de TDP-43 hace que HDAC6 regulaci&#xF3;n a la baja y por lo tanto podr&#xED;a contribuir a la patog&#xE9;nesis.
