Vascular endothelial growth factor (VEGF) acting through VEGF receptor 2 (VEGFR2) on endothelial cells (ECs) is a key regulator of angiogenesis, a process essential for wound healing and tumor metastasis. Rap1a and Rap1b, 2 highly homologous small G proteins, are both required for angiogenesis in vivo and for normal EC responses to VEGF. Here we sought to determine the mechanism through which Rap1 promotes VEGF-mediated angiogenesis. Using lineage-restricted Rap1-knockout mice we show that Rap1-deficiency in endothelium leads to defective angiogenesis in vivo, in a dose-dependent manner. Using ECs obtained from Rap1-deficient mice we demonstrate that Rap1b promotes VEGF-VEGFR2 kinase activation and regulates integrin activation. Importantly, the Rap1b-dependent VEGF-VEGFR2 activation is in part mediated via integrin Î±(v)Î²(3). Furthermore, in an in vivo model of zebrafish angiogenesis, we demonstrate that Rap1b is essential for the sprouting of intersomitic vessels, a process known to be dependent on VEGF signaling. Using 2 distinct pharmacologic VEGFR2 inhibitors we show that Rap1b and VEGFR2 act additively to control angiogenesis in vivo. We conclude that Rap1b promotes VEGF-mediated angiogenesis by promoting VEGFR2 activation in ECs via integrin Î±(v)Î²(3). These results provide a novel insight into the role of Rap1 in VEGF signaling in ECs.

Rap1 promotes VEGFR2 activation and angiogenesis by a mechanism involving integrin Î±vÎ²â‚ƒ.

Facteur de croissance vasculaire endoth&#xE9;liale (VEGF) agissant par l&#x26;#39;interm&#xE9;diaire du r&#xE9;cepteur VEGF 2 (VEGFR2) sur les cellules endoth&#xE9;liales (CE) est un r&#xE9;gulateur cl&#xE9; de l&#x26;#39;angiogen&#xE8;se, un processus essentiel pour la gu&#xE9;rison des plaies et des m&#xE9;tastases tumorales. Rap1a et Rap1b, 2 hautement homologue petites prot&#xE9;ines G, sont tous deux n&#xE9;cessaires pour l&#x26;#39;angiogen&#xE8;se in vivo et des r&#xE9;ponses communautaires normales &#xE0; VEGF. Ici nous avons cherch&#xE9; &#xE0; d&#xE9;terminer le m&#xE9;canisme par lequel Rap1 favorise VEGF-angiogen&#xE8;se induite. Important, l&#x26;#39;Rap1b-d&#xE9;pendante du VEGF VEGFR2-activation est en partie m&#xE9;di&#xE9;e par l&#x26;#39;int&#xE9;grine &#x3B1; (v) &#x3B2; (3). En outre, dans un mod&#xE8;le in vivo de l&#x26;#39;angiogen&#xE8;se poisson z&#xE8;bre, nous d&#xE9;montrons que Rap1b est essentielle pour la germination des navires intersomitic, un processus connu pour &#xEA;tre d&#xE9;pendant de signalisation du VEGF. Vaskul&auml;ren endothelialen Wachstumsfaktor (VEGF) handelnd durch VEGF-Rezeptor 2 (VEGFR2) auf Endothelzellen (ECs) ist ein wichtiger Regulator der Angiogenese, ein Prozess wesentlich f&uuml;r Wunde heilen und Tumor Metastasen. Rap1a und Rap1b, 2 hochgradig homologe kleine G-Proteine, sind beide f&uuml;r Angiogenesis in vivo und normalen EG Antworten zu VEGF erforderlich. Wir wollten hier den Mechanismus zu bestimmen, durch den welche Rap1 VEGF-vermittelten Angiogenese f&ouml;rdert. Mit Linie kalorienarme Rap1-Knockout-M&auml;use zeigen wir, dass Rap1-Mangel im Endothel zu defekten Angiogenesis in vivo eine dosisabh&auml;ngige Weise f&uuml;hrt. Mit ECs gewonnenen Rap1-defizienten M&auml;usen zeigen wir, dass Rap1b VEGF-VEGFR2 Kinase Aktivierung f&ouml;rdert und reguliert die Integrin Aktivierung. Wichtig ist, ist die Aktivierung von Rap1b abh&auml;ngigen VEGF-VEGFR2 teilweise durch Integrin &alpha;(v)&beta;(3) vermittelt. Dar&uuml;ber hinaus in einem in-vivo-Modell von Zebrafish Angiogenesis beweisen wir, dass Rap1b wesentlich f&uuml;r das sprie&szlig;en von intersomitic Schiffen, eines Prozess namens VEGF Signalisierung abh&auml;ngig zu sein. Mit 2 unterschiedliche pharmakologische VEGFR2 Hemmnissen zeigen wir, dass Rap1b und VEGFR2 Additiv handeln, um Angiogenesis in vivo zu steuern. Wir schlie&szlig;en, dass Rap1b VEGF-vermittelten Angiogenese f&ouml;rdert durch die F&ouml;rderung von VEGFR2 Aktivierung in ECs &uuml;ber Integrin &alpha;(v)&beta;(3). Diese Ergebnisse liefern einen neuartigen Einblick in die Rolle von Rap1 bei VEGF signalisieren in ECs.

Vascular endothelial growth factor (VEGF) che agiscono attraverso recettori VEGF 2 (VEGFR2) sulle cellule endoteliali (ECs) &egrave; un regolatore chiave dell&#39;angiogenesi, un processo essenziale per metastasi di tumore e di guarigione ferita. Rap1a e Rap1b, 2 altamente omologhe piccole proteine G, sono che entrambi necessari per l&#39;angiogenesi in vivo e per le normali risposte CE di VEGF. Qui abbiamo cercato di determinare il meccanismo attraverso il quale Rap1 promuove l&#39;angiogenesi VEGF-mediata. Utilizzando topi knockout Rap1 lineage-restricted mostriamo che Rap1-deficit di endotelio conduce a difettoso angiogenesi in vivo, in modo dose-dipendente. Rap1 integrin &alpha;v&beta;&#x2083; &#xAD00;&#xB828; &#xB41C; &#xBA54;&#xCEE4;&#xB2C8;&#xC998;&#xC5D0; &#xC758;&#xD574; VEGFR2 &#xD65C;&#xC131;&#xD654; &#xBC0F; &#xC2E0;&#xC0DD;&#xC744; &#xCD09;&#xC9C4; &#xD569;&#xB2C8;&#xB2E4;.

Factor de crecimiento endotelial vascular (VEGF), actuando a trav&#xE9;s del receptor 2 de VEGF (VEGFR2) en las c&#xE9;lulas endoteliales (EC) es un regulador clave de la angiog&#xE9;nesis, un proceso esencial para la cicatrizaci&#xF3;n de heridas y la met&#xE1;stasis del tumor. Rap1a y Rap1b, 2 prote&#xED;nas G altamente hom&#xF3;loga peque&#xF1;os, ambos son necesarios para la angiog&#xE9;nesis in vivo y para respuestas normales CE a VEGF. Aqu&#xED; hemos tratado de determinar el mecanismo a trav&#xE9;s del cual Rap1 promueve la angiog&#xE9;nesis mediada por VEGF. Uso de linaje restringido Rap1 ratones knock-out nos muestran que la deficiencia de Rap1 en el endotelio lleva a un defecto en la angiog&#xE9;nesis in vivo, de una manera dosis-dependiente. Uso de EC de obtener Rap1 ratones deficientes que demuestran que Rap1b promueve la activaci&#xF3;n de VEGF-VEGFR2 cinasa y regula la activaci&#xF3;n de la integrina. Es importante destacar que la Rap1b dependiente de VEGF-VEGFR2 activaci&#xF3;n es en parte mediada a trav&#xE9;s de la integrina &#x3B1; (v) &#x3B2; (3). Adem&#xE1;s, en un modelo in vivo de la angiog&#xE9;nesis pez cebra, se demuestra que Rap1b es esencial para el crecimiento de vasos intersomitic, un proceso conocido por ser dependiente de la se&#xF1;alizaci&#xF3;n VEGF. Con 2 diferentes inhibidores farmacol&#xF3;gicos VEGFR2 nos muestran que Rap1b VEGFR2 y actuar de forma aditiva para controlar la angiog&#xE9;nesis in vivo. Se concluye que el VEGF Rap1b promueve la angiog&#xE9;nesis mediada por promover VEGFR2 activaci&#xF3;n en ECs a trav&#xE9;s de la integrina &#x3B1; (v) &#x3B2; (3). Estos resultados proporcionan una visi&#xF3;n novedosa sobre el papel de Rap1 de se&#xF1;alizaci&#xF3;n VEGF en EC.
