&#x52A8;&#x7269;&#x57FA;&#x56E0;&#x82AF;&#x7247;&#x5206;&#x6790;&#x63ED;&#x793A;&#x4E86;&#x4E0D;&#x540C; WNT &#x4FE1;&#x53F7;&#x901A;&#x8DEF;&#x5728; somitic &#x4E2D;&#x80DA;&#x5C42;&#x7684;&#x65B0;&#x76EE;&#x6807;&#x3002;

WNT signaling plays a major role in patterning the dermomyotome of the somitic mesoderm. However, knowledge of downstream target genes and their regulation is limited. To identify new genes involved in the development and early patterning of the somite, we performed a comparison of gene expression by microarray between the presomitic mesoderm and the 5 most recently formed somites of the mouse at embryonic day 9.5. We identified 207 genes upregulated and 120 genes downregulated in somite formation. Expression analysis and functional categorization of these genes demonstrate this to be a diverse pool that provides a valuable resource for studying somite development. Thus far, we have found three genes expressed in the dermomyotome of the early somite. Consistent with their expression patterns, these genes are transcriptional targets of WNT signals, but display differential activation by different WNTs. We further demonstrate that 1 of these genes, Troy, is a direct target of canonical WNT signaling, while the other 2 genes, Selp and Arl4, are not. Thus, our microarray study using microdissected tissues not only provides global information on gene expression during somite development, it also provides novel targets to study the inductive signaling pathways that direct somite patterning.

La signalisation WNT joue un r&ocirc;le majeur dans la structuration de la dermomyotome du m&eacute;soderme somitique. Cependant, la connaissance des g&egrave;nes cibles en aval et de leur r&eacute;gulation est limit&eacute;e. Afin d&#39;identifier de nouveaux g&egrave;nes impliqu&eacute;s dans le d&eacute;veloppement et la structuration au d&eacute;but de la somite, nous r&eacute;alis&eacute; une comparaison de l&#39;expression g&eacute;nique par microarray entre le m&eacute;soderme ont et les 5 somites plus r&eacute;cemment form&eacute;s de la souris &agrave; jour embryonnaire 9.5. Nous avons identifi&eacute; 207 g&egrave;nes surexprim&eacute;s et 120 g&egrave;nes sous-exprim&eacute;s dans formation somite. Segnalazione WNT gioca un ruolo importante nella creazione di una serie dermomyotome del mesoderma somitic. Tuttavia, conoscenza dei geni bersaglio a valle e il loro regolamento &egrave; limitato. Se&ntilde;alizaci&oacute;n de WNT desempe&ntilde;a un papel importante en el dibujo de la dermomyotome del mesodermo somitic. Sin embargo, el conocimiento de genes diana y su regulaci&oacute;n es limitado. Para identificar nuevos genes implicados en el desarrollo y los patrones tempranos de las somitas, realizamos una comparaci&oacute;n de la expresi&oacute;n g&eacute;nica por microarray entre el mesodermo de la presomitic y las 5 somitas recientemente formados del rat&oacute;n en el d&iacute;a embrionario 9.5. Select members of the Wnt family of secreted glycoproteins have been implicated in inducing the myogenic determinant genes Pax3, MyoD and Myf5 during mammalian embryogenesis, but the mechanism of induction has not been defined. We describe an unexpected role for protein kinase A (PKA) signalling via CREB in this induction. Using a combination of in vitro explant assays, mutant analysis and gene delivery into mouse embryos cultured ex vivo, we demonstrate that adenylyl cyclase signalling via PKA and its target transcription factor CREB are required for Wnt-directed myogenic gene expression. Wnt proteins can also stimulate CREB-mediated transcription, providing evidence for a Wnt signalling pathway involving PKA and CREB. Our findings raise the possibility that PKA/CREB signalling may also contribute to other Wnt-regulated processes in embryonic patterning, stem cell renewal and cancer.

Selectos miembros de la familia Wnt de glicoprote&iacute;nas secretadas se han implicado en la inducci&oacute;n de los genes determinantes mi&oacute;genos Pax3, MyoD y Myf5 durante la embriog&eacute;nesis mam&iacute;feros, pero no se ha definido el mecanismo de inducci&oacute;n. Usando una combinaci&oacute;n de ensayos in vitro del explante, an&aacute;lisis mutante y entrega de genes en embriones de rat&oacute;n cultivadas ex vivo, demostramos que adenylyl cyclase se&ntilde;alizaci&oacute;n v&iacute;a PKA y su factor de transcripci&oacute;n de destino CREB se requieren para expresi&oacute;n g&eacute;nica mi&oacute;gena Wnt-dirigido. We report here a novel approach to direct gene expression in the mouse somite based on the combined application of adenovirus-mediated gene delivery and whole embryo ex vivo culture. As proof of principle, we show functional analysis of somites microinjected with an engineered virus expressing an activated form of Smoothened, the signaling receptor for Sonic Hedgehog (SHH). As adenovirus can infect many embryonic tissues in the mouse, this method may provide an effective alternative to conventional transgenesis for targeted spatial and temporal gene expression.

In spite of the therapeutic importance of endoderm derivatives such as the pancreas, liver, lung, and intestine, there are few molecular markers specific for early endoderm. In order to identify endoderm-specific genes as well as to define transcriptional differences between definitive and visceral endoderm, we performed microarray analysis on E8.25 definitive and visceral endoderm. We have developed an early endoderm gene expression signature, and clarified the transcriptional similarities and differences between definitive and visceral endoderm. Additionally, we have developed methods for flow cytometric isolation of definitive and visceral endoderm. These results shed light on the mechanism of endoderm formation and should facilitate investigation of endoderm formation from embryonic stem cells.

En d&#xE9;pit de l&#x26;#39;importance th&#xE9;rapeutique des d&#xE9;riv&#xE9;s endodermiques tels que le pancr&#xE9;as, le foie, les poumons et l&#x26;#39;intestin, il ya peu de marqueurs mol&#xE9;culaires sp&#xE9;cifiques pour l&#x26;#39;endoderme d&#xE9;but. Afin d&#x26;#39;identifier des g&#xE8;nes sp&#xE9;cifiques endoderme ainsi que de d&#xE9;finir les diff&#xE9;rences entre la transcription endoderme d&#xE9;finitif et visc&#xE9;rale, nous avons effectu&#xE9; l&#x26;#39;analyse des micror&#xE9;seaux sur E8.25 endoderme d&#xE9;finitif et visc&#xE9;rale. Nous avons d&#xE9;velopp&#xE9; une signature endoderme d&#xE9;but l&#x26;#39;expression des g&#xE8;nes, et de clarifier les similitudes et les diff&#xE9;rences entre la transcription endoderme d&#xE9;finitif et visc&#xE9;rale. En outre, nous avons d&#xE9;velopp&#xE9; des m&#xE9;thodes pour l&#x26;#39;isolement de cytom&#xE9;trie en flux de l&#x26;#39;endoderme d&#xE9;finitif et visc&#xE9;rale. Ermittlung Endodermis-spezifische Gene als auch hinsichtlich transkriptionelle Unterschiede zwischen definitive und viszeralen Endodermis definieren, f&uuml;hrten wir Microarray-Analyse auf E8.25 definitive und viszeralen Endodermis. Wir haben eine fr&uuml;he Endodermis Gen-Ausdruck-Signatur entwickelt und pr&auml;zisiert die transkriptionelle &Auml;hnlichkeiten und Unterschiede zwischen definitive und viszeralen Endodermis. We report that DNA methyltransferase and histone deacetylase (HDAC) inhibitors improve reprogramming efficiency. In embryonic stem (ES) cells, a well-characterized transcriptional network promotes pluripotency and represses gene expression required for differentiation. In comparison, the transcriptional networks that promote differentiation of ES cells and the blastocyst inner cell mass are poorly understood. Here, we show that Sox17 is a transcriptional regulator of differentiation in these pluripotent cells. ES cells deficient in Sox17 fail to differentiate into extraembryonic cell types and maintain expression of pluripotency-associated transcription factors, including Oct4, Nanog, and Sox2. In contrast, forced expression of Sox17 down-regulates ES cell-associated gene expression and directly activates genes functioning in differentiation toward an extraembryonic endoderm cell fate. We show these effects of Sox17 on ES cell gene expression are mediated at least in part through a competition between Sox17 and Nanog for common DNA-binding sites. By elaborating the function of Sox17, our results provide insight into how the transcriptional network promoting ES cell self-renewal is interrupted, allowing cellular differentiation.

ES-Zellen einen Mangel an Sox17 nicht in extraembryonic Zelltypen differenzieren und Ausdruck der Pluripotenz-assoziierte Transkriptionsfaktoren, einschlie&szlig;lich Oct4, Nanog und Sox2 pflegen. Im Gegensatz dazu erzwungene Ausdruck der Sox17 nach unten-reguliert ES Zelle-assoziierten Genexpression und direkt aktiviert Gene funktionieren bei Differenzierung gegen&uuml;ber einem extraembryonic Endodermis Zelle Schicksal. Wir zeigen, dass diese Auswirkungen des Sox17 auf ES Zelle Genexpression zumindest teilweise durch einen Wettbewerb zwischen Sox17 und Nanog f&uuml;r gemeinsame DNA-Bindungsstellen vermittelt werden. En madre embrionarias (ES) las c&#xE9;lulas, una red bien caracterizado transcripcional promueve la pluripotencia y reprime la expresi&#xF3;n de genes necesaria para la diferenciaci&#xF3;n. En comparaci&#xF3;n, las redes de transcripci&#xF3;n que promueven la diferenciaci&#xF3;n de c&#xE9;lulas madre embrionarias de blastocisto y de la masa celular interna son poco conocidos. Aqu&#xED;, mostramos que Sox17 es un regulador transcripcional de la diferenciaci&#xF3;n de estas c&#xE9;lulas pluripotentes. C&#xE9;lulas madre embrionarias deficientes en Sox17 no se diferencian en tipos de c&#xE9;lulas extraembrionarias y mantener la expresi&#xF3;n de factores de transcripci&#xF3;n asociados a la pluripotencia, incluyendo Oct4, Nanog, y Sox2. Por el contrario, la expresi&#xF3;n forzada de Sox17 regula a la baja ES asociada a las c&#xE9;lulas la expresi&#xF3;n g&#xE9;nica y activa directamente los genes que funcionan en la diferenciaci&#xF3;n hacia un destino de c&#xE9;lulas del endodermo extraembrionario. Nos muestran estos efectos de Sox17 en la expresi&#xF3;n g&#xE9;nica de c&#xE9;lulas ES son mediadas, al menos en parte, a trav&#xE9;s de una competencia entre Sox17 y Nanog para el com&#xFA;n de uni&#xF3;n al ADN sitios. Fertilized mouse zygotes can reprogram somatic cells to a pluripotent state. Human zygotes might therefore be useful for producing patient-derived pluripotent stem cells. However, logistical, legal and social considerations have limited the availability of human eggs for research. Here we show that a significant number of normal fertilized eggs (zygotes) can be obtained for reprogramming studies. Using these zygotes, we found that when the zygotic genome was replaced with that of a somatic cell, development progressed normally throughout the cleavage stages, but then arrested before the morula stage. This arrest was associated with a failure to activate transcription in the transferred somatic genome. In contrast to human zygotes, mouse zygotes reprogrammed the somatic cell genome to a pluripotent state within hours after transfer. Our results suggest that there may be a previously unappreciated barrier to successful human nuclear transfer, and that future studies could focus on the requirements for genome activation.

Hier zeigen wir, dass eine betr&auml;chtliche Anzahl von normal befruchteten Eier (Zygoten) abgerufen werden kann, f&uuml;r die Neuprogrammierung Studien. Verwenden diese Zygoten, fanden wir, dass als zygotic Genom mit einer somatischen Zelle ersetzt wurde, Entwicklung normalerweise w&auml;hrend der Spaltung-Stadien fortgeschritten, aber dann vor dem Morulastadium verhaftet. Diese Verhaftung war verbunden mit einem Ausfall, Transkription im &uuml;bertragenen somatische Genom zu aktivieren. Im Gegensatz zu menschlichen Zygoten umprogrammiert Maus Zygoten somatische Genom in einen pluripotenten Zustand innerhalb von Stunden nach &Uuml;bertragung. Cigotos fertilizados de rat&#xF3;n se pueden reprogramar c&#xE9;lulas som&#xE1;ticas a un estado pluripotente. Cigotos humanos tanto, podr&#xED;a ser &#xFA;til para producir paciente derivados de c&#xE9;lulas madre pluripotentes. Sin embargo, las consideraciones log&#xED;sticas, legales y sociales han limitado la disponibilidad de &#xF3;vulos humanos para investigaci&#xF3;n. Aqu&#xED; nos muestran que un n&#xFA;mero significativo de los huevos fecundados normales (cigotos) se pueden obtener para la reprogramaci&#xF3;n de los estudios. El uso de estos cigotos, hemos encontrado que cuando el genoma cig&#xF3;ticos se reemplaz&#xF3; con el de una c&#xE9;lula som&#xE1;tica, el desarrollo progres&#xF3; normalmente a lo largo de las etapas de escisi&#xF3;n, pero entonces detenido antes de la fase de m&#xF3;rula. Esta detenci&#xF3;n se asoci&#xF3; con un fracaso para activar la transcripci&#xF3;n en el genoma transferido som&#xE1;tica.
