Development of inducible genetic switches for in vivo use with transgenic mice has revolutionized many areas in modern molecular biology. Combining two techniques, Cre/loxP-based genetic recombination and ligand-dependent activation of a chimeric protein, we generated transgenic mice which allow for the spatiotemporal control of expression and of activity of the proto-oncogene c-myc. To these ends, the gene encoding the tamoxifen-inducible c-mycER(T) fusion protein (mycER(T)) was inserted in the ubiquitously active ROSA 26 gene locus by gene targeting. In the resulting ROSAMER allele, generalized transcription of the mycER(T) gene is prevented by a preceding transcriptional stop sequence which is flanked by loxP sites. Crosses of ROSAMER transgenic mice with Mox2 cre transgenic mice revealed tight control of mycER(T) transcription in various tissues unless the transcriptional stop sequence was removed by cre-mediated excision. Furthermore, we were able to demonstrate tamoxifen-dependent activation of the MycER(T) protein in embryonic fibroblasts derived from such mice. As a proof of principle, we demonstrate that primary neural crest cultures established from ROSAMER mice maintain their proliferative capacity in a 4-OHT-dependent manner. Furthermore, we demonstrate that such neural crest cells retain their differentiation potential as shown by expression of NF 160, a marker of neuronal differentiation upon 4-OHT withdrawal. The transgenic mice produced may thus be valuable tools for studying the cell type-specific effects of c-myc activity in development and disease.

D&eacute;veloppement d&#39;inductibles interrupteurs g&eacute;n&eacute;tiques pour utilisation in vivo sur des souris transg&eacute;niques a r&eacute;volutionn&eacute; plusieurs domaines de la biologie mol&eacute;culaire moderne. Combinant les deux techniques, recombinaison Cre/loxP-bas&eacute; et ligand-d&eacute;pendants d&#39;activation d&#39;une prot&eacute;ine chim&eacute;rique, nous avons g&eacute;n&eacute;r&eacute; des souris transg&eacute;niques qui permettent le contr&ocirc;le spatio-temporelle d&#39;expression et de l&#39;activit&eacute; du proto-oncog&egrave;ne c-myc. &Agrave; ces fins, le g&egrave;ne codant pour la prot&eacute;ine de fusion c-mycER(T) inductible par le tamoxif&egrave;ne (mycER(T)) a &eacute;t&eacute; ins&eacute;r&eacute; dans le locus du g&egrave;ne 26 actif contiennent toutes des ROSA en ciblant les g&egrave;nes. Dans l&#39;all&egrave;le ROSAMER qui en r&eacute;sulte, g&eacute;n&eacute;ralis&eacute;e de transcription du g&egrave;ne mycER(T) est emp&ecirc;ch&eacute;e par une s&eacute;quence de stop transcriptionnelle pr&eacute;c&eacute;dent flanqu&eacute; de loxP sites. Croisements de souris transg&eacute;niques ROSAMER avec Mox2 cre des souris transg&eacute;niques ont r&eacute;v&eacute;l&eacute; un contr&ocirc;le rigoureux de la transcription mycER(T) dans divers tissus &agrave; moins que la s&eacute;quence d&#39;arr&ecirc;t de la transcription a &eacute;t&eacute; supprim&eacute;e par l&#39;excision induite par la cre. En outre, nous avons pu d&eacute;montrer le tamoxif&egrave;ne d&eacute;pendant d&#39;activation de la prot&eacute;ine MycER(T) dans des fibroblastes embryonnaires de ces souris. Une preuve de principe, nous d&eacute;montrons que les cultures primaires de cr&ecirc;te neurale cr&eacute;&eacute;s des souris ROSAMER maintiennent leur capacit&eacute; prolif&eacute;rative en fonction de 4 OHT. De plus, nous d&eacute;montrons que ces cellules neurales de cr&ecirc;te conservent leur potentiel de diff&eacute;renciation, comme indiqu&eacute; par l&#39;expression de NF 160, un marqueur de diff&eacute;renciation neuronale apr&egrave;s le retrait de 4-OHT. Les souris transg&eacute;niques produites peuvent ainsi &ecirc;tre de pr&eacute;cieuses outils pour &eacute;tudier les effets sp&eacute;cifiques au type de cellule de l&#39;activit&eacute; de c-myc dans le d&eacute;veloppement et la maladie.

Sviluppo di viscoelastico interruttori genetici per l&#39;uso in vivo con topi transgenici ha rivoluzionato molte aree della biologia molecolare moderna. Combinando le due tecniche di ricombinazione genetica basata su Cre/loxP e attivazione ligando-dipendente di una proteina chimerica, abbiamo generato topi transgenici che consentono il controllo spatiotemporal di espressione e di attivit&agrave; del proto-oncogene c-myc. Nell&#39;allele ROSAMER risultante, generalizzata trascrizione del gene della mycER(T) &egrave; impedito da una sequenza di transcriptional stop precedente fiancheggiata da siti loxP. Croci di topi transgenici ROSAMER con topi transgenici Mox2 cre rivelarono stretto controllo della trascrizione di mycER(T) in vari tessuti a meno che la sequenza di transcriptional stop &egrave; stato rimosso da escissione cre-mediata. Inoltre, siamo stati in grado di dimostrare il tamoxifene-dipendente dalla attivazione della proteina MycER(T) in fibroblasti embrionali derivate da tali topi. Come prova di principio, ci dimostrano che culture primarie cresta neurale stabilite da topi ROSAMER mantengano la loro capacit&agrave; proliferativa in modo 4-OHT-dipendente. Inoltre, noi dimostrare che tali cellule della cresta neurale conservano il loro potenziale di differenziazione come indicato dall&#39;espressione di NF 160, un marcatore di differenziazione neuronale al momento del ritiro 4-OHT. Desarrollo de inducibles interruptores gen&eacute;ticos para uso in vivo con ratones transg&eacute;nicos ha revolucionado muchas &aacute;reas de la biolog&iacute;a molecular moderna. La combinaci&oacute;n de dos t&eacute;cnicas, basadas en Cre/loxP recombinaci&oacute;n gen&eacute;tica y dependiente de ligando la activaci&oacute;n de una prote&iacute;na quim&eacute;rica, hemos generado ratones transg&eacute;nicos que permiten el control espacio-temporal de expresi&oacute;n y de actividad de la proto-oncog&eacute;nicas c-myc. Para estos fines, el gen que codifica la prote&iacute;na de fusi&oacute;n c-mycER(T) tamoxifeno-inducible (mycER(T)) fue insertado en el locus de gene 26 ROSA ubicuo activo por gene targeting. En el alelo ROSAMER resultante, transcripci&oacute;n generalizada del gen mycER(T) es prevenido por una secuencia de parada transcripcional anterior que est&aacute; flanqueada por sitios loxP. Cruces de ratones transg&eacute;nicos ROSAMER con Mox2 cre ratones transg&eacute;nicos revelaron un control estricto de la transcripci&oacute;n de mycER(T) en varios tejidos a menos que la secuencia transcripcional stop fue quitada por la supresi&oacute;n de cre-mediada. Adem&aacute;s, hemos sido capaces de demostrar tamoxifen-dependiente de la activaci&oacute;n de la prote&iacute;na MycER(T) en los fibroblastos embrionarios derivados de esos ratones. Melanopsin is the photopigment of the retinal ganglion cells, which are involved in the synchronization of the biological clock in the suprachiasmatic nucleus (SCN) of mammals with the exogenous photoperiod. So far, no information about the three-dimensional (3D) structure of melanopsin is available. Here we report the predicted structure based on the protein-coding region of the nucleotide sequence of the gene for melanopsin, originating from isolated mRNA from the eyes of Djungarian hamsters (Phodopus sungorus). The nucleotide sequence shares the largest homologies with melanopsin of mice and rats (each 89%) and humans (84%). Based on the amino-acid sequence, and in comparison with the known structure of bovine rhodopsin, the three-dimensional melanopsin protein structure was modeled by using automated homology modeling approaches that were subsequently refined. Melanopsin consists of highly conserved seven-transmembrane domains and a long cytoplasmatic tail with multiple putative phosphorylation sites. In the binding site of the chromophore, a 11-cis-retinal is likely to be bound to lysine at position 336 as Schiff's base. The modeling results may indicate different photoisomerization within the melanopsin molecule compared with bovine rhodopsin.

Hasta ahora, ninguna informaci&oacute;n sobre la estructura tridimensional (3D) de la melanopsina est&aacute; disponible. Aqu&iacute; divulgamos la estructura prevista basada en la regi&oacute;n codificante de la prote&iacute;na de la secuencia de nucle&oacute;tidos del gen de la melanopsina, originarios de mRNA aislado de los ojos de Djungarian h&aacute;msters (Phodopus sungorus). La secuencia de nucle&oacute;tido comparte las homolog&iacute;as m&aacute;s grandes con la melanopsina de ratones y ratas (cada 89%) y los seres humanos (84%). Basado en la secuencia de amino&aacute;cidos y en comparaci&oacute;n con la estructura conocida de la rodopsina bovina, la estructura de la prote&iacute;na de la melanopsina tridimensional fue modelada por uso automatizado homolog&iacute;a modelado enfoques que fueron posteriormente refinados. Melanopsina consiste en dominios transmembrana de siete altamente conservados y una cola larga citoplasm&aacute;tica con m&uacute;ltiples sitios de fosforilaci&oacute;n putativos. En el sitio de uni&oacute;n del crom&oacute;foro, un 11-cis-retinal es probable que la obligaci&oacute;n de lisina en la posici&oacute;n 336 como base de Schiff. Murine neural crest stem cells (NCSCs) are a multipotent transient population of stem cells. After being formed during early embryogenesis as a consequence of neurulation at the apical neural fold, the cells rapidly disperse throughout the embryo, migrating along specific pathways and differentiating into a wide variety of cell types. In vitro the multipotency is lost rapidly, making it difficult to study differentiation potential as well as cell fate decisions. Using a transgenic mouse line, allowing for spatio-temporal control of the transforming c-myc oncogene, we derived a cell line (JoMa1), which expressed NCSC markers in a transgene-activity dependent manner. JoMa1 cells express early NCSC markers and can be instructed to differentiate into neurons, glia, smooth muscle cells, melanocytes, and also chondrocytes. A cell-line, clonally derived from JoMa1 culture, termed JoMa1.3 showed identical behavior and was studied in more detail. This system therefore represents a powerful tool to study NCSC biology and signaling pathways. We observed that when proliferative and differentiation stimuli were given, enhanced cell death could be detected, suggesting that the two signals are incompatible in the cellular context. However, the cells regain their differentiation potential after inactivation of c-MycER(T). In summary, we have established a system, which allows for the biochemical analysis of the molecular pathways governing NCSC biology. In addition, we should be able to obtain NCSC lines from crossing the c-MycER(T) mice with mice harboring mutations affecting neural crest development enabling further insight into genetic pathways controlling neural crest differentiation.

Establishment and controlled differentiation of neural crest stem cell lines using conditional transgenesis.

Les cellules souches neurales de cr&ecirc;te murine (SNCLC) sont une population nomade multipotente de cellules souches. Apr&egrave;s avoir &eacute;t&eacute; form&eacute; au cours de l&#39;embryogen&egrave;se pr&eacute;coce par suite de la neurulation au niveau du pli apicale de neurones, les cellules se dispersent rapidement tout au long de l&#39;embryon, migrant le long de voies sp&eacute;cifiques et de diff&eacute;rencier dans une grande vari&eacute;t&eacute; de types de cellules. In vitro la multipotentialit&eacute; est perdue rapidement, rendant difficile d&#39;&eacute;tudier le potentielle de diff&eacute;renciation ainsi que les d&eacute;cisions de destin cellulaire. &Agrave; l&#39;aide d&#39;une lign&eacute;e de souris transg&eacute;niques, permettant une commande spatio-temporelle de la transformation oncog&egrave;ne c-myc, que nous provenant d&#39;une lign&eacute;e cellulaire (JoMa1), qui a exprim&eacute; les marqueurs NCSC d&#39;une mani&egrave;re d&eacute;pendante du transg&egrave;ne-activit&eacute;. Murine Neuralleiste Stammzellen (NCSCs) sind fl&uuml;chtige Einwohner multipotenten Stammzellen. Nach w&auml;hrend der fr&uuml;hen Embryogenese infolge Neuralplatte an der apikalen neuralen Falten gebildet, zerstreuen die Zellen schnell w&auml;hrend der Embryo, auf bestimmten wegen migrieren und in einer Vielzahl von Zelltypen differenzieren. In-vitro ist die Multipotenz verloren rasch, es erschwert, um m&ouml;gliche Differenzierung sowie Zelle Schicksal Entscheidungen zu studieren. Mit Hilfe einer transgenen Maus-Linie, die r&auml;umlich-zeitliche Steuerung der transformieren-c-Myc-Onkogen, zulassend, wir eine Zelllinie (JoMa1), abgeleitet ausgedr&uuml;ckt, das NCSC Marker in gewissem Sinne Transgene-Aktivit&auml;t abh&auml;ngig. JoMa1 Zellen schnelles fr&uuml;hen NCSC-Marker und angewiesen werden, dass in Neuronen, Glia, glatten Muskelzellen, Melanozyten und auch Chondrozyten differenzieren. Eine Zelllinie klonal abgeleitet JoMa1 Kultur, genannt JoMa1.3 zeigte identisches Verhalten und wurde genauer untersucht. Dieses System stellt somit ein leistungsf&auml;higes Tool zum NCSC Biologie und Signalisierung Wege zu studieren. C&eacute;lulas murinas cresta neural (NCSCs) son una poblaci&oacute;n transitoria multipotent de c&eacute;lulas madre. Despu&eacute;s de formarse durante la embriog&eacute;nesis temprana como consecuencia de la neurulaci&oacute;n en el pliegue neural apical, las c&eacute;lulas se dispersan r&aacute;pidamente a lo largo del embri&oacute;n, migrar a lo largo de v&iacute;as espec&iacute;ficas y diferenciar a una gran variedad de tipos de c&eacute;lulas. In vitro la multipotencia se pierde r&aacute;pidamente, lo que hace dif&iacute;cil estudiar el potencial de diferenciaci&oacute;n as&iacute; como decisiones de destino celular. Utilizando un cable de rat&oacute;n transg&eacute;nico, lo que permite control espacio-temporal del oncog&eacute;n c-myc transformadora, que derivamos una l&iacute;nea celular (JoMa1), que expresan marcadores NCSC en forma dependiente del transgen-actividad. RNA expression data reveals that human embryonic stem (hES) cells differ from mouse ES (mES) cells in the expression of RNAs for keratin intermediate filament proteins. These differences were confirmed at the cellular and protein level and may reflect a fundamental difference in the epithelial nature of embryonic stem cells derived from mouse and human blastocysts. Mouse ES cells express very low levels of the simple epithelial keratins K8, K18 and K19. By contrast hES cells express moderate levels of the RNAs for these intermediate filament proteins as do mouse stem cells derived from the mouse epiblast. Expression of K8 and K18 RNAs are correlated with increased c-Jun RNA expression in both mouse and human ES cell cultures. However, decreasing K8 and K18 expression associated with differentiation to neuronal progenitor cells is correlated with increasing expression of the Snai2 (Slug) transcriptional repression and not decreased Jun expression. Increasing K7 expression is correlated with increased CDX2 and decreased Oct4 RNA expression associated with the formation of trophoblast derivatives by hES cells. Our study supports the view that hES cells are more similar to mouse epiblast cells than mouse ES cells and is consistent with the epithelial nature of hES cells. Keratin intermediate filament expression in hES cells may modulate sensitivity to death receptor mediated apoptosis and stress.

Augmente l&#39;expression de la K7 est en corr&eacute;lation avec une augmentation CDX2 et a diminu&eacute; l&#39;expression Oct4 ARN associ&eacute;e &agrave; la formation de d&eacute;riv&eacute;s de trophoblaste de CSEh. Notre &eacute;tude confirme que les CSEh ressemblent davantage &agrave; des cellules de souris &eacute;piblaste que souris ES cellules et est compatible avec la nature &eacute;pith&eacute;liale de CSEh. Maternal embryonic leucine zipper kinase (MELK) is expressed in several developing tissues, in the adult germ line, and in adult neural progenitors. MELK expression is elevated in aggressive undifferentiated tumors, correlating with poor patient outcome in human breast cancer. To investigate the role of MELK in mammary tumorigenesis in vivo, we used a MELK-green fluorescent protein (GFP) reporter mouse, which allows prospective isolation of MELK-expressing cells based on GFP fluorescence. We found that in the normal mammary gland, cells expressing high levels of MELK were enriched in proliferating cells that express markers of mammary progenitors. The isolation of cells with high levels of MELK in mammary tumors from MMTV-Wnt1/MELK-GFP bitransgenic mice resulted in a significant enrichment of tumorsphere formation in culture and tumor initiation after transplantation into mammary fat pads of syngeneic mice. Furthermore, using lentiviral delivery of MELK-specific shRNA and limiting dilution cell transplantations, we showed that MELK function is required for mammary tumorigenesis in vivo. Our findings identify MELK as a potential target in breast tumor-initiating cells.

In the adult organism, cell migration is required for physiological processes such as angiogenesis and immune surveillance, as well as pathological events such as tumor metastasis. The adaptor protein and Src substrate Tks5 is necessary for cancer cell migration through extracellular matrix in vitro and tumorigenicity in vivo. However, a role for Tks5 during embryonic development, where cell migration is essential, has not been examined. We used morpholinos to reduce Tks5 expression in zebrafish embryos, and observed developmental defects, most prominently in neural crest-derived tissues such as craniofacial structures and pigmentation. The Tks5 morphant phenotype was rescued by expression of mammalian Tks5, but not by a variant of Tks5 in which the Src phosphorylation sites have been mutated. We further evaluated the role of Tks5 in neural crest cells and neural crest-derived tissues and found that loss of Tks5 impaired their ventral migration. Inhibition of Src family kinases also led to abnormal ventral patterning of neural crest cells and their derivatives. We confirmed that these effects were likely to be cell autonomous by shRNA-mediated knockdown of Tks5 in a murine neural crest stem cell line. Tks5 was required for neural crest cell migration in vitro, and both Src and Tks5 were required for the formation of actin-rich structures with similarity to podosomes. Additionally, we observed that neural crest cells formed Src-Tks5-dependent cell protrusions in 3-D culture conditions and in vivo. These results reveal an important and novel role for the Src-Tks5 pathway in neural crest cell migration during embryonic development. Furthermore, our data suggests that this pathway regulates neural crest cell migration through the generation of actin-rich pro-migratory structures, implying that similar mechanisms are used to control cell migration during embryogenesis and cancer metastasis.

Im erwachsenen Organismus ist Zellmigration f&uuml;r physiologische Prozesse wie Angiogenese und immun-&Uuml;berwachung sowie pathologische Ereignisse wie Tumormetastasierung erforderlich. Das Adapterprotein und Src-Substrat ist Tks5 f&uuml;r Krebs Zelle Migration durch extrazellul&auml;re Matrix in-vitro- und in-vivo Tumorigenicity notwendig. Jedoch wurde eine Rolle f&uuml;r Tks5 w&auml;hrend der Embryonalentwicklung, wo Zellmigration entscheidend ist, nicht untersucht. Wir verwendeten Morpholinos Tks5 Ausdruck in den Zebrafish Embryos zu reduzieren und Entwicklungsst&ouml;rungen M&auml;ngel, vor allem in der Neuralleiste abgeleiteten Gewebe wie kraniofaziale Strukturen und Pigmentierung beobachtet. Der Tks5-Morphant-Ph&auml;notyp wurde gerettet, durch Ausdruck der S&auml;ugetier-Tks5, aber nicht durch eine Variante des Tks5, in denen die Src-Phosphorylierung-Sites mutiert wurde haben. Wir weiter evaluiert die Rolle des Tks5 in Neuralleistenzellen und Neuralleiste abgeleiteten Gewebe und festgestellt, dass der Verlust der Tks5 ihre ventrale Migration beeintr&auml;chtigt. Hemmung der Src Familie Kinasen, f&uuml;hrte auch zu abnormen ventralen Musterung Neuralleistenzellen und ihre Derivate. Wir best&auml;tigten, dass diese Effekte wurden wahrscheinlich von ShRNA-vermittelte Niederschlag von Tks5 in einem murinen Neuralleiste Stammzellen Linie autonome Zelle sein. Tks5 war bei der Neuralleiste Zellmigration in-vitro- und Src und Tks5 waren f&uuml;r die Bildung von Aktin-reiche Strukturen mit &Auml;hnlichkeit mit Podosomes erforderlich. Zus&auml;tzlich beobachteten wir, dass Neuralleistenzellen Src-Tks5-abh&auml;ngigen Zelle Vorspr&uuml;nge in 3-d Kulturbedingungen und in-vivo gebildet. Diese Ergebnisse zeigen eine wichtige und neuartige Rolle f&uuml;r die Src-Tks5-Pathway in Neuralleiste Zellmigration w&auml;hrend der Embryonalentwicklung. Dar&uuml;ber hinaus schl&auml;gt unsere Daten, dass diese Option regelt Neuralleiste Zellmigration durch die Erzeugung von Pro-wandernde Aktin-reiche Strukturen, was impliziert, dass &auml;hnliche Mechanismen zur Steuerung der Zellmigration w&auml;hrend der Embryogenese und Krebs Metastasen sind.

Ein Src-Tks5-Weg ist f&uuml;r Neuralleiste Zellmigration w&auml;hrend der Embryonalentwicklung erforderlich.

Nell&#39;organismo adulto, migrazione cellulare &egrave; necessaria per i processi fisiologici come l&#39;angiogenesi e la sorveglianza immunitaria, cos&igrave; come eventi patologici quali metastasi tumorali. La proteina dell&#39;adattatore ed il substrato Src Tks5 &egrave; necessario per la migrazione delle cellule di cancro attraverso la matrice extracellulare in vitro e in vivo tumorigenicity. Tuttavia, un ruolo per Tks5 durante lo sviluppo embrionale, dove la migrazione delle cellule &egrave; essenziale, non &egrave; stato esaminato. Abbiamo usato morpholinos per ridurre Tks5 espressione negli embrioni di zebrafish e osservati difetti di sviluppo, pi&ugrave; visibile nei tessuti derivati da cresta neurale come strutture cranio-facciali e pigmentazione. Il fenotipo morphant Tks5 fu salvato dall&#39;espressione di Tks5 dei mammiferi, ma non da una variante di Tks5 in cui sono stati mutati i siti di fosforilazione Src. Abbiamo ulteriormente valutato il ruolo di Tks5 in cellule della cresta neurale e cresta neurale-derivato tessuti e ha trovato che la perdita di Tks5 alterato loro migrazione ventrale. Inibizione della famiglie chinasi Src ha portato anche alla patterning ventrale anomala delle cellule della cresta neurale e loro derivati. Abbiamo confermato che questi effetti erano suscettibili di essere autonoma da shRNA-mediata atterramento di Tks5 in una linea di cellule staminali murine cresta neurale delle cellule. Tks5 &egrave; stato richiesto per cresta neurale migrazione cellulare in vitro, e sia Src e Tks5 sono stati richiesti per la formazione delle strutture ricche di actina con somiglianza a podosomes. En el organismo adulto, migraci&oacute;n celular es necesaria para los procesos fisiol&oacute;gicos tales como angiog&eacute;nesis y vigilancia inmunol&oacute;gica, as&iacute; como eventos patol&oacute;gicos como met&aacute;stasis del tumor. La prote&iacute;na adaptadora y sustrato de fuente Tks5 es necesario para la migraci&oacute;n de c&eacute;lulas de c&aacute;ncer a trav&eacute;s de la matriz extracelular in vitro y en vivo de la colonizaci&oacute;n. Sin embargo, un papel para Tks5 durante el desarrollo embrionario, donde la migraci&oacute;n celular es esencial, no ha sido examinado. Se utilizan morfolinos para reducir la expresi&oacute;n Tks5 en embriones de pez cebra y observaron defectos de desarrollo, principalmente en los tejidos derivados de la cresta neural como estructuras craneofaciales y pigmentaci&oacute;n. El fenotipo de morphant de Tks5 fue rescatado por la expresi&oacute;n de mam&iacute;feros Tks5, pero no por una variante del Tks5 en que los sitios de fosforilaci&oacute;n de la fuente han sido mutados.
