CLAVATA1 (CLV1), CLV2, CLV3, CORYNE (CRN), BAM1 and BAM2 are key regulators that function at the shoot apical meristem (SAM) of plants to promote differentiation by limiting the size of the organizing center that maintains stem cell identity in neighboring cells. Previous results have indicated that the extracellular domain of the receptor kinase CLV1 binds to the CLV3-derived CLE ligand. The biochemical role of the receptor-like protein CLV2 has remained largely unknown. Although genetic analysis suggested that CLV2, together with the membrane kinase CRN, acts in parallel with CLV1, recent studies using transient expression indicated that CLV2 and CRN from a complex with CLV1. Here, we report detection of distinct CLV2-CRN heteromultimeric and CLV1-BAM multimeric complexes in transient expression in tobacco and in Arabidopsis meristems. Weaker interactions between the two complexes were detectable in transient expression. We also find that CLV2 alone generates a membrane-localized CLE binding activity independent of CLV1. CLV2, CLV1 and the CLV1 homologs BAM1 and BAM2 all bind to the CLV3-derived CLE peptide with similar kinetics, but BAM receptors show a broader range of interactions with different CLE peptides. Finally, we show that BAM and CLV1 overexpression can compensate for the loss of CLV2 function in vivo. These results suggest two parallel ligand-binding receptor complexes controlling stem cell specification in Arabidopsis.

CLAVATA1 (CLV1), CLV2, CLV3, Coryne (CRN), BAM1 et BAM2 sont des r&#xE9;gulateurs cl&#xE9;s de cette fonction au m&#xE9;rist&#xE8;me apical de la tige (SAM) de plantes pour favoriser la diff&#xE9;renciation en limitant la taille du centre d&#x26;#39;organisation qui maintient l&#x26;#39;identit&#xE9; des cellules souches dans les pays voisins cellules. R&#xE9;sultats ant&#xE9;rieurs ont indiqu&#xE9; que le domaine extracellulaire de la kinase du r&#xE9;cepteur CLV1 lie au ligand CLV3 d&#xE9;riv&#xE9; CLE. Le r&#xF4;le biochimique du r&#xE9;cepteur-like protein CLV2 a demeur&#xE9; largement m&#xE9;connu. Enfin, nous montrons que la surexpression de BAM et CLV1 peut compenser pour la perte de CLV2 fonction in vivo. CLAVATA1 (CLV1), CLV2, CLV3, CORYNE (CRN), BAM1 und BAM2 sind wichtige Regulatoren, die Funktion auf das schie&szlig;en Apical Meristem (SAM) von Pflanzen zur F&ouml;rderung der Differenzierung durch Beschr&auml;nken der Gr&ouml;&szlig;e des veranstaltenden Zentrums, die Stammzellen Identit&auml;t in benachbarten Zellen verwaltet. Vorherige Ergebnisse gaben an, dass die extrazellul&auml;re Dom&auml;ne des Rezeptor-Kinase CLV1 an den CLV3 abgeleitete CLE-Liganden bindet. Die biochemische Rolle des Rezeptor-&auml;hnlichen Proteins CLV2 ist weitgehend unbekannt geblieben. Genetische Analyse oft empfohlen, CLV2, zusammen mit der Membran-Kinase CRN, parallel CLV1 fungiert, neuere Studien mit transienten Ausdruck darauf hingewiesen, dass CLV2 und CRN aus einem Komplex mit CLV1. Schw&auml;chere Interaktionen zwischen den beiden komplexen waren in vor&uuml;bergehender Ausdruck nachweisbar. Wir finden auch, dass CLV2 allein eine Membran lokalisiert CLE Bindung Aktivit&auml;t unabh&auml;ngig von CLV1 generiert. CLV2, CLV1 und die CLV1 UCP1 BAM1 und BAM2 alle an das CLV3 abgeleitete CLE-Peptid mit &auml;hnlichen Kinetik binden, aber BAM-Rezeptoren zeigen ein breiteres Spektrum von Interaktionen mit verschiedenen CLE-Peptide. Schlie&szlig;lich zeigen wir, dass der Verlust der CLV2 Funktion in-vivo BAM und CLV1 &Uuml;berexpression kompensieren kann. Diese Resultate schlagen zwei parallele Ligand-Bindung-Rezeptor-komplexe Steuerung Stammzell-Spezifikation in Arabidopsis.

CLAVATA1 (CLV1), CLV2, CLV3, CORYNE (CRN), BAM1 e BAM2 sono regolatori chiave che funzionano presso il meristema apicale di sparare (SAM) di piante per promuovere la differenziazione limitando la dimensione del centro organizzativo che mantiene l&#39;identit&agrave; delle cellule staminali in cellule vicine. CLAVATA1 (CLV1), CLV2, CLV3, coryne (CRN), y BAM1 BAM2 son los principales reguladores que funcionan en el meristemo brote apical (SAM) de las plantas para promover la diferenciaci&#xF3;n mediante la limitaci&#xF3;n del tama&#xF1;o del centro de la organizaci&#xF3;n que mantiene la identidad de las c&#xE9;lulas madre en el vecino c&#xE9;lulas. Los resultados previos han indicado que el dominio extracelular de la CLV1 quinasa del receptor se une al ligando CLV3 derivado CLE. El papel bioqu&#xED;mico del receptor de la prote&#xED;na similar a CLV2 se ha mantenido en gran parte desconocido. Aunque el an&#xE1;lisis gen&#xE9;tico sugiere que CLV2, junto con el CRN membrana quinasa, act&#xFA;a en paralelo con CLV1, estudios recientes que utilizan la expresi&#xF3;n transitoria indic&#xF3; que CLV2 y CRN de un complejo con CLV1. Aqu&#xED; reportamos la detecci&#xF3;n de distintos CLV2-CRN heteromultimeric y CLV1-BAM complejos multim&#xE9;ricos en la expresi&#xF3;n transitoria en tabaco y en los meristemos de Arabidopsis. Interacciones m&#xE1;s d&#xE9;biles entre los dos complejos se detectaron en la expresi&#xF3;n transitoria. Tambi&#xE9;n encontramos que CLV2 por s&#xED; sola genera una actividad de la membrana localizada CLE uni&#xF3;n independiente de CLV1. CLV2, CLV1 y los CLV1 hom&#xF3;logos BAM1 BAM2 y todos se unen al p&#xE9;ptido derivado de CLV3 CLE con la cin&#xE9;tica similares, pero los receptores de BAM muestran un rango m&#xE1;s amplio de las interacciones con los diferentes p&#xE9;ptidos CLE. Finalmente, mostramos que el BAM y la sobreexpresi&#xF3;n CLV1 puede compensar la p&#xE9;rdida de CLV2 funci&#xF3;n in vivo. Estos resultados sugieren dos paralelas de uni&#xF3;n al ligando del receptor de los complejos que controlan la especificaci&#xF3;n de c&#xE9;lulas madre en Arabidopsis.
