In Drosophila, like in humans, Dystrophin Glycoprotein Complex (DGC) deficiencies cause a life span shortening disease, associated with muscle dysfunction. We performed the first in vivo genetic interaction screen in ageing dystrophic muscles and identified genes that have not been shown before to have a role in the development of muscular dystrophy and interact with dystrophin and/or dystroglycan. Mutations in many of the found interacting genes cause age-dependent morphological and heat-induced physiological defects in muscles, suggesting their importance in the tissue. Majority of them is phylogenetically conserved and implicated in human disorders, mainly tumors and myopathies. Functionally they can be divided into three main categories: proteins involved in communication between muscle and neuron, and interestingly, in mechanical and cellular stress response pathways. Our data show that stress induces muscle degeneration and accelerates age-dependent muscular dystrophy. Dystrophic muscles are already compromised; and as a consequence they are less adaptive and more sensitive to energetic stress and to changes in the ambient temperature. However, only dystroglycan, but not dystrophin deficiency causes extreme myodegeneration induced by energetic stress suggesting that dystroglycan might be a component of the low-energy pathway and act as a transducer of energetic stress in normal and dystrophic muscles.

Chez la drosophile, comme chez les humains, la dystrophine glycoprot&#xE9;ine complexes (DGC) carences causer une maladie raccourcissement la dur&#xE9;e de vie, associ&#xE9;e &#xE0; une dysfonction musculaire. Nous avons effectu&#xE9; le premier dans l&#x26;#39;&#xE9;cran vivo interaction g&#xE9;n&#xE9;tique dans le vieillissement des muscles dystrophiques et des g&#xE8;nes identifi&#xE9;s qui n&#x26;#39;ont pas &#xE9;t&#xE9; indiqu&#xE9;es avant d&#x26;#39;avoir un r&#xF4;le dans le d&#xE9;veloppement de la dystrophie musculaire et d&#x26;#39;interagir avec la dystrophine et / ou dystroglycane. Des mutations dans plusieurs g&#xE8;nes qui interagissent trouv&#xE9;s provoquer fonction de l&#x26;#39;&#xE2;ge morphologiques et de la chaleur induite par les d&#xE9;fauts physiologiques dans les muscles, ce qui sugg&#xE8;re leur importance dans le tissu. La majorit&#xE9; d&#x26;#39;entre eux est phylog&#xE9;n&#xE9;tiquement conserv&#xE9;s et mis en cause dans des maladies humaines, principalement les tumeurs et les myopathies. When the main components of the DGC, Dystrophin (Dys) and Dystroglycan (Dg) are affected cognitive impairment and mental retardation in addition to muscle degeneration can occur. Previously we performed an array of genetic screens using a Drosophila model for muscular dystrophy in order to find novel DGC interactors aiming to elucidate the signaling role(s) in which the complex is involved. Since the function of the DGC in the brain and nervous system has not been fully defined, we have here continued to analyze the DGC modifiers' function in the developing Drosophila brain and eye.

Cuando los componentes principales de la DGC, la distrofina (Dys) y distroglicano (DG) se ven afectados el deterioro cognitivo y retraso mental, adem&#xE1;s de la degeneraci&#xF3;n muscular puede ocurrir. Previamente se realiz&#xF3; una serie de pantallas gen&#xE9;ticos usando un modelo de Drosophila para la distrofia muscular con el fin de encontrar nuevos interactores DGC destinados a dilucidar el papel de se&#xF1;alizaci&#xF3;n (s) en que est&#xE1; implicado el complejo. Mammalian musculature is a very robust and dynamic tissue that goes through many rounds of degeneration and regeneration in an individual's lifetime. There is a biological program that maintains muscle progenitor cells that, when activated, give rise to intermediate myoblast progeny that consequently differentiate into mature muscle cells. Recent works have provided a picture of the role that microRNAs (miRNAs) play in maintaining aspects of this program. Intriguingly, a subset of these miRNAs is de-regulated in muscular dystrophies (MDs), a group of fatal inherited neuromuscular disorders that are often associated with deficiencies in the Dystrophin (Dys) complex. Apparently, transcriptional expression of many of the muscle specific genes and miRNAs is dependent on chromatin state regulated by the Dys-Syn-nNOS pathway. This puts Dystrophin at the epicenter of a highly regulated program of muscle gene expression in which miRNAs help to coordinate networking between multiple phases of muscle maintenance, degeneration, and regeneration. Therefore, understanding the role of miRNAs in physiology of normal and diseased muscle tissue could be useful for future applications in improving the MD therapies and could open new clinical perspectives.

C&#39;&egrave; un programma biologico che mantiene le cellule progenitrici muscolari che, quando attivato, danno origine a progenie myoblast intermedi che di conseguenza differenziarsi in cellule muscolari maturo. Recenti lavori hanno fornito un quadro del ruolo che i microRNA (miRNAs) gioca nel mantenere gli aspetti di questo programma. Curiosamente, un sottoinsieme di questi miRNAs &egrave; sregolato nelle distrofie muscolari (MDs), un gruppo di fatale malattie neuromuscolari ereditarie che sono spesso associati con carenze nel complesso distrofina (dis). A quanto pare, transcriptional espressione di molti geni specifici del muscolo e miRNAs dipende della cromatina stato regolato con il sentiero dis-Syn-nNOS. Questo mette distrofina l&#39;epicentro di un programma altamente regolamentato di espressione del gene muscolo in cui miRNAs aiutano a coordinare la creazione di reti tra molteplici fasi di mantenimento muscolare, degenerazione e rigenerazione. Musculatura de los mam&#xED;feros es un tejido muy s&#xF3;lido y din&#xE1;mico que pasa a trav&#xE9;s de muchas rondas de la degeneraci&#xF3;n y la regeneraci&#xF3;n en la vida del individuo. No es un programa biol&#xF3;gico que mantiene las c&#xE9;lulas musculares progenitoras que, cuando se activa, dar lugar a progenie de mioblastos intermedia que, por tanto se diferencian en c&#xE9;lulas musculares maduras. Trabajos recientes han proporcionado una imagen de la funci&#xF3;n que los microRNAs (miRNAs) juegan en el mantenimiento de los aspectos de este programa. Curiosamente, un subgrupo de estos miRNAs es de las reguladas en las distrofias musculares (MDS), un grupo de mortales enfermedades hereditarias neuromusculares que son a menudo asociados con deficiencias en la distrofina (Dys) compleja. Aparentemente, la expresi&#xF3;n transcripcional de muchos de los genes espec&#xED;ficos del m&#xFA;sculo y miRNAs es dependiente en estado cromatina regulada por la v&#xED;a de Dys-Syn-nNOS.
