&#x5170;&#x5C3C;&#x78B1;&#x53D7;&#x4F53;&#x5728;&#x54FA;&#x4E73;&#x52A8;&#x7269;&#x8033;&#x8717;&#x4E2D;&#x7684;&#x672C;&#x5730;&#x5316;&#xFF1A; &#x8D85;&#x5FAE;&#x7ED3;&#x6784;&#x7684;&#x7814;&#x7A76;&#x3002;

Calcium-induced calcium release (CICR) in the mammalian cochlea has been suggested to enhance neurotransmitter release from inner hair cells and facilitate the efferent response in outer hair cells. Light microscopic evidence exists for the presence of ryanodine receptors in the organ of Corti but there is so far no information about their ultrastructural localisation. We have therefore used post-embedding immunogold labeling with antibodies that predominantly recognise ryanodine receptor isoforms 1 (RyR1) and 2 (RyR2) to investigate their distribution in rat cochleae. In inner hair cells, the highest levels of labeling were observed over an area of rough endoplasmic reticulum that lies in the cytoplasmic region beneath the nucleus; in outer hair cells, the cytoplasmic region above the nucleus displayed most labeling. Labeling was also associated with the subsurface cisternae adjacent to the lateral membranes of both types of hair cell, with the efferent terminals on the outer hair cells and was observed in adjacent supporting cells. Labeling in outer hair cells was significantly higher than that in inner hair cells or in the supporting cells. Our results support the presence of RyR1 in the cochlea but do not rule out the presence of other isoforms. CICR may be involved in the control of calcium levels in the base of the inner hair cells and supporting cells, and in the cholinergic efferent response and motile behaviour of the outer hair cells.

Ryanodine receptor localisation in the mammalian cochlea: an ultrastructural study.

Lib&eacute;ration du calcium induite par le calcium (CICR) dans la cochl&eacute;e de mammif&egrave;res a &eacute;t&eacute; sugg&eacute;r&eacute;e d&#39;am&eacute;liorer la lib&eacute;ration des neurotransmetteurs de cellules cili&eacute;es internes et de faciliter la r&eacute;ponse eff&eacute;rente dans les cellules cili&eacute;es externes. Il existe une l&eacute;g&egrave;re preuve microscopique pour la pr&eacute;sence de r&eacute;cepteurs de la ryanodine dans l&#39;organe de Corti, mais il n&#39;y a jusqu&#39;&agrave; pr&eacute;sent aucune information sur leur localisation ultrastructurale. Nous avons donc utilis&eacute; post-embedding immunogold marquage avec des anticorps qui reconnaissent principalement des isoformes du r&eacute;cepteur de la ryanodine (RyR1) 1et 2 (RyR2) pour enqu&ecirc;ter sur leur distribution en lima&ccedil;on de rat. Dans les cellules cili&eacute;es internes, les plus hauts niveaux de l&#39;&eacute;tiquetage ont &eacute;t&eacute; observ&eacute;s sur une superficie de r&eacute;ticulum endoplasmique rugueux qui se trouve dans la r&eacute;gion cytoplasmique sous le noyau ; dans les cellules cili&eacute;es externes, la r&eacute;gion cytoplasmique au-dessus du noyau affiche un &eacute;tiquetage plus. Marquage a &eacute;galement &eacute;t&eacute; associ&eacute;e aux sous-surface saccules adjacentes aux membranes lat&eacute;rales des deux types de cellules cili&eacute;es, munis de connexions eff&eacute;rentes sur les cellules cili&eacute;es externes et a &eacute;t&eacute; observ&eacute;e dans les cellules adjacentes de soutien. Marquage de cellules cili&eacute;es externes est significativement plus &eacute;lev&eacute;e que dans les cellules cili&eacute;es internes ou dans les cellules de soutien. Nos r&eacute;sultats soutiennent la pr&eacute;sence de RyR1 dans la cochl&eacute;e, mais n&#39;excluent pas la pr&eacute;sence d&#39;autres isoformes. CICR peut-&ecirc;tre &ecirc;tre impliqu&eacute; dans le contr&ocirc;le du taux de calcium dans la base des cellules cili&eacute;es internes et cellules de soutien et dans la r&eacute;ponse eff&eacute;rente cholinergique et comportement motile des cellules cili&eacute;es externes.

Kalzium Kalzium-induzierte Freisetzung (CICR) in der S&auml;ugetier-Cochlea ist vorgeschlagen worden, zur Verbesserung der Neurotransmitter-Freisetzung aus innerer Haarzellen und erleichtern die efferente Antwort im &auml;u&szlig;eren Haarzellen. Leichte mikroskopische Belege f&uuml;r die Anwesenheit von Ryanodin-Rezeptoren in den Organ von Corti gibt es aber bisher gibt es keine Informationen &uuml;ber ihre ultrastrukturelle Lokalisierung. Wir haben daher post-embedding Immunogold Kennzeichnung mit Antik&ouml;rper, die &uuml;berwiegend Ryanodin-Rezeptor-Isoformen erkennen 1 (RyR1) und 2 (RyR2) verwendet, um ihre Verteilung im Ratte Cochleae zu untersuchen. Im Inneren Haarzellen wurden optimale Kennzeichnung auf einer Fl&auml;che von rauen endoplasmatischen Reticulum beobachtet, die sich in der zytoplasmatischen Region unterhalb des Kerns befindet; im &auml;u&szlig;eren Haarzellen angezeigt die zytoplasmatische Region oberhalb des Kerns die meisten beschriften. Beschriftung war ebenfalls eng verkn&uuml;pft mit dem Untergrund Cisternae angrenzend an den seitlichen Membranen beider Typen der Haarzelle mit den efferenten Klemmen auf der &auml;u&szlig;eren Haarzellen und in angrenzenden unterst&uuml;tzenden Zellen festgestellt. Kennzeichnung im &auml;u&szlig;eren Haarzellen war deutlich h&ouml;her als die im Inneren Haarzellen oder in den unterst&uuml;tzenden Zellen. Unsere Ergebnisse unterst&uuml;tzen die Anwesenheit des RyR1 in der Cochlea aber nicht das Vorhandensein von anderen Isoformen ausschlie&szlig;en. ZPK kann in der Steuerung der Kalziumspiegel in der Basis der inneren Haarzellen und unterst&uuml;tzenden Zellen und in der cholinergen ableitendes Antwort und st&auml;bchenf&ouml;rmiges Verhalten von der &auml;u&szlig;eren Haarzellen beteiligt sein.

Rilascio di calcio indotto da calcio (CICR) nella coclea dei mammiferi &egrave; stato suggerito per migliorare neurotrasmettitore rilascio dalle cellule ciliate interne e facilitare la risposta efferente in cellule ciliate esterne. Luce microscopico prova esiste per la presenza di recettori rianodinico nell&#39;organo del Corti, ma finora non non c&#39;&egrave; alcuna informazione circa la loro localizzazione ultrastrutturale. Quindi abbiamo usato post-embedding immunogold etichettatura con anticorpi che riconoscono principalmente isoforme del recettore rianodinico (RyR1) 1 e 2 (RyR2) per indagare la loro distribuzione in cochleae ratto. In cellule ciliate interne, sono stati osservati i pi&ugrave; alti livelli di etichettatura su un&#39;area di reticolo endoplasmatico rugoso che si trova nella regione citoplasmatica sotto il nucleo; in cellule ciliate esterne, la regione citoplasmatica sopra il nucleo visualizzata la maggior parte delle etichette. Etichettatura era anche associato con il destino di sottosuolo adiacente alle membrane laterali di entrambi i tipi di capelli cella, con i terminali efferenti le cellule ciliate esterne ed &egrave; stata osservata in cellule adiacenti di supporto. Etichettatura in cellule ciliate esterne era significativamente pi&ugrave; alta rispetto a quella in cellule ciliate interne o le cellule di supporto. I nostri risultati supportano la presenza di RyR1 nella coclea, ma non esclude la presenza di altre isoforme. CICR possono essere coinvolti nel controllo dei livelli di calcio nella base delle cellule ciliate interne e cellule di supporto e nella risposta efferente colinergica e motili comportamento delle cellule dei capelli esterne.

Por lo tanto hemos utilizado post-embedding immunogold etiquetado con anticuerpos que reconocen predominante isoformas del receptor de rianodina (RyR1) de 1 y 2 (RyR2) para investigar su distribuci&oacute;n en c&oacute;cleas de rata. Ca(V)1.3 channels comprise a vital subdivision of L-type Ca2+ channels: Ca(V)1.3 channels mediate neurotransmitter release from auditory inner hair cells (IHCs), pancreatic insulin secretion, and cardiac pacemaking. Fitting with these diverse roles, Ca(V)1.3 channels exhibit striking variability in their inactivation by intracellular Ca2+. IHCs show generally weak-to-absent Ca2+-dependent inactivation (CDI), potentially permitting audition of sustained sounds. In contrast, the strong CDI seen elsewhere likely provides critical negative feedback. Here, we explore this mysterious CDI malleability, particularly its comparative weakness in hair cells. At baseline, heterologously expressed Ca(V)1.3 channels exhibit intense CDI, wherein each lobe of calmodulin (CaM) contributes a distinct inactivation component. Because CaM-like molecules (bearing four recognizable but not necessarily functional Ca2+-binding EF hands) can perturb the Ca2+ response of molecules regulated by CaM, we asked whether such CaM-like entities could influence CDI. We find that CaM-like calcium-binding protein (CaBP) molecules are clearly expressed within the organ of Corti. In particular, the rare subtype CaBP4 is specific to IHCs, and CaBP4 proves capable of eliminating even the potent baseline CDI of Ca(V)1.3. CaBP4 thereby represents a plausible candidate for moderating CDI within IHCs.

The sensory hair cells of the mammalian cochlea transduce acoustic stimuli into auditory nerve activity. The biomechanical and molecular details of hair cell mechanotransduction are being acquired at an ever-finer level of resolution. In this review, we discuss how selected mouse mutants and transgenic models have contributed to, and will continue to shape, our understanding of the molecular basis of hair cell mechanotransduction. Functional and structural discoveries made originally in hair cells of nonmammalian vertebrates have been further pursued in the mouse inner ear, where transgenic manipulation can be applied to test molecular mechanisms. Additional insights have been obtained from mice bearing mutations in genes underlying deafness in humans. Taken together, these studies emphasize the elegance of mechanotransduction, enlarge the team of molecular players, and begin to reveal the remarkable adaptations that provide the sensitivity and temporal resolution required for mammalian hearing.

Modulation of voltage-gated calcium channels was studied in inner hair cells (IHCs) in an ex vivo preparation of the apical turn of the rat organ of Corti. Whole cell voltage clamp in the presence of potassium channel blockers showed inward calcium currents with millisecond activation and deactivation kinetics. When temperature was raised from 22 to 37 degrees C, the calcium currents of immature IHCs [<12 days postnatal (P12)] increased threefold in amplitude, and developed more pronounced inactivation. This was determined to be calcium-dependent inactivation (CDI) on the basis of its reliance on external calcium (substitution with barium), sensitivity to internal calcium-buffering, and voltage dependence (reflecting the calcium driving force). After the onset of hearing at P12, IHC calcium current amplitude and the extent of inactivation were greatly reduced. Although smaller than in prehearing IHCs, CDI remained significant in the mature IHC near the resting membrane potential. CDI in mature IHCs was enhanced by application of the endoplasmic calcium pump blocker, benzo-hydroquinone. Conversely, CDI in immature IHCs was reduced by calmodulin inhibitors. Thus voltage-gated calcium channels in mammalian IHCs are subject to a calmodulin-mediated process of CDI. The extent of CDI depends on the balance of calcium buffering mechanisms and may be regulated by calmodulin-specific processes. CDI provides a means for the rate of spontaneous transmitter release to be adjusted to variations in hair cell resting potential and steady state calcium influx.

During this time, capacitance measurements of vesicular fusion have continued to be refined, optical methods have added insights regarding vesicle trafficking, and paired intracellular recordings have established the transfer function of the afferent synapse at high resolution. Further, genes have been identified with forms of deafness known as auditory neuropathy, and their role in afferent signaling explored in mouse models. With these advances, our view of the hair cell afferent synapse has continued to be refined, and surprising properties have been revealed that emphasize the unique role of this structure in neural function.

Multivesicular release at individual IHC ribbon synapses activates AMPA-mediated EPSCs with widely ranging amplitudes. The underlying mechanisms and specific role for multivesicular release in encoding sound are not well understood. Here we characterize the waveforms of individual EPSCs recorded from afferent boutons contacting IHCs and compare their characteristics in immature rats (postnatal days 8-11) and hearing rats (postnatal days 19-21). Two types of EPSC waveforms were found in every recording: monophasic EPSCs, with sharp rising phases and monoexponential decays, and multiphasic EPSCs, exhibiting inflections on rising and decaying phases. Multiphasic EPSCs exhibited slower rise times and smaller amplitudes than monophasic EPSCs. Both types of EPSCs had comparable charge transfers, suggesting that they were activated by the release of similar numbers of vesicles, which for multiphasic EPSCs occurred in a less coordinated manner. On average, a higher proportion of larger, monophasic EPSCs was found in hearing compared to immature rats. In addition, EPSCs became significantly faster with age. The developmental increase in size and speed could improve auditory signaling acuity. Multiphasic EPSCs persisted in hearing animals, in some fibers constituting half of the EPSCs. The proportion of monophasic versus multiphasic EPSCs varied widely across fibers, resulting in marked heterogeneity of amplitude distributions. We propose that the relative contribution of two modes of multivesicular release, generating monophasic and multiphasic EPSCs, may underlie fundamental characteristics of auditory nerve fibers.

Hier wir charakterisieren die Wellenformen der einzelnen EPSCs registrate afferente Boutons Kontaktaufnahme mit IHCs und vergleichen ihre Merkmale in unreifen Ratten (postnatale Tage 8-11) und Anh&ouml;rung Ratten (postnatale Tage 19-21).
